Single-cycle parainfluenza virus type 5 vectors for producing recombinant proteins, including a humanized anti-V5 tag antibody.

Parainfluenza virus type 5 (PIV5) can cause either persistent or acute/lytic infections in a wide range of mammalian tissue culture cells. Here, we have generated PIV5 fusion (F)-expressing helper cell lines that support the replication of F-deleted viruses. As proof of the principle that F-deleted single-cycle infectious viruses can be used as safe and efficient expression vectors, we have cloned and expressed a humanized (Hu) version of the mouse anti-V5 tag antibody (clone SV5-Pk1). We show that multiple different cell lines can be infected and express high levels of the Hu anti-V5 antibody

Fine-Needle Aspiration Features of BCOR-CCNB3 Sarcoma.

OBJECTIVES: BCOR-CCNB3 sarcoma (BCS) is an undifferentiated tumor that has some clinical and morphologic similarity to classic Ewing sarcoma, but it is characterized by a distinct BCOR-CCNB3 gene fusion. There are no reports describing its cytomorphologic findings. METHODS: We describe cytologic findings of five molecularly proven BCS cases (four males and one female, aged 8.5-22 years). RESULTS: Smears showed mainly round cells, but some spindle cells and rhabdoid-like cells were also observed. Dispersed cells dominated in smears, but also distinct pseudopapillary structures with vascular c

Docetaxel enhances the cytotoxic effects of imatinib on Philadelphia positive human chronic myeloid leukemia cells.

Chronic myelogenous leukemia (CML) results from a translocation between chromosomes 9 and 22 which generates BCR/ABL fusion protein and characterized by uncontrolled proliferation of immature white blood cells. Imatinib, a molecularly targeting anticancer agent, is used widely for the treatment of CML and showed significant activity in chronic and accelerated phases but much less in blast crisis phase. The resistance to imatinib especially in blast crisis phase is recognized as a major problem in the treatment of CML patients. Docetaxel is shown to arrest cells in G2/M phase of the cell cycle

OX40 ligation on activated T cells enhances the control of Cryptococcus neoformans and reduces pulmonary eosinophilia.

Pulmonary eosinophilia induced in C57BL/6 mice after Cryptococcus neoformans infection is driven by CD4(+) Th2 cells. The immunological mechanisms that protect against eosinophilia are not fully understood. Interaction of OX40 (CD134) and its ligand, OX40L, has been implicated in T cell activation and cell migration. Unlike CD28, OX40 is only expressed on T cells 1-2 days after Ag activation. Manipulation of this pathway would therefore target recently activated T cells, leaving the naive repertoire unaffected. In this study, we show that engagement of OX40 by an OX40L:Ig fusion protein drives

Nerve growth factor-induced differentiation changes the cellular organization of regulated Peptide release by PC12 cells.

PC12 cells, like endocrine chromaffin cells, undergo neuronal-like differentiation in response to nerve growth factor (NGF). Here we report that this phenotype conversion produces major changes in release of a green fluorescent protein-tagged neuropeptide-hormone. First, the spatial distribution of the releasable pool is altered; peptide release from untreated cells is supported predominantly by membrane-proximal vesicles, whereas a diffuse pool at the ends of processes is used by NGF-treated cells. Second, the time course of release evoked by photolysis of caged Ca(2+) is faster after differe

Retinoic acid promotes conjunctival epithelium differentiation and goblet cell regeneration: evidence from novel 3D conjunctival organoids and whole-mount PAS staining.

PURPOSE: To investigate the differentiation effects of retinoic acid on primary conjunctival epithelium using both 2D and 3D models, and to … te the effects of retinoic acid eye drops. RESULTS: In the 2D culture model, retinoic acid induced cell fusion, decreased stemness marker expression, and increased goblet cell differentiation markers. In the 3D organoid model, retinoic acid treatment led to elevated expression of goblet cell markers, including Muc5ac, Tff1, and Gcnt3, as confirmed by qRT-PCR …

Molecular pathogenesis of lymphomas of mucosa-associated lymphoid tissue--from (auto)antigen driven selection to the activation of NF-κB signaling.

