Human dental pulp stem cell response to a novel photoinitiator for resin-based dental materials.

Predicting the likelihood parents will become physically abusive towards their children remains a high priority. Early parenting stress may increase the extent to which parents negatively appraise their children's behaviours, which in turn may influence their abuse risk. This study explored whether parental negative attributions mediated the association between early parenting stress and later abuse risk. A sample of 186 first-time mothers and 146 of their male partners reported on their parenting stress when infants were 6 months old and their negative attributions regarding child behaviours

Sibling relationships as a mediator and a moderator between parental attachment and depression among adolescents.

Depression threatens adolescents' psychological health and is influenced by negative interpersonal relationships. Previous studies have explored the effect of a negative parent-child relationship on adolescent depression but have ignored the frequent coexistence of these relationships with other family relationships, for example, sibling relationships. This study examined how parent-child and sibling relationships co-impact adolescent depression, by testing the mediating and moderating roles of sibling relationships between parental attachment and adolescent depression. A sample of 5,896 adole

Intersections Among Attachment, Empathy and Emotion Regulation in Middle Childhood.

Within secure attachment relationships, caregivers function as primary sources of self-regulation and foster empathy. Affective empathy refers to the direct sharing of others' emotional states, whereas cognitive empathy involves understanding the meaning of others' affective displays. Although robust evidence links secure attachment to empathy, research examining the mediating role of emotion regulation (ER) during middle childhood remains limited. Participants were 214 children (108 girls) aged 8-12 years (M = 9.28 ± 1.09) and their parents (86.3% mothers). Children completed the Security Sca

Maternal and perinatal risk factors for necrotizing enterocolitis.

Necrotizing enterocolitis (NEC) remains among the most devastating complications of prematurity, carrying significant mortality and lifelong morbidity despite decades of mechanistic and translational research. Although NEC is classically framed as an inflammatory disease of intestinal immaturity, driven by microbial dysbiosis and exaggerated immune signaling, this model does not fully explain the uneven distribution of disease burden across infants, hospitals and communities. In this review, we reframe NEC within a broader maternal-perinatal systems context grounded in the Developmental Origin

Long-read sequencing reveals pre-meiotic gene conversion in sperm.

Meiotic recombination is a fundamental process that generates genetic diversity by creating new combinations of existing alleles1. Whereas crossovers in humans are well characterized2, the more frequent non-crossovers that lead to gene conversion remain challenging to study. Here we show that single high-fidelity long sequencing reads from sperm can capture both crossovers and non-crossovers, which enables effectively arbitrary sample sizes for analysis from a single male. We analysed 2,382 candidate non-crossovers in 15 sperm samples from 13 donors, and identified a consistent component with

Long noncoding RNA H19 in liver development and disease.

Liver disease is a global health problem responsible for more than two million deaths annually. Metabolic dysfunction-associated steatotic liver disease (MASLD) and alcohol-associated liver disease (ALD) are major contributors to chronic liver disease-related morbidity and mortality. Factors like diet and alcohol consumption have become key drivers of liver pathologies including steatosis, fibrosis/cirrhosis, and hepatocellular carcinoma. To date very few treatments are available, hence there is a critical need for the development of novel therapies to slow down the development/progression of

Intergenerational consequences of valproic acid exposure: Dose- and sex-specific effects in Drosophilamelanogaster.

Valproic acid (VPA) is a widely used anticonvulsant and mood stabilizer that is notable for its role as a histone deacetylase (HDAC) inhibitor and its potential impact on epigenetic modulation. However, its intergenerational impacts are not fully understood. In this study, we investigated the dose- and sex-dependent intergenerational effects of VPA using Drosophila melanogaster as a model organism. Parental (F0) flies - both male and female - were exposed to varying VPA concentrations, while their offspring (F1) were raised on standard food. We assessed several phenotypic outcomes such as life

First-Trimester Molecular Screening for Fetal and Neonatal Alloimmune Thrombocytopenia: Early Identification of At-Risk Pregnancies.

OBJECTIVE: The aim of this study was to evaluate the feasibility and clinical value of implementing first-trimester molecular HPA screening to identify pregnancies at risk for FNAIT. METHODS: In this prospective cohort study, pregnant women were genotyped during the first trimester for HPA-1, -2, -3, -4, -5, -6, -9, and -15 using molecular methods. When low-frequency maternal genotypes were identified, paternal genotyping and risk stratification were performed to assess maternal-fetal incompatibility. At-risk pregnancies underwent structured clinical monitoring, and neonatal platelet counts w

Reconsidering the Selection Strategy in a Flemish Honey Bee Breeding Program: Towards Selection by Exclusion.

Selective breeding is widely used to improve honey bee health and colony performance, commonly applying selection based on colony traits. Since 2017, a mass selection program in Flanders, Belgium, has ranked queens according to resilience, productivity and behavior traits. Using longitudinal data (2017-2024), we evaluated the effectiveness of this strategy by assessing colony performance across years and generations. Behavioral traits showed the clearest improvement over time, whereas progress in productivity and especially resilience-related traits remained limited. Offspring of top-ranking q

Autophagy as an architect of embryogenesis: Understanding the new mechanisms, emerging paradigms, and future directions.

Autophagy is no longer considered just a generic degradation pathway but is rather acknowledged as a programmed developmental mechanism whose roles in coordination across embryogenesis are still not very well understood. Various mammalian and non-mammalian models present evidence, where autophagy is switched on at fertilization, maintains proteostasis during cleavage, allocates lineage during blastocyst formation, and enforces genomic as well as cellular quality control by the selective removal of abnormal cells or paternal organelles. Moreover, following gastrulation, which gives rise to the

Case Report: Novel compound heterozygous mutations in SLC34A2 gene: a case of pulmonary alveolar microlithiasis in a child.

