Physiology and immunology of the cholinergic antiinflammatory pathway.

Cytokine production by the immune system contributes importantly to both health and disease. The nervous system, via an inflammatory reflex of the vagus nerve, can inhibit cytokine release and thereby prevent tissue injury and death. The efferent neural signaling pathway is termed the cholinergic antiinflammatory pathway. Cholinergic agonists inhibit cytokine synthesis and protect against cytokine-mediated diseases. Stimulation of the vagus nerve prevents the damaging effects of cytokine release in experimental sepsis, endotoxemia, ischemia/reperfusion injury, hemorrhagic shock, arthritis, and

Modeling Rapid Contextual Learning in the Visual Cortex with Fast-Weight Deep Autoencoder Networks.

Recent neurophysiological studies have revealed that the early visual cortex can rapidly learn global image context, as evidenced by a sparsification of population responses and a reduction in mean activity when exposed to familiar versus novel image contexts. This phenomenon has been attributed primarily to local recurrent interactions, rather than changes in feedforward or feedback pathways-supported by both empirical findings and circuit-level modeling. Recurrent neural circuits capable of simulating these effects have been shown to reshape the geometry of neural manifolds, enhancing robust

Spatial and temporal connectivity of brain resting-state fMRI during music-induced analgesia in fibromyalgia patients.

INTRODUCTION: Music-induced analgesia (MIA) has significant clinical value for patients with fibromyalgia (FM) and serves as a key model for understanding the complex neural mechanisms underlying the effects of music on physical and mental states. However, previous research offers limited interpretation of the broader neural network characteristics underlying pain regulation through music, particularly regarding spatial synchronization and temporal connectivity. METHODS: The present study examined the neural correlates of MIA in FM patients using resting-state functional magnetic resonance im

Neuroprotective Effects of Valproic Acid on Blood-Brain Barrier Disruption and Apoptosis-Related Early Brain Injury in Rats Subjected to Subarachnoid Hemorrhage Are Modulated by Heat Shock Protein 70/Matrix Metalloproteinases and Heat Shock Protein 70/AKT Pathways.

BACKGROUND: Blood-brain barrier (BBB) disruption and neural apoptosis are thought to promote early brain injury (EBI) after subarachnoid hemorrhage (SAH). Previous studies have demonstrated that valproic acid (VPA) decreased brain injury in a prechiasmatic injection model of SAH in mice. It should be noted that the beneficial effects of VPA and the underlying mechanisms have not been fully elucidated. OBJECTIVE: To characterize the effects of VPA on BBB disruption and neural apoptosis and to determine mechanisms involved in EBI after SAH. METHODS: An endovascular perforation model was used t

Jaw and branchial arch mutants in zebrafish I: branchial arches.

Jaws and branchial arches together are a basic, segmented feature of the vertebrate head. Seven arches develop in the zebrafish embryo (Danio rerio), derived largely from neural crest cells that form the cartilaginous skeleton. In this and the following paper we describe the phenotypes of 109 arch mutants, focusing here on three classes that affect the posterior pharyngeal arches, including the hyoid and five gill-bearing arches. In lockjaw, the hyoid arch is strongly reduced and subsets of branchial arches do not develop. Mutants of a large second class, designated the flathead group, lack se

The Harmonious Dance: A Narrative Review on Psychedelics and Music in Therapeutic Settings.

The integration of psychedelics and music in therapeutic settings is gaining recognition for its potential to enhance mental health outcomes. This review synthesizes current evidence on the neurobiological mechanisms underlying this synergy, focusing on receptor-level pathways (e.g., 5-HT2A receptor agonism, BDNF-TrkB signaling) and neural circuit dynamics (e.g., default mode network desynchronization, thalamo-cortical connectivity) that mediate psychedelic action and mu-sic-induced emotional processing. By examining how music, acting as a "hidden therapist," ampli-fies the emotional and cogni

Neuroimmune Dynamics in Diseases of the Arterial Wall: Mechanistic Foundations and Translational Prospects.

