5-fluorouracil-related gene expression levels in primary colorectal cancer and corresponding liver metastasis.

Gene expression levels of thymidylate synthase (TS), dihydropyrimidine dehydrogenase (DPD), thymidine phosphorylase (TP) and orotate phosphoribosyl transferase (OPRT) have been shown to be associated with response to 5-fluorouracil-based therapies. Analyzing these gene expression levels in liver metastases is important to obtain the best prediction of therapy. Our aim was to determine how TS, DPD, TP and OPRT gene expression levels in primary colorectal cancer (CRC) were related to those in liver metastases. Formalin-fixed, paraffin-embedded tumor specimens from 31 pairs of primary CRC and cor

DNA microarray screening of differential gene expression in bone marrow samples from AML, non-AML patients and AML cell lines.

This study used cDNA microarray technology to compare gene expression profiles in acute myeloblastic leukaemia (AML) with cDNA dot-blot and real time PCR analysis of cDNA transcripts to confirm array data. Patient AML marrow samples and AML cell lines were compared with normal/non-AML samples. Screening revealed five particular genes to be significantly differentially expressed across the sample groups. The migration-inhibitory factor-related-proteins 8 and 14 (MRP-8 and MRP-14) genes, the products of which inhibit cell migration and differentiation were the most highly expressed in non-malign

Expression of inducible nitric oxide synthase in cutaneous adnexal tumours of the head and neck.

Adnexal tumours are a rare heterogeneous group of neoplasms, most commonly found in the head and neck region. Although most are benign, malignant adnexal tumours often behave aggressively, resulting in early metastasis. There is increasing interest in the role that nitric oxide (NO) plays in the behaviour of many cancers. It is thought that NO, produced by the enzyme inducible NO synthase (iNOS), facilitates tumour growth and dissemination. iNOS has been studied in the common skin cancers, where its expression correlates with tumour behaviour, but it has not been previously investigated in cut

Regulation of fatty acid synthase expression by cholesterol in human cultured cells.

The regulation of fatty acid synthase (FAS) expression by sterols in a cultured human hepatoblastoma cell line, Hep G2, was studied. When cells were treated with compactin in a medium containing lipoprotein deficient serum, FAS mRNA level increased 1.6-fold. A squalene synthase inhibitor, TAN1607A, decreased both free and esterified cholesterol contents in Hep G2 cells and increased mRNA levels for FAS, HMG-CoA reductase, squalene synthase and LDL receptor. However, for the increment of FAS mRNA, a 10-fold higher concentration of this inhibitor was needed. These results demonstrate that the co

Regulation of ionic conductances and gene expression by hypoxia in an oxygen sensitive cell line.

We have shown that the PC12 cell line is an excellent model system for investigations of the molecular and cellular processes involved in O2-chemosensitivity. We have identified an O2-sensitive K channel in this cell line that mediates membrane depolarization, an increase in intracellular free Ca2+, and dopamine release during hypoxia. We also presented evidence which shows that expression of the gene for tyrosine hydroxylase, the rate-limiting enzyme in dopamine biosynthesis, is stimulated by reduced O2 tension in PC12 and type I carotid body cells. In addition, we have successfully identifie

Genome-wide analysis of long noncoding RNA (lncRNA) expression in hepatoblastoma tissues.

Long noncoding RNAs (lncRNAs) have crucial roles in cancer biology. We performed a genome-wide analysis of lncRNA expression in hepatoblastoma tissues to identify novel targets for further study of hepatoblastoma. Hepatoblastoma and normal liver tissue samples were obtained from hepatoblastoma patients. The genome-wide analysis of lncRNA expression in these tissues was performed using a 4×180 K lncRNA microarray and Sureprint G3 Human lncRNA Chips. Quantitative RT-PCR (qRT-PCR) was performed to confirm these results. The differential expressions of lncRNAs and mRNAs were identified through fol

Overexpression of HOXB7 and homeobox genes characterizes multiple myeloma patients lacking the major primary immunoglobulin heavy chain locus translocations.

Homeobox (HOX) gene transcription factors are frequently deregulated in hematologic malignancies and involved in leukemogenic transformation [1]. Moreover, their overexpression has been associated with tumoral-induced neoangiogenesis in solid cancer [2]. The expression and the role of these genes have not yet been completely elucidated in multiple myeloma (MM). Recently, we reported that a small fraction of MM patients shows a HOXB7 overexpression as compared with normal samples and that HOXB7 expression correlates with bone marrow angiogenesis and the production of the proangiogenic factors b

Overexpression of human aspartyl(asparaginyl)beta-hydroxylase in hepatocellular carcinoma and cholangiocarcinoma.

