Sex hormone binding globulin and non-protein-bound oestradiol in postmenopausal patients with primary biliary cirrhosis and normal controls.

Several studies have suggested that patients with primary biliary cirrhosis (PBC) have an increased risk of a number of medical conditions related to their oestrogen levels. This study has measured sex hormone binding globulin (SHBG) binding capacity, total oestradiol levels and percentage non-protein-bound (NPB) oestradiol and calculated the concentration of NPB oestradiol, in postmenopausal subjects in the following groups; normal controls, early PBC, advanced PBC and advanced PBC who were receiving therapy. Mean SHBG levels were higher in all groups of patients with PBC than in controls. No

In vitro production of cytokines by bone surface-derived osteoblastic cells in normal and osteoporotic postmenopausal women: relationship with cell proliferation.

To evaluate the role of cytokines produced by osteoblasts in the pathophysiology of bone lesions in postmenopausal osteoporosis (PMOP), we have determined by RIA and immunoradiometric assays the levels of prostaglandin E2 (PGE2), interleukin-1 beta (IL-1), tumor necrosis factor-alpha (TNF alpha), and IL-6 released by cultured bone surface-derived osteoblastic (OB) cells isolated from 24 untreated PMOP women with high, low, or normal bone turnover on bone biopsy. OB cells isolated from patients with high bone formation had a 2-fold increased proliferation rate in vitro compared to OB cells from

Type of postmenopausal hormone use and risk of breast cancer: 12-year follow-up from the Nurses' Health Study.

We prospectively examined the use of hormone replacement therapy in relation to breast cancer incidence in a cohort of women 30 to 55 years of age in 1976. During 12 years of follow-up (480,665 person-years) among postmenopausal women, 1,050 incident cases of breast cancer were documented. Overall, past users of replacement estrogen were not at increased risk. After adjustment for established risk factors, type of menopause, age at menopause, and current age, the rate ratio (RR) was 0.91, 95 percent confidence interval (CI) = 0.78-1.07. The risk of breast cancer was elevated significantly amon

Reduction of cardiovascular disease-related mortality among postmenopausal women who use hormones: evidence from a national cohort.

A national sample of 1944 white menopausal women greater than or equal to 55 years old from the epidemiologic follow-up of participants in the National Health and Nutrition Examination Survey was reviewed to investigate the role of hormone therapy in altering the risk of death from cardiovascular disease. Women in the study were observed for up to 16 years after the baseline survey in 1971 to 1975. By 1987 631 women had died; 347 of these deaths were due to cardiovascular disease. History of diabetes (relative risk, 2.38; 95% confidence interval 1.73 to 3.26), previous myocardial infarction (r

Bone cell responsiveness to transforming growth factor beta, parathyroid hormone, and prostaglandin E2 in normal and postmenopausal osteoporotic women.

We have shown previously that the decreased trabecular bone formation in osteoporotic postmenopausal women results from a reduced ability of osteoblastic cells to proliferate. In this study we have tested the possibility that bone cells from osteoporotic women with low bone formation have an abnormal responsiveness to hormonal or local mitogenic factors. Primary cultures of bone cells with osteoblastic characteristics were obtained by migration from the trabecular bone surface in osteoporotic postmenopausal women with high (n = 7) or low (n = 7) bone formation as evaluated histomorphometricall

Long-term prevention of postmenopausal osteoporosis by oestrogen. Evidence for an increased bone mass after delayed onset of oestrogen treatment.

Treatment of 63 oophorectomised women with oestrogen for five years prevented the reduction in bone mineral content observed in 57 women treated with a placebo preparation. When onset of therapy was delayed for three to six years there was a highly significant increase in bone mineral content mainly during the first three years of treatment. During the next two years there was no further increase in bone mineral, while the placebo-treated groups continued to lose bone at about 1% per annum. In association with the changes in bone mass, the expected biochemical effects of oestrogen therapy also

Issues for mature women with epilepsy.

Specific concerns regarding mature women with epilepsy (WWE), specifically epilepsy-associated issues during perimenopause, menopause, and postmenopause, have been emerging in the epilepsy community. This chapter presents evidence that for WWE, seizure frequency may increase during perimenopause and decrease at postmenopause, especially if a catamenial epilepsy pattern was present during the reproductive years. This finding implies that, as in other age groups, a subset of mature WWE are particularly susceptible to endogenous reproductive hormonal changes. An adverse effect on seizure frequenc…

Gut Microbiota-Derived Short-Chain Fatty Acids Mediate the Bone-Protective Effects of Metformin Against Postmenopausal Osteoporosis.