Lymphomas of mucosa-associated lymphoid tissue (MALT) are typically present at sites such as the stomach, lung or urinary tract, where lymphoid tissues scatter in mucosa lamina propria, intra- or sub-epithelial cells. The infection of certain pathogens, such as Helicobacter pylori, Chlamydophila psittaci, Borrelia burgdorferi, hepatitis C virus, or certain autoantigens cause these sites to generate a germinal center called the "acquired lymphoid tissue". The molecular pathogenesis of MALT lymphoma is a multi-step process. Receptor signaling, such as the contact stimulation of B cell receptors

Optimization of Affibody Affinity to Improve Distribution in Tumor Models.

Poor distribution is often a limiting factor when using affinity targeted treatments. High affinity creates a barrier to transport because proteins cannot diffuse throughout the tissue while bound to the cell surface. Lowering the affinity can help with transport throughout a tumor but can also result in a loss of total delivery and specificity for the target. In this research, we used point mutations to create affibodies with a range of different affinities for epidermal growth factor receptor (EGFR) and measured their penetration through tumor spheroids. Mutants in the range of ∼7-12 nM KD w

Hyalinizing Clear Cell Carcinoma of the Lung With Uncommon Distant Cutaneous Metastasis and Aggressive Clinical Course.

Hyalinizing clear cell carcinoma (HCCC) is a rare tumor. Most lesions arise in the salivary gland; however, other primary sites include the nasal and oral cavities, and rarely lung. Lesions are characterized by tumor lobules with pale or clear cytoplasm, arranged in nests or cords, surrounded by a mixed hyalinized and fibrocellular stroma. EWSR1-ATF1 gene fusion is often reported in these lesions. Typically, HCCC bears a good prognosis. We report a unique case of primary lung HCCC (PLHCCC) with cutaneous metastasis and aggressive clinical course. To the best of our knowledge, cutaneous metasta

Toward Safer Biotherapeutics: Expression and Characterization of a Humanized Chimeric L-Asparaginase in E. coli.

Acute lymphoblastic leukemia (ALL) is the most common cancer affecting children, making up about 80% of all acute leukemia cases in the pediatric population. While treatment with L-asparaginase (ASNase) has greatly improved survival rates, its bacterial origin often causes immune reactions in some patients, which can reduce how well the therapy works. To overcome this challenge, previous in silico studies designed a humanized chimeric ASNase by swapping out the predicted immunogenic parts of the bacterial enzyme with similar, less immunogenic segments from the human version-while keeping the e

Teardrop shapes minimize bending energy of fusion pores connecting planar bilayers.

A numerical gradient flow procedure was devised to characterize minimal energy shapes of fusion pores connecting two parallel planar bilayer membranes. Pore energy, composed of splay, tilt, and stretching, was obtained by modeling each bilayer as two monolayers and treating each monolayer of a bilayer membrane as a freely deformable surface described with a mean lipid orientation field. Voids between the two monolayers were prevented by a steric penalty formulation. Pore shapes were assumed to possess both axial and reflectional symmetry. For fixed pore radius and bilayer separation, the gradi

Intestinal epithelial CD98 synthesis specifically modulates expression of colonic microRNAs during colitis.

The transmembrane glycoprotein CD98 is known to be involved in intestinal inflammation. In the present study, we found that CD98 overexpression in intestinal epithelial cells does not normally affect the expression of colonic (epithelial and immune cell) microRNAs (miRNAs), small noncoding RNAs that posttranscriptionally regulate a wide variety of biological processes. However, upon dextran sulfate sodium (DSS) treatment, the expression of several colonic miRNAs, but not miRNAs from other tissues such as liver and spleen, were differentially regulated in mice overexpressing CD98 in epithelial

Endogenous retroviruses in the human genome sequence.

