CD8+ T cells directly target and kill cancer cells and are required for effective antitumor immunity. However, CD8+ T cells may become dysfunctional when exposed to chronic antigen stimulation during cancer. Dysfunction is characterized by reduced proliferation, increased expression of inhibitory receptors, and impaired effector function, yet the genetic mechanisms controlling dysfunction remain poorly understood. Therefore, we developed a murine ex vivo chronic stimulation model that recapitulates features of CD8+ T cell dysfunction observed within the tumor microenvironment. By coupling this

Determining how chemotherapeutic drugs alter the MHC class I-associated immunopeptidome is essential for designing rational combinatorial therapies to enhance antitumor CD8+ T-cell activity, such as immune checkpoint inhibitor Abs. We investigated the impact of a first-in-class inhibitor of cellular NEDDylation, MLN4924, on self-peptide presentation using 2 human tumor cell lines: HeLa cells secreting the human MHC class I protein HLA-A*11:01 and the glioblastoma cell line U87-MG. MLN4924 indirectly inactivates the cullin-RING ligases (CRLs), a large family of E3 ubiquitin ligases responsible

Immune checkpoint inhibitor (ICPI) therapy, targeting Programmed cell death protein 1 (PD-1) and its ligands PD-L1 and PD-L2, have revolutionized cancer care. However, ICPI is linked to immune-related adverse events, including nephropathy manifesting as acute kidney injury (AKI), a rapid decline in kidney function, often progressing to renal failure. Regulatory T cells (Tregs) could mitigate AKI, caused by ischemia-reperfusion injury (IRI). Treg stability and function rely on cellular metabolism, with a distinct preference for oxidative phosphorylation (OXPHOS) over glycolysis. Although PD-1 i

Regulatory T cell (Treg)-biased IL-2/anti-IL-2 monoclonal antibody complexes (IL-2c) can preferentially deliver IL-2 to CD25+ Tregs, causing proliferation of Tregs that is potentially advantageous in transplantation. We tested the ability of IL-2c to prolong murine cardiac allograft survival. C57BL/6 (H-2b) mice received fully major histocompatibility complex-mismatched BALB/c (H-2d) or hemi-allogeneic B6 × BALB/c (H-2b/d) F1 allografts. Recipients were treated prior to transplantation with IL-2c or control. Graft survival, anti-donor T cell priming, and donor-specific antibody production were

Chronic graft-versus-host disease (cGVHD) develops with complex interactions between immune cells and cytokines, leading to irreversible fibrosis. Severe cGVHD impairs quality of life and is associated with nonrelapse mortality; however, effective treatments are limited. Recombinant human soluble thrombomodulin (rTM), a novel anticoagulant consisting of the extracellular domains of thrombomodulin, has shown efficacy in patients with transplantation-associated coagulation disorders and, recently, anti-inflammatory properties in animal studies. Here, we investigated the effects of rTM on cGVHD u

Phosphoinositide 3-kinase (PI3K) mediates signaling downstream of many receptors expressed by B cells. Studies in both mice and humans have shown that PI3K plays a critical role in B-cell development, activation, and tolerance. Indeed the key role of PI3K in regulating activation versus tolerance is demonstrated by patients with genetic variants that cause increased PI3K signaling, which results in an immune dysregulatory disorder characterized by a range of manifestations including lymphoproliferation and autoantibody production. Interestingly, PI3K signaling also plays an essential role in m

Tumor-infiltrating lymphocyte (TIL) therapy is a promising approach, earning U.S. Food and Drug Administration approval in patients with anti-PD-1-resistant melanoma. Extending TIL therapy to patients with cholangiocarcinoma (CCA), an aggressive and largely immune-refractory cancer, is an emerging area of interest. However, cost and manufacturing complexity constrain clinical scalability of TIL therapy for CCA, underscoring the need for a murine model to optimize efficacy. Here, we established a novel orthotopic model of TIL therapy for CCA and tested a new ex vivo expansion strategy. We first

The retina is a central nervous system tissue with immune-privileged status, protected by the blood-retina barrier and maintained by specialized resident tissue macrophages: microglia, hyalocytes, and perivascular macrophages. These cells exhibit specific ontogeny, spatial localization, and immunologic functions. Each population contributes to homeostasis, phagocytosis, and immunoregulation in their local microenvironment. Microglia are the most abundant immune cells in the retina, and their key functions include vascular and synaptic regulation. Hyalocytes are abundant in the vitreous and at

The cause of inflammatory bowel disease (IBD) is not known, but both innate and adaptive immunity contribute to IBD pathogenesis. To better understand the contributions of innate immunity to IBD, we have generated a mouse model of IBD that occurs spontaneously in the absence of adaptive immunity. This model is 100% penetrant, early onset, and driven by microbes. Here, we report that the colitis in this model exhibits extensive cell death in the gut mucosa, including cell death with features of apoptosis, necroptosis, and pyroptosis. This innate colitis and mucosal cell death was dependent on t