BACKGROUND: Dysbiosis-induced inflammation has been implicated in postmenopausal osteoporosis (PMOP). Although metformin has shown benefits for bone health, the exact role of the gut microbiota in the mechanism underlying this relationship remains unclear. HYPOTHESIS/PURPOSE: We hypothesised that metformin could alleviate systemic inflammation in PMOP by augmenting the synthesis of short-chain fatty acids (SCFAs) by gut microbes. METHODS: Micro-computed tomography (μCT) was used to analyse the femurs. Faecal samples were collected for 16S rRNA sequencing and SCFAs detection. Quantitative rea

Phthalate mixture exposure is associated with oxidative imbalance in postmenopausal women: a cross-sectional study using WQS, qgcomp, and BKMR.

BACKGROUND: Phthalates are widespread environmental contaminants known for their endocrine-disrupting properties. Due to the decline in estrogen's antioxidant effects, postmenopausal women may be particularly vulnerable to oxidative stress; however, the effect of phthalate mixture exposure on systemic redox balance in this population remains uncertain. OBJECTIVE: This study aimed to investigate the associations between urinary phthalate metabolites and oxidative balance score (OBS) among postmenopausal women and to identify major contributors within phthalate mixtures. METHODS: We conducted

Menopausal duration and relative skeletal muscle index predict metabolic inflexibility during walking in postmenopausal women with overweight/obesity.

Menopause predisposes a diminished capacity to alter oxidative substrate during exercise-a risk factor for cardiometabolic diseases (CMDs). Since walking behavior influences CMD, identifying factors associated with substrate use during walking may inform strategies to attenuate risk. This study assessed changes in lipid (ΔFATox) and carbohydrate (ΔCHOox) oxidation during walking and identified physiological predictors. Thirty-six women [64 ± 7 yr; body mass index (BMI): 29.5 ± 4.9 kg/m2] completed a treadmill task (0.9 m/s, 0% grade) with indirect calorimetry to quantify oxygen uptake (V̇o2) k

A Pilot Study to Describe Cardiometabolic Health Indicators and Prescription Medication Use in Postmenopausal Women with a Self-Reported History of PMOS.

Background/Objectives: Relatively little is known about how the aberrations in cardiometabolic health linked with polyendocrine metabolic ovarian syndrome (PMOS, formally called polycystic ovary syndrome or PCOS) persist beyond the menopause transition. The study aim was to explore and contrast cardiometabolic health indicators in postmenopausal women with PMOS with postmenopausal women without PMOS. This study employed a cross-sectional analysis; comparisons between groups were completed with a two-tailed t-test. Methods: Postmenopausal females with a self-reported history of PMOS/PCOS (n = 1

A case study comparing VMAT treatment techniques to reduce dose to femoral heads for postmenopausal women during pelvic radiotherapy.

Volumetric modulated arc therapy (VMAT) is a common radiation treatment technique that delivers radiation to the planning target volume (PTV) while sparing the organs at risk (OAR). One limitation of VMAT on Varian linear accelerators is the limited distance the multileaf collimators (MLCs) can travel across the x-axis of the treatment field. The researchers in this case study sought to evaluate whether a split-field technique offers improved sparing of OAR, particularly dose to the femoral heads, in postmenopausal women undergoing treatment for the endometrium and pelvic lymph nodes. Serum-based biomarker development for dietary macronutrient densities and their association with breast and colorectal cancer risk in a cohort of postmenopausal females.

BACKGROUND: Associations of the macronutrient composition of the diet with risks of postmenopausal breast and colorectal cancer (CRC) are uncertain, partly because of reliance on self-reported dietary data. OBJECTIVES: This study aims to study biomarker development for several macronutrient component densities using serum and spot urine metabolomics and, when appropriate, to assess their associations with breast cancer and CRC risks in a Women's Health Initiative (WHI) cohort of postmenopausal United States females. METHODS: We explored linear biomarker equations for log-transformed macronut
