A robust high-throughput screening system to assess bacterial tyrosine ammonia lyase activity in the context of tyrosine inherited metabolic disorders.

Inborn errors of tyrosine metabolism result in patient's inability to degrade tyrosine. Current treatment consists of a phenylalanine and tyrosine restricted diet and nitisinone, causing a block in the tyrosine degradation pathway. However, tyrosine levels will increase, leading to acquired hypertyrosinemia, implying the need for an add-on treatment. Tyrosine ammonia lyases (TAL) can provide such an add-on treatment as they catalyze the deamination of tyrosine into p-coumaric acid and ammonia. In this study, we developed a robust high-throughput screening (HTS) assay to assess the capacity of

Sun protection and antibacterial activities of carotenoids from the soft coral Sinularia sp. symbiotic bacteria from Panjang Island, North Java Sea.

Carotenoids have shown beneficial applications in cosmetology, pharmacology, and medicine. However, environmental stress in the marine environment can trigger the production of unique secondary metabolites, such as carotenoids. These compounds can also be sustainably produced by symbiotic bacteria. We hypothesized that the soft corals in tropical regions may produce diverse biological secondary metabolites, including carotenoids, both by the host organism and their bacterial symbiont. The unique carotenoids may provide promising biological activity such as antioxidant, UV photoprotector, and a

Engineering a photosynthetic bacteria-incorporated hydrogel for infected wound healing.

Treating wounds with multidrug-resistant bacterial infections remains a huge and arduous challenge. In this work, we prepared a "live-drug"-encapsulated hydrogel dressing for the treatment of multidrug-resistant bacterial infections and full-thickness skin incision repair. Our live dressing was comprised of photosynthetic bacteria (PSB) and extracellular matrix (ECM) gel with photothermal, antibacterial and antioxidant properties, as well as good cytocompatibility and blood compatibility. More interestingly, live PSB could be regarded as not only photothermal agents but also as anti-inflammato

Determination of 15N/14N of Ammonium, Nitrite, Nitrate, Hydroxylamine, and Hydrazine Using Colorimetric Reagents and Matrix-Assisted Laser Desorption Ionization-Time-of-Flight Mass Spectrometry (MALDI-TOF MS).

In the nitrogen (N) cycle, nitrogenous compounds are chemically and biologically converted to various aqueous and gaseous N species. The 15N-labeling approach is a powerful culture-dependent technique to obtain insights into the complex nitrogen transformation reactions that occur in cultures. In the 15N-labeling approach, the fates of supplemented 15N- and/or unlabeled gaseous and aqueous compounds are tracked by mass spectrometry (MS) analysis, whereas MS analysis of aqueous N species requires laborious sample preparation steps and is performed using isotope-ratio mass spectrometry, which re

Bacterial metabolic-immune reprogramming in cancer: from living vectors to engineered biofactories.

Tumor immunotherapy has transformed modern oncology, yet its clinical efficacy against solid tumors remains severely restricted by complex physiological and immunosuppressive barriers. Distinct from passive delivery systems, live bacteria can actively navigate into hypoxic tumor cores and intrinsically modulate local immune networks, making them promising intelligent therapeutic vehicles. In this review, we comprehensively trace their evolution from living vectors to engineered biofactories. We discuss how colonizing bacteria reprogram the tumor microenvironment (TME) via metabolic interventio

Spatial organization of cutaneous microbiomes reveals putative microbial contributions to host chemical defenses in the American toad.

Chemical defenses are widely evolved throughout the tree of life. Animals can exploit mutualisms with toxin-producing symbionts as a mechanism of chemical defense. However, this has only begun to be explored in depth, and how these mutualisms may relate to how animals synthesize or acquire their toxins has been even less studied. True toads synthesize their own toxins and offer a novel system to study the interplay between the cutaneous skin microbiome and how it may contribute to toxin synthesis or biotransformation. In this study, we investigated whether the cutaneous microbiome of the Ameri

Antibacterial and Antibiofilm Activity of Titanium Treated with Hybrid Phospholipid Films Containing Carbonate Hydroxyapatite and Silver Nanoparticles.

