Integrative multi-omics analysis identifies stromal-immune crosstalk as a determinant of immunotherapy efficacy and establishes a prognostic signature in gastric cancer.

Immune checkpoint inhibitors like pembrolizumab exhibit variable efficacy in metastatic gastric cancer (GC). This study aimed to identify molecular drivers of pembrolizumab response, explore mechanisms of immune checkpoint inhibitors (ICIs) efficacy, and develop a prognostic signature. Transcriptomic analysis of pembrolizumab-treated GC (TIGER database) identified 165 response-associated differentially expressed genes (DEGs). Functional annotation and single-cell RNA sequencing (scRNA-seq) data from the Gene Expression Omnibus (GEO) revealed that responder-upregulated genes (R-DEGs) were enric

Advancing lung cancer therapy - leveraging cell biology to improve immunotherapy and anti-angiogenic strategies.

The standard of care for lung cancer typically involves a multidisciplinary approach including surgery, chemotherapy, radiotherapy, targeted therapy and immunotherapy, with treatment strategies tailored to the cancer subtype, disease stage and patient condition. Despite these efforts, the overall 5-year relative survival rate remains low, at ∼27%, with outcomes varying significantly by stage at diagnosis. These limited survival rates highlight the urgent need for more effective therapeutic strategies and innovative treatment approaches. A combination therapy involving immune checkpoint inhibit

Targeting purinergic signaling and MTAP-associated metabolism in cancer: A functional immunometabolic convergence framework.

Cancer progression is closely associated with metabolic reprogramming and immune evasion. Extracellular adenosine (eADO) metabolism regulates adenosine receptor activation within the tumor microenvironment (TME). Predominantly through the adenosine A2A receptor, and, in some contexts through the adenosine A2B receptor, eADO signaling suppresses antitumor immune responses and contributes to resistance to immunotherapy. Production of eADO is largely driven by ecto-5'-nucleotidase (CD73), which catalyzes the conversion of eADO monophosphate (AMP) into eADO. In parallel, methylthioadenosine phosph…

Microbiota as a modulator of drug response: targeting microbial-drug crosstalk in cancer therapy.

In recent years, accumulating evidence has established mechanistic connections between the microbiota and the efficacy of anticancer treatments, encompassing microbial communities residing in the gut, within tumors, and across other tissue sites. Across diverse malignancies, distinct compositional and functional profiles of these microbial ecosystems have been associated with differential responses to chemotherapy, immunotherapy, and targeted agents, as observed in both clinical settings and experimental systems. It is now understood that microbial communities exert influence on drug activity

Navigating the frontier of CAR-T cell resistance: From underlying mechanisms to innovative therapeutic solutions.

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable success in the treatment of relapsed or refractory hematological malignancies, particularly acute B-cell lymphoblastic leukemia (B-ALL), B-cell lymphoma (BCL), and multiple myeloma (MM). However, resistance in hematological malignancies and limited immune responses in solid tumors remain major challenges for CAR-T cell therapy. Tumor antigen loss or modulation, dysregulated apoptotic signaling, CAR-T cell-intrinsic dysfunction, and immunosuppressive components within the tumor microenvironment (TME) significantly compromise…

CellFuse Enables Multimodal Integration of Single-Cell and Spatial Proteomics Data for Systems-Level Analysis in Cancer.

UNLABELLED: Analysis of tumors using single-cell and spatial modalities is critical to advance our understanding of cancer. The growth of technologies that enable these studies provides an increasing number of single cell datasets. Integrating such data across studies will increase the impact of individual studies and speed cancer research. Most existing integration approaches are tailored to transcriptomic data and assume large sets of shared features, an assumption that fails for lower dimensional proteomic measurements. Here, we developed CellFuse, a deep learning-based integration framewor

Physiological, patho-physiological, and potential therapeutic roles for neutrophils in cancer & beyond.

Neutrophils are the most common type of white blood cell in the body, acting as crucial mediators of innate immunity. They initiate immune responses against invading microorganisms and threats like cancerous cells. Neutrophil infiltration is observed across inflammatory and autoimmune diseases and many cancers. Neutrophils orchestrate innate and adaptive immune responses to return the body to homeostasis, including after infections and preventing the development of tumours. While both protective and deleterious functions of neutrophils have been reported, this dual functionality reflects the h

Cancer-associated fibroblasts and tertiary lymphoid structure orchestrate the spatial architecture of tumor immunity.

Tumor immunity is shaped not only by the cellular composition of the tumor microenvironment (TME), but also by how stromal and immune elements are spatially organized within it. Among these regulators, cancer-associated fibroblasts (CAFs) and tertiary lymphoid structures (TLS) have emerged as two major determinants of the spatial niche of anti-tumor immunity. CAFs comprise heterogeneous and plastic stromal populations that can remodel the extracellular matrix, restrict lymphocyte infiltration, sustain chronic inflammation, and promote therapeutic resistance, while selected subsets may also sup…
