Preparation and immunological effects of a bivalent subunit candidate vaccine based on the PCV-Cap-2-3 gene engineered from baculovirus.

INTRODUCTION: Porcine circoviruses (PCVs), belonging to the genus Circovirus in the family Circoviridae, are prevalent worldwide and pose a serious threat to the pig industry. Although vaccines against PCV2 are available, PCV3 vaccine development, prevention strategies, and surveillance for co-infections remain limited, leading to delayed outbreak responses. Therefore, the aim of this study was to develop a bivalent subunit vaccine targeting the prevalent strains of PCV2 and PCV3. METHODS: Recombinant baculoviruses, Baculovirus-PCV2-Cap and Baculovirus-PCV3-Cap, were constructed to express th

A porcine circovirus type 2d-based virus-like particle subunit vaccine effectively protects pigs against homologous challenge.

Porcine circovirus-associated disease (PCVAD), caused by pathogenic porcine circovirus type 2 (PCV2), remains one of the most economically devastating diseases in the global swine industry, and the continued emergence and predominance of the PCV2d genotype have raised concerns regarding the efficacy of currently available vaccine. In this study, a virus-like particle (VLP) subunit vaccine based on the PCV2d capsid protein was developed using a prokaryotic expression system, and its immunogenicity and protective efficacy were systematically evaluated in pigs. Following immunization and subseque

Elucidating the role of Toxoplasma gondii EF-hand containing protein 1 in parasite biology and its potential as a subunit vaccine antigen.

Toxoplasma gondii (T. gondii) poses a major threat to human and animal health; however, no human vaccine is available. Parasite egress, which links successive intracellular cycles, is essential for parasite maintenance and is strictly controlled by calcium signaling. In particular, the EF-hand containing protein (Efhc) in T. gondii displays strong Ca²⁺ binding capacity. Here, we aimed to explore the role of TgEfhc1 in the growth and development of T. gondii tachyzoites, as well as the potential immunoprotective effects of recombinant TgEfhc1 (rTgEfhc1) as a subunit vaccine in infected mice. Construction of a subunit vaccine based on Mycobacterium tuberculosis EsxE and evaluation of its immunogenicity and ex vivo mycobacterial growth inhibitory activity.

Tuberculosis (TB) remains one of the world's deadliest infectious diseases and is the leading infectious disease killer globally. Bacillus Calmette-Guérin (BCG) is currently the only licensed vaccine for TB prevention, but it has limitations in preventing latent infection and TB reactivation. To overcome these limitations, this study selected the secreted antigen Rv3904c (EsxE) to construct the recombinant prokaryotic expression plasmid pET21b-Rv3904c, and the recombinant protein rEsxE was expressed and purified. The antigen specificity of rEsxE was further confirmed by measuring cytokine leve

Liposomal simvastatin potentiates intranasal H1N1 influenza subunit vaccine via increasing transcytosis of antigen and submucosal dendritic cell recruitment.

Intranasal vaccines specifically eliciting mucosal immunity in the upper respiratory tract have shown advantages in protecting against respiratory virus invasion. Yet, no clinically licensed intranasal adjuvant remains a major hurdle for the development of intranasal vaccines with low immunogenic antigens like subunit vaccines. Here, we show that liposomes loading simvastatin (Lipo-SV) serve as potent mucosal adjuvants for the intranasal liposomal subunit vaccine encapsulating the hemagglutinin 1 (HA1) glycoprotein of A/PR/8/34 (PR8) H1N1 influenza (Lipo-HA1), providing robust protection again

Development of a cholera toxin B subunit-adjuvanted Helicobacter pylori multi-epitope vaccine and evaluation of its immunization effect in mice.

This study used epitope screening technology to identify dominant T/B cell epitopes from the Helicobacter pylori virulence factors UreB, CagA, VacA and HpaA, (UCVH) and develop an effective vaccine. These epitopes were fused in series with the cholera toxin B subunit to develop a multivalent fusion vaccine. The immunogenicity of the recombinant vaccine was systematically evaluated after it was administered to C57BL/6 mice. The serum titers of UCVH-specific IgG, IgG1, and IgG2a antibodies and the mucosal sIgA concentration increased significantly. The splenic lymphocytes also increased their se

Adjuvant combination and antigen multimerization shape neutralizing antibody and T cell responses to a SARS-CoV-2 RBD subunit vaccine.

