BACKGROUND: Spinal cord injury (SCI) leads to severe and often permanent neurological deficits. The inhibitory Nogo/NgR signaling pathway constitutes a major barrier to axonal regeneration. Transplantation of bone marrow mesenchymal stem cells (BMSCs) and modulation of microRNAs, such as miR-let-7b, represent promising therapeutic strategies for neural repair.
OBJECTIVES: This study aimed to investigate the role and mechanism of miR-let-7b-modified BMSCs in repairing SCI, focusing on their interaction with the Nogo/NgR signaling pathway.
METHODS: In vitro, an oxygen-glucose deprivation (OGD)

Peripheral nerves contribute to tumor progression, but the mechanisms by which neural signals regulate cancer cell plasticity and immune resistance remain unclear. Using a paired murine model of intraneural and nonintraneural tumor growth with single-cell RNA sequencing, we identified a nerve-associated cancer cell state marked by increased expression of secretory leukocyte protease inhibitor (SLPI). SLPI was elevated in intraneural tumors and in human tumors with perineural invasion. Sensory neuron-derived substance P (SP) induced SLPI secretion through tumor cell TACR1, whereas sensory dener

Glycoprotein sialylation represents a critical posttranslational modification with diverse biological roles in animals. This review explores its multifaceted functions in the nervous system, with particular emphasis on neurophysiology, homeostasis, and associated neurological disorders. The sialylation pathway modulates key neural processes through effects on glycoprotein stability, localization, activity, and molecular interactions. Examples include its crucial role in the regulation of neuronal excitability by modulating the functions of voltage-gated ion channels. Recent studies have uncove

Receptor tyrosine kinase (RTK) signaling is a central mechanism by which local tissue interactions are translated into coordinated morphogenesis during eye development. The vertebrate eye provides a uniquely tractable system for studying this logic because it is assembled from neural epithelium, surface ectoderm, neural crest-derived mesenchyme, vascular endothelium, and specialized epithelial and endothelial derivatives across a prolonged developmental timeline. In this review, we synthesize how major RTK families, including FGF-FGFR, EGF-ErbB, PDGF-PDGFR, VEGF-VEGFR, Eph-ephrin, ANGPT-TEK/Ti

Nowadays, extracellular vesicles (EVs) such as exosomes participate in cell-cell communication and gain attention as a new approach for cell-free therapies. Recently, various studies have demonstrated the therapeutic ability of exosomes, while the biological effect of human endometrial stem cell (hEnSC)-derived small EVs such as exosomes is still unclear. Herein, we obtained small EVs from hEnSC and indicated that these small EVs activate the vital cell signaling pathway and progress neurite outgrowth in PC-12 cell lines. For this purpose, hEnSC-derived small EVs were extracted by ultracentrif

Spinal muscular atrophy (SMA) is an inherited neuromuscular disease resulting from a recessive mutation in the SMN1 gene. This disease affects multiple organ systems with varying degrees of severity. Exploration of the molecular pathological changes occurring in different cell types in SMA is crucial for developing new therapies. This study collected 39 human microarray datasets from ArrayExpress and GEO databases to build an integrative transcriptomic analysis for recognizing novel SMA targets. The transcriptomic analysis was conducted through combining weighted correlation network analysis (

A heightened sympathetic tone accelerates the development of lethal arrhythmias after myocardial infarction (MI) and the progression of heart failure (HF). Cardiomyocytes control their local neural milieu by expression of nerve growth factor (NGF), which triggers sympathetic neural growth (sympathetic nerve sprouting: SNS). The molecular mechanisms that regulate NGF expression are largely unknown. During HF or MI the myocytes are exposed to increased mechanical load and adrenergic stimulation. Both stimuli induce myocyte hypertrophy. The angiotensin-II-calcineurin-NFAT (nuclear factor of activ

Tourette syndrome (TS) is a neurodevelopmental disorder characterized by involuntary motor and phonic tics. It is hypothesized that excess dopamine leads to an imbalance in the pathways through the basal ganglia, resulting in unchecked movements via thalamic disinhibition. It has been unclear whether TS is associated with cognitive control deficits as well as pure motor control deficits, or whether cognitive deficits are associated with the presence of comorbid conditions. Furthermore, little is known about the neural underpinnings of TS in childhood, prior to the long-term effects of medicati

Intraoperative neuromonitoring (IONM) involves repeated electrical stimulation of the nervous system to assess integrity of neural pathways. Transcranial motor evoked potentials (tcMEPs) require high levels of electrical stimulation and are not recommended for patients with neurologic stimulators in situ, such as a deep brain stimulator (DBS) system, due to the risk of device damage and patient harm. We describe a case of a patient with bilateral DBS undergoing a surgery with IONM, in which direct electrical response wave (D-wave) monitoring via an epidural electrode was used, rather than tcME…

