Pathogenesis of acute infection in rhesus macaques with a lymphocyte-tropic strain of simian immunodeficiency virus.

The simian immunodeficiency virus, SIVmac, causes disease affecting multiple organ systems in macaques similar to human immunodeficiency virus infection in humans. Molecularly cloned SIVmac with a strong lymphocyte tropism was used in pathogenesis experiments to correlate viral cell tropism with disease. In 5 animals, exhaustive analyses on viruses from tissues and identification of infected precursor cells were done at multiple times during infection to ensure the virus had not mutated into a macrophage-tropic variant. Viral replication was measured by infectivity, infectious center assays, a…

Superior In Vivo Efficacy for T Cell Engineering via Citronellol-Tailored mRNA-tLNPs.

Efficient in vivo generation of CAR T cells via mRNA-LNP delivery necessitates high-efficiency T cell engineering, yet current systems remain suboptimal. While lymphoid tropism is essential, it is demonstrated that high tropism alone is insufficient for effective in vivo engineering-some highly tropic LNPs frequently exhibit suboptimal T cell transfection. Crucially, intrinsic T cell transfection potency emerges as an orthogonal and equally critical determinant, consistent with the need for both T cell specificity and transfection efficiency. Through screening a tail-varied ionizable lipid lib…

Modelling Between-Cell-Type Heterogeneity in Within-Host Influenza Virus Infection.

Cell tropism, or the preference of a virus for particular cell types, has major implications for viral transmission, pathogenesis, and evolution. Historically, changes in cell tropism for particular viruses have been observed to correlate to changes in viral fitness, in the form of increased within-host replication and increased transmission between hosts. This is illustrated in the context of influenza, where adaptation to infect cells expressing α 2-6 linked sialic acid receptors enhances human-to-human transmissibility. Target cell populations differ not only in abundance but also in intrin…

Deep-Sequencing Analysis of the Dynamics of HIV-1 Quasiespecies in Naive Patients during a Short Exposure to Maraviroc.

In this study, we have characterized quasispecies dynamics and the evolution of viral tropism in naive HIV-1-infected patients treated with a short course of maraviroc monotherapy (ClinicalTrials.gov registration no. NCT01060618) independently of the tropism of the infecting virus. We randomly selected 20 patients infected with viruses displaying different basal tropisms-10 carrying R5 and 10 carrying dual/mixed X4 (DM/X4) viruses-at recruitment as determined by phenotypic assay (Trofile). Evolution of viral quasiespecies at the end of treatment was determined by ultradeep sequencing of the V3…

Presence of HIV-1 R5 viruses in cerebrospinal fluid even in patients harboring R5X4/X4 viruses in plasma.

BACKGROUND: HIV-1 viruses have the ability to use CCR5 or CXCR4 coreceptors either solely (R5 or X4) or in combination (R5X4). The CCR5 antagonists block HIV entry into the cell and are specifically active against HIV-1 R5 strains. The objectives of this study were to investigate the predicted tropism of viruses present in paired cerebrospinal fluid (CSF) and plasma in a group of 22 HIV-1-infected patients with neurological disorders and to search for eventual discordance of predicted virus tropism between both compartments. METHODS: Paired CSF and plasma samples were selected from subjects h…

The Murine Neuronal Receptor NgR1 Is Dispensable for Reovirus Pathogenesis.

Engagement of host receptors is essential for viruses to enter target cells and initiate infection. Expression patterns of receptors in turn dictate host range, tissue tropism, and disease pathogenesis during infection. Mammalian orthoreovirus (reovirus) displays serotype-dependent patterns of tropism in the murine central nervous system (CNS) that are dictated by the viral attachment protein σ1. However, the receptor that mediates reovirus CNS tropism is unknown. Two proteinaceous receptors have been identified for reovirus, junctional adhesion molecule A (JAM-A) and Nogo-66 receptor 1 (NgR1)…

Echinococcus granulosus sensu lato in animal intermediate hosts: What is with the organ location?

Organ or tissue tropism/preference in many parasites is essential for their establishment, survival and pathogenesis. In protozoan parasites e.g. Trypanosoma and Plasmodium, tissue tropism is associated with many important aspects such as transmission, treatment, and pathogenesis. In these parasites, tissue tropism is achieved by mechanisms such as sequestration, extravasation, transcellular migration, and vascular permeability. In contrast, little is known about the mechanism behind organ tropism/preference and the potential implication in parasitic metazoans like Echinococcus. Since intraspe…

HIV-1 chemokine coreceptor utilization in paired cerebrospinal fluid and plasma samples: a survey of subjects with viremia.

BACKGROUND: Chemokine receptors serve as coreceptors for human immunodeficiency virus type 1 (HIV-1) entry, influence cell tropism, and may critically determine central nervous system infection pathogenesis. Using an in vitro functional entry assay, we examined utilization of 2 principal coreceptors in cerebrospinal fluid (CSF) and plasma in 46 subjects. METHODS: Paired CSF and plasma samples were selected from subjects with a range of CD4 T cell counts. Amplified populations of env sequences were characterized as using CCR5 (R5), CXCR4 (X4), or both receptors (R5+X4). Individual clones deriv…

Rabies Virus Pseudotyped with CVS-N2C Glycoprotein as a Powerful Tool for Retrograde Neuronal Network Tracing.

