The influence of body temperature on tissue stiffness, blood perfusion, and water diffusion in the mouse brain.

While hypothermia of the brain is used to reduce neuronal damage in patients with conditions such as traumatic brain injury or stroke, little is known about how temperature affects the biophysical properties of in vivo brain tissue. Therefore, we measured shear wave speed (SWS), apparent diffusion coefficient (ADC), and cerebral blood flow (CBF) in the mouse brain at different body temperatures to investigate the relationship between temperature and tissue stiffness, water diffusion, and blood perfusion in the living brain. Multifrequency magnetic resonance elastography (MRE), diffusion-weight

Epstein-Barr Virus-Specific CD8 T Cells Selectively Infiltrate the Brain in Multiple Sclerosis and Interact Locally with Virus-Infected Cells: Clue for a Virus-Driven Immunopathological Mechanism.

Epstein-Barr virus (EBV) is a ubiquitous herpesvirus strongly associated with multiple sclerosis (MS), a chronic inflammatory disease of the central nervous system (CNS). However, the mechanisms linking EBV infection to MS pathology are uncertain. Neuropathological and immunological studies suggest that a persistent EBV infection in the CNS can stimulate a CD8 T-cell response aimed at clearing the virus but inadvertently causing CNS injury. Inasmuch as in situ demonstration of EBV-specific CD8 T cells and their effector function is missing, we searched for EBV-specific CD8 T cells in MS brain

Erythropoietin as an add-on treatment for cognitive side effects of electroconvulsive therapy: a study protocol for a randomized controlled trial.

BACKGROUND: Electroconvulsive therapy (ECT) is the most effective treatment for severe depression, but its use is impeded by its cognitive side effects. Novel treatments that can counteract these side effects may therefore improve current treatment strategies for depression. The present randomized trial investigates (1) whether short-term add-on treatment with erythropoietin (EPO) can reduce the cognitive side -effects of ECT and (2) whether such effects are long-lasting. Further, structural and functional magnetic resonance imaging (MRI) will be used to explore the neural underpinnings of suc

Predicting hourly air pollutant levels using artificial neural networks coupled with uncertainty analysis by Monte Carlo simulations.

Recent progress in developing artificial neural network (ANN) metamodels has paved the way for reliable use of these models in the prediction of air pollutant concentrations in urban atmosphere. However, improvement of prediction performance, proper selection of input parameters and model architecture, and quantification of model uncertainties remain key challenges to their practical use. This study has three main objectives: to select an ensemble of input parameters for ANN metamodels consisting of meteorological variables that are predictable by conventional weather forecast models and varia

Pilot Study Employing a Machine Learning Approach as a Potential Method for Predicting Parkinson's Disease Using Voice as a Digital Biomarker and the SHAP Approach for Feature Engineering.

Introduction: Voice-based digital biomarkers have emerged as a promising approach for distinguishing individuals with neurodegenerative disorders, particularly Parkinson's disease (PD), from healthy subjects (HS). With the increasing availability of smartphone and web-based recording tools, voice data can be collected efficiently in both clinical and remote settings. However, further validation is required before such approaches can be translated into routine clinical practice. Methods: This study used a cross-sectional analysis at the recording level, treating repeated recordings from the sam

Early Prediction of Cardiomyopathy in Pediatric Cancer Survivors Using Echocardiographic Video-Based Deep Learning.

PURPOSE: Cardiomyopathy (CM) is a leading cause of late mortality in childhood cancer survivors, but it is often identified only after irreversible cardiac remodeling has already occurred. In this proof-of-concept study, we assessed whether deep learning applied to routine echocardiographic videos can identify features predictive of future CM. METHODS: We analyzed parasternal short-axis clips from cancer survivors ≤21 years old at cancer diagnosis. 3D deep convolutional neural networks were trained to classify CM-positive versus CM-negative studies from echocardiograms conducted up to five ye

Deep Learning-Based Automated Computed Tomography Classification of Posterior Malleolar Fractures for a Standardized Diagnostic Framework.

