Rapid assessment of mosquitoes and arbovirus activity after floods in southeastern Kansas, 2007.

A rapid assessment was conducted in July-August 2007 to determine the impact of heavy rains and early summer floods on the mosquitoes and arbovirus activity in 4 southeastern Kansas counties. During 10 days and nights of collections using different types and styles of mosquito traps, a total of 10,512 adult female mosquitoes representing 29 species were collected, including a new species record for Kansas (Psorophora mathesoni). High numbers of Aedes albopictus were collected. Over 4,000 specimens of 4 Culex species in 235 species-specific pools were tested for the presence of West Nile, St. L

Ivermectin as a rodent feed-through insecticide for control of immature sand flies (Diptera: Psychodidae).

Ivermectin was evaluated as a potential rodent feed-through for the control of immature stages of Phlebotomus papatasi. The survival of sand fly larvae fed feces of Syrian hamsters (Mesocricetus auratus) that had been fed a diet containing 0, 2, 6, 10, 20, 60, or 100 ppm ivermectin was measured. Sand fly larvae fed the feces of ivermectin-treated hamsters had significantly reduced survival, with 100% mortality of larvae fed feces of hamsters fed a diet containing 20, 60, and 100 ppm ivermectin. The results of this study suggest that a control strategy using rodent baits containing ivermectin t

Resistance to permethrin in Culex tarsalis in northeastern Colorado.

Resistance to permethrin and the activity of metabolic enzymes were monitored in field-collected Culex tarsalis from northeastern Colorado during 2005 and 2006 and compared with a lab strain from Bakersfield, CA. Collections were made from 13 different sites within and outside of the mosquito abatement area. Resistance levels by year and early vs. late season were examined and median lethal time values and 95% confidence intervals were compared with the lab strain. Mosquitoes collected in 2005 were more resistant to permethrin than either the lab strain or mosquitoes collected in 2006. Glutath

Entomological studies along the Colorado Front Range during a period of intense West Nile virus activity.

To better understand the ecology of West Nile virus transmission in Northern Colorado, field studies were conducted in Larimer and Weld counties from September 2003 through March 2005. During summer studies, 18,540 adult mosquitoes were collected using light traps and gravid traps. West Nile virus RNA was detected in 24 of the 2,140 mosquito pools tested throughout the study area in 2003 and 2004. Culex tarsalis had the highest minimum infection rate (MIR) in both 2003 (MIR = 34.48) and in 2004 (MIR = 8.74). During winter studies, 9,391 adult mosquitoes were collected by aspirator from various

A two-year evaluation of elevated canopy trapping for Culex mosquitoes and West Nile virus in an operational surveillance program in the northeastern United States.

The effectiveness of CO2-baited Centers for Disease Control and Prevention miniature light traps elevated in the tree canopy (approximately 7.6 m) was compared with light traps placed at ground level (approximately1.5 m) and grass-sod infused gravid traps for collecting Culex pipiens, Culex restuans, and Culex salinarius and detecting West Nile virus (WNV) activity in an operational surveillance program that encompassed 12 ecologically diverse sites in Connecticut in 2004 and 2005. More than twice as many Cx. pipiens were collected on average in light traps suspended in the tree canopy than in

Evidence of laboratory vector competence of Culex modestus for West Nile virus.

During the past 10 years, West Nile virus (WNV) has been responsible for large and severe human outbreaks and horse epizootics through the Old and the New World. Since WNV was first isolated from field-collected mosquitoes of Culex modestus in 1964 in France, this species, which aggressively feeds on birds and mammals, is considered a putative WNV vector in Europe. We report on the first evidence on the laboratory vector competence of Cx. modestus for WNV. To assess this trait, F3 and F4 females from southern France were fed through a membrane with a strain of WNV isolated from a horse in the

Associations between two mosquito populations and West Nile virus in Harris County, Texas, 2003-06.

Associations between Culex quinquefasciatus, Aedes albopictus and West Nile virus (WNV) activity, temperature, and rainfall in Harris County, Texas 2003-06 are discussed. Human cases were highly correlated to Cx. quinquefasciatus (r = 0.87) and Ae. albopictus (r = 0.78) pools, blue jays (r = 0.83), and Ae. albopictus collected (r = 0.71), but not Cx. quinquefasciatus collected (r = 0.45). Human cases were associated with temperature (r = 0.71), not rainfall (r = 0.29), whereas temperature correlated with Ae. albopictus and Cx. quinquefasciatus collections (r = 0.88 and 0.70, respectively) and

Reduced susceptibility to deltamethrin and kdr mutation in Anopheles stephensi Liston, a malaria vector in India.

The Indian urban malaria vector Anopheles stephensi Liston was selected for deltamethrin resistance for 25 generations (F25) at larval and adult stages separately in the laboratory. There was roughly a 151-fold increase in the lethal concentration (LC)50 and a 99-fold increase in the LC90 in larval selection, when the F25 was compared with the parent colony. Similarly, adult selection resulted in a 39-fold increase in the LC50 and a 31-fold increase in the LC90 in the adults. Knockdown bioassays conducted on adults (selected at the larval and adult stages) against the diagnostic concentration

New tools for surveillance of adult yellow fever mosquitoes: comparison of trap catches with human landing rates in an urban environment.

A novel mosquito trapping system, the BG-Sentinel trap, was evaluated as a monitoring tool for adult Aedes aegypti in field tests in the city of Belo Horizonte, Brazil. Human landing/biting collections, a gas-powered CO2 trap, and a Fay-Prince trap with only visual cues serving as references to evaluate the efficacy of the new trap. The BG-Sentinel is a simple suction trap that uses upward-directed air currents as well as visual cues to attract mosquitoes. The trap was tested with a new dispenser system (BG-Lure) that releases artificial human skin odors and needs no CO2. In comparison with th

Quantification of West Nile virus in the saliva of Culex species collected from the southern United States.

