HIF-1α impairs NK cell differentiation-maturation and cytotoxicity in myelodysplastic syndrome via JAK1/STAT5/SOCS2 pathway.

Myelodysplastic syndrome (MDS) is a heterogeneous group of myeloid neoplasms characterized by treatment difficulties and a propensity to progress to acute myeloid leukemia. Impaired natural killer (NK) cell surveillance is a hallmark of MDS, yet the underlying molecular mechanisms remain poorly understood. This study aims to elucidate the mechanism by which HIF-1α regulates NK cell differentiation disorders and its impact on NK cell cytotoxicity in MDS. Flow cytometry was employed to compare HIF-1α expression and NK cell differentiation between wild-type and NUP98/HOXD13 (NHD13) mice. Our resu

A novel monocyte-derived antigen presenting cell-T regulatory cell axis contributes to skin wound healing and is impaired in diabetic mice.

Despite a vast literature on the role of macrophages in wound healing, the role of dermal monocyte (Mo)-derived antigen presenting cells (APC) has received scant attention. Using scRNAseq and flow cytometry, we identify a population of APC that is prominent in wounds of non-diabetic mice but is reduced in wounds of diabetic mice. Using adoptive transfer experiments and Ccr2 knockout mice, we demonstrate that wound APC are derived primarily from Mo and that the diabetic wound environment inhibits differentiation of Mo into APC. We also show that Mo-specific Irf4 knockout mice exhibit reduced di

PI3Kδ inhibition alters CD8 T cell differentiation and reprograms the tumor microenvironment following adoptive immunotherapy.

T cell exhaustion remains a significant barrier to effective adoptive cell therapy in solid tumors. Here, we demonstrate that in vitro treatment with the PI3Kδ inhibitor CAL-101 generates T cells with enhanced stemness and metabolic fitness. These cells show increased mitochondrial dependence and spare respiratory capacity while maintaining normal basal metabolism. Under chronic antigen stimulation, CAL-101-treated T cells, including human T cells, resist terminal exhaustion and maintain stem-like properties. Using single-cell RNA sequencing and spatial transcriptomics of B16 melanoma tumors,

Increased ULBP1 by doxorubicin sensitizes neuroblastoma to γδT-cell cytotoxicity.

Neuroblastoma (NB) is the most common extracranial solid tumor in children and remains a major therapeutic challenge in high-risk patients because of frequent recurrence and metastasis. γδT cells are a unique lymphocyte subset with potent antitumor activity and are widely distributed in NB tumors. However, the immunosuppressive tumor microenvironment limits γδT-cell effector function and promotes immune evasion. In this study, we found that ULBP1 expression was frequently down-regulated in NB tumor tissues and cell lines and that low ULBP1 expression was associated with high-risk clinical feat

Distinct immune trajectories after severe trauma identify a Th17-biased immunophenotype in chronic critical illness.

Severe trauma triggers a dynamic disturbance of immune function that can progress from early hyperinflammation to prolonged dysregulation, increasing patient vulnerability to infection and chronic critical illness (CCI). To identify immune features linked to different postinjury clinical trajectories, we prospectively evaluated severely injured trauma patients and categorized them as rapid recovery, intermediate, or CCI based on clinical outcome by day 14 of intensive care unit admission. CCI patients developed infections earlier and more frequently than rapid recovery or intermediate patients

MHC class II on melanoma cells regulates the anti-tumor T cell response.

In human melanoma, tumor cell-specific MHC class II expression is associated with improved response to programmed death 1 and programmed death-ligand 1 blockade; yet the direct effect of melanoma cell-specific MHC class II expression on anti-melanoma T cell responses remains largely unknown. In the clinically relevant Yale University Mouse Melanoma Exposed to Radiation (YUMMER) cell lines, MHC class II expression was IFN-γ-inducible in a subset of cells. Transduction of melanoma cells with CIITA resulted in uniformly high, constitutive expression of MHC class II. YUMMER.G cells transduced with

The scaffolding protein AKAP79/150 shapes innate immune responses to allergen.