Fear is essential for survival; however, its dysregulation may lead to various neuropsychiatric disorders. Understanding the neural networks involved in the expression of fear is useful for elaborating therapeutic interventions (pharmacological or psychoeducation). We aimed to summarize suspected roles of amygdala, but also of other structures such as insula, frontal and temporal associative cortices, and brainstem. Three pathways can be distinguished leading to the expression of fear: indirect, direct and interoceptive, comprising the amygdala as a "hub of fear". Moreover, temporal epilepsies

Advances in theoretical understanding are frequently unlocked by access to large, diverse experimental datasets. Our understanding of olfactory neuroscience and psychophysics remain years behind the other senses, in part because rich datasets linking olfactory stimuli with their corresponding percepts, behaviors, and neural pathways remain scarce. Here we present a concerted effort to unlock and unify dozens of stimulus-linked olfactory datasets across species and modalities under a unified framework called Pyrfume. We present examples of how researchers might use Pyrfume to conduct novel anal…

Optic nerve injury induces optic nerve degeneration and retinal ganglion cell (RGC) death that lead to visual disturbance. In this study, we examined if topical ripasudil has therapeutic potential in adult mice after optic nerve crush (ONC). Topical ripasudil suppressed ONC-induced phosphorylation of p38 mitogen-activated protein kinase and ameliorated RGC death. In addition, topical ripasudil significantly suppressed the phosphorylation of collapsin response mediator protein 2 and cofilin, and promoted optic nerve regeneration. These results suggest that topical ripasudil promotes RGC protect

Leg pain is a very common complaint in clinical medicine which deserves thorough investigation. All tissues of the lower leg are able to cause pain, each of them by different pathomechanisms. In the current review, all the different types of tissue, i.e., spine, neural plexus, peripheral nerves, muscles, and vasculature, are systematically covered. The different disease entities are explained in terms of pathophysiology and clinical picture. Diagnostic measures and pathways are sketched, as well as therapeutic approaches in some instances. Diseases of the bone and joint are omitted since they

Many candidate genes are now thought to confer susceptibility to autism spectrum disorders (ASDs). Here we review four interrelated complexes, each composed of multiple families of genes that functionally coalesce on common cellular pathways. We illustrate a common thread in the organization of glutamatergic synapses and suggest a link between genes involved in Tuberous Sclerosis Complex, Fragile X syndrome, Angelman syndrome and several synaptic ASD candidate genes. When viewed in this context, progress in deciphering the molecular architecture of cellular protein-protein interactions togethe

Several neurocutaneous syndromes have been reported in association with tuberous sclerosis complex. The coexistence of intracranial features of Sturge-Weber syndrome in patients with a confirmed diagnosis of tuberous sclerosis complex has been recently described. Several hypotheses can be raised to explain this association. The coexistence of signs of both diseases in the same patient could be explained by abnormal neural crest development, and by mutually enhanced common altered pathways. Mutation characterization of tuberous sclerosis complex and a better definition of the clinical and neuro

An emerging theme in neurodegenerative diseases is the aggregation of proteins as inclusions in neural cells. Their presence is a useful tool in the differential diagnosis of the particular illness, although in no instance is the specific role of the inclusions in disease pathogenesis understood at present. However, apart from their role in the disease mechanism, the inclusions themselves may contain important molecular clues as to the mechanism(s) behind the specific inclusion-associated disease. Thus, isolation and analysis of the composition of the inclusions is likely to yield biochemical

Intercellular signaling is an important factor in cardiovascular regulation and the development of disease. The endothelins (ETs) are a recently described family of signaling peptides that appear to be important in cardiovascular regulation. Initial studies showed ET to be a potent vasoconstrictor of vascular smooth muscle. Subsequently, ET binding and activity have been demonstrated in a wide variety of tissues, including cardiac, venular, adrenal, and neural. Numerous studies have implicated the ETs in cardiovascular diseases such as hypertension, heart failure, and atherosclerosis. This art

We examined the effect of naloxone on plasma oxytocin levels during sexual activity in men. Eight normal men took part in a double-blind, two-period, cross-over design. Mean plasma oxytocin rose to 362% of baseline values at orgasm with placebo (saline) but showed no increase with naloxone (P less than 0.01). While naloxone had no effect on heart rate or blood pressure at orgasm, a decrease in the level of subjective arousal and pleasure at orgasm was noted. We conclude that opioid receptor blockade with naloxone has an inhibitory effect on the neural pathways mediating the oxytocin response a…
