Vaginal use of estradiol is associated with a reduced risk of rectal cancer in postmenopausal women: A Finnish nationwide case-control study.

OBJECTIVE: We assessed the impact of vaginal estradiol on the risk of rectal cancer. STUDY DESIGN: We identified in this nationwide case-control study primary rectal cancer cases with five age-matched control cases in 1994-2019 from our dataset of 1.1 million Finnish postmenopausal women. We excluded users of systemic hormone therapy. Then we traced users of vaginal estradiol (10-25 μg twice a week) from the reimbursement register in 1994-2013, a period in which users could be accurately traced in the database (which was not the case for 2014-2019). MAIN OUTCOME MEASURES: Odds ratios with 95

Effectiveness and safety of electroacupuncture and manual acupuncture in postmenopausal women with overactive bladder: a multicenter, randomized, controlled, parallel clinical trial.

BACKGROUND: Overactive bladder (OAB) is a common disease in menopausal women that can negatively affect a patient's quality of life and cause high stress levels. Despite several studies of the effects of electroacupuncture (EA) and manual acupuncture (MA) on OAB, to our knowledge, there has been no direct comparison between EA and MA for postmenopausal OAB in a single trial. The object of this study was to evaluate the effectiveness and safety of EA at two frequencies (2 and 16 Hz) and MA for OAB in postmenopausal women. METHODS: In this multicenter, randomized, controlled, parallel clinical

Diagnostic workup of patients with benign or inconclusive reports on office endometrial biopsy after first episode of postmenopausal blood loss.

BACKGROUND: Postmenopausal bleeding carries a risk of endometrial cancer, and office endometrial sampling (OES) is often the first diagnostic approach. Whether further diagnostic procedures such as hysteroscopy or saline infusion sonography (SIS) are required to rule out polyps and malignancies is uncertain. The objective of this study is to evaluate whether clinicians perform further diagnostic tests when OES is benign or inconclusive. Secondary outcomes include the patient characteristics associated with the likelihood of performing further diagnostic tests and the incidence of endometrial i

Dual disease co-expression analysis reveals potential roles of estrogen-related genes in postmenopausal osteoporosis and Parkinson's disease.

INTRODUCTION: The deficiency of estrogen correlates with a range of diseases, notably Postmenopausal osteoporosis (PMO) and Parkinson's disease (PD). There is a possibility that PMO and PD may share underlying molecular mechanisms that are pivotal in their development and progression. The objective of this study was to identify critical genes and potential mechanisms associated with PMO by examining co-expressed genes linked to PD. METHODS: Initially, pertinent data concerning PMO and PD were obtained from the GWAS database, followed by conducting a Bayesian colocalization analysis. Subsequen

A Metal-Phenolic Network-Coated Coacervate System Mediates Mitochondrial Modulation and Bone Homeostasis Restoration for Postmenopausal Osteoporosis Therapy.

Postmenopausal osteoporosis (PMOP) manifests as a systemic skeletal disorder arising from disrupted bone metabolic equilibrium, characterized by a marked reduction in bone mineral density and compromised microarchitectural integrity, ultimately leading to increased bone fragility and greater fracture susceptibility. Current delivery therapeutic strategies frequently fail to address mitochondrial dysfunction arising from estrogen depletion and senescence-associated processes. We developed a multifunctional coacervate system, in which peptide coacervates were used to encapsulate estradiol (E2) a

Catalpol attenuates postmenopausal osteoporosis by activating SIRT1-mediated P53 deacetylation and inhibiting CASPASE-3-driven osteoblast apoptosis.

BACKGROUND: Postmenopausal osteoporosis (PMOP) is primarily characterized by a disruption in the balance between bone formation and resorption, predominantly due to osteoblast apoptosis induced by estrogen deficiency. Sirtuin 1 (SIRT1), a pivotal deacetylase known for inhibiting P53-mediated apoptosis, is observed to be downregulated in PMOP. Catalpol (CAT), an iridoid glycoside derived from Rehmannia glutinosa, has demonstrated antiapoptotic and antioxidative properties; however, its potential role in modulating the SIRT1-P53 signaling pathway in the context of PMOP remains to be elucidated.
