Contralateral trigeminal dysaesthesias associated with second cervical nerve compression: a case report.

This 70-year-old woman presented with a left C2 solitary metastatic lesion producing ipsilateral occipital pain associated with contralateral fronto-orbital dysaesthesias. Examination revealed analgesia in the left C2 dermatome and hyperaesthesia in the right forehead. These symptoms and findings resolved following a course of radiation therapy to the C2 metastasis. Ipsilateral trigeminal dysaesthesias produced by cervical lesions have been described, however, contralateral cervicogenic trigeminal dysaesthesias have not. Relevant experimental data are analysed; neural pathways are suggested by…

Susceptibility to motion sickness and the development of anticipatory nausea and vomiting in cancer patients undergoing chemotherapy.

There are neural pathways between the vestibular system and an area of the brain in the fourth ventricle that appear to coordinate vomiting. Based on this and clinical observations, we theorized a relationship between a susceptibility to motion sickness and the intensity of nausea and vomiting from chemotherapy. A significant relationship (P less than 0.01) between a susceptibility to motion sickness and anticipatory nausea/vomiting in cancer patients undergoing chemotherapy was found. Antimotion drugs such as scopolamine may have a greater antiemetic effect on chemotherapy patients susceptibl…

Galactorrhea secondary to chest wall surgery in an adolescent.

Although nonpuerperal galactorrhea in youth may be a sign of pituitary prolactinoma, the etiology may be benign and extensive neurologic or endocrinologic evaluation and treatment may be unnecessary. An 18-year-old female with an unusual but benign form of transient galactorrhea due to chest wall surgery is reported. It is hypothesized that the sectioning of the intercostal nerves may result in reflex stimulation of hypothalamic centers controlling lactation through the same neural pathways involved in puerperal lactation. The patient experienced galactorrhea for two months and transient ameno…

Nogo-Nogo receptor signalling in PNS axon outgrowth and pathfinding.

The Nogo/Nogo66 receptor signaling pathway has been characterized as inhibitory for axon growth, regeneration, and structural plasticity in the adult mammalian central nervous system. Nogo and its receptor are highly expressed when axon growth is abundant, however, the function of this pathway in neural development is unclear. We have characterized zebrafish Nogo pathway members and examined their role in the developing nervous system using anti-sense morpholinos that inhibit protein synthesis. Depletion of the Nogo66 receptor or a Nogo isoform causes truncated outgrowth of peripheral nervous

Neurovascular coupling, cognitive reserve, and cognitive performance in mild cognitive impairment.

BACKGROUND: Neurovascular coupling (NVC) impairment contributes to mild cognitive impairment (MCI), but its relationship with NVC in MCI remains unclear. OBJECTIVE: To explore the associations between CR, NVC, and cognition in MCI patients, and to clarify their potential correlational patterns. MATERIALS AND METHODS: We enrolled 113 MCI and 83 healthy controls, all undergoing resting-state fMRI and arterial spin labeling to calculate CBF/neural activity ratios (ALFF, fALFF, ReHo, DC) as NVC measures. Cognitive reserve was assessed using the Cognitive Reserve Index Questionnaire. RESULTS: Co

[Environmental modulation of musical emotion: frequency-specific analysis based on virtual reality and electroencephalography].

Musical emotion perception, as a key pathway to decoding the essence of human emotions, requires the analysis of its regulatory mechanisms for the development of precise neural modulation strategies. Although electroencephalography (EEG) signals can be used to capture the dynamic neural activities associated with music emotion processing, and virtual reality (VR) technology can offer immersive enhancement effects for emotion regulation, the interaction mechanism among the VR environment, music emotion and neural activity remains unclear. This study established a multimodal experimental paradig

EGFRvIII Reduces Neural Stem/Progenitor Marker Expression in GFAP-Negative iNSCs: Evidence for a Context-Dependent Cellular Response.

Epidermal growth factor receptor variant III (EGFRvIII) is a constitutively active EGFR deletion variant found in a subset of glioblastomas (GB). Its association with stem-like tumor populations is well documented, but the outcome of EGFRvIII expression may vary with the developmental and molecular state of the recipient cell. Here, we used human induced pluripotent stem cell (iPSC)-derived, glial fibrillary acidic protein (GFAP)-negative-induced neural stem cells (iNSCs) to explore how this receptor variant affects a defined neural stem/progenitor background. EGFRvIII was introduced either co

Molecular Aspects of Melatonin Treatment in Tinnitus: A Review.