Despite advances in computed tomography (CT)-based classification systems for posterior malleolar fractures (PMFs), diagnostic consistency remains limited because these systems are still highly dependent on the observer's experience. This study aimed to develop a deep learning-based automated model capable of classifying PMFs according to the Haraguchi, Mason-Molloy, and Bartoníček-Rammelt clasification systems using CT scans, and to compare the classification performance of different convolutional neural network architectures. Retrospectively collected preoperative CT data from 480 adult pati

Safety signals of tocilizumab in pediatric patients: A real-world pharmacovigilance analysis based on the FAERS database with network pharmacology insights.

Tocilizumab, a humanized monoclonal antibody targeting the interleukin-6 receptor, is widely used in immune-mediated diseases, yet its real-world safety in pediatric populations remains insufficiently characterized. This retrospective pharmacovigilance study analyzed adverse event (AE) reports from the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS) database (2010-2025), focusing on pediatric cases where tocilizumab was the primary suspect drug. After data cleaning, 1,485 reports were included. Four classical disproportionality methods - reporting odds ratio (ROR), pro

Preoperative Angiographic Washout and Coronary Artery Bypass Graft Patency.

BACKGROUND: Coronary artery bypass graft (CABG) failure occurs in 3% to 30% of grafts by 1 year. Because graft flow depends on distal outflow, elevated microvascular resistance may predispose target vessels to graft failure. OBJECTIVES: The objective of the study was to determine whether quantitative target-vessel contrast washout from standard preoperative coronary angiography is associated with subsequent CABG patency. METHODS: This single-center retrospective study included 107 patients undergoing CABG from 2021 to 2024 with preoperative coronary angiography and postoperative graft imagin

Cuproptosis-related molecular alterations in a neonatal rat model of hypoxic-ischemic brain damage.

BACKGROUND: Copper homeostasis imbalance is closely linked to the progression of various neurological disorders. Cuproptosis, a recently identified form of cell death caused by abnormal intracellular copper accumulation, has an unclear relationship with hypoxic-ischemic brain damage (HIBD). This study aims to characterize cuproptosis-associated molecular changes in the pathological process of HIBD in neonatal rats. METHODS: A neonatal rat HIBD model was established using the modified Rice-Vannucci method. Cerebral blood flow was measured with laser Doppler flowmetry, while infarct volume was

Real-time dynamic prediction of mortality and renal replacement therapy after cardiac surgery using a time-series deep learning model.

OBJECTIVE: Predicting postoperative deterioration following cardiac surgery remains challenging. Conventional risk scores rely on static variables and fail to capture evolving physiologic trajectories. We developed a time-series deep learning model (DLM) using serial intensive care unit (ICU) measurements to dynamically predict mortality and continuous renal replacement therapy (CRRT) following cardiac surgery. METHODS: Using the Medical Information Mart for Intensive Care database, we analyzed ICU admissions of patients undergoing coronary artery bypass grafting, isolated or combined with va

Disease-associated RNA and protein signatures in iPSC-derived microglia model of Alzheimer's disease.

INTRODUCTION: Microglia, the resident immune cells of the central nervous system, play a critical role in maintaining neural homeostasis and regulating inflammatory responses in the brain. Increasing evidence suggests that microglial dysfunction contributes to the progression of neurodegenerative diseases, including Alzheimer's disease (AD). However, the molecular mechanisms underlying these alterations remain incompletely understood. This study aimed to characterize disease-associated molecular changes in microglia derived from induced pluripotent stem cells (iPSCs) of sporadic AD patients an

The Endogenous Opioid System in Compulsive Eating.

Background/Objectives: The rates of obesity and binge-eating disorder (BED) have increased markedly over the last few decades. The onset of these conditions has been associated in part with the disruption of neural pathways that regulate food reward. Existing literature has implicated the endogenous opioid system as an important mediator of pleasure and reinforcing behaviors associated with food intake. While the relationship between opioids and food intake has been studied extensively, how dysregulated opioid signaling contributes to compulsive eating still remains unclear. Therefore, the aim
