We have previously reported that the administration of metrazole (MTZ) produces a sequential, dose-dependent induction of c-fos and proenkephalin (Penk) gene expression in the rat hippocampus and adrenal. The adrenal is more sensitive to induction of these genes by MTZ. In the present study, we have compared the induction of c-fos and Penk in the hippocampus and adrenal, and examined the consequences of selected pharmacological manipulations. Treatment with LY274614, a competitive NMDA-receptor antagonist, blocked MTZ-induced convulsions and the MTZ-induction of c-fos and PPenk mRNAs in the hi

Lymphotoxin (LT) can activate human neutrophils. Using a hemolytic plaque assay to detect secretion of lactoferrin and myeloperoxidase (MPO) from single adherent neutrophils, we showed that LT induced secretion from both primary and secondary granules. Incubation of cells with cytochalasin B was required for MPO secretion, and it enhanced lactoferrin secretion. Pertussis toxin, which blocks a G-protein in the plasma membrane, inhibited LT-induced exocytosis of MPO, but not of lactoferrin. Incubation with LT did not induce any detectable changes of the cytoplasmic free [Ca2+] in neutrophils. On

Exposure of synaptosomes to microwave radiation at a power density of 10 mW/sq cm or more produced stimulation of the 32Pi-incorporation into phosphoinositides. The extent of 32Pi incorporation was found to be much more pronounced in phosphatidylinositol-4-phosphate (PIP), and phosphatidylinositol-4,5-bisphosphate (PIP2) as compared to phosphatidylinositol (PI) and phosphatidic acid (PA). Other lipids were also found to incorporate 32Pi but no significant changes in their labeling were seen after exposure to microwave radiation. Inclusion of 10 mM lithium in the medium reduced the basal labeli

Pulmonary fibrosis is a progressive fibrotic lung disorder in which chemical injury, inflammation, extracellular matrix deposition, and endothelial dysfunction converge to remodel lung architecture. Baicalein, a chemically defined flavonoid, has reported anti-inflammatory and antifibrotic activity, but the endothelial mechanotransduction events linked to its effects in bleomycin-induced fibrosis remain insufficiently defined. Here, we evaluated whether baicalein modulates endothelial Piezo1-associated signaling during experimental pulmonary fibrosis. A bleomycin-induced mouse model, endothelia

Photobiomodulation (PBM) is a non-thermal therapeutic modality in which low-intensity red or near-infrared light is used to modulate biological activity. Experimental inner-ear studies report preservation of auditory neurons, attenuation of vestibular injury, improved survival of transplanted stem cells, and changes in otic organoid differentiation. These observations raise the possibility that PBM may influence differentiation-associated cellular states in addition to providing cytoprotection. The classical mechanistic model proposes that photons interact with mitochondrial chromophores, part

INTRODUCTION: Huntington's Disease (HD) is an autosomal dominant neurodegenerative disorder that is caused by expansion of Cytosine-Adenine-Guanine (CAG) repeats in the Huntingtin (HTT) gene and leads to progressive motor, cognitive, and psychiatric dysfunction. Mitochondrial dysfunction is a major pathological feature of HD. Mitochondrial Rho GTPase 1 (Miro1) controls mitochondrial trafficking, calcium homeostasis, mitophagy, and neuronal signaling, and its dysregulation may contribute significantly to the progression of HD.
METHODS: The literature search to identify relevant studies on Miro

Plant-derived extracellular vesicles (PDEVs) are emerging as bioactive dietary particles with the capacity to modulate mammalian physiology. Here, we characterize the structure and functional properties of apple-derived extracellular vesicles (ADEVs) and evaluate their relevance to neuroimmune and gut-brain communication. ADEVs exhibit canonical PDEV features and elicit rapid IP₃-dependent Calcium (Ca²⁺) signaling in fibroblasts while preserving blood-brain barrier integrity. Neural assays reveal marked cell-type specificity: ADEVs are efficiently internalized by glial cells and activate glial

Selenium-independent peroxiredoxin 6 (Prdx6) is a unique member of the peroxiredoxin family, which protects cells from various stressors by regulating reactive oxygen species (ROS) and maintaining survival signaling. As a multifunctional "moonlighting" protein, Prdx6 exhibits glutathione peroxidase (GPx), acidic calcium-independent phospholipase A2, and lysophosphatidylcholine acyltransferase activities, enabling it to reduce ROS. Loss of Prdx6, owing to dysregulation of its transactivator nuclear factor erythroid 2-related factor 2 or aberrant oxidative post-translational modifications from a

Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer with limited targeted therapeutic options, underscoring the urgent need for novel treatment strategies. Feilike (FLK), a Traditional Chinese Medicine formula with heat-clearing and detoxifying properties, aligns with key pathological features implicated in breast cancer progression. In addition, several of its components have demonstrated anti-tumor activity, positioning FLK as a potential therapeutic candidate for TNBC. In this study, we employed an integrated approach combining network pharmacology, transcriptomic