Tinnitus is a hearing disorder characterized by the perception of sound without external acoustic stimuli, which is caused by damage to the auditory system in response to excessive levels of noise, ototoxic agents and aging. Neural plasticity, oxidative/nitrosative stress and apoptosis play important roles in the pathogenesis of tinnitus. The expression of neural plasticity related to excessive glutamatergic neurotransmission leads to generation of abnormal sound in one's ears or head. Furthermore, hyperactivation and over-expression of NMDA receptors in response to excessive release of glutam

tp53-dependent and independent signaling underlies the pathogenesis and possible prevention of Acrofacial Dysostosis-Cincinnati type.

Ribosome biogenesis is a global process required for growth and proliferation in all cells, but disruptions in this process surprisingly lead to tissue-specific phenotypic disorders termed ribosomopathies. Pathogenic variants in the RNA Polymerase (Pol) I subunit POLR1A cause Acrofacial Dysostosis-Cincinnati type, which is characterized by craniofacial and limb anomalies. In a zebrafish model of Acrofacial Dysostosis-Cincinnati type, we demonstrate that polr1a-/- mutants exhibit deficient 47S rRNA transcription, reduced monosomes and polysomes and, consequently, defects in protein translation.

Selective reduction in neural responses to high calorie foods following gastric bypass surgery.

OBJECTIVE: To investigate changes in neural activation and desire to eat in response to appetitive cues from pre- to postbariatric surgery for obesity. BACKGROUND: Roux-en-Y gastric bypass (RYGB) is the most common bariatric procedure. However, the mechanisms of action in RYGB are not well understood. A significant proportion of the resulting reduction in caloric intake is unaccounted for by the restrictive and malabsorptive mechanisms and is thought to be mediated by neuroendocrine function. Numerous investigations of postsurgical changes in gut peptides have resulted; however, changes in ne

Neutrophil responses to injury or inflammation impair peripheral gustatory function.

The adult peripheral taste system is capable of extensive functional plasticity after injury. Sectioning the chorda tympani (CT), a primary sensory afferent nerve, elicits transient changes in the uninjured, contralateral population of taste receptor cells. Remarkably, the deficits are specific to the sodium transduction pathway. Normal function is quickly restored in the intact nerve, in parallel with an influx of macrophages to both the denervated and uninjured sides of the tongue. However, changing the dietary environment by restricting sodium blocks the macrophage response and prolongs fun

Mutations in Traf3ip1 reveal defects in ciliogenesis, embryonic development, and altered cell size regulation.

Tumor necrosis factor alpha receptor 3 interacting protein 1 (Traf3ip1), also known as MIPT3, was initially characterized through its interactions with tubulin, actin, TNFR-associated factor-3 (Traf3), IL-13R1, and DISC1. It functions as an inhibitor of IL-13-mediated phosphorylation of Stat6 and in sequestration of Traf3 and DISC1 to the cytoskeleton. Studies of the Traf3ip1 homologs in C. elegans (DYF-11), Zebrafish (elipsa), and Chlamydomonas (IFT54) revealed that the protein localizes to the cilium and is required for ciliogenesis. Similar localization data has now been reported for mammal

The cholinergic anti-inflammatory system limits T cell infiltration into the neurodegenerative CNS, but cannot counteract complex CNS inflammation.

Stimulation of the nicotinic alpha7 acetylcholine receptor (nAChRalpha7) by nicotine or acetylcholine initiates the cholinergic anti-inflammatory pathway, a mechanism for neural inhibition of inflammation. The action of this pathway was initially discovered in animal models of endotoxemia and septic shock, and later described in a number of other diseases. Moreover, the action of this pathway is also implied in human degenerative diseases of the central nervous system (CNS) like amyotrophic lateral sclerosis or Alzheimer's disease. In spite of this general interest, little is known about its i

Identification and functional analysis of novel facial patterning genes in the duplicated beak chicken embryo.

