Neurodevelopmental disorders following fetal reduction of triplet pregnancies: a nationwide cohort study.

BACKGROUND: Triplet pregnancies carry high risks of prematurity and subsequent neurodevelopmental disorders. Fetal reduction from triplets to twins improves short-term obstetric outcomes, but long-term neurodevelopmental outcomes among surviving children remain uncertain. OBJECTIVE: To compare the long-term risk of neurodevelopmental disorders among liveborn children from trichorionic triamniotic triplet pregnancies managed with fetal reduction from 3 to 2 fetuses vs no reduction. STUDY DESIGN: We conducted a nationwide, population-based cohort study using the Danish Fetal Medicine Database,…

Efficient Organic Fluorescence Scintillation by Engineering Anti-Kasha Triplet Energy Levels.

Scintillation, the phenomenon of luminescence under bombardment of x-rays, excites multiple areas, including ionizing radiation detection, biomedical imaging, and electron microscopy. For organic scintillators, employing triplet excitons represents a useful strategy to increase the radioluminescence intensity. However, utilization of the triplet excitons generally conflicts with the development of fast scintillators, which are key to fast-timing medical radiography. Here, we design a series of efficient fluorophores with anti-Kasha energy levels, enabling fast scintillation by effectively util

Effective Near-Infrared Triplet Emitter Based on Hetero-Metal-Metal Interaction.

Incorporating metal-metal (M-M) interactions into excited states of closed-shell d8 and d10 metal complexes is an effective strategy in the design of near-infrared (NIR) phosphorescent materials. While extensive studies have focused on homometallic M-M-bonded excited states, the potential of heterometallic interactions remains relatively underexplored. Herein, we report a series of heterometallic Rh(I)-Pt(II) double salt complexes that achieve efficient NIR phosphorescence, with emission peak energy spanning 830-980 nm and room-temperature quantum yield up to 23%. In this system, the Rh(I) cen

Organic Diradicals Bridged by Inverted Singlet-Triplet Units for Optical-Spin Interfaces.

Molecular platforms for optically addressable spin states are emerging as fascinating alternatives to solid-state spin centers, offering scalable synthesis, structural tunability, and chemical versatility. Here, we present a molecular design strategy for achieving photoinduced spin polarization in organic diradicals bridged by systems featuring an inverted singlet-triplet (InveST) energy gap. These InveST units possess HOMO and LUMO orbitals localized on complementary atomic sites. By covalently linking the non-SOMO-bearing positions of alternant hydrocarbon radicals to the LUMO-localized atom

Optimizing lower intensity triplet therapy in acute myeloid leukemia: a practical guide.

Venetoclax-based doublets with azacitidine or low dose cytarabine are the standard of care for the treatment of acute myeloid leukemia (AML) … ling the transition to allogeneic stem cell transplantation for eligible patients. While effective, triplets are associated with myelosuppression and cytopenia-related toxicities, which can affect treatment tolerability and quality of life. In this review, we summarize the available evidence for triplet therapy in AML and offer our recommendations on the p …

Temperature-Dependent Photoreactivity of 2-Azidomethylbenzophenone: Insights into the Triplet Imine Biradical Pathway.

Photoenols, formed through photoinduced intramolecular H atom abstraction in o-alkyl-substituted arylketones, typically have limited utility as reactive intermediates owing to fast reversion to the starting material. Herein, we introduced an azido group on the o-alkyl substituent to render the photoreaction irreversible. Irradiation of 2-azidomethylbenzophenone (1) in methanol yielded 2-(hydroxy(phenyl)methyl)benzonitrile (2). Laser flash photolysis of 1 revealed the formation of biradical 3Br1 followed by intersystem crossing to photoenols Z-3 (τ ∼ 3.3 μs) and E-3 (τ > 45 μs), both of which r

Triplet Excited Nitroarene Converts Aryl Alkynes to Ketones by Deleting a Carbon Atom.

In this study, triplet excited nitroarenes were used to convert internal alkynes to ketone fragments through simultaneous oxygen insertion and carbon deletion. The approach was effective across 33 diverse substrates, compatible with various functional groups, and applicable to bioactive molecule derivatives and large-scale synthesis. UV-vis spectroscopy revealed the role of visible light in exciting nitroarenes, while mechanistic experiments identified ketene and nitrosoarene as key intermediates, with the deleted carbon released as CO and CO2.

Targeted triplet therapies incorporating FLT3 or IDH inhibitors: ready for prime time?

