Product of arachidonic acid oxidation inhibits neutrophil migration by forming the inflammatory substrate for integrin αMβ2.

Oxidation of arachidonic acid generates biologically active derivatives that play a critical role in shaping the inflammatory response. One such derivative, carboxypropylpyrrole (CPP), is a stable end product of arachidonic acid oxidation that has been previously described and detected in inflamed tissues; however, its pathophysiological contribution to inflammation has not been investigated. CPP forms covalent adducts with the extracellular matrix and plasma proteins, altering their biochemical properties and receptor-binding characteristics. Here, we examined how CPP formation impacts neutro

Immune checkpoints in a genetically engineered mouse model of spontaneous autoimmune uveitis.

Engineered mutant AireGW/+Lyn-/- C57BL/6 mice are a model of spontaneous autoimmunity in which 50% of mice develop highly destructive uveitis due to compromised central and peripheral T cell tolerance. The key retinal autoantigen in these mice is interphotoreceptor retinoid-binding protein (IRBP). The CD4+ T cells recognizing the dominant epitope of IRBP, called P2, from eye-draining lymph nodes of mice with or without uveitis and from the retinas of mice with uveitis, were characterized by single-cell RNA sequencing (scRNA-seq) and flow cytometry. Mice with uveitis had autoantigen-specific T

CXCL13/CXCR5 chemokine axis promotes antiviral CXCR5+CD19+ B Cells and follicular/effector CXCR5+CD4+ T Cells in the lungs associated with protection from severe and fatal COVID-19 following infection with pathogenic SARS-CoV-2 Delta variant.

Chemokines play a crucial role in the lung's immune responses to infections and diseases. The role of CXC ligand 13 (CXCL13), a chemokine produced homeostatically by various lung cell types, in protecting against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and disease remains controversial. Some studies reported that patients who survived severe coronavirus disease 2019 (COVID-19) had CXCL13-dominated lung immune responses early in infection. In contrast, other studies reported that high CXCL13 levels were associated with severe COVID-19. In this study, to determine

Sustained antigen-specific CD8+ T cell immunity post-mRNA booster requires notch pathway activation.

Messenger RNA (mRNA) vaccines effectively induce protective immunity, but antigen-specific CD8+ T cell responses exhibit limited persistence. In this study, we aimed to assess CD8+ T cell responses following a third dose of the Pfizer BNT162b2 COVID-19 vaccine and identify factors contributing to their longevity. Using HLA tetramers, we analyzed antigen-specific CD8+ T cells in 141 vaccinated individuals (86.5% female) and identified 2 groups: those with strong responses (strong group) and those with weak responses (weak group) 6 mo after the third mRNA vaccination. Transcriptomic analysis rev

BPTF is essential for vaccine-induced germinal center B cell responses.

Germinal centers (GCs) are microanatomical structures in which antigen-specific B cells undergo proliferation, somatic hypermutation, and affinity-based competition to select high-affinity clones that differentiate into memory B cells and plasma cells (PCs). B cell progression through the GC is tightly regulated, and the molecular determinants that modulate GC B cell proliferation and survival are still under investigation. Here, we use a conditional deletion mouse model to demonstrate that bromodomain PHD finger transcription factor (BPTF), a subunit of the nucleosome remodeling factor chroma

Differential stem-like potential of naïve and memory CD8 T cells after chronic infection.

Memory CD8 T cells respond rapidly upon antigen re-encounter and are considered advantageous for protective immunity. However, they undergo a swift decline under chronic antigen stimulation. In this study, we found that memory CD8 T cells' heightened activation sensitivity promotes terminal differentiation and impairs the formation of CXCR5+Tim-3- progenitor subsets, resulting in reduced persistence. This defect was commonly observed in memory CD8 T cells generated by diverse immunization strategies. Mechanistically, their inability to generate progenitor cells was not due to insufficient expr

Restriction of IgA secretion in gut plasma cells is driven by a tissue-specific glycolytic program.

IgA-secreting plasma cells (PCs) provide durable humoral immunity by supplying critical antibodies to mucosal and systemic sites. These cells are found in large numbers in the gut lamina propria and the bone marrow (BM). In this study, we found that IgA+ PCs in the gut secrete significantly fewer antibodies on a per-cell basis compared to BM PCs in B6 mice. While the cell-intrinsic and -extrinsic signals responsible for regulating BM PC function have been extensively studied, these regulatory signals are understudied in gut PCs. Recent studies have established that metabolism is a critical com

Development of genomic resources and assays for immune repertoire profiling in Syrian hamsters.