INTRODUCTION: The rapid development and deployment of multiple safe and effective COVID-19 vaccines were critical cornerstones of pandemic control. However, vaccine inequity and the emergence of new variants of concern (VOCs) highlighted major gaps in the global strategy to control SARS-CoV-2 infection. Despite the use of distinct platforms, most approved vaccines utilize the Spike protein as the main antigen due to its pivotal role in virus entry, mediated by the receptor binding domain (RBD). In this context, RBD stands out as a promising antigen for a subunit vaccine candidate, as it is the

Exploring varicella zoster virus proteome for construction and validation of a multi-epitope based subunit vaccine using multifaceted immunoinformatics approaches.

Varicella zoster (VZ) is a viral disease caused by varicella zoster virus (VZV) that is related to alphaherpesvirus subfamily. VZV causes a neurotropic disease in humans. The aim of this work was to stimulate the human immune system by developing a multi-epitope vaccine based on five VZV surface proteins. Multiple immunoinformatics techniques were applied to assess B-cell and T-cell epitopes. The population coverage of each T-cell epitope was analyzed and the results showed high population coverage scores. The vaccine consists of 615 amino acids and it was antigenic and non-allergenic. The vac

A BCG-boosting subunit vaccine RS2/DMI induces Th1-polarized immunity by targeting RD6 and secretory antigens of Mycobacterium tuberculosis.

Bacillus Calmette-Guérin (BCG) vaccine has limited protection against tuberculosis (TB) in adults, and there is an urgent need to develop novel TB vaccines. Currently, booster vaccines based on initial immunization with BCG are important research hotspots. In this study, we constructed recombinant protein RS2 by innovatively fusing Mycobacterium tuberculosis (M. tb) region of difference (RD) antigen Rv1506c with secretory phase antigen Rv3204, and developed a novel composite adjuvant DMI (dimethyl dioctadecylammonium bromide [DDA] + colloidal manganese salt [Mn jelly, MnJ] + imiquimod) to prep

Evaluation of immunogenicity and protective efficacy of bacteriophage conjugated haemagglutinin based subunit vaccine against equine influenza virus in a murine model.

Equine influenza (EI) is a highly contagious acute respiratory disease of equines caused by the H3N8 subtype of Influenza A virus i.e. equine influenza virus (EIV). Vaccination is an important and effective tool for the control of EI in equines. Most of the commercial influenza vaccines are produced in embryonated hen's eggs which has several inherent disadvantages. Hence, subunit vaccine based on recombinant haemagglutinin (HA) antigen, being the most important envelope glycoprotein has been extensively exploited for generating protective immune responses, against influenza A and B viruses. A subunit-based influenza/SARS-CoV-2 Omicron combined vaccine induced potent protective immunity in BALB/c mice.

Infection with influenza A virus (IAV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) poses a significant risk to human life, health, and the global economy. Vaccination is one of the most effective strategies in the fight against infectious viruses. In this study, we, for the first time, have evaluated the immunogenicity and protective effect of an influenza/SARS-CoV-2 Omicron subunit combined vaccine adjuvanted with MF59 and administered to BALB/c mice. Results showed that the combined vaccine induced high levels of IgG, IgG1 , and IgG2a antibodies, as well as influenza A H

Porvac® subunit vaccine induces neutralizing antibodies against all three main classical swine fever virus genotypes.

Classical swine fever (CSF) is endemic in Cuba and is one of the major health problems of the Cuban swine industry. The current efforts to control the disease in Cuba include vaccination with Porvac®, a subunit marker vaccine. Although the efficacy of Porvac against CSF virus (CSFV) subgenotype 1.4 has been extensively documented, little is known about the ability of the antibodies induced by this vaccine to neutralize other CSFV genotypes. In this study, sera collected from three pigs vaccinated with Porvac were able to efficiently neutralize CSFV strains belonging to genotypes 1, 2, and 3. T

Stability of closed and needle-punctured vials of Porvac® subunit vaccine against classical swine fever subjected to thermal stress.