To characterize genes that become upregulated with malignant transformation of human hepatocytes, a library of monoclonal antibodies was produced against the FOCUS hepatocellular carcinoma cell line. Antibody FB-50 reacted with an antigen that was highly expressed in 4 of 10 primary hepatocellular carcinomas, in all 20 cholangiocarcinomas we studied, and in a variety of transformed cell lines. This antigen was also highly expressed in neoplastic epithelial cells of breast and colon carcinomas in contrast to its low level of expression in normal hepatocytes and in non-neoplastic epithelial cell

Loss of Numb promotes hepatic progenitor expansion and intrahepatic cholangiocarcinoma by enhancing Notch signaling.

Numb, a stem cell fate determinant, acts as a tumor suppressor and is closely related to a wide variety of malignancies. Intrahepatic cholangiocarcinoma (iCCA) originates from hepatic progenitors (HPCs); however, the role of Numb in HPC malignant transformation and iCCA development is still unclear. A retrospective cohort study indicated that Numb was frequently decreased in tumor tissues and suggests poor prognosis in iCCA patients. Consistently, in a chemically induced iCCA mouse model, Numb was downregulated in tumor cells compared to normal cholangiocytes. In diet-induced chronic liver inj

A multi-omics convergence on CA2 as a prognostic hub and potential molecular target of benzo[a]pyrene diol epoxide in lung cancer.

BACKGROUND: Benzo[a]pyrene diol epoxide (BPDE), a key carcinogenic metabolite of the ubiquitous environmental pollutant benzo[a]pyrene, is a well-established etiological agent in lung cancer. However, a systems-level understanding of its molecular targets and pathogenic mechanisms remains incomplete. This study aimed to identify and characterize the core toxicity targets linking BPDE exposure to lung carcinogenesis using an integrative network toxicology approach. METHODS: Putative BPDE targets were retrieved from SwissTargetPrediction and TargetNet. Lung cancer-associated genes were compiled

Mechanistic insights into TXNDC17-mediated ferroptosis suppression in glioblastoma progression.

BACKGROUND: Glioblastoma (GBM) is a lethal brain tumor with limited treatment options. Molecular targeted therapy is considered safe and promising for GBM. Ferroptosis is regulated by lipid, iron and cysteine metabolism. Induction of ferroptosis is a promising strategy. METHODS: Integrated bioinformatics analysis of GBM datasets (TCGA) and RNA-seq data from human gliomas was used to identify candidate genes. TXNDC17 expression was validated in glioma tissues and cell lines by qPCR, western blotting and immunofluorescence. In vitro functional assays (CCK-8, wound healing, Transwell) were perfo

Macrophage- or neutrophil-expressed UPP1 drives tumor growth and immune suppression in the tumor microenvironment.

Uridine phosphorylase 1 (UPP1) has recently emerged as a metabolic regulator with critical roles in tumor cell biology. However, its expression pattern and functional significance in nonmalignant components of the tumor microenvironment (TME) remain largely unexplored. Here, we performed a comprehensive pan-cancer single-cell transcriptomic analysis revealing that UPP1 is highly expressed not only in tumor cells but also in tumor-associated macrophages (TAMs) and neutrophils (TANs). Clinically, UPP1 expression in TAMs and TANs correlates with poor prognosis and diminished response to immunothe

A Multistep Immune-Competent Genetically Engineered Mouse Model Reveals Phenotypic Plasticity in Uveal Melanoma.

UNLABELLED: Uveal melanoma is a highly aggressive intraocular malignancy with limited therapeutic options for metastatic disease. Existing transgenic uveal melanoma mouse models inadequately recapitulate human disease progression, whereas transplant models lack immune competence for studying the tumor immune microenvironment and therapeutic interventions. To address these limitations, we developed a genetically engineered mouse model incorporating stepwise genetic alterations implicated in human uveal melanoma progression. Spatiotemporally controlled expression of mutant GNAQQ209L from the end

Exosome-derived hsa-miR-21-5p stimulates malignant progression of colorectal cancer by regulating HRK.

Exosome-mediated microRNAs (miRNA) delivery is increasingly recognized as a central mechanism in tumor microenvironment (TME) remodeling. While the pro-tumorigenic role of tumor cell-derived exosomal miRNAs in colorectal cancer (CRC) is established, their specific regulatory mechanisms require further elucidation. In this study, we demonstrate that exosomes originating from CRC cells not only markedly enhance the proliferation, migration, and invasion capacities of recipient CRC cells but also lead to the suppression of apoptosis in the recipient cells. By means of integrated public database a

Mechanism of action of MiR-330-5p targeting ITGA5 in the regulation of proliferation, migration, and invasionof gastric cancer.

BACKGROUND: ITGA5 is an oncogene that performs its biological function by integrating the intracellular structure and extracellular matrix. We found that ITGA5 is highly expressed in gastric tumors and is closely related to proliferation and metastasis. Multiple miRNAs regulate the ITGA5 gene during the occurrence and development of tumors. This study aimed to explore the role of targeting miRNAs upstream of ITGA5 in the regulation of the proliferation, invasion, and migration of gastric cancer cells. METHODS: Target miRNA molecules regulating the ITGA5 gene were predicted using four bioinfor
