Brain function and network features in patients with chronic epilepsy before and after antiseizure medication withdrawal.

OBJECTIVES: A considerable proportion of epilepsy patients who achieved long-term seizure freedom with standardized treatment of antiseizure medication will attempt to withdraw medications. Epilepsy is currently considered as a network disease, however, the characteristics of brain function and neural network before and after medication withdrawal remain to be discovered. METHODS: Resting-state functional magnetic resonance imaging was obtained for 32 healthy controls, 32 seizure-free patients initiating medication tapering (PG1 group), and 16 seizure-free patients that had completely discont

Neural Correlates of Verb Fluency Performance in Cognitively Healthy Older Adults and Individuals With Dementia: A Pilot fMRI Study.

Currently there are ~6 million Americans who are affected by dementia. Verbal fluency tasks have been commonly and frequently utilized to document the disease progression in many forms of dementia. Verb fluency has been found to display substantial potential to detect and monitor the cognitive declines of individuals with dementia who have fronto-striatal involvement. The neural substrates underlying verb fluency task performance, however, have remained unclear so far, especially in individuals with dementia. Therefore, in the current study, brain activation patterns of seven individuals with

Amyloid precursor protein intracellular domain-dependent regulation of FOXO3a inhibits adult hippocampal neurogenesis.

The amyloid precursor protein (APP) intracellular domain (AICD) is a metabolic by-product of APP produced through sequential proteolytic cleavage by α-, β-, and γ-secretases. The interaction between AICD and Fe65 has been reported to impair adult neurogenesis in vivo. However, the exact role of AICD in mediating neural stem cell fate remains unclear. To identify the role of AICD in neuronal proliferation and differentiation, as well as to clarify the molecular mechanisms underlying the role of AICD in neurogenesis, we first generated a mouse model expressing the Rosa26-based AICD transgene. Atypical Dorsolateral Prefrontal Activity in Female Adolescents With Conduct Disorder During Effortful Emotion Regulation.

BACKGROUND: Conduct disorder (CD), which is characterized by severe aggressive and antisocial behavior, is linked to emotion processing and regulation deficits. However, the neural correlates of emotion regulation are yet to be investigated in adolescents with CD. Furthermore, it remains unclear whether CD is associated with deficits in emotional reactivity, emotion regulation, or both. METHODS: We used functional magnetic resonance imaging to study effortful emotion regulation by cognitive reappraisal in 59 female adolescents 15 to 18 years of age (30 with a CD diagnosis and 29 typically dev

An exploration of neural dynamics of motor imagery for people with amyotrophic lateral sclerosis.

OBJECTIVE: Studies of the neuropathological effects of amyotrophic lateral sclerosis (ALS) on the underlying motor system have investigated abnormalities in the magnitude and timing of the event-related desynchronization (ERD) and synchronization (ERS) during motor execution (ME). However, the spatio-spectral-temporal dynamics of these sensorimotor oscillations during motor imagery (MI) have not been fully explored for these patients. This study explores the neural dynamics of sensorimotor oscillations for ALS patients during MI by quantifying ERD/ERS features in frequency, time, and space. A

Effects of Different Light Sources on Neural Activity of the Paraventricular Nucleus in the Hypothalamus.

Background and Objectives: Physical function is influenced by light irradiation, and interest in the influence of light irradiation on health is high. Light signals are transmitted from the retina to the suprachiasmatic nucleus (SCN) via the retinal hypothalamic tract as non-image vision. Additionally, the SCN projects a nerve to the paraventricular nucleus (PVN) which acts as a stress center. This study examined the influences of three different light sources on neural activity in the PVN region using two different color temperatures. Materials and Methods: Experiments were conducted using tw

In Utero Exposure to Valproic Acid Induces Neocortical Dysgenesis via Dysregulation of Neural Progenitor Cell Proliferation/Differentiation.

UNLABELLED: Valproic acid (VPA), a widely used antiepileptic drug, is an inhibitor of histone deacetylases, which epigenetically modify cell proliferation/differentiation in developing tissues. A series of recent clinical studies in humans reported that VPA exposure in utero impaired histogenesis and the development of the central nervous system, leading to increased risks of congenital malformation and the impairment of higher brain functions in children. In the present study conducted in mice, we report that VPA exposure in utero (1) increases the amount of acetylated histone proteins, (2) a

Multimodal Navigation in Endoscopic Transsphenoidal Resection of Pituitary Tumors Using Image-Based Vascular and Cranial Nerve Segmentation: A Prospective Validation Study.

BACKGROUND: Transsphenoidal surgery (TSS) is the most common approach for the treatment of pituitary tumors. However, misdirection, vascular damage, intraoperative cerebrospinal fluid leakage, and optic nerve injuries are all well-known complications, and the risk of adverse events is more likely in less-experienced hands. This prospective study was conducted to validate the accuracy of image-based segmentation coupled with neuronavigation in localizing neurovascular structures during TSS. METHODS: Twenty-five patients with a pituitary tumor underwent preoperative 3-T magnetic resonance imagi

Cortical hubs and subcortical cholinergic pathways as neural substrates of poststroke dementia.

