The effect of active immunization against vasoactive intestinal peptide (VIP) and inhibin on reproductive performance of aging White Leghorn roosters.

Decreasing fertility in aging domestic roosters is a well-known phenomenon. Aging is manifested by a decrease in plasma testosterone level, testis function, and spermatogenesis, resulting in a low level of fertility. The roles of vasoactive intestinal peptide (VIP) and testicular inhibin in this aging process are not clear. The effects of active immunization against VIP, inhibin, or the combination of both hormones on the reproduction of aging White Leghorn (WL) roosters were assayed. In experiment 1a, 60 White Leghorn roosters (67 wk of age) were divided into 4 groups (n = 15/group). The firs

Host-pathogen interaction in invasive Salmonellosis.

Salmonella enterica infections result in diverse clinical manifestations. Typhoid fever, caused by S. enterica serovar Typhi (S. Typhi) and S. Paratyphi A, is a bacteremic illness but whose clinical features differ from other Gram-negative bacteremias. Non-typhoidal Salmonella (NTS) serovars cause self-limiting diarrhea with occasional secondary bacteremia. Primary NTS bacteremia can occur in the immunocompromised host and infants in sub-Saharan Africa. Recent studies on host-pathogen interactions in Salmonellosis using genome sequencing, murine models, and patient studies have provided new in

Global analysis of gene expression in response to L-Cysteine deprivation in the anaerobic protozoan parasite Entamoeba histolytica.

BACKGROUND: Entamoeba histolytica, an enteric protozoan parasite, causes amebic colitis and extra intestinal abscesses in millions of inhabitants of endemic areas. E. histolytica completely lacks glutathione metabolism but possesses L-cysteine as the principle low molecular weight thiol. L-Cysteine is essential for the structure, stability, and various protein functions, including catalysis, electron transfer, redox regulation, nitrogen fixation, and sensing for regulatory processes. Recently, we demonstrated that in E. histolytica, L-cysteine regulates various metabolic pathways including ene

The common hepatic branch of the vagus is not required to mediate the glycemic and food intake suppressive effects of glucagon-like-peptide-1.

The incretin and food intake suppressive effects of intraperitoneally administered glucagon-like peptide-1 (GLP-1) involve activation of GLP-1 receptors (GLP-1R) expressed on vagal afferent fiber terminals. Central nervous system processing of GLP-1R-driven vagal afferents results in satiation signaling and enhanced insulin secretion from pancreatic-projecting vagal efferents. As the vast majority of endogenous GLP-1 is released from intestinal l-cells following ingestion, it stands to reason that paracrine GLP-1 signaling, activating adjacent GLP-1R expressed on vagal afferent fibers of gastr

Gastrin response to candidate messengers in intact conscious rats monitored by antrum microdialysis.

We monitored gastrin release in response to locally applied candidate messengers in intact conscious rats. Earlier studies have been performed on anaesthetized animals, isolated pieces of antrum, or purified preparations of gastrin cells. In this study we created an experimental situation to resemble physiological conditions, using reverse microdialysis to administer regulatory peptides and amines that might affect gastrin secretion. Microdialysis probes were implanted in the submucosa of the antrum of the rat stomach. Three days later, putative messenger compounds were administered via the pr

Key importance of the Helicobacter pylori adherence factor blood group antigen binding adhesin during chronic gastric inflammation.

Helicobacter pylori has been assigned as a class I carcinogen because of its relation to gastric adenocarcinoma. Chronic H. pylori infection may lead to severe gastritis, glandular atrophy (AT), and intestinal metaplasia (IM). Strains secreting the vacuolating toxin VacA and producing the cytotoxin-associated antigen CagA (type 1 strains), as well as the blood group antigen binding adhesin (BabA) targeting Lewis(b) antigens, have been associated previously with distal gastric adenocarcinoma (M. Gerhard et al., Proc. Natl. Acad. Sci. USA, 96: 12778-12783, 1999) and may therefore also be related

Preclinical and clinical pharmacology of the 5-HT3 receptor antagonists.