Diabetic cardiomyopathy (DCM) is a major contributor to heart failure in patients with diabetes. Although berberine (BBR) exhibits broad metabolic and cardiovascular benefits, its mechanistic role in DCM remains incompletely defined. Cardiovascular function was assessed using arterial blood pressure (ABP), electrocardiography (ECG), and echocardiography. Biochemical analyses included glycemic indices, lipid profile, oxidative stress markers, inflammatory cytokines, and cardiac injury biomarkers. Cardiac and aortic tissues were evaluated for calcium (Ca2⁺) content, gene expression by RT-PCR, an

AIMS: Nephrolithiasis, a prevalent and recurring kidney disease is associated with oxidative stress, inflammation, apoptosis, necroptosis, and fibrosis. Nesfatin-1, a bioactive peptide, is known to have antioxidant, anti-inflammatory, and antifibrotic properties, however, its role in nephrolithiasis associated pathological changes remains unexplored. Moreover, the receptor by which nesfatin-1 exerts its protective effects is unknown. Thus, the present study aimed to elucidate the role and underlying molecular mechanisms of nesfatin-1 in a mouse model of glyoxylate-induced nephrolithiasis/nephr

PURPOSE OF REVIEW: We evaluate the converging evidence positioning lithium as a systemic modulator of bone and brain health through shared molecular pathways. This review examines the molecular basis, preclinical data, and clinical observations suggesting that lithium-long established as first-line therapy for bipolar disorder-may simultaneously protect against osteoporosis and neurodegeneration as two clinical conditions increasingly recognized to share biological substrates.
RECENT FINDINGS: Lithium inhibits glycogen synthase kinase-3β (GSK-3β), stabilizes β-catenin, and activates Wnt signa

Type 2 diabetes mellitus (T2DM) and coronary artery disease (CAD) are closely linked cardiometabolic disorders that share common inflammatory, metabolic, and vascular mechanisms. However, the molecular mediators underlying their interconnected pathophysiology remain incompletely understood. Orosomucoid 2 (ORM2), a hepatocyte-derived acute-phase glycoprotein, has emerged as a potential mediator at the interface of metabolic and vascular dysfunction. Experimental and clinical evidence suggests that ORM2 plays important roles in hepatic lipid metabolism, insulin sensitivity, and immune regulation

Pathological cardiac hypertrophy is a critical pathological process involved in the development of various cardiovascular diseases. Isochlorogenic acid A (ICAA) is a phenolic compound found in a wide spectrum of herbal medicines and plants, and several pharmacological properties of ICAA have been examined. However, there is currently no report on whether ICAA has therapeutic effect on pathological cardiac hypertrophy. Thus, the aim of this study was to explore the protective effects of ICAA on cardiac hypertrophy and clarify its potential mechanism. In our research, we found that ICAA effectiv

Metabolic diseases are jointly driven by insulin resistance, chronic inflammation, lipotoxicity, and disrupted organelle homeostasis arising from sustained nutrient overload. These disorders substantially increase the risk of cardiovascular, renal, and other multi-organ complications and have become a major global public health burden. Mitochondria are central organelles that integrate energy metabolism with stress signaling. They participate in fatty acid β-oxidation, the tricarboxylic acid cycle, and oxidative phosphorylation, while also regulating reactive oxygen species generation, mitocho

Calcific aortic valve disease (CAVD) is a progressive fibrocalcific illness for which no effective pharmaceutical treatment exists. This study investigated whether carbamoylated high-density lipoprotein (C-HDL), a defective type of HDL that can develop during inflammation, contributes to CAVD progression and the involved molecular pathways. Male ApoE-/- mice were divided into three groups: CAVD model, cyanate-treated, and inhibitor, and analyzed after 12 weeks. C57BL/6 mice on a regular diet served as blank controls. Serum paraoxonase-1 (PON1), aortic valve calcification, cluster of differenti

BACKGROUND: Chemical injury of the cornea, particularly alkali-induced corneal injuries, triggers a self-amplifying cascade of inflammation, extracellular matrix disruption, and pathological neovascularization that often resists conventional therapy. Transglutaminase-2 (TG2) is a calcium-dependent enzyme whose activation in injured tissues orchestrates crosslinking of matrix proteins and modulation of inflammatory signaling, positioning it as a mechanistic hub in tissue damage.
MAIN TEXT: Here we review the molecular and cellular mechanisms underpinning TG2's role in corneal injury, focusing

Mesenchymal stem cell (MSC) osteodifferentiation involves adaptations of lipid metabolism but the specific regulatory roles of lipid species remain underexplored. We use a global metabolomics approach, comprising LC-MS lipidomics and NMR metabotolomics, to identify mechanistic features of human adipose-derived MSC (hAMSC) osteodifferentiation and new markers of osteogenic progression. Results show that, upon differentiation, cellular metabolism progresses through an early osteocommitment phase (day 7) followed by active osteodifferentiation (day 21), with underlying proliferation contributing

Fine particulate matter (PM2.5) is toxic to reproduction and can cause a range of reproductive disorders. However, the mechanisms underlying female reproductive impairment due to PM2.5 exposure are unclear. In this study, we aimed to investigate the effects of PM2.5 on rat ovaries and the underlying molecular mechanisms. Animal models showed that PM2.5 exposure induced abnormal ovarian function and disturbed ovarian-related hormone homeostasis in rats, significantly increasing the proportion of follicular atresia. This process was accompanied by ovarian inflammation, oxidative stress, apoptosi
