Chromosome-scale assembly of wheat cultivar Sumai 3, a major germplasm source for Fusarium head blight resistance.

Fusarium head blight (FHB) is a devastating disease that severely impacts global wheat (Triticum aestivum L.) production. Sumai 3, a wheat cultivar widely used in breeding programs for its strong FHB resistance, has not been fully resolved at the chromosome level. Here, we present a high-quality chromosome-scale assembly of Sumai 3 using PacBio HiFi reads and chromosome conformation capture sequencing. The 14.6 Gb assembly consists of 832 contigs, with the longest contig being 245.2 Mb and a contig N50 of 41.90 Mb, which were scaffolded into 21 pseudomolecules. De novo annotation identified 10…

The Equine Reproductive Microbiota: Composition, Dynamics, Dysbiosis, and Implications for Fertility in Mares and Stallions.

The equine reproductive microbiota has emerged as an important factor influencing reproductive health and fertility in both mares and stalli … micus, Escherichia coli, and Pseudomonas aeruginosa in association with endometritis and persistent breeding-induced endometritis, while sequencing-based studies suggest broader community-level dysbiosis rather than simple pathogen presence. In stallions, semen is not sterile and commonly contains taxa such as Porphyromonas, Corynebacterium, Peptoniphilus, …

TaGAD2 is a potential downstream effector of Rht5 in controlling wheat plant height.

Plant height is a key determinant of wheat plant architecture that affect lodging resistance and grain yield. The GA-responsive (GAR) dwarfing gene Rht5 was previously shown to decrease plant height without compromise of wheat seedling vigor and considered as a promising candidate gene for breeding wheat varieties in water-limited conditions. However, the mechanisms underlying Rht5-mediated dwarfism are unclear. In this study, we investigated the genetic effects of Rht5 on wheat growth and development using recombinant inbred lines (RILs) and found that Rht5 reduces plant height through inhibi…

Leave-One-Chromosome-Out Association Testing Decodes Uncapping Behaviour in Honeybees During Varroa destructor Infestation.

BACKGROUND: Uncapping behaviour of Varroa‑infested brood cells is a critical social‑immunity mechanism in honeybees, enabling colonies to detect and remove parasitized brood. This trait reduces mite reproduction and pathogen transmission and is increasingly recognized as a valuable phenotypic marker for breeding Varroa‑resistant colonies. OBJECTIVES: This study aimed to identify robust genomic markers associated with uncapping behaviour by applying a statistically rigorous GWAS framework that overcomes the potential inflated associations and limited resolution of earlier fixed‑model approache…

Yield Stability and Adaptability of Black Soybean Line BSCM41 Support Bi-Directional Seed Rotation System in Northeast Thailand.

Black soybean is a high-value alternative crop that can offset declining yellow soybean prices in Thailand. Currently, "Sukhothai3" is the only commercial variety produced through conventional breeding over the past three decades. Its low first-pod height (FPH) limits mechanized harvesting and reduces production efficiency. Recently developed elite RILs have shown improved FPH, grain yield, and protein content (34.88-41.22%), but their yield stability and adaptability across environments remain unevaluated. This information is essential for establishing an effective bi-directional seed rotatio…

Tuberculosis Susceptibility and Inbreeding Depression Hinder Ex Situ Conservation in a Critically Endangered Rainforest Bird.

Captive breeding can be a key component of species conservation strategies, but also exposes these rare species to novel environments including the pathogen landscape. The critically endangered white-winged wood duck (WWWD) Asarcornis scutulata has experienced substantial population declines, local extirpations and fragmentation of its former range in Southeast Asia, making it one of the rarest birds in the world. As in other rare species, WWWD declines have led to the initiation of captive breeding programs, but these have been hampered by the WWWDs' high susceptibility to Mycobacterium avium…

Association Between Back Scent Gland Development and Reproductive Status in Male Brown Bears (Ursus arctos).

Cutaneous scent glands secrete chemical signals for communication, a significant portion of which is to convey information about male reproductive status. This might be particularly important to animals with intraspecific variation in male reproductive activity, such as bears. Male brown bears mark trees with their scent secreted from back apocrine and sebaceous glands during the breeding season and thus might convey information about reproductive status. Therefore, we speculated that the development of scent glands in the back reflects reproductive performance. We investigated the effects of …

Research Note: A novel immunization approach for chicken coccidiosis vaccine: waterline immunization and its application effect.

Coccidiosis is a prevalent disease in poultry industry, causing significant economic losses. Vaccine immunization has been developed as an effective method to prevent coccidiosis. In China, the traditional method for coccidiosis vaccination in the poultry industry involves the using water troughs and powder drinking water adjuvants for chick immunization via drinking. However, as the breeding industry evolves rapidly, water troughs are progressively being replaced by water lines. This article introduces an innovative immunization approach-waterline immunization-utilizing a novel microencapsula…

Diet composition and trophic ecology of two Antarctic storm-petrel species.