Inhalation of house dust mite-derived allergens is a major factor leading to the development of asthma both in children and adults. Allergens stimulate Ca2+-dependent pro-inflammatory cytokine and chemokine gene expression through the opening of ORAI1 Ca2+ channels in the surface membrane of various immune cell-types within the lung. ORAI1 participates in a signalosome with the scaffolding protein A-kinase anchoring protein (AKAP)79/150 (encoded by the Akap5 gene), which binds numerous protein kinases, calcineurin and transcription factors required for cytokine expression. However, the role of

Long-term survivors of melanoma after anti-PD-1 therapy demonstrate on-treatment increase in abundance of 4-1BB+ CD4 T cells in blood.

Anti-PD-1 monotherapy has shown a significant survival benefit for patients with advanced-stage melanoma. In this study, we explored whether long-term clinical benefit is determined by abundance or changes in such abundances of CD4 and CD8 T-cell subsets in blood. Peripheral blood mononuclear cells from 58 patients with advanced-stage melanoma treated with anti-PD-1 monotherapy were collected prior to first, second, and third administration. Overall survival (OS) was categorized into 3 groups: <1 yr (short term); 1-3 yr; and >3 yr (long term). The abundance of different T-cell subsets was dete

Getting on your last nerve: IFNs and resistance to infection.

Interferons (IFNs) affect almost all nucleated cells and induce transcriptional events that are associated with control of intracellular infections as well as regulatory pathways that influence inflammatory processes. How different hematopoietic and non-hematopoietic cell types interpret these signals is a reflection of cellular context and function. For example, neurons are often viewed as immunologically inert and have muted responses to IFNs. However, because neurons can be targeted by several classes of pathogens, there is a need for these cells to sense infection and respond to IFNs to en

The transcription factor SREBP2 supports the activation and homeostasis of CD4+ Foxp3+ regulatory T cells.

Periodic activation of regulatory T cells (Tregs) is necessary to restrict autoreactive T cells and maintain immune homeostasis. IL-2-STAT5 signaling instructs several metabolic processes underlying Treg activation, including the mevalonate pathway, which is crucial for optimal Treg responses. However, direct links between IL-2-STAT5, mevalonate, and Treg function have yet to be established. To explore this relationship, we deleted sterol regulatory binding element protein 2 (Srebp2), a critical mediator of mevalonate metabolism, selectively in Tregs. Here, we show that when Srebp2-deficient T

Fluorescence tracking CD8 T cells from tumor-draining lymph nodes to irradiated tumors and their activation in the tumor environment.

Radiation is an effective site-specific cytotoxic therapy for cancer cells. In preclinical models, CD8 T cells are essential for optimum tumor control by radiation therapy, yet CD8 T cells are radiosensitive and depleted within the treatment field. Irradiated tumors can be refilled with T cells that were outside the field at the time of treatment. Using the Kaede photoconvertible mouse model we tracked CD8 T cells that originated in the tumor-draining lymph node and entered the tumor following radiation therapy. We show that radiation induces CD69 expression in newly infiltrating CD8 T cells w

FTR85 negatively regulates type I IFN antiviral signaling pathway by promoting K48-linked polyubiquitination of IRF3.

The finTRIM (FTR) subfamily, a group of fish-specific tripartite motif proteins, has arisen through gene duplication events specific to particular genera or species. However, the regulatory mechanisms of FTR in antiviral immune response remains largely unknown. In the present study, we identified a fish novel tripartite motif member (finTRIM, FTR) in common carp (Cyprinus carpio L.) and named it CcFTR85, which contains a RING (really interesting new gene) domain, a coiled-coil region, and a PRY/SPRY domain. Its expression can be induced following spring viremia of carp virus (SVCV) infection a

CD153 promotes B-cell responses to immunization in aged mice.

We and others have described homeostatic dysregulation of the CD4+ memory T-cell compartment with age. To gain greater insights into this dysregulation, we performed comprehensive single-cell genomic analysis of endogenous memory CD4+ T cells from young and aged mice. This analysis revealed 16 populations, composed of Th1, Th17, several subsets of regulatory T (Treg) cells, memory T cells, CD4+ CTLs, and T follicular helper (Tfh) cells. One of the most highly expressed genes in aged Tfh cells was Tnfsf8 (CD153 or CD30L), and flow cytometric analysis confirmed age-increased expression of CD153

Kaempferol's potential in targeting IL-17-mediated inflammatory pathways for colorectal cancer treatment.

