MFG-E8 glycoprotein in inflammatory tissue regeneration: molecular mechanisms, therapeutic potential and advanced polymeric biomaterial delivery strategies.

Milk fat globule epidermal growth factor 8 (MFG-E8) is a multifunctional glycoprotein that coordinates efferocytosis, inflammatory resolution, angiogenesis, and matrix remodeling during tissue repair. In ischemic and inflammatory microenvironments, endogenous MFG-E8 is functionally deficient due to proteolytic cleavage and oxidative inactivation, while free recombinant protein suffers from rapid clearance and poor targeting. Polymeric biomaterials, including nanoparticles, hydrogels, and extracellular vesicle carriers, offer protection, sustained release, and targeted delivery. This review sys

Investigation into the Impact of Hollow Cylinder Inner Diameter on the Mechanical Properties of Biomimetic Voronoi Scaffolds.

The current study investigates the mechanical performance of the hollow cylindrical scaffolds with varying inner radii designed for bone tissue engineering. The biomimetic Voronoi-based structures with distinct inner diameters were manufactured using the stereolithography (SLA) technique and eco-friendly plant-based resin. The experimental results revealed that increasing the inner radius significantly reduces the mechanical properties of the biological scaffolds. Conversely, designs with a smaller inner diameter exhibited higher resistance to compressive stress and superior energy absorption.…

A dual-pathway Wnt-IL-13 fusion protein enhances human intestinal regeneration through tuft cell activation.

Compromised intestinal regenerative responses drive severe inflammatory conditions, such as inflammatory bowel disease and graft-versus-host disease, affecting millions of people worldwide each year. Despite extensive research, effective therapies remain limited, and no curative treatments are currently available. We recently discovered that human intestinal tuft cells promote tissue repair following injury through Wnt and IL-4/IL-13 signaling pathways. Building on this discovery, here, we report the engineering and functional validation of a synthetic Wnt-IL-13 fusion protein that simultaneou

Effect of quince seed gum (QSG) on the performance of injectable hyaluronic acid hydrogels in terms of the rheological, morphological, and mechanical aspect.

Injectable hydrogels play an important role in tissue engineering as a filling and repairing material. This study aimed to develop a new injectable hydrogel based on hyaluronic acid (HA) and quince seed gum (QSG) and investigate the effect of QSG on hydrogel performance. The amount of unreacted 1,4-Butanediol diglycidyl ether is maintained at an undetectable level for HA-QSG hydrogels. Amino acid analysis showed that the HA-QSG hydrogel had rich amino acid concentrations of leucine, arginine, and valine. After thermal sterilization, the elastic modulus of HA-QSG gels for dermal and intraarticu…

The application and progress of high-density porous polyethylene in the repair of orbital wall defect.

High-density porous polyethylene is a type of polymeric biomaterial. When used to efficiently fill the extensive orbital volume and correct enophthalmos caused by orbital wall defect, it has a significant advantage of biocompatibility, which results in a low rate of postoperative exposure and infection. The major disadvantage of this material is its radiolucency. However, with the development of imaging techniques, it is now possible to use multidetector computed tomography to directly contour the implant and describe its position. The use of tissue engineering involving high-density porous po…

Biomechanical modeling of the rectum for the design of a novel artificial anal sphincter.

This paper discusses biomechanical issues that are related to the option of a novel artificial anal sphincter around the human rectum. The prosthesis consists of a compression cuff system inside and a reservoir cuff system outside, which is placed around the debilitated sphincter muscle. The micropump shifts fluid between the cuffs and thus takes over the expansion and compression function of the sphincter muscle. However, the human rectum is not a rigid pipe, and motion in it is further complicated by the fact that the bowel is susceptible to damage. With the goal of engineering a safe and re

Re: "Clinical evaluation of a modified coronally advanced flap alone or in combination with a platelet-rich fibrin membrane for the treatment of adjacent multiple gingival recessions: a 6-month study".