The human genome contains many endogenous retroviral sequences, and these have been suggested to play important roles in a number of physiological and pathological processes. Can the draft human genome sequences help us to define the role of these elements more closely?

First-Line Lorlatinib for ALK-Rearranged Pulmonary Large-Cell Neuroendocrine Carcinoma with Brain Metastases and Suspected Leptomeningeal Involvement: A Case Report and Literature Review.

We report a case of ALK-rearranged pulmonary large-cell neuroendocrine carcinoma (LCNEC) with brain metastases and radiologically suspected leptomeningeal involvement, successfully treated with first-line lorlatinib. A 58-year-old woman was diagnosed with LCNEC, and molecular testing revealed an EML4-ALK fusion. Lorlatinib achieved a partial response, with regression of the primary lesion, brain metastases, and suspected leptomeningeal lesions. Our literature review showed that ALK-positive LCNEC occurs more often in younger, female, and never- or light-smoking patients. In such cases, molecul

Structure-guided identification of a PrPC-binding peptide derived from Brucella abortus Hsp60 for receptor-mediated mucosal targeting.

Here, we report the structure-guided identification pipeline of a novel PrPC-binding peptide, PEP1, derived from Brucella abortus Hsp60 through computational docking. Computational modeling predicted that PEP1 engages the PrPC binding interface with favorable binding energetics, motivating its experimental evaluation as a receptor-targeting ligand. Fusion of PEP1 to a model protein enabled receptor-mediated targeting to intestinal M-cell-associated regions and resulted in increased mucosal IgA and systemic IgG responses relative to the untargeted protein. Together, these findings establish PEP

Spatiotemporal control of myonuclear identity during muscle regeneration.

Skeletal muscle exhibits an unusually complex architecture, in which large multinucleated myofibers accommodate a small population of resident muscle stem cells (MuSCs) along their surface. In addition, myofibers contain molecularly and functionally specialized domains at junctions with motor neurons and tendons. Regeneration of this tissue, therefore, requires a coordinated series of events, spanning MuSC activation, proliferation, fusion to restore myofiber mass, as well as reconstruction of specialized domains. Recent advances have begun to reveal how myogenic nuclei undergo dynamic state t

A Case of Primary Endobronchial Hyalinizing Clear Cell Carcinoma Presenting as an Oncologic Emergency Due to Complete Left Main Bronchus Obstruction Successfully Managed with Airway Intervention.

A 66-year-old man presented with dyspnea and severe hypoxemia, requiring mechanical ventilation. Computed tomography revealed an obstruction of the left main bronchus by a bronchial tumor, thus prompting airway intervention to relieve the stenosis. A histopathological examination of the tumor revealed clear cytoplasm and hyalinized stroma, and rearrangement of EWSR1 was detected by a fluorescence in situ hybridization analysis. Comprehensive genomic profiling was performed because of the rarity of this case. GenMineTOP, capable of detecting the EWSR1::ATF1 fusion, contributed to a definitive d

Concurrent Bone Marrow Acute Undifferentiated Leukemia and Mediastinal T-Lymphoblastic Lymphoma With Identical SET::NUP214 Fusion and PHF6 and EZH2 Mutations.

Acute undifferentiated leukemia (AUL) is a rare hematologic malignancy lacking lineage-specific markers. Concurrent, clonally related AUL and T-lymphoblastic lymphoma (T-LBL) has not been reported previously. Here we describe a patient who was diagnosed with AUL in the bone marrow and T-LBL in the mediastinum after a thorough immunophenotyping by flow cytometry and immunohistochemistry. Despite their immunophenotypic differences, the AUL and T-LBL showed identical genetic alterations: SET::NUP214 fusion, PHF6, and EZH2 mutations. The patient achieved and remained in complete remission after ch
