Construction and preclinical evaluation of an anti-CD19 chimeric antigen receptor.

T cells can be engineered to express the genes of chimeric antigen receptors (CARs) that recognize tumor-associated antigens. We constructed and compared 2 CARs that contained a single chain variable region moiety that recognized CD19. One CAR contained the signaling moiety of the 4-1BB molecule and the other did not. We selected the CAR that did not contain the 4-1BB moiety for further preclinical development. We demonstrated that gammaretroviruses encoding this receptor could transduce human T cells. Anti-CD19-CAR-transduced CD8+ and CD4+ T cells produced interferon-gamma and interleukin-2 s

Proteasome regulation of ULBP1 transcription.

Killer lymphocytes recognize stress-activated NKG2D ligands on tumors. We examined NKG2D ligand expression in head and neck squamous cell carcinoma (HNSCC) cells and other cell lines. HNSCC cells typically expressed MHC class I chain-related gene A (MICA), MICB, UL16-binding protein (ULBP)2, and ULBP3, but they were uniformly negative for cell surface ULBP1 and ULBP4. We then studied how cancer treatments affected NKG2D ligand expression. NKG2D ligand expression was not changed by most cancer-relevant treatments. However, bortezomib and other proteasome inhibitor drugs with distinct mechanisms

Interferon-beta expressed by a rabies virus-based HIV-1 vaccine vector serves as a molecular adjuvant and decreases pathogenicity.

Type I interferon is important in anti-viral responses and in coordinating the innate immune response. Here we explore the use of interferon-beta to adjuvant the response to a rabies virus (RV) vaccine vector expressing both HIV-1 Gag and IFN-beta. Viral load and immune responses of immunized mice were analyzed over time. Our results indicate that the RV expressing IFN-beta (IFN+) is highly attenuated when compared to control RV and demonstrate that the expression of IFN-beta reduces viral replication approximately 100-fold. Despite the decrease in replication, those mice immunized with the IF

Glycyrrhizin and its metabolite inhibit Smad3-mediated type I collagen gene transcription and suppress experimental murine liver fibrosis.

AIMS: Glycyrrhizin has been widely used for the treatment of chronic hepatitis C. It decreases the serum levels of aminotransferases, and suppresses progression of liver fibrosis as well as subsequent occurrence of hepatocellular carcinoma. Although previous studies have shown that glycyrrhizin and its metabolite inhibit collagen gene expression, its underlying mechanisms are virtually unknown. This study was aimed to explore molecular mechanisms responsible for the inhibitory effect of glycyrrhizin on type I collagen gene transcription. MAIN METHODS: Effects of glycyrrhizin and its metabolit

The orphan nuclear receptor NUR77 regulates hormone-induced StAR transcription in Leydig cells through cooperation with Ca2+/calmodulin-dependent protein kinase I.

Cholesterol transport in the mitochondrial membrane, an essential step of steroid biosynthesis, is mediated by a protein complex containing the steroidogenic acute regulatory (StAR) protein. The importance of this transporter is underscored by mutations in the human StAR gene that cause lipoid congenital adrenal hyperplasia, male pseudohermaphroditism, and adrenal insufficiency. StAR transcription in steroidogenic cells is hormonally regulated and involves several transcription factors. The nuclear receptor NUR77 is present in steroidogenic cells, and its expression is induced by hormones know

Disruption of ROBO2 is associated with urinary tract anomalies and confers risk of vesicoureteral reflux.

Congenital anomalies of the kidney and urinary tract (CAKUT) include vesicoureteral reflux (VUR). VUR is a complex, genetically heterogeneous developmental disorder characterized by the retrograde flow of urine from the bladder into the ureter and is associated with reflux nephropathy, the cause of 15% of end-stage renal disease in children and young adults. We investigated a man with a de novo translocation, 46,X,t(Y;3)(p11;p12)dn, who exhibits multiple congenital abnormalities, including severe bilateral VUR with ureterovesical junction defects. This translocation disrupts ROBO2, which encod

Melatonin, an endogenous-specific inhibitor of estrogen receptor alpha via calmodulin.

