Evaluation of ethylglucuronide and cocaethylene as hair markers of alcohol consumption.

BACKGROUND AND AIMS: The concurrent use of alcohol and cocaine is known to be a harmful condition in terms of behaviour and organ damage. The identification of this condition requires the use of objective biomarkers, such as hair ethylglucuronide (hEtG) and hair cocaethylene (hCE). The aim of the present work was to investigate their presence and their possible mutual relationship in a group of cocaine users to evaluate the different potentials offered by the two analytes in the retrospective investigation of alcohol intake. MATERIALS AND METHODS: 209 hair specimens, previously collected from

Defects of Cortical Microcircuits Following Early Exposure to Alcohol.

The interplay between excitatory pyramidal neurons and GABAergic interneurons is the basic building block of neocortical microcircuits and plays a critical role in carrying out higher cognitive functions. Cortical circuits are deeply and permanently disrupted by exposure to alcohol during brain development, the main non-genetic cause of intellectual disability. Here, I review experimental studies of fetal alcohol spectrum disorders, dealing with permanent cellular and molecular alterations of neocortical neurons and their connections.

The role of DNA methylation in alcohol-mediated neurodevelopmental toxicity.

Alcohol induces neurodevelopmental toxicity through multiple biological processes, including DNA methylation and histone modifications in epigenetic regulation. Epigenetic mechanisms involving DNA chemical modifications represent crucial molecular pathways that regulate gene expression during neurodevelopment, exhibiting high sensitivity to adverse lifestyle factors such as alcohol consumption, smoking, and stress. Prenatal alcohol consumption is a primary cause of fetal neurodevelopmental disorders. Alcohol alters DNA methylation and histone modification levels in the brain, which in turn dis

Smoking, alcohol and risk of sarcopenia: a Mendelian randomisation study.

OBJECTIVE: Observational studies have found that cigarette smoking increased the prevalence and incidence of sarcopenia, whereas alcohol consumption appeared to decrease the risk. These findings, however, may be susceptible to either confounding bias or reverse causation. We conducted a Mendelian randomisation (MR) study to appraise the causal relation of cigarette smoking and alcohol consumption to the risk of sarcopenia. METHODS: Genetic instruments associated with cigarette smoking (cigarettes per day) and alcohol consumption (drinks per week) were retrieved from the publicly available gen

Current Therapeutic Targets for Alcohol-Associated Liver Disease.

Alcohol-associated liver disease (ALD) is a progressive liver disorder induced by chronic excessive alcohol consumption, affecting approximately 3.5% of the global population. The clinical spectrum of ALD encompasses simple steatosis, alcoholic hepatitis, fibrosis, cirrhosis, and the potential development of hepatocellular carcinoma. The pathogenesis of ALD involves a complex interplay of factors, including direct cell damage caused by alcohol and its metabolites, hepatic inflammation, immune dysregulation, and oxidative stress. Additionally, alcohol-induced dysbiosis and the subsequent imbala

Biosynthesis of fatty aldehydes and alcohols in the eye and their role in meibogenesis.

Fatty alcohols (FAlc) and aldehydes (FAld) are essential intermediates/precursors in the biosynthesis of lipids. However, elevated FAld levels were shown to be geno- and cytotoxic, thus requiring conversion into less toxic FAlc and fatty acids (FA). An increase in FAlc and FAld in tissues of patients with Sjögren-Larsson syndrome was reported before and repeatedly linked to inactivation of ALDH3A2, which oxidizes FAld in FA. Recently, we hypothesized that another group of enzymes, namely SDR16C5/SDR16C6 (EC 1.1.1.105), could control the balance between FA, FAlc, and FAld via a separate mechani

Non-alcoholic steatohepatitis incidence in patients with psoriasis vulgaris.

