Impact of Prenatal Alcohol Exposure on Cerebral Cortex Development.

Fetal alcohol spectrum disorders (FASD) represent a significant global health concern and are the leading cause of preventable birth defects, intellectual disabilities, and persistent behavioral deficits. The neurodevelopmental mechanisms underlying FASD are not yet fully understood, but there is growing interest in the role of cortical development in the manifestation of these disorders. The cerebral cortex serves as the master regulator of higher-order functions, including sensory perception, motor planning, decision-making, and general intelligence-processes that are all adversely affected

An assessment of the suitability of online blood alcohol concentration calculators.

OBJECTIVES: The increasing popularity of online blood alcohol concentration (BAC) calculators which estimate alcohol levels based on user-entered variables such as sex, weight, and alcohol consumption data raises concerns about their reliability and their influence on the decision to drive. The aim of this study is to assess the accuracy and usability of 36 online BAC calculators in Polish and English. MATERIAL AND METHODS: The calculated BAC values were compared with reference values obtained from 8 female, 8 male in both fasted and post-meal states. Blood alcohol concentration was evaluated

Effect of cannabinol, tetrahydrocannabivarin and cannabidiol on voluntary alcohol consumption.

AIMS: Previous studies have demonstrated that the endocannabinoid system plays a significant role in the development of alcohol use disorder (AUD), and CB1 receptor antagonists/inverse agonists show promise as a novel AUD pharmacotherapy. However, these compounds failed in clinical trials due to the severe psychiatric side effects. Non-psychoactive phytocannabinoids may have a better safety profile and could be used as an alternative approach to treat AUD. The aim of this study was to test the potential of three phytocannabinoids in reducing alcohol consumption: CB1 receptor partial agonist ca

An Exploration of Alcohol Withdrawal Protocols, with a Focus on the Use of Phenobarbital.

OBJECTIVE: Severe alcohol withdrawal (SAW) is a common cause of hospital admission in the United States. There has been increased interest in phenobarbital use for SAW treatment. We aimed to investigate variability in alcohol withdrawal management protocols at different hospitals within a geographic region, with a focus on phenobarbital use. METHODS: We e-mailed a survey to intensive care unit physician leadership to all New York City acute care hospitals as well as large acute care hospitals in Massachusetts regarding their protocols for SAW. RESULTS: Of 13 respondents, eight have SAW proto

The Role of the Hedgehog Pathway in Alcohol-Induced Birth Defects.

In this chapter, we review the current literature that focuses on the role of Sonic hedgehog (Shh) signaling, a major morphogenic pathway regulating embryonic development, in the pathology of fetal alcohol spectrum disorder (FASD), caused by prenatal alcohol exposure (PAE). Ethanol exposure disrupts Shh signaling during development across a range of species including in mice, chick, and zebrafish. Ethanol appears to interfere with Shh signaling through multiple mechanisms, both direct and indirect, including reduced Shh ligand production, impaired post-translational processing, and inducing ap

FGF21-inducing rare sugars for addressing obesity and alcoholism.

Obesity and alcoholism are major global health burdens driven in part by maladaptive ingestive behaviors. Accordingly, functional foods that curb overeating and excessive alcohol consumption may help prevent these disorders. Ingestion of simple sugars or alcohol increases plasma fibroblast growth factor 21 (FGF21), which activates oxytocin (OXT) neurons in the paraventricular nucleus of the hypothalamus (PVH) and suppresses the specific appetite for sugar and alcohol. D-Allulose and D-tagatose are generally recognized as safe rare sugars and serve as potent inducers of FGF21. Oral administrati

Adipokine Profile in Non-alcoholic Fatty Liver Disease.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is commonly linked with metabolic disturbances such as central obesity, insulin resistance, dyslipidaemia and low-grade inflammation. Adipokines released from adipose tissue may influence hepatic fat accumulation, metabolic dysfunction and progression of liver injury. This study was conducted to assess the serum adipokine profile in patients with NAFLD, compare it with healthy controls and evaluate its relationship with anthropometric and biochemical parameters. MATERIALS AND METHODS: This case-control study included 100 participants, cons

Alcoholic cirrhosis with Wernicke encephalopathy: Clinical case report.

RATIONALE: Early differentiation between hepatic encephalopathy and Wernicke encephalopathy (WE) is clinically critical in patients with decompensated alcoholic cirrhosis, as vitamin B1 deficiency can produce overlapping neurological symptoms that obscure the correct diagnosis and delay life-saving treatment. PATIENT CONCERNS: A 55-year-old male with a 2-year history of alcoholic cirrhosis was admitted to the hospital due to progressively worsening abdominal distension, diarrhea, and fatigue over the month prior to presentation. DIAGNOSES: WE was established based on the classic clinical tri

Application of tobacco control experience to alcohol consumption measures.

