Accumulating evidence suggests that neurons are key regulators of skeletal tissues. A recent study by Chen et al. demonstrated that aged brain neurons export WD repeat and FYVE domain containing 1 (WDFY1) to bone via extracellular vesicles. After uptake, WDFY1 engages in retromer-dependent endosome-to-Golgi recycling, shifting bone stem cell fate from osteogenesis toward adipogenesis, revealing a novel mechanism whereby brain aging regulates bone aging.

A table of molecularly defined chondrodysplasias with short limbs is provided. Several are discussed in detail, including osteogenesis imperfecta and type I collagen mutations, Jansen metaphyseal chondrodysplasia and parathyroid hormone/parathyroid hormone-related protein receptor mutation, and chondrodysplasias caused by fibroblast growth factor receptor 3 mutations. The latter group includes achondroplasia, hypochondroplasia, thanatophoric dysplasia (types 1 and 2), San Diego platyspondylic dysplasia, and SADDAN.

Sutures are fibrous joints in the vertebrate skull. They consist of two bone ends and intervening fibrous tissue which differentiates from embryonic mesenchyme. Sutures are not merely articulations between bones they are primary sites of osteogenesis mediating much of the growth of the face and skull vault. In this chapter the development of sutures will be described including the origin of sutural tissues, the determinants of suture location, and suture morphology. Also, the main functions of sutures will be explained.

The authors describe the progressive correction of a deformed forearm and an ulnar deviation of the hand as a result of a bullet wound. The injury comprised a fracture of the two bones of the forearm, with deformed callus and after effects of a probable Volkmann syndrome. The correction was obtained after 10 months. This report illustrates the interest and the difficulty presented by osteogenesis by distraction and osteotomy with a minimal cutaneous wound. This allows the reduction of risk to the cutaneum and vessels involved in extemporaneous corrections.
