Constitutively activated Jak-STAT pathway in T cells transformed with HTLV-I.

Human T cell lymphotropic virus I (HTLV-I) is the etiological agent for adult T cell leukemia and tropical spastic paraparesis (also termed HTLV-I-associated myelopathy). HTLV-I-infected peripheral blood T cells exhibit an initial phase of interleukin-2 (IL-2)-dependent growth; over time, by an unknown mechanism, the cells become IL-2-independent. Whereas the Jak kinases Jak1 and Jak3 and the signal transducer and activator of transcription proteins Stat3 and Stat5 are activated in normal T cells in response to IL-2, this signaling pathway was constitutively activated in HTLV-I-transformed cel

Identification of a dimerization domain in the C-terminal segment of the IE110 transactivator protein from herpes simplex virus.

The 775-amino-acid IE110 (or ICP0) phosphoprotein of herpes simplex virus (HSV) functions as an accessory transcription factor during the lytic cycle and plays a critical role in reactivation from latent infection. By immunofluorescence analysis, IE110 localizes in a novel pattern consisting of several dozen spherical punctate granules in the nuclei of DNA-transfected cells. We constructed a hybrid version of IE110 that contained an epitope-tagged domain from the N terminus of the HSV IE175 protein and lacked the IE110 N-terminal domain that confers punctate characteristics. This hybrid IE175(

Additive effects of ultraviolet B and crude coal tar on cutaneous carcinogen metabolism: possible relevance to the tumorigenicity of the Goeckerman regimen.

The effect of cutaneous exposure to ultraviolet B (UVB) radiation alone, to crude coal tar (CCT) alone, and to the combination of UVB and CCT on the inducibility of the microsomal cytochrome P-450-dependent carcinogen-metabolizing enzyme aryl hydrocarbon hydroxylase (AHH) and other monooxygenases such as 7-ethoxyresorufin O-deethylase (ERD) and 7-ethoxycoumarin O-deethylase (ECD) activities in the skin of neonatal rats was studied. Exposure of the animals to UVB (400-1600 mJ/cm2) alone resulted in a dose-dependent increase in cutaneous enzyme activities. At a UVB dose of 1200 mJ/cm2 increases

Highly sensitive detection of exosomal miR-122 with CRISPR-Dx for precise HCC diagnosis.

Hepatocellular carcinoma (HCC), one of the most common malignancies worldwide, poses a major threat to the global health. Its prognosis depends heavily on early detection. Currently, alpha-fetoprotein (AFP) is the widely used biomarker for HCC diagnosis. Nevertheless, approximately 40% of the HCC cases do not produce AFP. It has been reported that the downregulation of miR-122 is strongly associated with the development and progression of HCC. Here, employing miR-122 as a biomarker, we developed a cascade amplification-based CRISPR-Dx platform for HCC diagnosis. Owing to the strong trans-cleav

Marigold flavonoids coordinate emulsion stabilization, epithelial transport and gut microbiota modulation to enhance intestinal lutein absorption.

BACKGROUND: Flavonoids naturally coexisting with lutein in marigold (Tagetes erecta L.) extracts have been suggested to influence carotenoid bioavailability; however, their coordinated roles in intestinal lutein absorption remain insufficiently understood. This study systematically explored the synergistic effects of six major marigold flavonoids on lutein absorption via a multilevel strategy integrating emulsion behavior, intestinal epithelial transport and gut microbiota modulation. RESULTS: In vitro, flavonoids stabilized lutein-containing mixed emulsion, reduced particle size, increased ζ

The end of 'leaky gut': pathway-specific mechanistic approaches to barrier restoration.

PURPOSE OF REVIEW: For over 50 years, increased intestinal permeability has been associated with diverse inflammatory and systemic diseases. Yet the oversimplified concept of 'leaky gut' as a singular phenomenon has limited both mechanistic understanding and therapeutic development. RECENT FINDINGS: It is now recognized that intestinal permeability occurs via two molecularly distinct, differentially regulated trans-tight junction pathways, while a tight junction-independent unrestricted pathway allows flux at sites of epithelial damage. The pore pathway is a high-conductance, size-selective a

Inference of spatial chromatin accessibility via integration of spatial transcriptomics and single-cell multi-omics data.