Bacterial contamination and low osteogenic activity are the major causes of dental implant failure. The development of titanium coatings has provided new research directions to improve both antibacterial and osteogenic activity. In this study, we constructed phospholipidic films containing carbonate hydroxyapatite added or not to silver nanoparticles. The purpose was to reduce the biofilm formation while maintaining the osteogenic potential of surfaces. Phospholipid monolayers were transferred to titanium by Langmuir-Blodgett (LB) technique and resulted in dense, ordered and uniform films. Mod

Pathogen-Centric Activation of an Azoreductase-Responsive Antibody-Antibiotic Conjugate for the Targeted Eradication of MRSA.

Antibody-antibiotic conjugates (AACs) provide a transformative platform for eradicating intracellular methicillin-resistant Staphylococcus aureus (MRSA). However, current AACs largely follow antibody-drug conjugates (ADCs) design principles and rely on host-lysosomal proteases for activation, rendering them ineffective against extracellular (planktonic) bacteria. Furthermore, we demonstrate that high bacterial burden induces host-lysosomal dysfunction, fundamentally compromising the efficacy of host-dependent AACs against intracellular reservoirs. To address these limitations, we developed a p

Early signaling of inflammatory bursts by using estimated C-reactive Protein velocity (eCRPv): a model of acute meningitis and iso-CRPinemia.

PURPOSE: C-Reactive Protein (CRP) is often used to support the differentiation between acute bacterial and viral infections, including acute meningitis. However, this is not always possible due to similar CRP concentrations upon presentation in these two cohorts. We have improved the diagnostic yield of this biomarker by adjusting its concentrations to the estimated time from disease onset, transforming a single, static value into a dynamic measurement, expressed in milligrams per liter per hour. METHODS: Our study included 46 matched pairs of patients with acute bacterial and acute viral meni

Gentamicin-Loaded Chitosan Nanocoating on Polyurethane Prostatic Stents to Combat Biofilm Formation and Urogenital Device-Associated Infections.

Prostatic obstruction is a blockage of the urethra caused by benign prostatic hyperplasia, a common condition in aging men that often results in significant urinary complications. Polyurethane prostatic stents are widely used to alleviate this obstruction. However, their susceptibility to biofilm formation and subsequent bacterial infections by pathogens such as Escherichia coli and Proteus mirabilis remains a major clinical challenge. This study investigates the polyurethane prostatic stents (PS) that are surface-immobilized with gentamicin-loaded chitosan nanoparticles (GMCSNPs) to combat th

Glutamine alleviates Lipopolysaccharide-induced corneal epithelial inflammation and oxidative stress in dogs.

Pseudomonas aeruginosa is a common pathogenic bacteria in canine ophthalmology. Lipopolysaccharide (LPS), a component in the cell wall of gram-negative bacteria, is released following bacterial lysis and causes pathology and inflammation of the cornea. Antibiotics are used to treat bacterial keratitis, and the reuse of antibiotics can easily cause bacterial resistance. Research has shown that glutamine (GLN) has anti-inflammatory and antioxidant biological functions. Herein, we explored the effects and underlying mechanisms of GLN and established an LPS-induced cornea inflammation model. Treat

Antibacterial and photocatalytic activity of hydrothermally synthesized SnO2 doped GO and CNT under visible light irradiation.

Bacterial and dye pollution are major problems with wastewater treatment. An increasing number of photocatalysts are being used in industry to kill bacterial and reduce pollution. In the present study, highly stable SnO2-doped nanocomposites have been prepared successively by a hydrothermal method. The synthesized nanocomposite was characterized using a range of techniques, such as X-ray diffraction, field emission scanning electron microscopy with energy dispersive X-ray spectroscopy and electron probe micro analysis, ultraviolet-visible spectroscopy, Fourier transform infrared spectroscopy,

Production of human metabolites by gastrointestinal bacteria as a potential source of post-mortem alteration of antemortem drug/metabolite concentrations.

