Rapid multiplex detection of 10 foodborne pathogens with an up-converting phosphor technology-based 10-channel lateral flow assay.

The rapid high-throughput detection of foodborne pathogens is essential in controlling food safety. In this study, a 10-channel up-converting phosphor technology-based lateral flow (TC-UPT-LF) assay was established for the rapid and simultaneous detection of 10 epidemic foodborne pathogens. Ten different single-target UPT-LF strips were developed and integrated into one TC-UPT-LF disc with optimization. Without enrichment the TC-UPT-LF assay had a detection sensitivity of 10(4) CFU mL(-1) or 10(5) CFU mL(-1) for each pathogen, and after sample enrichment it was 10 CFU/0.6 mg. The assay also sh

The mycotoxin deoxynivalenol facilitates allergic sensitization to whey in mice.

Intestinal epithelial stress or damage may contribute to allergic sensitization against certain food antigens. Hence, the present study investigated whether impairment of intestinal barrier integrity by the mycotoxin deoxynivalenol (DON) contributes to the development of whey-induced food allergy in a murine model. C3H/HeOuJ mice, orally exposed to DON plus whey once a week for 5 consecutive weeks, showed whey-specific IgG1 and IgE in serum and an acute allergic skin response upon intradermal whey challenge, although early initiating mechanisms of sensitization in the intestine appeared to be

Experimental and natural warming elevates mercury concentrations in estuarine fish.

Marine food webs are the most important link between the global contaminant, methylmercury (MeHg), and human exposure through consumption of seafood. Warming temperatures may increase human exposure to MeHg, a potent neurotoxin, by increasing MeHg production as well as bioaccumulation and trophic transfer through marine food webs. Studies of the effects of temperature on MeHg bioaccumulation are rare and no study has specifically related temperature to MeHg fate by linking laboratory experiments with natural field manipulations in coastal ecosystems. We performed laboratory and field experimen

Hospital-acquired listeriosis associated with sandwiches in the UK: a cause for concern.

BACKGROUND: Hospital-acquired outbreaks of listeriosis are not commonly reported but remain a significant public health problem. AIM: To raise awareness of listeriosis outbreaks that have occurred in hospitals and describe actions that can be taken to minimize the risk of foodborne listeriosis to vulnerable patients. METHODS: Foodborne outbreaks and incidents of Listeria monocytogenes reported to the Health Protection Agency national surveillance systems were investigated and those linked to hospitals were extracted. The data were analysed to identify the outbreak/incident setting, the food

Combining nutrition, food science and engineering in developing solutions to Inflammatory bowel diseases--omega-3 polyunsaturated fatty acids as an example.

The Inflammatory bowel diseases, Crohn's disease and ulcerative colitis, are debilitating conditions, characterised by lifelong sensitivity to certain foods, and often a need for surgery and life-long medication. The anti-inflammatory effects of long chain omega-3 polyunsaturated acids justify their inclusion in enteral nutrition formulas that have been associated with disease remission. However, there have been variable data in clinical trials to test supplementary omega-3 polyunsaturated fatty acids in inducing or maintaining remission in these diseases. Although variability in trial design

Occurrence of enterotoxigenic Staphylococcus aureus in food.

Gastroenteritis is one of the most frequent microbial diseases, which is caused by the ingestion of food contaminated with staphylococcal enterotoxins. In our study, the production of staphylococcal enterotoxins A, B (SEA, SEB) and the presence of respective staphylococcal enterotoxin genes were investigated in the field S. aureus isolates obtained from foods and food industry manufactures in East Slovakia. Radioimmunoassay (RIA), polymerase chain reaction (PCR) and dot-blot hybridisation were used for examination. The ability to synthesise enterotoxins was found in 20 (39.2%) of the total num

[Bovine spongiform encephalopathy and food safety].

The governments of Great Britain and France disagree on the safety of British beef with respect to bovine spongiform encephalopathy (BSE). Eventually the consumer might have the burden to decide whether beef from Britain is safe to eat with regard to the new variant of Creutzfeldt-Jakob disease (vCJD). Outside Britain the incidence of BSE increases. Probably in continental Europe, the use of high risk material, derived from non-British cattle in the subclinical stage of BSE, poses a greater threat than beef imported from Great Britain. The risk to contract vCJD depends on two unknown factors:

The ethnophysiology of digestion and diarrhoea in a Bangladeshi hospital population.

The results presented in this paper are drawn from a study of the acceptability of the weaning food, ARGC. The study aimed to investigate attitudes and practices surrounding weaning and the dietary management of diarrhoea. One hundred and twenty mothers of children aged between six months and 24 months suffering from mild diarrhoea and admitted to the ICDDR,B treatment centre were randomly selected. Diarrhoea was attributed by mothers to a number of causes; most common were contaminated food and breastmilk. Breastmilk was understood to have been spoiled either by the mother's diet or mystical

A single enzyme PCR-RFLP assay targeting V1-V3 region of 16S rRNA gene for direct identification of Alicyclobacillus acidoterrestris from other Alicyclobacillus species.

Alicyclobacillus acidoterrestris is considered to be one of the most important target microorganisms in quality control of heat-processed acidic foods. This species can cause spoilage by forming spores with very high heat resistance; however, no gas production or visible changes occur in the contaminated product during storage. Thus, the differentiation of A. acidoterrestris from other Alicyclobacillus species is of great importance. Τhe present study aims to find a rapid method for the identification of A. acidoterrestris. To achieve this, 78 Alicyclobacillus isolates were subjected to PCR-RF

Living in poisoning environments: Invisible risks and human adaptation.