Pulmonary alveolar microlithiasis (PAM), a rare disorder caused by SLC34A2 mutations, demonstrates clinical-radiological discordance. Pediatric cases face heightened diagnostic challenges; nonspecific presentations often result in misattribution to recurrent pneumonia or delayed diagnosis, especially in children ≤5 years. We present a case of a 3-year-old girl hospitalized for persistent cough (11-day duration) and intermittent fever (9-day history). Her recurrent productive cough had been misattributed to recurrent pneumonia in prior healthcare encounters. Diagnostic imaging revealed extensiv

Ataxia With Vitamin E Deficiency Syndrome and a Novel TTPA Variant: A Paired Case Report.

OBJECTIVES: Ataxia with vitamin E deficiency (AVED) is a chronic progressive syndrome with low vitamin E levels, caused by biallelic pathogenic variants in the alpha-tocopherol transfer protein (TTPA) gene on chromosome 8q13.1. According to family segregation studies, we describe a novel putative variant in compound heterozygosity with a known pathogenic variant. METHODS: Clinical and instrumental evaluations were performed at our Neuromuscular Unit. RESULTS: Two siblings had been experiencing clumsiness, which later evolved into progressive gait unsteadiness, with falls and dysarthria. Brai

Environmental influences on seminal plasma: Molecular and functional insights.

Seminal plasma is a pivotal regulator of reproductive success that contributes to fertility and fecundity beyond its traditionally recognized function as a vehicle for spermatozoa. Rich in soluble and extracellular vesicle-encased signaling molecules, seminal plasma influences sperm integrity and function, whilst simultaneously driving profound physiological changes in the female reproductive tract. These functions are broadly conserved across vertebrate and invertebrate species and help to optimize fertilization and create an immunological environment that supports implantation and fetal deve

Trained Memory of Uterine Macrophages Improves Subsequent Pregnancy Outcomes.

A previously successful pregnancy promotes the fitness of subsequent pregnancies, of which the pregnancy-induced memory is ascribed exclusively to adaptive lymphocytes. However, whether macrophages at maternal-fetal interface acquire pregnancy-specific immune memory remains unclear. Using human decidual samples and complementary mouse models, we identify human leukocyte immunoglobulin-like receptor subfamily B3+ (LILRB3+) and murine paired immunoglobulin-like receptor B+ (PIR-B+) macrophages as a uterine memory subset, which expands progressively with gravidity and gestational age, and exhibit

DLK1 is a GATA1s-driven dependency and therapeutic target in Down syndrome-associated myeloid leukemia.

Children with Down syndrome have a markedly increased risk of developing myeloid leukemia. Although having an excellent prognosis, 10% to 20% develop relapsed or refractory disease with poor survival, highlighting the need for new targeted approaches. The pathogenesis of myeloid leukemia of Down syndrome (ML-DS) is tightly linked to fetal hematopoiesis and mutations in GATA1, generating the truncated GATA1 short (GATA1s) isoform. We identified Delta-like noncanonical Notch ligand 1 (DLK1) as a direct GATA1s target. DLK1, a paternally imprinted transmembrane protein, is highly expressed in feta

Inflammation and Oxidative Stress in Male Infertility: A Narrative Review.

BACKGROUND: Male infertility affects 8%-15% of couples, with male factors implicated in about half of cases; many remain idiopathic despite improved diagnostics. This narrative review addresses this gap by examining chronic inflammation as an underappreciated driver of sperm dysfunction that forms a bidirectional, self-reinforcing cycle with oxidative stress (OS). METHODS: We synthesized studies from 2015 to 2025 identified via PubMed, Scopus, and Web of Science, examining links between inflammation and OS, key immune pathways, and related risk factors through qualitative narrative synthesis.

De novo GNAS-Gsα variant (p.Thr55Ala) with constitutive gain-of-function effects on AVPR2 and PTH1R signalings.

Recent studies have revealed de novo or germline-derived GNAS-Gsα variants with constitutive ligand-independent gain-of-function (GOF) effects on specific G-protein-coupled receptor signalings in patients with nephrogenic syndrome of inappropriate antidiuresis (NSIAD), osteolytic bone disorder with metaphyseal dysplasia, and peripheral precocious puberty. We encountered a Japanese girl with NSIAD and osteolytic bone disorder with metaphyseal dysplasia. Whole genome sequencing identified a de novo ″likely pathogenic″ heterozygous GNAS-Gsα missense variant (NM_000516.7:c.163 A > G:p.(Thr55Ala))

E3 Ubiquitin Ligases in Neurodevelopmental Disorders.

Neurodevelopmental disorders (NDDs) comprise a heterogeneous group of conditions that may present with developmental delay, intellectual disability, language impairment, epilepsy, autism-like features, motor abnormalities, or syndromic manifestations. Ubiquitination is an essential post-translational mechanism that regulates protein stability, localization, trafficking, and signaling. Because E3 ubiquitin ligases determine substrate specificity in this process, abnormalities in E3 ligases or E3-associated complexes can selectively affect proteins required for brain development and neuronal fun

Targeted Epigenetic Reactivation strategies as new treatments for Prader-Willi Syndrome: Achievements and Challenges.

Prader-Willi Syndrome (PWS) is a genetic neurodevelopmental disorder caused by the loss of expression of paternal genes in the 15q11-q13 locus. The same genes on the maternal allele are present but are epigenetically silenced. This genetic alteration causes a spectrum of symptoms associated with hypothalamic dysfunction. Research is increasingly focused on setting strategies aiming to reactivate the maternal locus and restore the expression of that critical gene cluster. Early therapeutic efforts explored broad-spectrum epigenetic drugs, such as DNA- and histone-methyltransferase inhibitors. W
