Peptide-drug conjugates in tumor therapy: Current advances and future perspectives.

As a novel targeted therapeutic approach, peptide-drug conjugates (PDCs) integrate peptides, drug payloads, and linkers to establish an effi … rties, PDCs could induce immunogenic cell death and synergistically reprogram the immunosuppressive tumor microenvironment, thereby enabling precise intervention in tumors. This strategy exhibits a favorable efficacy-to-toxicity profile and remarkable potential in immunotherapy, ultimately leading to an improved therapeutic index against tumors. Advancing glioblastoma immunotherapy: Molecular pathways and innovative therapeutic strategies.

Glioblastoma (GBM) has been an aggressive brain tumor and it demonstrates poor prognosis and resistance to the traditional therapeutics. GBM is characterized by abnormal proliferation, high growth, and a significantly immunosuppressive tumor microenvironment (TME), making immunotherapy as an ideal treatment for GBM. The recent advances in the field of immunotherapy such as vaccines, and CAR-T cell treatments, have demonstrated significant efficacy in accelerating anticancer immune responses and improving survival outcomes. Immune checkpoint drugs targeting CTLA-4 and PD-1/PD-L1 have demonstrat…

Immune snapshots along the inflammation-to-cancer road in bladder urothelium.

The inflammatory microenvironment formed by chronic inflammation is not only a major risk factor for cancer but also a well-recognized precursor to bladder cancer. However, the immunological transitions that occur along the inflammation-to-cancer continuum remain incompletely understood. This mini-review synthesizes recent advances in understanding how the immune microenvironment evolves from an inflamed yet non-malignant urothelium to invasive carcinoma. First, we discuss how persistent stimuli-such as chronic infection or exposure to carcinogens-disrupt immune homeostasis, leading to sustain

Bidirectional Regulatory Mechanisms of Disulfidptosis Govern Precision Targeting in Degenerative Bone Diseases and Osteosarcoma.

Disulfidptosis is a newly identified form of programmed cell death closely associated with cystine metabolism abnormalities and cytoskeletal damage. Orthopedic diseases, such as degenerative conditions including intervertebral disc degeneration, osteoporosis, osteoarthritis, and malignant bone tumors like osteosarcoma, all involve imbalances in the immunometabolic microenvironment. The triggering conditions for disulfidptosis, such as high expression of SLC7A11 and glucose deprivation, are highly correlated with the pathaological features of orthopedic diseases and associated immune dysregulat

Cancer-associated fibroblasts: The "Achilles' Heel" in thyroid tumor progression and therapy.

Growing evidence indicates that the complex and heterogeneous tumor microenvironment (TME) beyond the tumor cells plays a pivotal role in cancer development and therapy. Notably, cancer-associated fibroblasts (CAFs)-a key stromal cell population within the TME-are closely associated with malignant tumor progression and therapeutic resistance and have progressively emerged as a research focus in oncology. As a major mesenchymal cell population in thyroid cancer (TC), CAFs exhibit high heterogeneity and dynamic plasticity. They secrete numerous cytokines and non-cytokine factors that abnormally …

Role of the bone marrow microenvironment in multiple myeloma: Impact of niches on drug resistance mechanisms.

Multiple myeloma (MM) is a blood cancer characterized by the uncontrolled growth of plasma cells in the bone marrow. These malignant plasma cells can proliferate locally and spread to other tissues and organs. The M-protein they produce can lead to various clinical symptoms, including anemia, hypercalcemia, bone pain, bone destruction, and kidney dysfunction. Despite significant advancements in treatment over the past two decades that have improved survival and outcomes for many patients, drug resistance remains a significant therapeutic challenge. This resistance is largely driven by the comp

An updated understanding of follicular lymphoma transformation.

Although most patients with follicular lymphoma (FL) follow an indolent disease course, some patients experience a critical inflection point when FL transforms into an aggressive lymphoma. Historically, FL transformation is marked by poor outcomes, particularly for patients with previous FL-directed treatment. Compared with FL, transformed FL (tFL) is marked by numerous additional genetic changes, upregulates novel signaling pathways, and arises from an ancestral FL clone with shared FL-initiating mutations. Prediction of tFL risk remains a high-priority area of disease research, with recent w

Malignant tumors in vagal-innervated organs: Exploring its homeostatic role.

Cancer remains a significant global health challenge, with its progression shaped by complex and multifactorial mechanisms. Recent research suggests that the vagus nerve could play a critical role in mediating communication between the tumor microenvironment and the central nervous system (CNS). This review highlights the diversity of vagal afferent receptors, which could position the vagus nerve as a unique pathway for transmitting immune, metabolic, mechanical, and chemical signals from tumors to the CNS. Such signaling could influence systemic disease progression and tumor-related responses…

Roles of Peripheral Nerves in Tumor Initiation and Progression.