Integrating spatial transcriptomics, which maps gene expression location within tissues, with single-cell multi-omics data, profiling gene expression and chromatin accessibility (or other epigenomic data) for the same cell, offers powerful insights into gene regulation. However, commercially available kits for simultaneous spatial multi-omics profiling are currently unavailable, hindering widespread data generation. Here, we present ISON (Integrated Spatial Omics Network), a unified computational method for integrative spatial multi-omics analysis from single cell multiome data and spatial tra

Portable CRISPR/Cas12a-Based Aptasensor System for Point-of-Care Detection of Cytokine Tumor Necrosis Factor-Alpha (TNF-α).

Tumor necrosis factor-α (TNF-α) is an important inflammatory cytokine whose rapid and quantitative detection is essential for disease diagnosis and monitoring. However, conventional methods are often limited by bulky instrumentation and are not well suited for analysis in complex biological environments. Herein, a portable CRISPR/Cas12a-based aptasensor system was developed for detection of TNF-α by integrating aptamer recognition, Cas12a trans-cleavage signal amplification, and a miniaturized fluorescence detection device. An aptamer for TNF-α was designed to regulate the accessibility of the

Serum-compatible aptamer-regulated RCA-CRISPR/Cas12a assay for fluorescence and lateral flow detection of myeloperoxidase.

Myeloperoxidase (MPO), a neutrophil-derived heme peroxidase involved in oxidative inflammatory responses, has been investigated as a biomarker for cardiovascular, hematological, and other inflammation-associated diseases. In this study, an aptamer-regulated rolling circle amplification (RCA)-CRISPR/Cas12a fluorescence assay was established for MPO detection in serum samples. Two aptamers, Apt-02 and Apt-14, were obtained with dissociation constants (KD) of 57.76 and 56.88 nM, respectively, using MPO-depleted human serum as the counter-selection matrix during MB-SELEX. Apt-14 exhibited relative

Temperature-tunable anisotropic absorption for photo-modulated gas sensing.

This study presents a temperature-tunable and photo-modulated sensing platform based on a polyurethane-azobenzene-carbon black (PAC) composite that supports temperature-dependent narrow molecular absorption features. PAC films fabricated under UV and visible light curing exhibit distinct thermally responsive absorption features arising from strong light-matter coupling within the composite. UV curing promotes azobenzene trans-cis isomerization and molecular asymmetry, leading to narrow, temperature-dependent absorption features associated with azobenzene molecular transitions, whereas visible

Identification of the cellular transcription factor KLF16 as a novel repressive epigenetic repressor of HIV-1 transcription.

Despite antiretroviral therapy, human immunodeficiency virus type 1 (HIV-1) persists in latently-infected cells through epigenetic and transcriptional mechanisms. Latency-reversing agents have failed clinically, partly due to incomplete understanding of HIV-1 latency reversal. Here, using DNA-affinity capture and mass spectrometry on the HIV-1 5' long terminal repeat (5'LTR) enhancer-core promoter, we identify KLF16 (Krüppel-like Factor 16) as a novel regulator of HIV-1 gene expression. KLF16 binds to the HIV-1 5'LTR in vivo at Sp1 binding sites, and KLF16 depletion reactivates latent HIV-1 in

Emulsion-template oleogels: Formation mechanisms, structural regulation, and potential as solid fat substitutes.

Solid fats, including saturated and trans fats, provide desirable functionality, texture, and palatability to foods; however, they are associated with adverse health effects. Food industry is actively seeking alternative ingredients to replace solid fats while preserving the sensory and texture attributes crucial for consumer acceptance. The emulsion-template approach for gelling vegetable oils is a novel strategy to replace solid fats, offering advantages such as the use of safe and plentiful raw materials, mild processing conditions, a healthier fatty acid profile, controllable viscoelastici

Advances on protein-based particles as Pickering emulsion stabilizers: Preparation, stabilization mechanism, modification, and applications.

