Methodologic considerations for investigating the diet-cancer link.

Evidence from human ecological studies and experimental animal studies suggest that a number of dietary factors may have a role in the etiology of cancers of various sites. When associations are examined within populations on the level of the individual, they often weaken or disappear. Although in some cases the suspect nutrient may have no real carcinogenic effect, it is proposed that there are at least three important methodologic problems that could prevent the observation of a true association between dietary factors and human cancer. First, diet assessment methods are inadequate to estima

Characterization of a human ovarian adenocarcinoma line, IGROV1, in tissue culture and in nude mice.

A cell line, IGROV1, originating from an ovarian carcinoma of a 47-year-old woman was established in tissue culture and in nude mice. Maintained in monolayer cultures, IGROV1 cells exhibited a 20-h doubling time and highly tumorigenic properties. The s.c. injection of 2 X 10(6) cultured cells into nude mice gave rise to fast growing tumors, while the i.p. route induced a peritoneal carcinomatosis with ascites which killed the animals in 2 months. The epithelial morphology of IGROV1 cells was retained during in vitro and in vivo passages, as judged by both the light and the electron microscopes

Parasympathetic cardiovascular control in human disease: a critical review of methods and results.

A variety of experimental approaches have been used to alter reflex parasympathetic cardiovascular control in normal human subjects and in patients with cardiovascular diseases. This review critically appraises several of these methods (the Valsalva maneuver, upright tilt, lower body negative pressure, pressor drug infusion, neck suction, and coronary arteriography) and summarizes the normal response patterns of healthy subjects and the abnormal response patterns of some patients with cardiovascular disease. Tests of parasympathetic function tend to be heterogeneous; they are not standardized,

Factors affecting sperm motility. III. Influence of visible light and other electromagnetic radiations on human sperm velocity and survival.

Specimens of semen from fertile and infertile patients were exposed to different electromagnetic radiations, including visible light, ultraviolet (UV) light, x-rays, and high-frequency radio waves. Sperm motility was analyzed before, during, and after irradiation by the multiple exposure photography (MEP) method. No significant difference was found between controls and specimens exposed to various doses of visible and UV light and x-rays either immediately or several hours after exposure. In contrast to spermatozoa of other species that were reported to be adversely affected by visible and UV

Kinetics of iron hexacyanide release from the ferric hemes of partially oxidized intermediates of human deoxy hemoglobin.

A new experiment is presented in which the kinetics of iron hexacyanide release is measured from partially oxidized intermediates of human deoxy hemoglobin (T state). The intermediates were generated by briefly exposing a solution of hemoglobin and dithionite to various concentrations of ferricyanide at 6 degrees C in 0.1 M phosphate buffer, pH 7. The reaction of the residual dithionite with the partially oxidized intermediates was limited by the release of ferrocyanide and was strongly biphasic with about 75% fast phase and 25% slow and with the two phases differing in rate by a factor of 26.

Human spongiform encephalopathies in marmoset monkeys (Saguinus sp.).

Brain homogenates (10% w/v) from five of seven kuru patients inoculated intracerebrally (i.c., 0.1 ml) into marmosets (Saguinus sp.) induced a rapidly progressive CNS disease 26, 31, 36, 76 and 94 months postinoculation. Serial marmoset passages of kuru were accomplished by i.c. inoculation of neonatal marmosets with brain homogenates from marmosets with experimentally induced kuru. The incubation periods ranged from 1.5 to 11 months (average 7.3) in 19 animals, 20-25 months in four animals and greater than 26 months in two animals. Symptoms and brain lesions of the induced disease were compat

[Pharmacological studies on the cough reflex].

The artificially induced cough reflex in experimental animals should necessarily be similar to the cough which occurs naturally in humans. The curve demonstrated during a cough of each patient with respiratory disease was characteristic. Compared with the curve of a cough-like reflex (CLR) induced by DMPP, the curve of a cough induced by electrical stimulation of the tracheal mucosa was similar to that of a cough in humans. In dogs and cats anesthetized with alpha-chloralose (80 similar to 100 mg/kg i.p.), the number of CLR with DMPP was dose dependent. Tachphylaxis did not occur despite repea

Dose optimization of inhaled tigecycline in humans to overcome inherent adverse events and maximize bacterial clearance using a physiologically-based pharmacokinetic modeling approach.

INTRODUCTION: Intrapulmonary delivery of tigecycline has been highlighted as an optimal strategy for enhancing local drug concentrations at the site of infection in the treatment of Mycobacterium abscessus pulmonary infections. Therefore, determining an appropriate inhaled dose is imperative to optimize therapeutic use of tigecycline. In this study, we aimed at establishing a human dose rationale for inhaled tigecycline by leveraging various preclinical experimental datasets through physiologically-based pharmacokinetic (PBPK) modeling. METHODS: The PBPK model developed to predict plasma and

Limited dissemination and shedding of the UL128 complex-intact, UL/b'-defective rhesus cytomegalovirus strain 180.92.