Kawasaki disease (KD) is the most common childhood vasculitis. Approximately 25% of KD patients are refractory to standard intravenous immunoglobulin (IVIG) therapy and frequently develop coronary artery lesions (CAL) that result in long-term complications. Transcriptome studies utilizing blood cells from KD patients and reported animal model studies had identified interleukin (IL)-1β as a crucial component of an essential immune pathway in the formation of CAL. We previously reported that high-dose immunoglobulin G (IgG) treatment completely inhibited tumor necrosis factor (TNF)-α-stimulated

The polarization of naive CD4+ T cells into Th2 cells is initiated in lymphoid organs and completed as the cells become tissue resident, where they express ST2, the receptor for the alarmin interleukin (IL)-33, which may be a key signal for tissue integration. Cellular metabolic requirements associated with this transition remain poorly understood. To address this, we compared the response of lymphoid tissue (LT) Th2 cells from helminth parasite-infected mice to stimulation by IL-33 versus through the T cell receptor via anti-CD3/CD28. We found that IL-33, but not anti-CD3/CD28, induced the de

Peritoneal cavity (PerC) B cells can be classified into distinct subpopulations; however, their differential antigen-presenting capabilities and roles in antitumor immune responses remain largely unexplored. This study aimed to elucidate the properties of PerC B cell subpopulations in antitumor immune responses by using ovalbumin (OVA) peptides as neoantigen mimics and employing OT-I mice, which express a transgenic T cell receptor specific to OVA peptides. We found that OVA-pulsed PerC B cells effectively stimulated CD8+ T cell proliferation, although their antigen-presenting capacity was low

The importance of NAD metabolism in T cell differentiation and function has gained attention in recent years. However, technical limitations impede the specific interrogation of NAD dynamics in living immune cells. In this report, we present the redox index and capacity analysis (RICA) assay, a novel technique for measuring mitochondrial NAD content and redox balance. The RICA assay is a flow cytometry-based technique that uses NADH autofluorescence and mitochondrial inhibitors to assess NAD within specific phenotypic subsets of immune cells. We validated this technique using metabolic modulat

Transformed cells frequently adapt to immune pressure by modulating expression of ligands for PD-1 and CTLA-4. Despite the success of therapies targeting these interactions, only a minority of patients experience a clinical response, highlighting the critical need for alternative targets. Prior work from our group showed that the inhibitory receptor KLRG1 binds to tumor-derived E-cadherin. However, the therapeutic potential of targeting KLRG1 signaling remains incompletely understood. To address this, we generated a library of cancer cell lines engineered to express varying levels of E-cadheri

Interleukin 22 (IL-22) produced by CD4+ T cells plays an important role in regulating intestinal immune responses during inflammation and infection, but the mechanisms controlling IL-22 expression in T cells remain incompletely understood. MicroRNA-10a (miR-10a) is known to regulate CD4+ T-cell function, but its role in IL-22 production has not been defined. Here, using mouse CD4+ T cell-specific miR-10a knockout models, we examined how miR-10a regulates IL-22 expression and the underlying metabolic mechanisms. MiR-10a deficiency led to increased IL-22 production in CD4+ T cells both in vitro

Mucosal-associated invariant T (MAIT) cells are unconventional T cells, which upon activation can display potent cytotoxic and cytokine-producing capabilities. Together, these features make MAIT cells promising candidates for cancer immunotherapy. In this study, we show that MAIT cells can be efficiently amplified in vitro, and these amplified MAIT cells are armed with potent anticancer functions, including the ability to produce significant amounts of effector molecules such as IFNγ and granzyme B. Excitingly, we demonstrate that MAIT cells can be redirected to potently kill cancerous cells u

Genetically modified chimeric antigen receptor (CAR) T cells eliminate tumors by recognizing specific antigens on the cell surface. T cell ion channels (e.g. Kv1.3, KCa3.1) influence activation, proliferation, and effector functions such as target cell killing, through the regulation of Ca2+ signaling. We showed that CAR-expressing cells (Jurkat) specifically eliminate tumor cells (Raji B cells) in monolayer culture and inhibition of KCa3.1 by TRAM34 increased the tumor cell-killing ability of KCa3.1+ Jurkat CARs. Blockage of KCa3.1 facilitated the migration of KCa3.1+ Jurkat CARs (mean speed,

The central nervous system (CNS) includes a uniquely regulated immune response that supports homeostasis, response to injury, and response to pathogens. Recent work has shown that virus-associated immune responses in the CNS may contribute to neuronal injury and long-term outcomes such as neurocognitive decline. However, the fundamental mechanisms that regulate acute infiltration of immune cells from vascular compartments into the CNS are not well defined. Using an attenuated Venezuelan equine encephalitis virus TC83 (referred to as TC83) to inoculate via olfactory and intracranial injections,

Regulatory T cells (Tregs) play a crucial role in mediating recovery from acute lung injury (ALI). However, the complex roles of functionally heterogeneous Treg subsets in the lung during the resolution of acute inflammation remain unclear. To investigate the role of peripherally induced Tregs, we utilized mice lacking conserved noncoding sequence 1 (CNS1) of the Foxp3 locus, a genetic deletion that impairs peripheral Treg induction. We found that CNS1-deficient mice exhibit greater mortality and delayed resolution during ALI. Tregs induced via CNS1 modulated antiviral and proinflammatory immu