In recent years, many studies have focused on Pickering emulsions (PEs) stabilized by protein particles/nanoparticles (NPs) because of their excellent nutritional value, biodegradability, easy extraction, high surface activity, and modification capability. Despite growing interest in this area, the relationships among protein particle morphology, fabrication techniques, surface modification strategies, and stabilization performance have not always been discussed in an integrated manner, synthesizing the goal of this paper. First, a brief history of PEs is presented. Second, the article provide

Reconfigurable Supramolecular Hydrogel for Light-Triggered Pesticide Release via Reversible Gel-Sol-Gel Transition.

Conventional pesticide application often results in the premature loss of active ingredients through runoff, degradation, and nontarget dispersion, highlighting the need for delivery systems capable of externally regulated release. Herein, we report a proof-of-concept reconfigurable supramolecular hydrogel based on the self-assembly of cetyltrimethylammonium bromide (CTAB) and azobenzene-4,4'-dicarboxylic acid (ADA), enabling light-triggered release through a reversible gel-sol-gel transition. The hydrogel network was systematically tuned using composition and concentration, establishing well-

Development of a replication-restricted, esterase-deficient influenza D virus as a live vaccine candidate.

Influenza D virus (IDV) is an emerging bovine respiratory pathogen associated with bovine respiratory disease complex (BRDC), highlighting the need for a safe and effective live attenuated vaccine. In this study, we developed a replication-restricted IDV vaccine candidate by introducing a point mutation (S57Q) into the esterase active site of the hemagglutinin-esterase-fusion (HEF) protein, rendering the virus incapable of replication in conventional host cells. To enable efficient virus production, we established a swine testis (ST) cell line expressing wild-type HEF under a doxycycline-induc

Genome-wide association study of childhood B-cell acute lymphoblastic leukemia reveals novel African ancestry-specific susceptibility loci.

B-cell acute lymphoblastic leukemia (B-ALL) is the most common pediatric malignancy. Given racial/ethnic differences in incidence and outcomes, B-ALL genome-wide association studies among children of African ancestry are needed. Leveraging multi-institutional datasets with 840 African American children with B-ALL and 3360 controls, nine loci achieved genome-wide significance (P < 5 × 10-8) after meta-analysis. Two loci were established trans-ancestral susceptibility regions (IKZF1, ARID5B), while the remaining novel loci were specific to African populations. Five-year overall survival among ch

Low-Molecular-Weight Collagen Peptide Improves Skin Dehydration and Barrier Dysfunction in Human Dermal Fibrosis Cells and UVB-Exposed SKH-1 Hairless Mice.

Ultraviolet B (UVB), a component of solar ultraviolet light, is a major contributor to skin photodamage. UVB exposure primarily affects the epidermis, which leads to wrinkle formation, loss of skin elasticity, oxidative stress, and inflammation. Prolonged or intense UVB exposure can increase the risk of skin cancer. Collagen peptides are known as functional foods that improve skin dryness and wound healing. In this study, we aimed to investigate the protective and ameliorative effects of a low-molecular-weight collagen peptide (LMWCP) with a high absorption rate and photodamage. In vitro analy

A dual-mode RNA-splitting aptamer biosensor for sensitive HIV Tat peptide detection via colorimetry and fluorescence.

Early diagnosis of human immunodeficiency virus (HIV) is critical for effective treatment; however, traditional antibody methods encounter challenges during the infection window, and nucleic acid tests require specialized equipment. In this study, a dual-mode ribonucleic acid (RNA)-splitting aptamer biosensor was developed to target the HIV trans-activator of transcription (Tat) protein, a key HIV biomarker for viral replication throughout the infection cycle. The biosensor integrates colorimetric and fluorescent detection techniques by utilizing gold nanoparticles (AuNPs) and two types of apt

Molecular and structural basis for nitrosoglutathione-dependent redox regulation of triosephosphate isomerase from Chlamydomonas reinhardtii.

Protein S-nitrosylation is a reversible redox-based post-translational modification that plays an important role in cell signaling by modulating protein function and stability. At the molecular level, S-nitrosylation consists of the formation of a nitrosothiol (-SNO) and is primarily induced by the trans-nitrosylating agent nitrosoglutathione (GSNO). Triosephosphate isomerase (TPI), which catalyzes the interconversion of dihydroxyacetone phosphate and glyceraldehyde-3-phosphate, has been identified as a putative target of S-nitrosylation in both plant and non-plant systems. Here we investigate