PURPOSE OF REVIEW: Peripheral nerves are increasingly recognized as regulators of vascular pathology, shaping arterial tone, inflammation, and structural remodeling. This review delineates how neural circuits interface with the aortic wall, considers sympathetic and sensory pathways in atherosclerosis and aneurysm pathophysiology, and discusses emerging experimental approaches and therapeutic strategies. RECENT FINDINGS: Autonomic and sensory neural circuits are increasingly recognized as modulators of arterial disease. In experimental atherosclerosis, neuroimmune crosstalk shapes vascular in

Pharmacological and Psychological Treatment Have Common and Specific Effects on Brain Activity in Obsessive-Compulsive Disorder.

Initial treatment for obsessive-compulsive disorder (OCD) consists of pharmacological treatment with selective serotonin reuptake inhibitors (SSRIs) and/or psychological treatment with cognitive behavioral therapy (CBT). The assumption is that both treatments have different neural working mechanisms, but empirical evidence is lacking. We investigated whether these treatments induce similar or different functional neural changes in OCD. We conducted a longitudinal nonrandomized controlled trial in which thirty-four OCD patients were treated with sixteen weeks of CBT or SSRIs. Functional magneti

Investigation of the neural discrimination of acoustic characteristics of speech sounds in normal-hearing individuals through Frequency-following Response (FFR).

PURPOSE: To evaluate how the auditory pathways encode and discriminate the plosive syllables [ga], [da] and [ba] using the auditory evoked Frequency-following Response (FFR) in children with typical development. METHODS: Twenty children aged 6-12 years were evaluated using the FFR for the [ga], [da] and [ba] stimuli. The stimuli were composed of six formants and were differentiated in the F2 to F3 transition (transient portion). The other formants were identical in the three syllables (sustained portion). The latencies of the 16 waves of the transient portion (<70ms) and of the 21 waves of th

Distinct neural correlates of social and object reward seeking motivation.

The "Choose-a-Movie-CAM" is an established task to quantify the motivation for seeking social rewards. It allows participants to directly assess both the stimulus value and the effort required to obtain it. In the present study, we aimed to identify the neural mechanisms of such cost-benefit decision-making. To this end, functional Magnetic Resonance Imaging data were collected from 24 typical adults while they completed the CAM task. We partly replicated the results from our previous behavioural studies showing that typical adults prefer social over object stimuli and low effort over higher e

Comparison of village dog and wolf genomes highlights the role of the neural crest in dog domestication.

BACKGROUND: Domesticated from gray wolves between 10 and 40 kya in Eurasia, dogs display a vast array of phenotypes that differ from their ancestors, yet mirror other domesticated animal species, a phenomenon known as the domestication syndrome. Here, we use signatures persisting in dog genomes to identify genes and pathways possibly altered by the selective pressures of domestication. RESULTS: Whole-genome SNP analyses of 43 globally distributed village dogs and 10 wolves differentiated signatures resulting from domestication rather than breed formation. We identified 246 candidate domestica

Pharmacologically-mediated reactivation and reconsolidation blockade of the psychostimulant-abuse circuit: a novel treatment strategy.

Psychostimulant abuse continues to present legal, socioeconomic and medical challenges as a primary psychiatric disorder, and represents a significant comorbid factor in major psychiatric and medical illnesses. To date, monotherapeutic drug treatments have not proven effective in promoting long-term abstinence in psychostimulant abusers. In contrast to clinical trials utilizing monotherapies, combinations of dopamine (DA) agonists and selective 5-HT(3), 5HT(2A/2C), or NK(1) antagonists have shown robust efficacy in reversing behavioral and neurobiological alterations in animal models of psycho

Challenges to normal neural functioning provide insights into separability of motion processing mechanisms.

There is a long history of attempts to disentangle different visual processing mechanisms for physically different motion cues. However, underlying neural correlates and separability of networks are still under debate. We aimed to refine the current understanding by studying differential vulnerabilities when normal neural functioning is challenged. We investigated effects of ageing and extrastriate brain lesions on detection thresholds for motion defined by either luminance- or contrast modulations, known as first- and second-order motion. Both approaches focus on extrastriate processing chang