Colorectal cancer (CRC) is a leading malignancy with limited treatment options at advanced stages. Interleukin (IL)-17 signaling promotes tumor progression and immune evasion. Kaempferol, a natural flavonol, has anti-inflammatory and antitumor effects, but its role in modulating IL-17 pathways in CRC is unclear. We used an immunocompetent MC38 xenograft model to evaluate kaempferol monotherapy (50/100 mg/kg), its combination with a PD-L1 inhibitor, and IL-17 overexpression. Tumor growth, immune infiltration, cytokines, and signaling pathways were assessed using immunohistochemistry, enzyme-lin

The aryl hydrocarbon receptor promotes the resolution of pulmonary neutrophilia via regulation of macrophage efferocytosis.

Cigarette smoke is the primary cause of chronic obstructive pulmonary disease (COPD), an incurable condition characterized by irreversible airflow obstruction and alveolar destruction driven by chronic inflammation of the lungs and airways. The inflammatory response caused by cigarette smoke is typified by the recruitment of innate and adaptive immune cells to the lung. Paradoxically, many of these immune cells are functionally impaired by smoke. Notable among these are lung macrophages, which have reduced ability to clear apoptotic lung epithelial cells and neutrophils by efferocytosis when e

Binding of death receptor 3 (DR3) to intracellular TRADD and TRAF2 is independent of its natural ligand, TL1A.

The TNF‑like cytokine TL1A is a key regulator of immune responses through engagement of its receptor, death receptor 3 (DR3), and has emerged as a therapeutic target in inflammatory diseases. Prevailing models propose that TL1A binding initiates recruitment of adaptor proteins to DR3, leading to downstream signaling. Here, we investigated proximal DR3 signaling events and found that the adaptor proteins TRADD and TRAF2 associate with DR3 independently of TL1A stimulation in both TF‑1 cells and primary human CD4+ T cells. In contrast, TL1A stimulation was strictly required for activation of NF‑

CD8+ T-cell effector programming is epigenetically regulated by DUSP2 through BAF complex-driven chromatin remodeling.

Effector gene expression in CD8+ T cells is tightly controlled at the epigenetic and transcriptional levels; however, crucial regulatory signals remain incompletely understood. In this study, we identify dual specificity phosphatase 2 (DUSP2) as a key epigenetic modulator of effector CD8+ T-cell state, by elucidating inhibition of BAF-mediated chromatin opening by DUSP2. Induced downregulation of DUSP2 in human CD8+ T cells enhances cytotoxicity and effector phenotypes by increasing chromatin accessibility through upregulation of BAF subunits, such as SMARCA4, and permitting binding of ETS1 to

Extracellular vesicles from Candidozyma (Candida) auris inhibit proliferation of CD4 T cells by disrupting the IL-2 axis.

Candidozyma auris (formerly Candida auris) is an emerging multidrug-resistant pathogenic fungus that has rapidly spread across the world. Due to the high frequency of multidrug-resistant strains and mortality rate, C. auris is considered a critical health threat by the Centers for Disease Control and Prevention and the World Health Organization. Like other pathogens, C. auris employs virulence factors that are delivered by extracellular vesicles (EVs). We have shown that EVs from C. auris (CauEVs) activate murine phagocytes, boosting innate immune mechanisms. However, the effect of fungal EVs

Innate recognition and phagocytosis of zymosan by human neutrophils require cell priming in a p38-dependent manner.

Neutrophils are the first line of defense against fungal infection and other microbial pathogens. Through alternative pathway of complement activation in innate immunity, complement fragments (opsonin) generated promote phagocytosis of the yeast forms of fungal pathogens such as Candida albicans. However, when complement levels are limited, through consumption in disease and inflammation or where microbial pathogens employ mechanisms to degrade or inhibit complement components, phagocytosis may still proceed via innate receptors, in the absence of effective opsonisation. Mechanisms of innate p

Enforcing mTORC1 activity in therapeutic CD4+ T cells promotes persistence but eventual immune exhaustion.

There is substantial interest in developing novel engineering strategies to promote the sustained metabolic fitness of therapeutic T cells. We previously showed that overexpression of RAS homologue enriched in brain (RHEB), a positive regulator of mammalian target of rapamycin complex 1 (mTORC1), promotes aerobic glycolysis and increases the anti-tumor functions of effector CD8+ T cells. To address whether these effects are conserved in CD4+ T cells, we have now examined how enforced activation of mTORC1 activity affects CD4+ T cell differentiation and function. Rheb overexpression induced a m