Efficient viral vectors for mapping and manipulating long-projection neuronal circuits are crucial in structural and functional studies of the brain. The SAD strain rabies virus with the glycoprotein gene deleted pseudotyped with the N2C glycoprotein (SAD-RV(ΔG)-N2C(G)) shows strong neuro-tropism in cell culture, but its in vivo efficiency for retrograde gene transduction and neuro-tropism have not been systematically characterized. We compared these features in different mouse brain regions for SAD-RV-N2C(G) and two other widely-used retrograde tracers, SAD-RV(ΔG)-B19(G) and rAAV2-retro. We f…

Differential ChAdOx1 and ChAdOx2 transduction efficiency and receptor specificity in human and mouse B cell lines in vitro.

Adenoviruses represent versatile platforms for vaccine development and gene therapy, owing to their broad tropism and capacity to transduce a wide range of cellular subsets, including antigen-presenting cells such as dendritic cells and B cells. Adenoviral tropism is strongly influenced by serotype and viral origin, which together determine the efficiency with which distinct cellular phenotypes are infected. Among currently used platforms, the replication-incompetent chimpanzee adenoviral vectors ChAdOx1 and ChAdOx2 have gained prominence because of their favourable safety profiles and robust …

Determination of Bacterial Adherence to Mouse Tissues Using a Hematogenous Model.

This procedure is used to determine the tissue tropism of bacteria to mouse tissues. Briefly, a high dose of bacteria is inoculated intravenously via the mouse tail vein. Bacteria are allowed to circulate through the blood for a short duration of time, during which they will adhere to the endothelium in tissues for which there is a tropism. After the desired time, sterile saline is perfused through the animal and any unbound bacteria are washed away. Tissues harvested after perfusion will contain any bound bacteria, and tissue-specific tropism can be determined qualitatively by culturing live …

Persistence of CXCR4-tropic virus in people living with four-class drug-resistant HIV and its clinical impact in the modern antiretroviral era.

BACKGROUND: CXCR4-tropic HIV seems to be associated with more clinical events than CCR5-tropic virus. OBJECTIVES: This study aims to describe the effect of the persistence of CXCR4-tropic virus on the occurrence of clinical events in people with four-class drug-resistant HIV. METHODS: This is a retrospective study on people with four-class drug-resistant HIV from the PRESTIGIO Registry, with at least two HIV-tropism determinations during follow-up. Follow-up accrued from the date of the first four-class drug resistance evidence (baseline) until death, loss to follow-up or freezing date (31 D…

Preferential Tissue Sites of Different Cancer-Risk Groups of Human Papillomaviruses.

The oncogenic potential of human papillomavirus (HPV) may be used to determine the tissue tropism of each HPV type. Cervical cancer develops in the squamo-columar junction of the cervices, and most lesions are induced by high-risk (HR) HPV types. This suggests that HR types preferentially infect the cervix, whereas the preferential infection site for low-risk (LR) types is not well defined. The determination of HPV tropism when using cytology samples can be uncertain since it is difficult to avoid contamination of cell samples between the cervix and the vagina. Herein, cell samples were carefu…

[Molecular mechanism of tissue-specific infection of poliovirus].

Poliovirus is the causative agent of an acute disease of the central nervous system, poliomyelitis. Poliovirus will be eradicated in the near future by a world-wide vaccination program. Poliovirus is a neurotropic virus that produces severe lesions selectively in the CNS. However, a basic question why poliovirus exhibits neurotropic property has not been elucidated. Poliovirus receptor and host factors involved in the translation initiation of viral protein, which are required for virus replication, play important roles in determining tissue tropism. We found that type I interferon response is…

Adaptation of the infectious bronchitis virus H120 vaccine strain to Vero cell lines.

Infectious bronchitis virus (IBV) has restricted cell and tissue tropism. IBVs, except the Beaudette strain, can infect and replicate in chicken embryos, primary chicken embryo kidneys, and primary chicken kidney cells, only. The limited viral cell tropism of IBV substantially hinders in vitro cell-based research on pathogenic mechanisms and vaccine development. Herein, the parental H120 vaccine strain was serially passaged for five generations in chicken embryos, 20 passages in CK cells and 80 passages in Vero cells. This passaging yielded a Vero cell-adapted strain designated HV80. To furthe…

Mapping the parameters of prion-induced neuropathology.

We present a theoretical framework that enables us to dissect out the parametric dependencies of the pathogenesis of prion diseases. We are able to determine the influence of both host-dependent factors (connectivity, cell density, protein synthesis rate, and cell death) and strain-dependent factors (cell tropism, virulence, and replication rate). We use a model based on a linked system of differential equations on a lattice to explore how the regional distribution of central nervous system pathology in Creutzfeldt-Jakob disease, Gerstmann-Sträussler-Scheinker syndrome, and fatal familial inso…

Host factors involved in resistance to retroviral infection.

Viral replication requires the help of host cell factors, whose species specificity may affect viral tropism. On the other hand, there exist host factors that restrict viral replication. The anti-viral system mediated by some of these restriction factors, which is termed intrinsic immunity and is distinguished from conventional innate and adaptive immunity, has been described as playing an important role in making species-specific barriers against viral infection. Here, we describe the current progress in understanding of such restriction factors against retroviral replication, focusing on TRI…
