Thermoregulation in multiple sclerosis.

Multiple sclerosis (MS) is a progressive neurological disorder that disrupts axonal myelin in the central nervous system. Demyelination produces alterations in saltatory conduction, slowed conduction velocity, and a predisposition to conduction block. An estimated 60-80% of MS patients experience temporary worsening of clinical signs and neurological symptoms with heat exposure. Additionally, MS may produce impaired neural control of autonomic and endocrine functions. This review focuses on five main themes regarding the current understanding of thermoregulatory dysfunction in MS: 1) heat sens

Taste buds: development and evolution.

The gustatory system in vertebrates comprises peripheral receptors (taste buds), innervated by three cranial nerves (VII, IX, and X), and a series of central neural centers and pathways. All vertebrates, with the exception of hagfishes, have taste buds. These receptors vary morphologically in different vertebrates but usually consist of at least four types of cells (dark, light, basal, and stem cells). An out-group analysis indicates that taste buds were restricted to the oropharynx, primitively, and that external taste buds, distributed over the head and, in some cases, even the trunk, evolve

The maternal gut-fetal brain axis: role of maternal microbial metabolome in early neurodevelopment.

Fetal neurodevelopment is a critical phase modulated by the maternal microbiome. While certain specific signaling pathways have been identified, the complete signaling network and the spatial and temporal complexity remain largely unknown. Maternal metabolites are essential for fetal growth and certain metabolites are subject to dynamic adaptation during pregnancy. Alteration of maternal microbe richness and diversity, caused by malnutrition, psycho-social distress, and or infection, are cues known to impair fetal neural developmental circuits concomitant with increased risk of cognitive dysfu

Heart and Brain: Anatomic and Physiologic Relationship.

The heart and brain are an integrated, bidirectional axis, communicating through neural and hormonal pathways. This article examines the neuroanatomy, physiologic interactions, and clinical implications of this vital system. Anatomically, the cardiac autonomic nervous system features extrinsic sympathetic (norepinephrine) and parasympathetic (acetylcholine) innervation, fine-tuned by the local intrinsic cardiac nervous system. Cardiovascular diseases impair brain function, raising the risk of cognitive decline and stroke. Conversely, neurologic disorders can trigger cardiac complications via a

Evaluating the Genetic Overlap Between Congenital Heart Disease and Neuroblastoma Risk.

Children with congenital heart disease (CHD) have elevated neuroblastoma (NB) risk, potentially due to shared neural crest origins. We analyzed rare exonic de novo single-nucleotide variants in 702 CHD and 454 NB trios from the Neuroblastoma Epidemiology in North America Study, Gabriella Miller Kids First Program, and a published cohort. Seven genes, including CHD risk genes POGZ and LZTR1 (linked to Noonan syndrome and Schwannomatosis), showed nominal (p < 0.05) enrichment in both cohorts. CIC, IREB2, POGZ, and PCDHG cluster are associated with neurodevelopmental disorders, supporting shared

Dysregulation of mTOR Signaling after Brain Ischemia.

In this review, we provide recent data on the role of mTOR kinase in the brain under physiological conditions and after damage, with a particular focus on cerebral ischemia. We cover the upstream and downstream pathways that regulate the activation state of mTOR complexes. Furthermore, we summarize recent advances in our understanding of mTORC1 and mTORC2 status in ischemia-hypoxia at tissue and cellular levels and analyze the existing evidence related to two types of neural cells, namely glia and neurons. Finally, we discuss the potential use of mTORC1 and mTORC2 as therapeutic targets after

Cognitive dysfunctions in high myopia: An overview of potential neural morpho-functional mechanisms.

Dementia and cognitive impairment (CIM) carry high levels of mortality. Visual impairment (VI) is linked with CIM risk. High myopia (HM) is a chronic disease frequently leading to irreversible blindness. Current opinion has shifted from retinal injury as the cause of HM to the condition being considered an eye-brain disease. However, the pathogenesis of this disease and the manner in which neural structures are damaged are poorly understood. This review comprehensively discusses the relationship between HM, the central nervous system, and CIM, together with the novel concept of three visual pa

Drugs on the Horizon for Chronic Pruritus.

Chronic pruritus is a common condition that has a detrimental impact on quality of life. As the molecular pathogenesis of itch is elucidated, novel therapies that disrupt itch pathways are being investigated. Emerging treatments include drugs targeting the neural system, drugs targeting the immune system, antihistamines, bile acid transport inhibitors, and topical drugs that work through a variety of mechanisms such as phosphodiesterase-4 inhibition or targeting of nerve ion channels. Many of these therapies show promising results in the treatment of chronic itch of various etiologies, such as

Histology and molecular aspects of central neurocytoma.