Cranial neural crest cells form the majority of the facial skeleton. However exactly when the pattering information and hence jaw identity is established is not clear. We know that premigratory neural crest cells contain a limited amount of information about the lower jaw but the upper jaw and facial midline are specified later by local tissue interactions. The environmental signals leading to frontonasal identity have been explored by our group in the past. Altering the levels of two signaling pathways (Bone Morphogenetic Protein) and retinoic acid (RA) in the chicken embryo creates a duplica

More than blindsight: Case report of a child with extraordinary visual capacity following perinatal bilateral occipital lobe injury.

Injury to the primary visual cortex (V1, striate cortex) and the geniculostriate pathway in adults results in cortical blindness, abolishing conscious visual perception. Early studies by Larry Weiskrantz and colleagues demonstrated that some patients with an occipital-lobe injury exhibited a degree of unconscious vision and visually-guided behaviour within the blind field. A more recent focus has been the observed phenomenon whereby early-life injury to V1 often results in the preservation of visual perception in both monkeys and humans. These findings initiated a concerted effort on multiple

Lipopolysaccharide signaling in the carotid chemoreceptor pathway of rats with sepsis syndrome.

In addition to their role in cardiorespiratory regulation, carotid body (CB) chemoreceptors serve as sensors for inflammatory status and as a protective factor during sepsis. However, lipopolysaccharide-induced sepsis (LPS) reduces CB responsiveness to excitatory or depressant stimuli. We tested whether LPS exerts a direct effect on the carotid chemoreceptor pathway, the CB and its sensory ganglion. We determined that the rat CB and nodose-petrosal-jugular ganglion complex (NPJgc) express TLR4, TNF-α and its receptors (TNF-R1 and TNF-R2). LPS administration (15mg/kg intraperitoneally) evoked M

Joint-brain communication in rheumatoid arthritis: glial mechanisms, pain phenotypes, and translational evidence.

Rheumatoid arthritis (RA) is a systemic autoimmune disease in which persistent pain, fatigue, cognitive impairment, depression, and anxiety may involve altered central nervous system (CNS) processing. This narrative review synthesizes preclinical and clinical evidence concerning proposed glial cell-mediated joint-brain communication in RA. Joint-derived neural inputs and systemic inflammatory signals may communicate with the CNS through humoral, cellular, neural, neuroendocrine, and neurovascular pathways. Preclinical studies implicate microglia and astrocytes in neuroinflammatory responses, s

Brain organoid models of SZT2-related disease reveal an overproduction of outer radial glial cells through mTORC1 activation.

Biallelic loss-of-function variants of Seizure Threshold 2 (SZT2) cause neurodevelopmental diseases with developmental delay, epilepsy, and macrocephaly. SZT2 forms the KICSTOR complex, which represses the mechanistic target of rapamycin complex 1 (mTORC1) amino acid-sensitive pathway. SZT2 dysfunction is thought to cause abnormal activation of the mTOR pathway, underlying the pathogenesis of SZT2-related diseases. We previously reported constitutive activation of mTORC1 in lymphoblastoid cell lines derived from patients with SZT2-related disease. However, the impact of SZT2 dysfunction on hum

De Novo-Designed Peptide-Engineered Multimodal Platform for Post-Ischemic Stroke Tissue Repair.

Orchestrating tissue regeneration in complex pathologies like post-ischemic stroke requires materials that can precisely regulate multiple signaling pathways. A central challenge is engineering a single platform integrating mechanical, electrical, and biochemical cues to redirect these pathological networks. Here, we present a computation-driven, multimodal hydrogel engineered to function as a programmable regulatory node. The system integrates a computationally screened de novo vasculogenic peptide scaffold and surface-engineered, inflammation-responsive conductive MXene nanosheets. This rati

Skeletal muscle lysate derived macroporous hydrogel repairs spinal cord injury via immunoregulation and axon regeneration.

Secondary damage following spinal cord injury (SCI), mediated by excessive local inflammation, oxidative stress, and the limited regenerative capacity of the central nervous system, can lead to irreversible neurological dysfunction. Inspired by the role of exercise rehabilitation in promoting neural recovery post-SCI and the neuroregulatory functions of muscle-derived bioactive proteins in modulating inflammation and promoting regeneration, we developed a bioactive hydrogel (MTLH) using skeletal muscle lysate as the primary matrix cross-linked with sodium alginate. MTLH features a macroporous