Low- and middle-income countries face a rapidly escalating, alcohol-related public health crisis spanning cancer, injury, liver disease, mental health disorders and the complications of human immunodeficiency virus infections. Despite compelling evidence that alcohol is a leading risk factor for premature death and disability, few countries include comprehensive alcohol control policies in their public health agenda. This gap between evidence and policy is critical because evidence-based interventions could decrease the alcohol-attributable disease burden by up to 30% within a decade. We condu

The brain age gap as a predictor of alcohol initiation in adolescence.

BACKGROUND: Growing evidence suggests regional and network-level brain imaging features in late childhood are predictive of alcohol use in adolescence. However, the directionality of these effects (i.e. whether they reflect accelerated or delayed neuromaturation) are mixed. We applied a Brain Age Gap Estimation (BrainAGE) model to examine whether overall deviations from typical brain aging trajectories are predictive of (1) alcohol initiation and (2) use behaviour (experimentation versus binge drinking) in adolescence. METHODS: Data from the Adolescent Brain Cognitive Development study releas

Gut microbiome in non-alcoholic fatty liver disease.

Non-alcoholic fatty liver disease (NAFLD) has a rapidly growing incidence worldwide, affecting approximately one-third of world population. The disturbance of gut commensal bacteria impacting host's homeostasis is referred to as gut dysbiosis. The gut microbiome contributes to the pathogenesis of NAFLD through various pathways. Gut microbiota is at constant interactions with the intestinal epithelial barrier and affects its integrity. Through gut-liver axis, gut microbiota may influence liver immune function. The release of lipopolysaccharides (LPS) from intestines to portal vein which are tra

Framing alcohol harm in the UN context - the importance of language.

BACKGROUND: Alcohol is a global health issue with a high level of controversy. After being absent from World Health Organization (WHO) global governing body discussions for about 20 years, alcohol re-entered the agenda in 2005. The expression 'harmful use of alcohol' became the compromise language after hard negotiations, an example of 'adopted language" that has remained for almost 20 years. This article analyses the background and use of the expression 'harmful use of alcohol' in the context of WHO governing bodies, current challenges and implications for public health. METHODS: The article

Discovery, production and characterization of bacterial aryl-alcohol oxidases.

Alcohol oxidation is a pivotal reaction in synthetic chemistry. Classical procedures typically involve the use of hypervalent reagents and organic solvents that often lead to poor selectivity, overoxidation issues and low atom-economy processes. The implementation of biocatalytic methods for this reaction is particularly appealing and several enzyme types have shown to catalyze this process although equilibrium issues, cofactor and coenzyme recycling or the use of external mediators limit the uptake of biocatalytic oxidations in large scale. Alcohol oxidases catalyze the non-reversible aerobic

Non-coding RNAs in alcohol-associated liver disease.

Non-coding RNAs (ncRNAs), encompassing microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs), have emerged as critical regulators of gene expression and cellular function. In alcohol-associated liver disease (ALD), chronic alcohol consumption disrupts the expression and function of ncRNAs in the liver and circulation, contributing to the disease's pathogenesis and progression. Dysregulated ncRNAs influence key pathways involved in hepatocyte injury, lipid metabolism, inflammation, and hepatic stellate cell (HSC) activation, thereby exacerbating steatosis, inflammatio

Association between alcohol consumption and mortality in Parkinson's disease.

Previous studies on the association between alcohol consumption and risk of Parkinson's disease (PD) have produced controversial results. However, the relationship between alcohol consumption and mortality in PD has scarcely been investigated. Among the nationwide population data from Korea National Health Insurance Service, newly diagnosed PD (ICD-10 code: G20 and a rare intractable disease registration code: V124), between 2009 and 2017, were selected. Alcohol consumption habit was obtained from a self-reported questionnaire on the National Health Screening Program. 32,419 individuals with P

Copper-Mediated Oxidative Chloro- and Bromodifluoromethylation of Aliphatic Alcohols.

The synthesis of chloro- and bromodifluoromethyl alkyl ethers remained a fundamental challenge in synthetic chemistry. Herein we report the efficient and direct synthesis of chloro- and bromodifluoromethyl alkyl ethers through copper-mediated oxidative chrolo- and bromodifluoromethylation of aliphatic alcohols with difluorocarbene-reagents. This difluorocarbene-involved oxidative coupling protocol exhibited broad functional group compatibility and was applicable to a wide range of primary and secondary alcohols.