Central neurocytoma (CN) is a well-differentiated tumor of neural cells occurring within the ventricles. It is composed of monomorphic cells with round, regular nuclei within clear cytoplasm and must be distinguished from other clear cell tumors. Immunohistochemical markers of CN that aid in diagnosis include synaptophysin and neuronal nuclear antigen. The molecular biology of these tumors is becoming increasingly elucidated, particularly with the use of microarray analyses. Several oncogenic pathways have been suggested by these studies. Although progress continues to be made, knowledge of CN

The Casey Holter lecture. Non-selective intervention in newborn babies with open spina bifida: the outcome 30 years on for the complete cohort.

The disability in neural tube defect is determined by the neurological deficit. The sensory level is a reliable measure of this deficit and predicts the disability. Achievement at the age of 30 related as well as to IQ and disability events in the history of CSF shunt. Episodes of raised intracranial pressure may damage the brain and visual pathways. They may also reduce achievement and enterprise. Blockage of the CSF shunt, when symptomatic, should be treated urgently, especially when it occurs after infancy. Blockage of the CSF shunt when asymptomatic was not usually associated with a low IQ

Signs of essential blepharospasm: a motion-picture analysis.

Motion pictures of 15 patients with essential blepharospasm were studied. Previously unrecognized signs indicated multiple cranial nerve involvement. These signs include impersistence of gaze, lid retraction, tongue thrust, head tilts, head jerks, vertical gaze spasms, and asymmetry. The sugns were also observed in a patient with bilateral blepharospasm who had a history of Bell's palsy suggesting facial nerve injury as a possible factor in blepharospasm. The presence of these signs can be explained by known neural pathways, but the site, or sites, of the lesion remains obscure. These signs ma…

Integrated signalling networks in the healthy lower urinary tract: A narrative review.

Lower urinary tract (LUT) function in health, is summarised in urine storage and voluntary voiding. They are mediated through finely coordinated structural signalling mechanisms. The histological structure of the bladder not only preserves a barrier function but also dynamically senses mechanical and chemical stimuli. Urothelial mechano-transduction comprises stretch-activated channels (ENaC, Piezo1/2, TRPV4) and neurotransmitters such as ATP, nitric oxide and acetylcholine to modulate adjacent afferent nerves. Cholinergic signalling through M3 predominantly muscarinic and nicotinic receptors

Elucidation of Relevant Neuroinflammation Mechanisms Using Gene Expression Profiling in Patients with Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder characterized by damage of motor neurons. Recent reports indicate that inflammatory responses occurring within the central nervous system contribute to the pathogenesis of ALS. We aimed to investigate disease-specific gene expression associated with neuroinflammation by conducting transcriptome analysis on fibroblasts from three patients with sporadic ALS and three normal controls. Several pathways were found to be upregulated in patients with ALS, among which the toll-like receptor (TLR) and NOD-like receptor (NLR

Nephrotic syndrome in a child with neurofibromatosis type 1: A case report and literature review.

Neurofibromatosis type 1 (NF1) is an autosomal dominant genetic disorder that caused by NF1 mutations. NF1 gene encodes neurofibromin (a GTPase-activating protein) and plays a regulatory role in many signalling pathway such as the Ras/MAPK pathway, which is important for regulating cell growth, proliferation and neural development. Therefore, NF1 gene mutations causes the excessive activation of signalling pathways and uncontrolled cell growth. NF1 exhibits complete genetic penetrance and clinical heterogeneity. Glomerular disease has rarely been reported in patients with NF1, especially in ch

[Pruritus associated with cholestasis].

Pruritus is commonly associated with cholestatic disorders and shows wide interindividual variability. The presence of skin lesions due to scratching and the application of a visual analogue scale are useful for clinical evaluation. Although the pathophysiology of this entity is not well understood, advances have recently been made in understanding of the pruritoceptive neural pathway, which shares certain similarities with the nociceptive pathway, although there are other distinguishing characteristics such as the action of a specific neurotransmitter, GPR, on the first synapsis at the poster

Melatonin Promotes Neurogenesis via the JAK2/STAT3 Pathway in Hypoxic-Ischemic Neonatal Rats.

Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological dysfunction and long-term disability in newborns. Lactate accumulation and metabolic disturbances after brain injury inhibit neurogenesis, while the restorative capacity of endogenous neural stem cells (NSCs) is essential for neural reconstruction. Melatonin (Mel) alleviates neonatal brain injury, but its effects on NSCs proliferation and migration remain unclear, and the visualization methods for dynamic monitoring of metabolic changes are inadequate. In this study, a neonatal rat model of HIBD was established, a

MicroRNA-489-3p inhibits neurite growth by regulating PI3K/AKT pathway in spinal cord injury.

Spinal cord injury (SCI) is caused by mechanical disruption of the spinal cord. This primary injury is followed by a devastating secondary SCI. It has been shown that various microRNAs (miRNAs) are involved in secondary SCI. The present study explored the role of miR-489-3p on secondary SCI, and its underlying mechanisms. First, we determined the expression of miR-489-3p in blood samples of SCI patients and healthy controls. Further experiments were performed on human neural cell lines, treated with bupivacaine to induce neuron damage. The cultured neural cells were transfected with miR-489-3p