In recent years, a long list of relevant studies has highlighted the engagement of the nervous system in the fine-tuning of tumor development and progression. Several authors have shown that different types of nerve fibres (sympathetic, parasympathetic/vagal or somatosensory fibres) may contribute to tumor innervation affecting cancer initiation, progression and metastasis. A large presence of nerve fibres is frequently observed in tumors with respect to the corresponding healthy tissues. In this regard, it is worth noting that in some cases a reduced innervation may associate with slow tumor

Immunotherapy in central nervous system tumors.

Immunotherapy, defined as strategies that enhance host antitumor immunity by activating innate and adaptive immune responses, has demonstrated remarkable efficacy in hematologic malignancies. However, its application in solid tumors, particularly those affecting the central nervous system (CNS) such as gliomas, brain metastases, primary CNS lymphoma (PCNSL), and medulloblastomas, has been considerably slower. Major barriers to immunotherapy in these contexts include the physical and functional constraints imposed by the blood-brain barrier (BBB), tumor immune evasion, immunosuppressive microen

Mapping immune trajectories from Helicobacter pylori gastritis to gastric cancer.

Helicobacter pylori-associated gastric tumorigenesis is typically staged by histopathology, yet clinical decisions often hinge on immune dynamics that histology alone cannot resolve. This mini review introduces a phase-specific "immune snapshot" framework that integrates three elements-who is present in the microenvironment, how key circuits are engaged, and where these features reside-to track immune trajectories across chronic gastritis, atrophy with intestinal metaplasia, and dysplasia or early cancer. Rather than cataloging markers, we emphasized a practical approach that maps compact tiss

Converging and evolving immuno-genomic routes toward immune escape in breast cancer.

The interactions between tumor and immune cells along the course of breast cancer progression remain largely unknown. Here, we extensively characterize multiple sequential and parallel multiregion tumor and blood specimens of an index patient and a cohort of metastatic triple-negative breast cancers. We demonstrate that a continuous increase in tumor genomic heterogeneity and distinct molecular clocks correlated with resistance to treatment, eventually allowing tumors to escape from immune control. TCR repertoire loses diversity over time, leading to convergent evolution as breast cancer progr

Increased matrix stiffness enhances pro-tumorigenic traits in a physiologically relevant breast tissue- monocyte 3D model.

High mammographic density, associated with increased tissue stiffness, is a strong risk factor for breast cancer per se. In postmenopausal women there is no differences in the occurrence of ductal carcinoma in situ (DCIS) depending on breast density. Preliminary data suggest that dense breast tissue is associated with a pro-inflammatory microenvironment including infiltrating monocytes. However, the underlying mechanism(s) remains largely unknown. A major roadblock to understanding this risk factor is the lack of relevant in vitro models. A biologically relevant 3D model with tunable stiffness

Unlocking estrogen receptor: Structural insights into agonists and antagonists for glioblastoma therapy.

Glioblastoma (GBM), a malignant brain tumor originating in glial cells, is one of the most common primary brain malignancies, affecting one in 100,000 people, typically in the frontal lobe. Estrogens, like estradiol-17 (E2), significantly influence GBM progression, metastasis, and angiogenesis. Estrogen receptors (ERs) are crucial in signal transduction and physiology, making them potential therapeutic targets. However, their roles in GBM pathogenesis remain unclear. This review explores ERs in GBM, focusing on their involvement in tumor immune evasion, modulation of the tumor microenvironment…

Unveiling the cancer risk nexus of the steatotic liver.

Steatotic liver, characterized by the accumulation of fat in the liver, poses significant health risks including metabolic dysfunction-associated steatotic liver disease (MASLD) and an elevated risk of primary liver cancer. Emerging evidence indicates a robust association between steatotic liver and increased susceptibility to extrahepatic primary cancers and their metastases. The deposition of fat induces dynamic changes in hepatic microenvironments, thereby fostering inflammation and immune responses that enhance liver metastasis from extrahepatic primary cancers. This review explores the im

A combinatorial culture strategy to develop pseudomyxoma peritonei organoid models.

BACKGROUND AND OBJECTIVES: Few preclinical models of pseudomyxoma peritonei (PMP) have been developed, probably due to the tumor's low incidence and its peculiar characteristics of slow growth. Therefore, there is a need to develop more refined PMP models that better reflect its characteristics. The aim of the study is to develop a culture strategy to generate organoid models derived from PMP patient samples. METHODS: We followed a strategy based on combinatorial culture conditions that include the different factors essential for PMP growth and that mimic the microenvironment present in the p