Culex quinquefasciatus, Cx. restuans, Cx. pipiens complex, and Cx. nigripalpus were collected as larvae or egg rafts from the southern USA. Adult female mosquitoes were intrathoracically inoculated with approximately 1,000 plaque-forming units of West Nile virus (WNV) and saliva was collected from them 5 days later. The amount of infectious WNV in the saliva samples was quantified by plaque assay and WNV RNA was detected by reverse transcriptase polymerase chain reaction (RT-PCR). More than 90% of the mosquitoes had either infectious virus or viral RNA in their saliva. The RT-PCR assay detecte

Vector competence of Peruvian mosquitoes (Diptera: Culicidae) for a subtype IIIC virus in the Venezuelan equine encephalomyelitis complex isolated from mosquitoes captured in Peru.

We evaluated mosquitoes collected in the Amazon Basin, near Iquitos, Peru, for their susceptibility to a subtype IIIC strain of the Venezuelan equine encephalomyelitis complex. This virus had been previously isolated from a pool of mixed Culex vomerifer and Cx. gnomatos captured near Iquitos, Peru, in 1997. After feeding on hamsters with viremias of about 10(8) plaque-forming units of virus per ml, Cx. gnomatos was the most efficient vector. Other species, such as Ochlerotatus fulvus and Psorophora cingulata, although highly susceptible to infection, were not efficient laboratory vectors of th

A comparison of two West Nile virus detection assays (TaqMan reverse transcriptase polymerase chain reaction and VecTest antigen assay) during three consecutive outbreaks in northern Illinois.

Mosquitoes identified as female Culex (Culex) species, primarily mixtures or uniform batches of Culex pipiens and Culex restuans, were collected daily from gravid traps by 2 mosquito abatement districts (MADs) in Cook County, Illinois. From 2002 through 2004, batches (pools) of mosquitoes were tested by the MADs for West Nile virus (WNV) by using VecTest WNV antigen assays and the same samples were retested, usually within 1-2 wk, for WNV RNA by the TaqMan reverse transcriptase polymerase chain reaction (RT-PCR). There were 952 TaqMan-positive pools out of 3,953 pools over the 3 years, and abo

Investigation of relationships between Aedes aegypti egg, larvae, pupae, and adult density indices where their main breeding sites were located indoors.

Aedes aegypti (L.) density indices obtained in a dengue fever (DF) endemic area were compared. One hundred and twenty premises, in an urban area of Colombia where dengue type-1 and type-2 virus cocirculated, were randomly selected and sampled for 7 months. The geometric mean monthly numbers (density index, DI) of Ae. aegypti eggs (ODI), 4th instar larvae (LDI), pupae (PDI), and adults (ADI) were calculated based on the use of ovitraps, nets, and manual aspirators, respectively. A negative temporal correlation was observed between the LDI and the ODI (r = -0.83, df = 5, and P < 0.01). Positive

Detection of West Nile virus RNA from the louse fly Icosta americana (Diptera: Hippoboscidae).

West Nile virus (WNV) was detected by Taqman reverse transcription-polymerase chain reaction in 4 of 85 (4.7%) blood-engorged (n = 2) and unengorged (n = 2) Icosta americana (Leach) hippoboscid flies that were collected from wild raptors submitted to a wildlife rehabilitation center in Mercer County, NJ, in 2003. This report represents an additional detection of WNV in a nonculicine arthropod in North America and the first documented detection of the virus in unengorged hippoboscid flies, further suggesting a possible role that this species may play in the transmission of WNV in North America.

Detection of chicken antibodies to mosquito salivary gland antigens by enzyme immunoassay.

Sentinel chickens are used to detect western equine encephalomyelitis, St. Louis encephalitis, and West Nile virus activity. Flocks that receive high mosquito exposure will be most effective for surveillance purposes. However, mosquito population indices at the flock sites may only provide an indirect measure of potential exposure. Therefore, we developed an indirect enzyme immunoassay to detect chicken antibodies to salivary gland antigens (SGAs) from Culex tarsalis, the primary arbovirus vector in California. Chickens fed upon by Cx. tarsalis developed an antibody response that was proportio

Surveillance and behavioral investigations of Aedes aegypti and Aedes polynesiensis in Moorea, French Polynesia, using a sticky ovitrap.

The effectiveness of the sticky ovitrap was assessed for the container-breeding Aedes aegypti and Aedes polynesiensis in Moorea, French Polynesia. These mosquitoes are the primary vectors of dengue viruses and Bancroftian filariasis, respectively, in the area. Both Ae. aegypti and Ae. polynesiensis were collected in greatest numbers in sticky ovitraps baited with water or grass infusions rather than leaf infusions. Sticky ovitrap collections were significantly higher for both species in the 12 h post-midday than pre-midday and in traps set in shaded compared with open locations. More females o

Resting behavior and malaria vector incrimination of Anopheles stephensi in Goa, India.

Malaria in Goa, India, has been endemic ever since an outbreak occurred in 1986. Anopheles stephensi Liston has always been suspected as a malaria vector in this area. Due to lack of knowledge on its resting behavior, sufficient adult females could not be collected and incriminated as vectors in the past. In this study mosquito collections were conducted in three endemic urban and suburban areas of Goa. In well-built houses, 67 h of collections did not yield a single An. stephensi mosquito, although other species were encountered. However, collections in construction sites and workers' huts fo