This article describes the hidden natural chemical contaminants present in a unique desert environment and their health consequences on ancient populations. Currently, millions of people are affected worldwide by toxic elements such as arsenic. Using data gathered from Atacama Desert mummies, we discuss long-term exposure and biocultural adaptation to toxic elements. The rivers that bring life to the Atacama Desert are paradoxically laden with arsenic and other minerals that are invisible and tasteless. High intake of these toxic elements results in severe health and behavioral problems, and e

An update of the Worldwide Integrated Assessment (WIA) on systemic insecticides. Part 1: new molecules, metabolism, fate, and transport.

With the exponential number of published data on neonicotinoids and fipronil during the last decade, an updated review of literature has been conducted in three parts. The present part focuses on gaps of knowledge that have been addressed after publication of the Worldwide Integrated Assessment (WIA) on systemic insecticides in 2015. More specifically, new data on the mode of action and metabolism of neonicotinoids and fipronil, and their toxicity to invertebrates and vertebrates, were obtained. We included the newly detected synergistic effects and/or interactions of these systemic insecticid

[Imported tropical fish causes ciguatera fish poisoning in Germany].

Ciguatera is a seafood-borne illness caused by consumption of tropical fish contaminated with ciguatoxins, lipophilic polyethers that are produced in benthic dinoflagellates and accumulate through the marine food chain. Ciguatera cases in Europe usually occur in travellers returning from tropical and subtropical regions of the Pacific and Carribean, where ciguatera is endemic. In 2012, several cases of ciguatera occurred in Germany due to sale of contaminated fish products originating from the Indian Ocean. Although the symptomatology in these cases were typical of ciguatera, with patients rep

Congenital toxoplasmosis.

Approximately 85 percent of women of childbearing age in the United States are susceptible to acute infection with the protozoan parasite Toxoplasma gondii. Transmission of T. gondii to the fetus can result in serious health problems, including mental retardation, seizures, blindness, and death. Some health problems may not become apparent until the second or third decade of life. An estimated 400 to 4,000 cases of congenital toxoplasmosis occur in the United States each year. Serologic tests are used to diagnose acute T. gondii infection in pregnant women. Because false-positive tests occur f

A case report of sporadic ovine listerial menigoencephalitis in Iowa with an overview of livestock and human cases.

A case of ovine listeriosis was examined in a flock of sheep. The index case was a male lamb, which was part of a flock of 85 sheep located in central Iowa. Because the sheep were raised on a premise where soybean sprouts were also cultivated for the organic foods market, the potential of a public health concern was addressed. To identify the source of contaminations, clinical and environmental samples were cultured for Listeria monocytogenes. Isolates were serotyped and analyzed using pulsed-field gel electrophoresis (PFGE). Listeria monocytogenes (serotype 1) was recovered from the brain of

Houseflies (Musca domestica) as mechanical vectors of shigellosis.

Houseflies (Musca domestica) are mechanical vectors of Shigella organisms. Seasons during which both flies and cases of dysentery are prevalent often coincide. Houseflies have an affinity for human excrement and, as documented by bacteriologic culture of trapped flies, become contaminated with Shigella organisms following contact with infected human feces. The inoculum required to transmit shigellosis is only 10-10(2). Since houseflies cohabit with humans, they can readily contaminate food and eating utensils. With the advent of insecticides in the 1940s, intervention studies in the United Sta

Microwave disinfection of soft contact lenses.

We evaluated the effectiveness and convenience of microwave irradiation as a method of disinfecting soft contact lenses. Soft contact lenses from each of the four Food and Drug Administration (FDA) categories were placed in sterile vials and immersed in 2 ml of saline which had been contaminated with one of three common species of bacteria. The contaminated lens vials were placed in a standard 600 W microwave oven and exposed to microwave irradiation times ranging from 30 to 180 s. Significant reductions in bacteria colony counts were found after 30 s of microwave irradiation. Few of the bacte

A Cross-Sectional Study of Dairy Cattle Metagenomes Reveals Increased Antimicrobial Resistance in Animals Farmed in a Heavy Metal Contaminated Environment.

The use of heavy metals in economic and social development can create an accumulation of toxic waste in the environment. High concentrations of heavy metals can damage human and animal health, lead to the development of antibiotic resistance, and possibly change in bovine microbiota. It is important to investigate the influence of heavy metals in food systems to determine potential harmful effects environmental heavy metal contamination on human health. Because of a mining dam rupture, 43 million cubic meters of iron ore waste flowed into the Doce river basin surrounding Mariana City, Brazil,

A Common Adverse Outcome Pathway for Metal(loid)s Inducing Nephrotoxicity to Advance Next-Generation Risk Assessment of Chemical Mixtures.

Chronic exposure to the metal(loid)s arsenic, cadmium, lead, and mercury via contaminated food or drinking water may induce kidney toxicity, but there is little consensus on the biological processes involved. Health risk assessment of these substances is further complicated by coexposures and the sometimes unclear causal interpretation of population studies. To address these issues, we developed a common adverse outcome pathway (AOP) describing how these metal(loid)s can induce kidney toxicity. Upon identification of renal dysfunction resulting from proximal tubular damage as a common adverse

Effect of Genotype on Cadmium and Trace Element Accumulation in Wheat from Weakly Alkaline Cadmium-contaminated Soil.

Cadmium (Cd) contamination of farmland soils leads to Cd accumulation in crops and reduced micronutrient uptake, posing grave risks to food safety. Herein, we investigated the enrichment and transportation patterns of Cd and trace elements in different parts of six wheat genotypes grown in weakly alkaline Cd-contaminated soils via pot experiments. The results revealed that the wheat grain variety with high Cd accumulation (Ningmai13) demonstrated a 1.94-fold increase compared to the variety with low accumulation (Yanong0428). The transfer factor of Cd from wheat straw to grain ranged from 0.31
