Low-dose ionizing radiation drives thyroid carcinogenesis via VPS53 enhanced EGFR Re-recycling.

Ionizing radiation is a confirmed independent environmental factor for thyroid cancer. Environmental and occupational exposure are exposed to low-dose ionizing radiation (LDR), which underscores the persistent need to decipher the mechanisms of LDR induced thyroid carcinogenesis. In addition to the canonical driver mutations or chromosomal rearrangements, there remains a gap in understanding the key drivers of LDR-induced thyroid cancer that needs to be clarified. VPS53, a core subunit of the Golgi-associated retrograde protein (GARP) complex, mediates retrograde vesicular trafficking, but its

Integrated Analysis Reveals ZNF184 as a Novel Regulator of Stemness-Associated Paclitaxel Resistance and Tumor Progression in Breast Cancer.

Chemotherapy resistance is the primary cause of clinical treatment failure and unfavorable prognosis among breast cancer patients. Consequently, the exploration of novel molecular targets for chemotherapy resistance is warranted. Here, we demonstrated that Zinc Finger Protein 184 (ZNF184) facilitates chemoresistance in breast cancer. Through integrated bioinformatics and experimental validation, we identified that ZNF184 was highly expressed in paclitaxel-resistant breast cancer cells. Knockdown of ZNF184 inhibited cell proliferation and re-sensitized resistant cells to paclitaxel in vitro and

KRT17 promotes triple negative breast cancer through activation of Wnt signaling and γδ T-cells recruitment.

Triple-negative breast cancer (TNBC) is a very aggressive form of breast cancer and Black American (BA) women face disproportionately higher mortality rates than White American (WA) women. The molecular mechanism behind this disparate clinical outcome remains poorly understood. We find that BA TNBC patients exhibit higher protein expression of KRT17 compared to WA TNBC and non-TNBC patients and correlates to poor distant metastasis-free survival. Mechanistic studies in metastatic mouse TNBC tumors with higher Krt17 demonstrates higher Wnt signaling targets, cancer stem cells (CSCs), which posi

Role of Latrophilin-1 and Latrophilin-2 as Downstream Effectors of Androgen Receptor Signaling in Urothelial Tumorigenesis.

BACKGROUND: Accumulating evidence indicates a critical role of androgen receptor (AR) in the pathogenesis of male-dominant urothelial cancer. Meanwhile, the functions of latrophilins (LPHNs), a subfamily of G protein-coupled receptors, remain largely uncharacterized in neoplastic conditions. AIMS: To determine the functional role of LPHN1 and LPHN2 in association with AR signaling in urothelial tumorigenesis. METHODS AND RESULTS: AR overexpression in immortalized human normal urothelial SVHUC cells or androgen treatment in SVHUC-AR cells markedly increased the expression levels of LPHN1 and

The LINC02041/SRSF1 Axis Facilitates Aerobic Glycolysis and Stemness Maintenance in Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) remains one of the most aggressive and lethal malignancies worldwide, with high rates of metastasis and recurrence contributing to its poor prognosis. There is an urgent need to elucidate the molecular mechanisms driving HCC progression and to develop effective therapeutic strategies. Metabolic reprogramming, especially aerobic glycolysis known as the Warburg effect, is a well-established hallmark of cancer. Concurrently, cancer stem cells (CSCs) play crucial roles in tumor initiation, therapy resistance, and recurrence. However, the involvement of long non-codin

MYCN reprograms postnatal cells toward embryonic-like transcriptional states and drives tumorigenesis.

The embryonic-like features of neuroblastoma are commonly explained by a developmental arrest model, in which neural crest-derived progenitors undergo transformation after failing to complete normal sympathoadrenal differentiation. Whether such features can instead be actively imposed on postnatal cells during tumor development remains unclear. Here, we show that Mycn, but not c-Myc, induced adrenal tumors after activation in adult tyrosine hydroxylase-positive cells, accompanied by repression of differentiated programs and acquisition of embryonic cell-like features. Similarly, in adult hepat

Pharmacological inhibition of the canonical Wnt/β-catenin signaling pathway sensitizes 5-fluorouracil-resistant colorectal cancer cells.