BACKGROUND AND AIMS: Non-alcoholic steatohepatitis (NASH) is a severe form of non-alcoholic fatty liver disease (NAFLD), characterized by hepatic inflammation and damage due to fat accumulation. Psoriasis patients show higher NASH incidence due to overlapping risk factors like obesity and insulin resistance. The study aimed to determine the incidence of non-alcoholic steatohepatitis in patients with psoriasis vulgaris. METHODS: This cross-sectional observational study included 80 adult patients diagnosed with psoriasis vulgaris. Psoriasis severity was assessed using the Psoriasis Area and Sev

Polyphenolic Activation of Basophils Explains Alcohol Hypersensitivity in AERD.

OBJECTIVE: Alcohol hypersensitivity (AH), an exacerbation of respiratory symptoms in response to alcohol consumption, is common in aspirin-exacerbated respiratory disease (AERD) and other forms of chronic rhinosinusitis (CRS). The mechanism of this is unknown. This study investigates the ability of polyphenolic compounds in alcoholic beverages to activate innate immune cells as a means of explaining AH in AERD. METHODS: Data were collected from 478 consecutive adults presenting to a tertiary care sinonasal clinic in whom the presence of AH and their CRS phenotype was determined. A subset of t

Non-alcoholic fatty liver disease: pathogenesis and models.

Non-alcoholic fatty liver disease (NAFLD) is a complex disease characterized by a massive accumulation of lipids in the liver, with a continuous progression of simple steatosis, non-alcoholic steatohepatitis (NASH), cirrhosis, and hepatocellular carcinoma. Non-alcoholic fatty liver disease is associated with obesity, insulin resistance, and metabolic syndrome; it is a severe public health risk and is currently the most common liver disease of the world. In addition to the fatty infiltration of the liver in non-alcoholic fatty liver disease patients, the field of liver transplantation faces sim

Interleukins: Pathogenesis in Non-Alcoholic Fatty Liver Disease.

Inflammatory cytokines have been implicated as crucial contributors to the onset and progression of non-alcoholic fatty liver disease (NAFLD). The exact mechanisms by which interleukins (ILs) contribute to NAFLD may vary, and ongoing research is aimed at understanding the specific roles of different ILs in the pathogenesis of this condition. In addition, variations in environmental factors and genetics in each individual can influence the onset and/or progression of NAFLD. The lack of clinical studies related to the potential therapeutic properties of IL-1 inhibitors currently does not allow u

Electrocatalytic Aromatic Alcohols Splitting to Aldehydes and H2 Gas.

Selective electrocatalytic transformation of alcohols to aldehydes offers an efficient and environmentally friendly platform for the simultaneous production of fine chemicals and pure hydrogen gas. However, traditional alcohol oxidation reactions (AORs) in aqueous electrolyte unavoidably face competitive reactions (e.g., water oxidation and overoxidations reactions) for the presence of active oxygen species from water oxidation, causing an unwanted decrease in final efficiency and selectivity. Here, we developed an integrated all-solid proton generator-transfer electrolyzer to trigger the pure

[Hodgkin lymphoma - pain at alcohol consumption as the only symptom].

The authors Lanke and Relander describe a patient with classical Hodgkin lymphoma (cHL) stage IIA, who had pain at alcohol consumption as the only symptom at diagnosis. The patient was treated with 4 cycles of ABVD chemotherapy and proton therapy 29.75 Gy (RBE). Apart from FDG-PET/CT the course of the disease was followed with serum-TARC. The case illustrates the value of knowing also rare symptoms at the general practice, the usefulness of TARC as a tumour marker in cHL, and the use of proton therapy in order to further reduce late radiotherapy side effects.

Dynamics of cellular plasticity in non-alcoholic steatohepatitis (NASH).

Non-alcoholic steatohepatitis (NASH) is a pathogenic stage of the broader non-alcoholic fatty liver disease (NAFLD). Histological presentation of NASH includes hepatocyte ballooning, macrophage polarization, ductular reaction, and hepatic stellate cell (HSCs) activation. At a cellular level, a heterogenous population of cells such as hepatocytes, macrophages, cholangiocytes, and HSCs undergo dramatic intra-cellular changes in response to extracellular triggers, which are termed "cellular plasticity. This dynamic switch in the cellular structure and function of hepatic parenchymal and non-paren