The Syrian hamster (Mesocricetus auratus) is an important model for human infectious diseases, particularly those that infect the respiratory tract, due to having similar disease progression and immune responses as humans. However, immune repertoire studies are extremely limited due to incomplete genomic characterization of hamster B and T cell receptor loci. To begin to develop hamster immunoglobulin (IG) and T cell receptor (TR) reference resources, we first performed long-read transcriptome sequencing of six tissue types. We obtained 43,090 full-length IG/TR transcripts which were used to c

CD19 expression associates with polyfunctionality and altered secretion dynamics in individual activated human B cells.

The concept of polyfunctional secretion, the ability of cells to secrete multiple, functionally distinct proteins, has received increased attention for B cells as they have been described to secrete both antibodies and cytokines. However, the functional analysis of B cells was mostly performed in bulk, and consequently, the dynamics, quantity, and temporality of poly-secreting B cells are not well understood on the level of the individual cell. Furthermore, data on the existence of B-cell polyfunctionality in healthy settings remain scarce. Therefore, in this work, we developed a microfluidic

Control of antibody class switch recombination by quantitative integration of antigen signaling.

Immunoglobulin class switch recombination by B lymphocytes requires the simultaneous expression of activation-induced cytidine deaminase (AID) and noncoding germline transcripts (GLT). Expression of both components is controllable through cytokine signalling and arise through stochastic mechanisms linked to cell division that can be predicted in mathematical models. Here we demonstrate that the same principles govern the mathematically predictable antigen-mediated regulation of isotype switching. We report both antigen affinity and antigen concentration regulated rates of AID expression and ge

The V158F polymorphism in human FcγRIIIa/CD16a defines opposing receptor responses when interacting with soluble immune complexes.

The V158F polymorphism found in the human Fc receptor CD16a (also known as FcγRIIIa) is thought to influence autoimmune disorders and responses to IgG-based therapies. V158F is known to influence the IgG affinity, and thereby triggering of the CD16a receptor expressed on various immune cells. In contrast to CD16 triggering by opsonized IgG, the mechanistic details of its interaction with soluble circulating IgG immune complexes (ICs) remain poorly understood. In this study we aimed to clarify the functional impact of the CD16a-V158F polymorphism in the context of IC-mediated diseases. We gener

Isotype-specific antibody secreting plasma cells harbor epigenetic and transcription factor architectures defining distinct cell lineages.

Antibody-secreting cells (ASCs) are critical effectors of humoral immunity, wherein their distinct isotypes play specialized roles in pathogen defense. Despite studies defining the transcriptional programs of selected ASC isotypes, specific mechanisms underlying isotype-specific gene regulation are largely unaddressed. Here, we performed an integrated multiomics analysis (RNA sequencing, ATAC-seq [assay for transposase-accessible chromatin using sequencing], DNA methylation) on surface-sorted IgM, IgG, and IgA ASCs isolated from the spleens and mediastinal lymph nodes of mice following influen

Context-dependent regulation of cytotoxic T lymphocytes by nuclear receptors.

Cytotoxic CD8⁺ T lymphocytes eliminate pathogen-infected and malignant cells through migration to affected tissues, cytokine production, and cytotoxic activity. A mounting body of evidence indicates that CD8 T cell differentiation into functionally different subsets, such as effector, memory, tissue-resident memory, and exhausted T cells, is controlled by a group of nuclear receptors (NRs). NRs sense signals from various physiological and environmental cues, such as nutritional, endocrine, stress/circadian, and metabolic signals, and translate them into gene-regulatory decisions. NRs act throu

C3-CD18-VATPase-ATG16L1 and C3-ATG16L1 axes restrict the escape of microbes through the autophagy-lysosome pathway.

As the key component of the complement system, C3 plays important roles in complement activation to regulate phagocytosis, lyse cells, mediate inflammation, and clear immune complexes. In the present study, CgC3 in the cell-free hemolymph of the Pacific oyster Crassostrea gigas was found to be able to bind various polysaccharides and microbes and then interacted with membrane receptor CgCD18 to mediate the entry of the CgC3-coated Vibrio into hemocytes. CgATPV1D in hemocytes sensed the CgC3-coated Vibrio vacuole and recruited CgATG16L1. The free CgC3 in hemocytes could also recognize and bind

SLC1A5 augmentation bypasses NK cell transduction barriers to deliver complex CAR payloads.