BACKGROUND: The activation of the Wnt/β-catenin pathway with chemoresistance in colorectal cancer has been hypothesized. However, the use of specific inhibitors for targeting this pathway has not been well explored. In the present study, we analyze the activation of this pathway and its role in the stemness phenotype acquisition to regulate chemoresistance using our long-term 5-fluorouracil (5-FU) - resistant model of colorectal cancer cells, known to present epithelial-mesenchymal transition and enhanced migration and invasion. METHODS AND RESULTS: Initially by bioinformatic analyses we demo

Nicotine-driven enhancement of tumor malignancy in triple-negative breast cancer via additive regulation of CHRNA9 and IGF1R.

Cigarette smoking is a significant risk factor for cancer development with complex mechanisms. This study aims to investigate the impact of nicotine exposure on the regulation of stemness- and metastasis-related properties via cholinergic receptor nicotinic alpha 9 subunit (CHRNA9) and insulin-like growth factor-1 receptor (IGF1R) and to evaluate their therapeutic potential in triple-negative breast cancer (TNBC). We performed Kaplan-Meier survival analysis of public databases and revealed that high expression of CHRNA9, IGF1R signaling molecules, and stemness genes was significantly associate

Photodynamic therapy boosts the anti-proliferative activity of oxaliplatin in cervical cancer cells by regulating stemness-related genes.

Cervical cancer is the fourth most prevalent cancer among women worldwide and remains a significant contributor to cancer-related mortality, particularly in low- and middle-income countries, largely due to treatment resistance and disease recurrence. Growing evidence indicates that cancer stem cells (CSCs) play a pivotal role in tumor initiation, progression, and therapeutic resistance. Consequently, the development of novel therapeutic strategies capable of eliminating cancer cells while simultaneously targeting CSC-associated pathways holds substantial clinical promise. This study aimed to e

CEBPB promotes transformation of endometrial complex atypical hyperplasia to endometrial cancer.

BACKGROUND: As the precursor malignancy of endometrial cancer (EC), about 50% of endometrial complex atypical hyperplasia (CAH) will eventually progress to EC. Elucidating the underlying transformation mechanisms could aid in disease management. METHODS: EC, CAH, reversed CAH and normal endometrium tissues were collected and sequenced to identify genes involved in the malignant transformation. CEBPB expression was then compared between endometrial CAH and normal endometrium. After evaluation of its effects on proliferation, apoptosis, EMT, migration and invasion by using primary culture, the

Cetuximab and Paclitaxel Drug Response in Head and Neck Tumor Stem Cells.

Head and neck cancer (HNC) is one of the most common types of cancer in the world, characterized by resistance to conventional therapies and an unfavorable prognosis due to the presence of tumor stem cells (TSCs). TSCs are cell subpopulations with high potential for invasion, migration, and metastasis, being responsible for the initiation and dissemination of cancer. This study aimed to evaluate the efficacy of treatments with cetuximab and paclitaxel, alone and in combination, in TSCs from oral cavity (SCC-28) and hypopharynx (FADU) cancer cell lines. In addition, the influence of the gene an

FDFT1 repression by piR-39980 prevents oncogenesis by regulating proliferation and apoptosis through hypoxia in tongue squamous cell carcinoma.

AIM: Tongue squamous cell carcinoma (TSCC) is one of the most aggressive tumors whose underlying molecular mechanism remains elusive. Previous studies have identified piR-39980, a non-coding RNA, as a tumour suppressor or oncogene in different malignancies and the cholesterogenic protein, Farnesyl-Diphosphate Farnesyltransferase 1 (FDFT1) playing critical roles in cancer. The present study investigates the role of piR-39980, and its target FDFT1, in regulating the malignancy of TSCC. MAIN METHODS: We performed qRT-PCR to determine the expression of FDFT1, piR-39980 and validated FDFT1 as a ta

Epigenetic effects of low-level sodium arsenite exposure on human liver HepaRG cells.