5-HT3-receptor antagonists are potent and highly selective competitive inhibitors of the 5-HT3-receptor with negligible affinity for other receptors. They are rapidly absorbed and penetrate the blood-brain barrier easily. 5-HT3-receptor antagonists are metabolized by diverse subtypes of the cytochrome P450-system, metabolites are excreted mainly in urine. Half-lifes in healthy subjects vary from 3-4 hours (ondansetron, granisetron) to 7-10 hours (tropisetron, hydrodolasetron). 5-HT3-receptor antagonists do not modify any aspect of normal behaviour in animals or induce remarkable changes of phy

The cea10 gene encodes a secreted member of the murine carcinoembryonic antigen family and is expressed in the placenta, gastrointestinal tract and bone marrow.

Although members of the carcinoembryonic antigen (CEA) family have been shown to convey cell adhesion in vitro, their location in some tissues contradicts such a function. As a basis for investigating their in vivo functions, we are characterizing the mouse CEA family. This paper describes the structure and expression of a new murine family member, cea10. Two full-length cDNA clones were isolated from a mouse colon library, whose deduced protein sequence comprises two immunoglobulin variable-like N-domains, directly followed by a short C-terminal domain indicating that this molecule is secrete

Preparation and in vitro pharmacology of 5-HT4 receptor ligands. Partial agonism and antagonism of metoclopramide analogous benzoic esters.

Alicyclic ester analogues of the gastroprokinetic benzamide metoclopramide (1) and its ester congener SDZ 205557 (2), a 5-HT4 receptor antagonist, were prepared by O-alkylation of 4-amino-5-chloro-2-methoxybenzoate with N-(2-chloroethyl) substituted alicyclic amines. The bromo and iodo analogue of compound 13b (2-(1-piperidinyl)ethyl 4-amino-5-chloro-2-methoxybenzoate) were obtained by halogenation of dechloro-13b with N-halogenated succinimides. The series was evaluated in functional in vitro assays with regard to affinity for serotoninergic 5-HT4, 5-HT3 and muscarinic M3 receptors. The affin

Beeturia and the biological fate of beetroot pigments.

Beeturia, the passage of pink or red urine after the ingestion of beetroot, is said to occur in 10-14% of the population, and is more common in iron deficiency and malabsorption. A specific HPLC assay for betacyanins, the red beetroot pigments, in biological fluids was developed to study the prevalence of this apparent polymorphism in humans, and to investigate its basis in rats. Two major peaks were observed in chromatograms of extracts of unpickled beetroot. They had identical UV absorption spectra (lambda max = 535 nm) by diode array analysis, and mass spectrometry indicated that one (betac

Chromogranin: widespread immunoreactivity in polypeptide hormone producing tissues and in serum.

Chromogranin A is the major soluble protein, co-stored and co-released with catecholamines from storage vesicles of adrenal medulla and sympathetic nerve, and has been used as an index of exocytotic sympathoadrenal neurosecretion. Since some neuropeptides have a rather wide distribution in nerve, gut, and endocrine glands, we used a sensitive chromogranin A radioimmunoassay to probe the occurrence of chromogranin in several polypeptide hormone producing tissues. Immunoreactive chromogranin was ubiquitous in such tissues, in rank order of concentration (microgram/g wet weight): adrenal medulla

Palmatine Ameliorates High-Temperature and High-Humidity-Induced Enteritis via Mitophagy and NLRP3 Inflammasome Degradation.

BACKGROUND: Previous studies have shown that a High-Temperature- and High-Humidity (HTH) environment leads to mild enteritis, accompanied by impaired mitophagy and activated NLRP3-IL-1β, as found in Inflammatory Bowel Disease (IBD). Therefore, whether Palmatine (PAL), a candidate for treating IBD, ameliorates HTH-induced enteritis via mitophagy-associated mechanisms was examined. This study aimed to investigate the protective effects of PAL against HTH-induced enteritis in mice and determine the underlying mechanisms. METHODS: BALB/c mice were used to model HTH-induced enteritis. The mice wer

Metabolic benefits of 1,3-diacylglycerol in type 2 diabetes mellitus and its association with gut microbiota-derived SCFAs-GPR41-GLP-1 signaling.