Choukroun's platelet-rich fibrin (PRF) is a fibrin biomaterial with a specific composition, three-dimensional architecture, and associated biology. This letter highlights some key issues related to the use of this complex material during periodontal surgery. First, a reproducible protocol for the production of PRF membranes must be followed to control the quantity and quality of the fibrin matrix, leukocytes, platelets, and growth factors. Second, its use in periodontology must follow two principles founded on tissue-engineering basic rules and classical periodontal concepts. Using this two-pr

Decellularized tendon-bone composite grafts for extremity reconstruction: an experimental study.

BACKGROUND: Restoration of biomechanical strength following surgical reconstruction of tendon or ligament insertion tears is challenging because these injuries typically heal as fibrous scars. The authors hypothesize that injuries at the tendon-bone interface would benefit from reconstruction with decellularized composite tendon-bone grafts. METHODS: Tendon-bone grafts were harvested from Sprague-Dawley rats. Grafts subjected to decellularization were compared histologically and biomechanically with untreated grafts ex vivo and in a new in vivo model. Wistar rats underwent Sprague-Dawley allo

Paracrine Signaling in Cell-Biomaterial Interactions in Scaffold Vascularization: A Mini Review.

Vascularization remains a fundamental bottleneck in tissue engineering, as the absence of functional vascular networks limits oxygen and nutrient delivery, resulting in necrotic cores and poor host integration. While structural scaffold design and cell sourcing have advanced considerably, emerging evidence indicates that paracrine signaling, rather than direct cell contact or scaffold architecture alone, is the primary driver of angiogenesis and vasculogenesis within engineered constructs. Key cell types, including endothelial cells (ECs) and mesenchymal stem cells (MSCs), engage in bidirectio…

Biocompatible Gelatinized Starch as a Functional Modulator to Expand the Scope of Injectable Hydrogels.

Injectable hydrogels used in tissue engineering and minimally invasive therapies are limited by insufficient biocompatibility, poor scalability, high costs, and inconsistent performance. Here, we introduce gelatinized starch, a common polysaccharide, as a versatile and economical building block to expand the existing repertoire of injectable hydrogel materials. Derived from native corn starch via heating and gelatinization, this biopolymer exhibits shear-thinning, thixotropic, and thermoreversible sol-gel transition properties, key rheological characteristics essential for syringeability and i…

Engineered Perfusable Hepatic Fibrosis Model via Embedded Sacrificial Bioprinting Recapitulates Stiffness-Driven Fibrogenesis.

Hepatic fibrosis, as the common pathological endpoint of chronic liver diseases, is characterized by a self-perpetuating vicious cycle comprising extracellular matrix (ECM) driven liver tissue stiffening and sustained hepatic stellate cell (HSC) activation. Although existing studies have simulated fibrotic microenvironments using 2D models with tunable matrix stiffness or static 3D cultures, these models lack engineered hepatic sinusoidal vasculature and dynamic mechanical stimulation within 3D ECM contexts. This study employed embedded sacrificial bioprinting to construct functional liver sin

Controllable preparation of magnesium-hybridized PLA-PEG-PLA porous microspheres with anti-inflammatory function.

Biodegradable polylactic acid-based microspheres have been widely used in biomedical applications such as drug delivery and tissue engineering, however, most of the microspheres typically possess simple surface structures, lacking bioactivity and the ability to promote cell adhesion. Our group previously synthesized poly (L-lactic acid) magnesium-doped microspheres (PMg) with immunomodulatory and osteogenic potential. However, several drawbacks of PMgs, such as high hydrophobicity, a narrow pore distribution and large average particle size, and limited sustainable Mg2+release, can affect cell …

A Fully Human Engineered Bone Niche With Endogenous Osteoclastogenesis Reveals Osteoclast-Dependent Osteomimicry in Prostate Cancer Cells.

Bone is the predominant site of metastasis in advanced prostate cancer (PCa), yet the mechanisms governing tumor-bone interactions remain incompletely understood, thanks in part to the scarcity of relevant models. The role played by osteoclasts in such interactions is especially obscure. A modular human three-dimensional (3D) in vitro bone niche model was developedThe model integrates osteoblasts and osteoclasts within a mineralized scaffold, recreating an endosteal-like microenvironment for co-culture with PCa cell lines and patient-derived organoids (PDOs). The engineered construct maintains