Chronic exposure to inorganic arsenic is associated with a variety of adverse health effects, including lung, bladder, kidney, and liver cancer. Several mechanisms have been proposed for arsenic-induced tumorigenesis; however, insufficient knowledge and many unanswered questions remain to explain the integrated molecular pathogenesis of arsenic carcinogenicity. In the present study, using non-tumorigenic human liver HepaRG cells, we investigated epigenetic alterations upon prolonged exposure to a noncytotoxic concentration of sodium arsenite (NaAsO2). We demonstrate that continuous exposure of

lncRNA XIST interacts with miR-140 to modulate lung cancer growth by targeting iASPP.

X-inactive specific transcript (XIST), one of the first found cancer-associated long non-coding RNAs (lncRNAs), is involved in the development and progression of many types of tumors. Aberrant expression of XIST has been observed in hepatocellular carcinoma, cervical, breast, ovarian and colorectal cancer. However, the exact effects and molecular mechanisms of XIST in lung cancer progression are still unknown to date. In the present study, we investigated the role of XIST in human lung cancer cell lines and clinical tumor samples in order to determine the function of this molecule. In our rese

Time-matched analysis of DNA adduct formation and early gene expression as predictive tool for renal carcinogenesis in methylazoxymethanol acetate treated Eker rats.

Genotoxic carcinogens pose great hazard to human health. Uncertainty of current risk assessment strategies and long latency periods between first carcinogen exposure and diagnosis of tumors have raised interest in predictive biomarkers. Initial DNA adduct formation is a necessary step for genotoxin induced carcinogenesis. However, as DNA adducts not always translate into tumorigenesis, their predictive value is limited. Here we hypothesize that the combined analysis of pro-mutagenic DNA adducts along with time-matched gene expression changes could serve as a superior prediction tool for genoto

Targeting CK2-driven non-oncogene addiction in B-cell tumors.

Genetic mutations of oncogenes often underlie deranged cell growth and altered differentiation pathways leading to malignant transformation of B-lymphocytes. However, addiction to oncogenes is not the only drive to lymphoid tumor pathogenesis. Dependence on non-oncogenes, which act by propelling basic mechanisms of cell proliferation and survival, has also been recognized in the pathobiology of lymphoid leukemias, lymphomas and multiple myeloma. Among the growing number of molecules that may uphold non-oncogene addiction, a key place is increasingly being recognized to the serine-threonine kin

Stem cell characteristics of cell sub-populations in cell lines derived from head and neck cancers of Fanconi anemia patients.

Increasing evidence indicates that cancer growth is driven by a sub-population of self-renewing cancer stem cells (CSCs) and that clinical problems of tumor recurrence after therapy may be related to differential resistance of CSCs to therapeutic elimination. Fanconi anemia (FA) is an autosomal recessive disorder associated with deficiencies of DNA repair and a greatly enhanced risk of hematopoietic malignancies and of head and neck squamous cell carcinoma (HNSCC). In FA patients, lack of DNA repair complicates therapies acting through DNA damage and alternative approaches, such as targeting s

HOXB13 promotes ovarian cancer progression.

Deregulated expression of HOXB13 in a subset of estrogen receptor-positive breast cancer patients treated with tamoxifen monotherapy is associated with an aggressive clinical course and poor outcome. Because the ovary is another hormone-responsive organ, we investigated whether HOXB13 plays a role in ovarian cancer progression. We show that HOXB13 is expressed in multiple human ovarian cancer cell lines and tumors and that knockdown of endogenous HOXB13 by RNA interference in human ovarian cancer cell lines is associated with reduced cell proliferation. Ectopic expression of HOXB13 is capable