Combination therapies in acute myeloid leukemia (AML) are an area of current investigation due to the potential of overcoming resistance by targeting multiple pathways simultaneously. Triplet therapies combining hypomethylating agents, venetoclax, and targeted inhibitors are emerging as a promising therapy for older patients with AML unfit for intensive chemotherapy. These regimens have predominantly been studied in FLT3- and IDH1/2-mutated AML with attainment of high rates of measurable residual disease-negative composite remissions. Notably, patients with FLT3-internal tandem duplication and

Emergency neurosurgical separation of pygopagus conjoined twins in a triplet pregnancy: illustrative case.

BACKGROUND: Conjoined twins are a rare type of congenital malformation. The point of attachment is the primary factor used to classify conjoined twins; typically, this is front to front, with thoracopagus and omphalopagus twins accounting for about 75% of cases and pygopagus twins only between 6% and 19%. OBSERVATIONS: This is a case report of 46-day-old female triplets born to a 37-year-old para 4 mother. Triplets A and B were conjoined at the lumbosacrococcygeal area dorsally. Triplet C was not conjoined. The conjoined triplets had a low birth weight; hence, it was decided to start nutritio…

Prediction of Adverse Pregnancy Outcomes Based on Maternal and Pregnancy Characteristics in Triplet Pregnancies.

Objective: Multifetal gestations are linked to an increased risk of pregnancy-related hypertensive disorders and other adverse outcomes. The probability positively correlates with the number of fetuses. Therefore, the objective of the study was to assess the use of various maternal and pregnancy-related characteristics for the prediction of adverse pregnancy outcomes in triplet pregnancies, dependent on different possible predictive factors such as maternal age, BMI, assisted reproductive technology (ART), parity, uterine artery Doppler (UtA-PI) measured and chorionicity. Methods: This was a s

When repetita no-longer iuvant: somatic instability of the CAG triplet in Huntington's disease.

Trinucleotide repeats in DNA exhibit a dual nature due to their inherent instability. While their rapid expansion can diversify gene expression during evolution, exceeding a certain threshold can lead to diseases such as Huntington's disease (HD), a neurodegenerative condition, triggered by >36 C-A-G repeats in exon 1 of the Huntingtin gene. Notably, the discovery of somatic instability (SI) of the tract allows these mutations, inherited from an affected parent, to further expand throughout the patient's lifetime, resulting in a mosaic brain with specific neurons exhibiting variable and often

Navigating triplet repeats sequencing: concepts, methodological challenges and perspective for Huntington's disease.

The accurate characterization of triplet repeats, especially the overrepresented CAG repeats, is increasingly relevant for several reasons. First, germline expansion of CAG repeats above a gene-specific threshold causes multiple neurodegenerative disorders; for instance, Huntington's disease (HD) is triggered by >36 CAG repeats in the huntingtin (HTT) gene. Second, extreme expansions up to 800 CAG repeats have been found in specific cell types affected by the disease. Third, synonymous single nucleotide variants within the CAG repeat stretch influence the age of disease onset. Thus, new sequen

Accidental triplet harvesting in donor-acceptor dyads with low spin-orbit coupling.

We present an accidental mechanism for efficient intersystem crossing (ISC) between singlet and triplet states with low spin-orbit coupling (SOC) in molecules having donor-acceptor (D-A) moieties separated by a Sigma bond. Our study shows that SOC between the lowest singlet excited state and the higher-lying triplet states, together with nuclear motion-driven coupling of this triplet state with lower-lying triplet states during the free rotation about a Sigma bond, is one of the possible ways to achieve the experimentally observed ISC rate for a class of D-A type photoredox catalysts. This mec

Tissue-specific TCF4 triplet repeat instability revealed by optical genome mapping.

BACKGROUND: Fuchs endothelial corneal dystrophy (FECD) is the most common repeat-mediated disease in humans. It exclusively affects corneal endothelial cells (CECs), with ≤81% of cases associated with an intronic TCF4 triplet repeat (CTG18.1). Here, we utilise optical genome mapping (OGM) to investigate CTG18.1 tissue-specific instability to gain mechanistic insights. METHODS: We applied OGM to a diverse range of genomic DNAs (gDNAs) from patients with FECD and controls (n = 43); CECs, leukocytes and fibroblasts. A bioinformatics pipeline was developed to robustly interrogate CTG18.1-spanning

The efficient triplet states formation of Se-modified PDI dimers and tetramers in solvents.

The triplet excited states of molecules play an important role in photophysical processes, which has attracted great research interest. Perylene diimide (PDI) is a widely studied material closely associated with the generation of triplet states, and it is highly anticipated to become an electron acceptor material for improving photovoltaic conversion efficiency. In this work, we prepared dimers and tetramers composed of selenium-modified PDI-C5 (N,N'-bis(6-undecyl) perylene-3,4,9,10-bis(dicarboximide)) units. We investigated the photophysical processes of these dimers and tetramers in chlorofo