BACKGROUND AND PURPOSE: A role of neural networks in the development of poststroke dementia has not been clearly established. We hypothesized that stroke-mediated disruption of subcortical cholinergic pathway or large-scale neural networks contributes to poststroke dementia. METHODS: A matched case-control study was conducted in a predetermined cohort with acute ischemic stroke. Cases were defined as newly developed dementia diagnosed >3 months after stroke using the Korean Vascular Cognitive Impairment Harmonization Standards. Each case was matched to 2 controls for age, education, and initi

Neural origin of evoked potentials during thalamic deep brain stimulation.

Closed-loop deep brain stimulation (DBS) systems could provide automatic adjustment of stimulation parameters and improve outcomes in the treatment of Parkinson's disease and essential tremor. The evoked compound action potential (ECAP), generated by activated neurons near the DBS electrode, may provide a suitable feedback control signal for closed-loop DBS. The objectives of this work were to characterize the ECAP across stimulation parameters and determine the neural elements contributing to the signal. We recorded ECAPs during thalamic DBS in anesthetized cats and conducted computer simulat

Therapeutic window of hyperbaric oxygen therapy for hypoxic-ischemic brain damage in newborn rats.

Previous studies showed that hyperbaric oxygen (HBO) promoted cell proliferation in hypoxic-ischemic (HI) neonate rats. Neural stem cells (NSC) existed in the brain lifelong and can be activated. This study was undertaken to assess whether HBO treatment promoted the proliferation of NSC and repaired the brain damage regardless of when it is started, thus to explore the therapeutic window of HBO treatment. Seven-day-old Sprague-Dawley rats underwent left carotid ligation followed by 2 h of hypoxic stress (8% O(2) at 37 degrees C). Hyperbaric oxygen therapy was administered 3, 6, 12, 24, and 72

The requirement of pax6 for postnatal eye development: evidence from experimental mouse chimeras.

PURPOSE: The small eye mouse mutant (Sey) is caused by a mutation of the Pax6 gene. Previous studies, in which aggregation chimeras were used, have demonstrated that Sey/Sey cells contribute poorly to the neural retina forming small clumps of cells restricted to the inner retina at embryonic day 16.5. In addition, Sey/+ cells are absent from the lens epithelium during this embryonic period and postnatally. This study was conducted to determine the fates of these Sey/Sey and Sey/+ cells with continued development in chimeric mouse eyes. METHODS: Observations were made on heterozygous and homoz

Reciprocal photolabile O2 consumption and chemoreceptor excitation by carbon monoxide in the cat carotid body: evidence for cytochrome a3 as the primary O2 sensor.

High carbon monoxide (CO) gas tensions (> 500 Torr) at normoxic PO2 (125-140 Torr) stimulates carotid chemosensory discharge in the perfused carotid body (CB) in the absence but not in the presence of light. According to a metabolic hypothesis of O2 chemoreception, the increased chemosensory discharge should correspond to a photoreversible decrease of O2 consumption, unlike a non-respiratory hypothesis. We tested the respiratory vs. non-respiratory hypotheses of O2 chemoreception in the cat CB by measuring the effect of high CO. Experiments were conducted using CBs perfused and superfused in v

An Electroconductive Hydrogel Scaffold with Injectability and Biodegradability to Manipulate Neural Stem Cells for Enhancing Spinal Cord Injury Repair.

Spinal cord injury (SCI) generally leads to long-term functional deficits and is difficult to repair spontaneously. Many biological scaffold materials and stem cell treatment strategies have been explored, but very little research focused on the method of combining exogenous neural stem cells (NSCs) with a biodegradable conductive hydrogel scaffold. Here, a NSC loaded conductive hydrogel scaffold (named ICH/NSCs) was assembled by amino-modified gelatin (NH2-Gelatin) and aniline tetramer grafted oxidized hyaluronic acid (AT-OHA). Desirably, the well-conducting ICH/NSCs can be simply injected in

Mechanisms of vagal plasticity influencing feeding behavior.

Sensory neurons of the vagus nerve receive many different peripheral signals that can change rapidly and frequently throughout the day. The ability of these neurons to convey the vast array of nuanced information to the brain requires neuronal adaptability. In this review we discuss evidence for neural plasticity in vagal afferent neurons as a mechanism for conveying nuanced information to the brain important for the control of feeding behavior. We provide evidence that synaptic plasticity, changes in membrane conductance, and neuropeptide specification are mechanisms that allow flexibility in

Intact recognition of facial expression, gender, and age in patients with impaired recognition of face identity.

We conducted a series of experiments to assess the ability to recognize the meaning of facial expressions, gender, and age in four patients with severe impairments of the recognition of facial identity. In three patients the recognition of face identity could be dissociated from that of facial expression, age, and gender. In one, all forms of face recognition were impaired. Thus, a given lesion may preclude one type of recognition but not another. We conclude that (1) the cognitive demands posed by different forms of recognition are met at different processing levels, and (2) different levels