BACKGROUND: Classical Swine Fever (CSF) is still one of the most economically important viral diseases of pigs. In endemic countries, the disease is controlled mostly through vaccination; hence, the availability of safe and effective vaccines is of utmost importance. Vaccines intended for application in developing countries must also be thermally stable, since the infrastructure needed to maintain a cold chain in those countries is usually lacking. Porvac® is a second-generation subunit marker vaccine against CSF that has demonstrates to be safe and protective. Previous studies have also shown

Cationic pH-sensitive liposomes as tuberculosis subunit vaccine delivery systems: Effect of liposome composition on cellular innate immune responses.

Tuberculosis (TB) is a major global health problem, and the development of effective and safe vaccines is urgently needed. CD8+ T-cells play an important role alongside CD4+ T-cells in the protective immune response against TB. pH-sensitive liposomes are hypothesized to boost CD8+ T-cell responses by promoting class I presentation through a mechanism involving pH-dependent endosomal escape and the cytosolic transfer of antigens. The aim of the study was to explore the potential of pH-sensitive liposomes as a novel delivery system for a multi-stage protein subunit vaccine against TB in primary

Exploring structural antigens of yellow fever virus to design multi-epitope subunit vaccine candidate by utilizing an immuno-informatics approach.

BACKGROUND: Yellow fever is a mosquito-borne viral hemorrhagic disease transmitted by several species of virus-infected mosquitoes endemic to tropical regions of Central and South America and Africa. Earlier in the twentieth century, mass vaccination integrated with mosquito control was implemented to eradicate the yellow fever virus. However, regular outbreaks occur in these regions which pose a threat to travelers and residents of Africa and South America. There is no specific antiviral therapy, but there can be an effective peptide-based vaccine candidate to combat infection caused by the v

An intranasally administrated SARS-CoV-2 beta variant subunit booster vaccine prevents beta variant replication in rhesus macaques.

Emergence of SARS-CoV-2 variants and waning of vaccine/infection-induced immunity pose threats to curbing the COVID-19 pandemic. Effective, safe, and convenient booster vaccines are in need. We hypothesized that a variant-modified mucosal booster vaccine might induce local immunity to prevent SARS-CoV-2 infection at the port of entry. The beta-variant is one of the hardest to cross-neutralize. Herein, we assessed the protective efficacy of an intranasal booster composed of beta variant-spike protein S1 with IL-15 and TLR agonists in previously immunized macaques. The macaques were first vaccin

A Recombinant Alpha-Like Protein Subunit Vaccine (GBS-NN) Provides Protection in Murine Models of Group B Streptococcus Infection.

BACKGROUND: Group B Streptococcus (GBS) transmission during pregnancy causes preterm labor, stillbirths, fetal injury, or neonatal infections. Rates of adult infections are also rising. The GBS-NN vaccine, engineered by fusing N-terminal domains of GBS Alpha C and Rib proteins, is safe in healthy, nonpregnant women, but further assessment is needed for use during pregnancy. Here, we tested GBS-NN vaccine efficacy using mouse models that recapitulate human GBS infection outcomes. METHODS: Following administration of GBS-NN vaccine or adjuvant, antibody profiles were compared by ELISA. Vaccine

Optimization of immunization procedure for SWCNTs-based subunit vaccine with mannose modification against spring viraemia of carp virus in common carp.

Immersion vaccination of single-walled carbon nanotubes loaded with mannose-modified glycoprotein (SWCNTs-MG) vaccine has been proved to be effective in preventing spring viraemia of carp virus (SVCV). Immunization procedure has immense consequence on the immune effect of the immersion vaccine. However, immunization procedure optimization for SWCNTs-MG vaccine against SVCV has not been reported. In this study, accordingly, a full-factor experiment was designed to optimize the immunization procedure of SWCNTs-MG vaccine by three aspects of vaccine dose (30 mg/L, 40 mg/L and 50 mg/L), immunizati