UNLABELLED: The UL128 complex of human cytomegalovirus (CMV) is a major determinant of viral entry into epithelial and endothelial cells and a target for vaccine development. The UL/b' region of rhesus CMV contains several open reading frames, including orthologs of the UL128 complex. We recently showed that the coding content of the rhesus CMV (RhCMV) UL/b' region predicts acute endothelial tropism and long-term shedding in vivo in the rhesus macaque model of CMV infection. The laboratory-passaged RhCMV 180.92 strain has a truncated UL/b' region but an intact UL128 complex. To investigate whe

Assist-as-Needed Hip Exoskeleton Control for Gait Asymmetry Correction via Human-in-the-Loop Optimization.

Gait asymmetry is a significant clinical characteristic of hemiplegic gait that most stroke survivors suffer, leading to limited mobility and long-term negative impacts on their quality of life. Although a variety of exoskeleton controls have been developed for robot-aided gait rehabilitation, little attention has been paid to correcting the gait asymmetry of stroke patients, and it remains challenging to properly share control between the exoskeleton and patients with partial motor control. In view of this, an assist-as-needed (AAN) hip exoskeleton control with human-in-the-loop optimization

Modality-specific reactive and predictive responses in the human superior colliculus.

Inspired by recent theoretical work emphasizing predictive neural processing, we tested whether a midbrain structure for multimodal sensory integration-the superior colliculus-would show predictive responses to both visual and somatosensory stimulation. A multi-stage experimental design allowed us to test predictive activity in the human superior colliculus while 7-Tesla functional magnetic resonance imaging (fMRI) allowed us to map its sensory response across superior colliculus layers (N = 80). As expected from non-human animal work, visual and somatosensory stimulation elicited distinct dep

Molecular pathogenesis, global epidemiological trends, and treatment strategies for pteropine orthoreoviruses: a narrative review.

Pteropine orthoreoviruses are emerging bat-borne zoonotic viruses of the genus Orthoreovirus (family Reoviridae), increasingly recognized as causes of acute respiratory disease in humans. Originally grouped with the largely non-pathogenic mammalian orthoreoviruses, they have challenged that view through their association with severe influenza-like illness, evidence of human-to-human transmission, and a broad geographic range across the Old World. Maintained primarily in fruit bats of the family Pteropodidae, they are now linked to neurological as well as respiratory disease. This narrative rev

Swarm-initialized adaptive controller with beetle antenna searching of wearable lower limb exoskeleton for sit-to-stand and walking motions.

In recent years, exoskeleton robots have attracted great interest from researchers in the area of robotics due to their ability to assist human functionality improvement. A wearable lower limb exoskeleton is aimed at supporting the limb functionality rehabilitation process and to assist physical therapists. Development of a stable and robust control system for multi-joint rehabilitation robots is a challenging task due to their non-linear dynamic systems. This paper presents the development of a Swarm-Initialized Adaptive (SIA) based controller, which is a combination of a swarm-based intellig

Quantification of mechanical-cytoskeletal coupling in human corneal cells across myopia severity.

Myopia is a prevalent refractive eye disorder closely associated with alterations in corneal biomechanical properties. As fundamental units of corneal tissue, corneal cells significantly influence myopia progression through their nanomechanical characteristics. However, the biophysical mechanisms underlying this process, particularly in human corneal cells, remain unclear. This study investigates the coupling between mechanical properties and cytoskeletal morphology in human corneal cells across varying myopia severity levels. Utilizing atomic force microscopy (AFM), the Young's modulus and ad

Health impacts of microplastic and nanoplastic exposure.

As evidence demonstrating the presence of micro- and nanoplastics (MNPs) in the human body accumulates, so do concerns about their potential health impacts. Multiple factors determine the properties and behavior of MNPs-including polymer type, size, shape and the presence of a biocorona, among others-which place high demands on analytical methodology and tools for assessment of potential adverse health effects. Experimental models have shown that MNPs can cross cell barriers in the human lung and intestine and reach systemic circulation and subsequently tissues such as reproductive organs, pla

Multiomic Landscape of Primary Hypothyroidism Induced by Subchronic Exposure to Low-Dose Novel PFOS Substitute OBS in Human and Murine Models.

Sodium p-perfluorous nonenoxybenzenesulfonate (OBS) as a novel surrogate for perfluorooctanesulfonate (PFOS) has been extensively utilized in industrial manufacturing and daily life. However, studies on OBS-induced environmental health risks of obstructive biosynthesis (OBS) are currently limited, particularly the risk for thyroid diseases. Following the construction of in vivo (mouse) and in vitro (normal human primary thyrocytes) models of subchronic low-dose OBS exposure, we explored the thyroid-disrupting effects of OBS through multiomics approaches and experimental validations. Our result

Engineering a Multilayered Skin Equivalent: The Importance of Endogenous Extracellular Matrix and Model Flexibility for Nuanced and Accurate Skin Studies.

Human skin equivalents (HSEs) are a valuable tool for both academic and industrial laboratories to further the understanding of skin physiology in a range of health and disease situations. There have been many advances in the development of HSEs that successfully recapitulate the structure of human skin in vitro; however a main limitation is variability due to the use of complex protocols and exogenous extracellular matrix (ECM) proteins. Previously, we have developed a robust, reproducible, and unique full thickness HSE, using a consistent scaffold, commercially available cells and defined lo