Availability and quality of food shape the distribution and movements of animal populations. In sympatric species, sharing limited resources, coexistence is typically achieved through niche segregation. However, information on dietary niches is lacking particularly for small and elusive seabirds, which often forage in remote oceanic areas. In this study, we aimed to characterize the trophic ecology of two highly pelagic seabird species, Wilson's storm-petrel Oceanites oceanicus and Black-bellied storm-petrel Fregetta tropica, breeding sympatrically on King George Island, maritime Antarctica, u…

Underpinning regions associated with nitrogen use efficiency in Brassica juncea L. Czern: using associative transcriptomics approach.

BACKGROUND: Breeding crops for enhanced understanding of apparent nitrogen use efficiency (NUE) requires understanding of phenotypic variations and underlying genetics. Improving NUE in Indian mustard is considered important to reduce nitrogen (N) fertilizer use without sacrificing crop yield. In the present study, 260 genotypes of Brassica juncea were field evaluated for seed yield and NUE-related traits at three N levels: N0 (no added nitrogen), N50 (50 kg/ha nitrogen), and N100 (100 kg/ha nitrogen) for two years. RESULTS: N response in plants varied depending on the dose of nitrogen applie…

IPENS: Interactive unsupervised framework for rapid plant phenotyping extraction via NeRF-SAM2 fusion.

Advanced plant phenotyping technologies are vital for trait improvement and accelerating intelligent breeding. Due to the species diversity of plants, existing methods heavily rely on large-scale high-precision manually annotated data. For self-occluded objects at the grain level, unsupervised methods often prove ineffective. This study proposes IPENS, an interactive unsupervised multi-target point cloud extraction method. It utilizes radiance field information to lift 2D masks, segmented by SAM2 (Segment Anything Model 2), into 3D space for target point cloud extraction. A multi-target collab…

Comparative transcriptome profiling of vanilla (Vanilla planifolia) capsule development provides insights of vanillin biosynthesis.

BACKGROUND: Vanillin is the most abundant volatile compound in natural vanilla extract and the primary metabolite from an economic perspective. Natural vanilla is the second most expensive spice in the world and the most profitable crop adapted to the warm tropics. Despite its global popularity, vanilla is mainly cultivated from vegetatively propagated clones and insufficient modern plant breeding has been achieved. One of the breeding objectives is to increase the vanillin concentration in the cured vanilla capsules. The vanillin biosynthesis pathway has not been thoroughly deciphered and mul…

GWAS identifies a molecular marker cluster associated with monoterpenoids in grapes.

Monoterpenoids are vital compounds that impart a distinctive floral flavor. They exist in both glycosidic and free forms in grapes. The breeding of improved monoterpenoid varieties has consistently been a topic of interest, yet only a limited number of molecular markers have been documented. This study employed a genome-wide association study (GWAS) on an F1 population crossed between a typical muscat variety ('Muscat of Alexandria') and a non-aromatic variety ('Christmas Rose'), conducted over two consecutive years. A total of 4089 significant single nucleotide polymorphism sites (sigSNPs) an…

Phenotypic Physiological and Metabolomic Analyses Reveal Crucial Metabolic Pathways in Quinoa (Chenopodium quinoa Willd.) in Response to PEG-6000 Induced Drought Stress.

Drought stress seriously threatens human food security, and enhancing crops' drought tolerance is an urgent problem to be solved in breeding. Quinoa is known for its high nutritional value and strong drought tolerance, but its molecular mechanism in response to drought stress is still unclear. In this study, we used drought-tolerant (D2) and drought-sensitive (ZK1) quinoa varieties, and PEG-6000 was used to simulate drought stress in quinoa seedlings. Phenotypic and physiological biochemical indicators were measured during the seedling stage, and LC-MS was used for a metabolite analysis of dro…

Fine mapping of the dominant female sterility gene and novel model of hybrid seed production in cabbage (Brassica oleracea L. var. capitata).

Hybrid breeding based on male sterility requires the removal of male parents, which is time- and labor-intensive; however, the use of female sterile male parent can solve this problem. In the offspring of distant hybridization between Brassica oleracea and Brassica napus, we obtained a mutant, 5GH12-279, which not only fails to generate gynoecium (thereby causing female sterility) but also has serrated leaves that could be used as a phenotypic marker in seedling screening. Genetic analysis revealed that this trait was controlled by a single dominant gene. Further analysis revealed that Bo2g005…