Melatonin is an indole hormone produced mainly by the pineal gland. We have previously demonstrated that melatonin interferes with estrogen (E(2)) signaling in MCF7 cells by impairing estrogen receptor (ER) pathways. Here we present the characterization of its mechanism of action showing that melatonin is a specific inhibitor of E(2)-induced ERalpha-mediated transcription in both estrogen response element- and AP1-containing promoters, whereas ERbeta-mediated transactivation is not inhibited or even activated at certain promoters. We show that the sensitivity of MCF-7 cells to melatonin depend

Decoding vectorial information from a gradient: sequential roles of the receptors Frizzled and Notch in establishing planar polarity in the Drosophila eye.

The Drosophila eye is composed of several hundred ommatidia that can exist in either of two chiral forms, depending on position: ommatidia in the dorsal half of the eye adopt one chiral form, whereas ommatidia in the ventral half adopt the other. Chirality appears to be specified by a polarizing signal with a high activity at the interface between the two halves (the 'equator'), which declines in opposite directions towards the dorsal and ventral poles. Here, using genetic mosaics, we show that this polarizing signal is decoded by the sequential use of two receptor systems. The first depends o

The promoter of the plant defensin gene PDF1.2 from Arabidopsis is systemically activated by fungal pathogens and responds to methyl jasmonate but not to salicylic acid.

The plant defensin PDF1.2 has previously been shown to accumulate systemically via a salicylic acid-independent pathway in leaves of Arabidopsis upon challenge by fungal pathogens. To further investigate the signalling and transcriptional processes underlying plant defensin induction, a DNA fragment containing 1184 bp and 1232 bp upstream of the transcriptional and translational start sites, respectively, was cloned by inverse PCR. To test for promoter activity this DNA fragment was linked to the beta-glucuronidase (GUS)-encoding region of the UidA gene as a translational fusion and introduced

UV-induced DNA damage is an intermediate step in UV-induced expression of human immunodeficiency virus type 1, collagenase, c-fos, and metallothionein.

UV irradiation of human and murine cells enhances the transcription of several genes. Here we report on the primary target of relevant UV absorption, on pathways leading to gene activation, and on the elements receiving the UV-induced signal in the human immunodeficiency virus type 1 (HIV-1) long terminal repeat, in the gene coding for collagenase, and in the cellular oncogene fos. In order to induce the expression of genes. UV radiation needs to be absorbed by DNA and to cause DNA damage of the kind that cannot be repaired by cells from patients with xeroderma pigmentosum group A. UV-induced

CHD1L promotes testicular cancer progression through epigenetic activation of the CXCR6/PI3K/AKT pathway.

Testicular cancer is the most common malignancy in young men, and treatment options for metastatic or refractory disease remain limited. Chromatin remodeling plays a critical role in tumorigenesis, yet its contribution to testicular cancer progression is poorly understood. This study aimed to elucidate the oncogenic and epigenetic functions of chromodomain helicase DNA-binding protein 1-like (CHD1L) in testicular cancer. Immunohistochemistry and TCGA transcriptomic analyses were performed to assess CHD1L expression and clinical relevance. Functional assays, RNA sequencing, and ATAC-seq were co

SLC11A1 Drives Ferroptosis in Acute Myocardial Infarction Via Sp1-mediated Transcriptional Repression of GPX4.

Ferroptosis is a critical contributor to cardiomyocyte injury in acute myocardial infarction (AMI). This study aimed to identify key ferroptosis-associated genes and elucidate their roles in AMI. Transcriptomic datasets from AMI patients and healthy controls were analyzed to identify differentially expressed genes (DEGs). The role of the solute carrier family 11 member 1 (SLC11A1) was validated in vitro using a hypoxia/reoxygenation (H/R) model in H9c2 cardiomyocytes and in vivo using a murine AMI model. SLC11A1 expression was elevated in H/R-treated cardiomyocytes and infarcted murine hearts.