Previous studies have demonstrated that bacterial species are capable of transforming complex chemical substances. Several of these species, native to the human gastrointestinal tract, are active in postmortem decomposition. They have potential to cause biotransformations affecting compound-to-metabolite ratios within the human body, especially after death. Investigation of postmortem effects could supply valuable information, especially concerning compound identification and confirmation. The purpose of this research was to investigate the effects of Escherichia coli, Bacteroides fragilis, an

Increased expression of type-1 fimbriae by nonpathogenic Escherichia coli 83972 results in an increased capacity for catheter adherence and bacterial interference.

BACKGROUND: In vitro, urinary catheter colonization by avirulent Escherichia coli 83972 impedes subsequent catheter colonization by a variety of uropathogenic organisms. However, E. coli 83972 shows a low efficacy of adherence to silicone urinary catheter material, possibly because the fim operon encoding adhesive type 1 fimbriae is incomplete. We hypothesized that improving the catheter adherence of E. coli 83972 would improve its bacterial interference properties. METHODS: We created adhesive mutants by transforming wild-type E. coli 83972 with fim(+) plasmids. Adherence to urinary catheter

Fast, easy and efficient: site-specific insertion of transgenes into enterobacterial chromosomes using Tn7 without need for selection of the insertion event.

BACKGROUND: Inserting transgenes into bacterial chromosomes is generally quite involved, requiring a selection for cells carrying the insertion, usually for drug-resistance, or multiple cumbersome manipulations, or both. Several approaches use phage lambda red recombination, which allows for the possibility of mutagenesis of the transgene during a PCR step. RESULTS: We present a simple, rapid and highly efficient method for transgene insertion into the chromosome of Escherichia coli, Salmonella or Shigella at a benign chromosomal site using the site-specific recombination machinery of the tra

Green alga Chlamydomonas reinhardtii can effectively remove diclofenac from the water environment - A new perspective on biotransformation.

The use of unicellular algae to remove xenobiotics (including drugs) from wastewaters is one of the rapidly developing areas of environmental protection. Numerous data indicate that for efficient phycoremediation three processes are important, i.e. biosorption, bioaccumulation, and biotransformation. Although biosorption and bioaccumulation do not raise any serious doubts, biotransformation is more problematic since its products can be potentially more toxic than the parent compounds posing a threat to organisms living in a given environment, including organisms that made this transformation.

Overexpression of Stenotrophomonas maltophilia major facilitator superfamily protein MfsA increases resistance to fluoroquinolone antibiotics.

BACKGROUND: Stenotrophomonas maltophilia is an opportunistic human pathogen causing nosocomial infections worldwide. S. maltophilia infection is of particular concern due to its inherent resistance to currently used antibiotics. Proton motive force-driven transporters of the major facilitator superfamily frequently contribute to the efflux of substances, including antibiotics, across cell membranes. METHODS: An mfsA expression plasmid (pMfsA) was constructed and transferred into bacterial strains by electroporation. The antibiotic susceptibility levels of S. maltophilia strains were determine

Two routes of MexS-MexT-mediated regulation of MexEF-OprN and MexAB-OprM efflux pump expression in Pseudomonas aeruginosa.

The NfxC-type mutant of Pseudomonas aeruginosa produces the MexEF-OprN efflux pump and down-regulates expression of the quorum-sensing-dependent efflux pump MexAB-OprM and production of virulence factors in the presence of an active transcriptional regulator, MexT. Consequently, these cells are resistant to chloramphenicol and hypersusceptible to β-lactam antibiotics. An upper negative regulator, MexS, has been assumed to inactivate MexT in wild-type strains, hence shutting down production of the MexEF-OprN pump. This observation was, however, reported in only one clinical strain and not confi

High-Pressure Homogenization Modifies the Structure of Pediococcus acidilactici S1 Exopolysaccharide and Enhances Its Bile Salts Binding Capacity.

Bile salts (BSs) binding capacity plays a crucial role in cholesterol reduction. This study investigated the enhancement of BSs binding capacity and structural characteristics of Pediococcus acidilactici S1 exopolysaccharide (EPS, designated S1-D1N1) through high-pressure homogenization (HPH) modification optimized using response surface methodology (RSM). The optimal modification parameters were determined to be 60 mg/mL S1-D1N1 concentration, 40 MPa pressure, and 3 homogenization cycles. Under these conditions, the modified polysaccharide (H-S1-D1N1) demonstrated significantly improved BSs b