1,3-Diacylglycerol (1,3-DAG) is a dietary lipid with known lipid-lowering effects, yet its role in type 2 diabetes mellitus (T2DM) and gut microbiota regulation remains unclear. This study investigated the effects of 1,3-DAG on glucose-lipid metabolism and gut microbiota- short chain fatty acid (SCFA)-G protein-coupled receptors (GPR41) signaling in T2DM rats. Male Wistar rats were fed a high-fat, high-sugar diet with weekly Streptozotocin (STZ) injections (30 mg kg-1) for 4 weeks to induce T2DM. After confirming stable hyperglycemia, rats were randomly divided into four groups: healthy contro

Dihydroartemisinin alleviates ulcerative colitis by modulating gut microbiota and butyrate to restore Treg/Th17 balance and intestinal barrier function.

Accumulating evidence suggests that dihydroartemisinin (DHA), a natural compound known for its anti-inflammatory and immunomodulatory properties, holds therapeutic potential for ulcerative colitis (UC). However, its mechanism of action, particularly concerning the gut microbiota and its metabolites, remains incompletely understood. In this study, we evaluated the therapeutic effects of dihydroartemisinin in a dextran sulfate sodium (DSS)-induced murine model of UC. Dihydroartemisinin was administered at doses of 10, 20, and 50 mg/kg, with 5-aminosalicylic acid (5-ASA, 50 mg/kg) serving as a po

Curtailment of Toll-like receptor signalling and cytokine production in dendritic cells by secreted products of Heligmosomoides polygyrus.

Dendritic cells (DCs) are the professional antigen-presenting cells responsible for recognition of pathogens and induction of appropriate adaptive immune responses. Many successful pathogens have evolved to release molecular products that can manipulate DC responses to dampen immunity. Here, we report that the excretory/secretory (ES) products of the intestinal helminth Heligmosomoides polygyrus bakeri profoundly repress the ability of murine and human DCs, differentiated under various conditions, to respond to canonical stimuli delivered through Toll-like Receptors (TLRs). H. polygyrus ES (HE

Genomic characterization, stress tolerance and cardiac transcriptomic responses of a virulent Edwardsiella piscicida strain from farmed largemouth bass.

Edwardsiella piscicida is an important intracellular pathogen in finfish aquaculture. During a mortality episode in farmed largemouth bass (Micropterus salmoides) in Foshan, South China, we isolated a virulent E. piscicida strain (B202203044) from diseased liver and characterized its pathogenicity, stress tolerance and genomic features. The strain was identified as E. piscicida based on biochemical profiling, species-specific PCR and 16S rRNA sequencing, and caused typical Edwardsiellosis signs in experimentally infected bass. Intraperitoneal challenge caused dose-dependent mortality, reaching

Primary Hyperparathyroidism in Celiac Disease: Diagnostic Challenges and Therapeutic Strategies.

Celiac disease (CD) is a primary immune-mediated enteropathy triggered by gluten ingestion in genetically predisposed individuals. Gluten ingestion results in villous atrophy of the small intestine and can lead to impaired absorption of multiple vitamins and nutrients, including vitamin D and calcium. This may result in vitamin D deficiency, a physiologic stimulus for parathyroid hormone (PTH) secretion, subsequently causing secondary hyperparathyroidism. Malabsorption and hyperparathyroidism in CD can contribute to metabolic bone disease (MBD), including osteopenia or osteoporosis. Correction

Modulation of hyperglycemia, insulin resistance responses and gut microbiota composition in type 2 diabetic mice by capsicum residue soluble dietary fiber.

Type 2 diabetes mellitus (T2DM) poses substantial health risks, emphasizing the need to identify natural ingredients from biological sources that can alleviate its symptoms. Capsicums produce a large amount of capsicum residue (CR) by-products during processing, which are underutilized. CR is rich in soluble dietary fiber (SDF) and has shown potential as an adjunctive hypoglycemic dietary supplement. This study investigated the structure of capsicum residue soluble dietary fiber (CR-SDF), its hypoglycemic effects in T2DM mice fed a high-fat diet combined with streptozotocin, and its hypoglycem