MiR-874-3p suppresses TNF-α-induced inflammation in adipocytes by targeting nucleolin.

Many investigations have indicated the significance of microRNAs (miRNAs) as potential biomarkers for early obesity in children. MiR-874-3p was revealed to be downregulated in overweight/obese children. However, the specific function and mechanism of miR-874-3p in the progression of childhood obesity remain unclear. Human Simpson-Golabi-Behmel syndrome (SGBS) adipocytes were stimulated with tumor necrosis factor α (TNF-α) to establish an in vitro cell model. CCK-8 assay and ELISA were used to assess cell viability and proinflammatory cytokine secretion, respectively. RT-qPCR was used for miR-8

Gene-Specific Endothelial Programs Drive AVM Pathogenesis in SMAD4 and ALK1 Loss-of-Function.

BACKGROUND: Hereditary hemorrhagic telangiectasia is a genetic disorder caused by loss-of-function mutations in components of the bone morphogenetic protein signaling pathway, leading to arteriovenous malformations. Most prior work has treated BMP (bone morphogenetic protein) component depletion as mechanistically interchangeable, yet whether distinct genes converge on a shared mechanism remains unclear. We aimed to understand the molecular relationship between BMP signaling and endothelial flow response that leads to arteriovenous malformation formation. METHODS: We expose human endothelial

Organ-specific transcriptional programs regulate cellular plasticity in lymphatic endothelial cells.

Lymphatic vessels play essential roles in maintaining fluid homeostasis and responding to inflammatory and tissue environmental changes. During these biological processes, lymphatic endothelial cells (LECs) lining lymphatic vessels adapt to external stimuli in various organs and alter their characteristics. While organ-specific molecular and functional heterogeneity has been well-characterized in blood vascular endothelial cells, the extent and functional significance of such heterogeneity in LECs remain poorly understood. In this study, we established LECs derived from the heart and lung tiss

Hypermethylation-mediated HNF4A silencing by Helicobacter pylori infection drives gastric cancer by disrupting epithelial cell polarity and activating EMT signaling.

Helicobacter pylori (Hp.) infection is one of the high-risk factors for gastric carcinogenesis (GC). However, the underlying mechanism remains largely unclear. In this study, we uncover an essential role of Hp. infection in mediating tumor suppressor gene silencing in gastric epithelial cells through promoter DNA hypermethylation. Hepatocyte nuclear factor HNF4A was downregulated in GC and predicted poor survival. The in vitro and in vivo assays together confirmed that HNF4A plays a tumor suppressive role in GC. Single-cell analysis showed that HNF4A was selectively expressed in gastric epithe

DNMT3a contributes to bone cancer pain by epigenetic silencing of Kcnq2/Kcnq3 in dorsal root ganglion neurons.

Metastatic bone tumors induce transcriptional changes in dorsal root ganglion (DRG) neurons, which may contribute to bone cancer pain. A key factor in this process is the downregulation of Kv7 (KCNQ)/M channels in DRG neurons, leading to neuronal hyperexcitability and pain hypersensitivity. However, the mechanisms underlying bone cancer-induced Kv7 channels suppression remain poorly understood. DNA methyltransferase (DNMT)-mediated DNA methylation is known to suppress gene expression. In this study, we demonstrate that DNMT3a plays a critical role in the genesis of bone cancer pain, likely thr

Mendelian randomization reveals the causal links between miRNAs and rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent joint inflammation and progressive structural damage, with early diagnosis remaining a significant clinical challenge. Circulating microRNAs (miRNAs) have emerged as promising biomarkers for disease diagnosis, prognosis, and therapeutic response due to their critical roles in gene regulation. However, the specific miRNAs causally involved in RA pathogenesis remain largely unidentified. We conducted a 2-sample Mendelian randomization (MR) analysis using summary-level data from the largest available genome-wide