Reproducible and efficient genetic engineering of human NK cells remains a primary challenge to next-generation chimeric antigen receptor (CAR) therapies, specifically for larger constructs with multiple plasmids and transgenes. Lentiviral vectors (LVs) pseudotyped with baboon envelope (BaEV) outperform vesicular stomatitis virus G protein in transducing hematopoietic stem cells, T cells, B cells, and NK cells, but suffer from fusogenic toxicity in producer cells and lower virion yields (BaEV-TR). Here, we engineer an integrated viral receptor platform that overcomes these constraints and enab

Live versus killed Listeria monocytogenes immunization differentially programs Hup-specific CD4+ T cell responses with distinct protective outcomes.

The histone-like protein Hup is a highly conserved DNA-stabilizing and compacting protein found across bacterial families. We previously demonstrated that the Hup protein of Staphylococcus and Streptococcus species harbors a shared CD4+ T cell epitope, and that Hup77-88-specific CD4+ T cells confer cross-species protection against pulmonary infection with these bacteria. Here, we investigated whether exposure to the Gram-positive intracellular pathogen Listeria monocytogenes, that expresses an orthologous Hup protein, induces a Hup-specific CD4+ T cell response. Live L. monocytogenes infection

ZFP335-MYBL2 transcriptional axis orchestrates Th17 cell differentiation and psoriatic inflammation.

T helper 17 (Th17) cell differentiation is governed by a complex transcriptional network centered on the lineage-specifying factor RORγt, accompanied by STAT3, IRF4, and SMAD2/3. However, the complete repertoire of transcriptional regulators controlling Il17a gene transcription and Th17 commitment remains incompletely defined. Here, we identify zinc finger protein 335 (ZFP335) as a critical regulator of Th17 differentiation. Mechanistically, ZFP335 directly binds and activates the Mybl2 promoter, and the resulting MYBL2 protein cooperates with RORγt to drive Il17a transcription. Genetic ablati

Improving peptide vaccine manufacturability without sacrificing immunogenicity: substitution of methionine and cysteine with oxidation-resistant isosteres.

Vaccines comprising peptide antigens for inducing T cell immunity are being developed for a broad range of therapeutic applications including prevention and treatment of cancer, autoimmunity, and infectious diseases. However, many peptide antigens contain cysteine and/or methionine, which are prone to form oxidation products that can present challenges to manufacturing and reduce biological activity. To address this challenge, we introduced oxidation resistant (OXR) antigens wherein the cysteine and methionine residues of naturally occurring, wild type (WT) peptide antigens are substituted wit

Effectiveness and immunogenicity of a nanoemulsion protein subunit vaccine against Pseudomonas aeruginosa: investigation in diet-induced obese mice.

Pseudomonas aeruginosa (Pa) is an opportunistic pathogen threatening individuals with obesity, a condition associated with chronic meta-inflammation and altered immunity. In this study, we evaluate the immunogenicity and protective efficacy of a nanoemulsion-based subunit vaccine (L-PaF/ME/BECC) targeting Pa in a diet-induced obese mouse model. Mice fed a high-fat diet (HFD; 60% fat) were compared to those on a low-fat diet (LFD; 10% fat) or regular chow (3% to 4% fat) to assess differences in vaccine responsiveness. Our results show that while L-PaF induces robust immune responses across all

Silencing of Zc3h13 attenuates LPS-induced inflammatory response in macrophages via m6A-dependent stabilization of Spic mRNA.

N6-methyladenosine (m6A) represents a reversible and ubiquitous posttranscriptional modification of mRNA. It plays a crucial role in immune cell development and is implicated in a range of pathological conditions. Nevertheless, the precise role of m6A in LPS-induced macrophage inflammatory responses remains elusive. In the present study, we investigated the role of the m6A writer Zc3h13 in LPS-induced liver inflammation. Macrophages, which are vital components of the immune system, play critical roles in liver injury. Our results demonstrate that Zc3h13 is highly expressed in liver macrophages
