Mania-like behavior induced by genetic dysfunction of the neuron-specific Na+,K+-ATPase α3 sodium pump.

Bipolar disorder is a debilitating psychopathology with unknown etiology. Accumulating evidence suggests the possible involvement of Na(+),K(+)-ATPase dysfunction in the pathophysiology of bipolar disorder. Here we show that Myshkin mice carrying an inactivating mutation in the neuron-specific Na(+),K(+)-ATPase α3 subunit display a behavioral profile remarkably similar to bipolar patients in the manic state. Myshkin mice show increased Ca(2+) signaling in cultured cortical neurons and phospho-activation of extracellular signal regulated kinase (ERK) and Akt in the hippocampus. The mood-stabili

The cation/H+ exchanger ChCAX3 fine-tunes fruit ripening by modulating cytosolic Ca2+ homeostasis in Cerasus humilis.

Cerasus humilis is a calcium-rich fruit crop with high nutritional value, but its synchronized ripening causes concentrated harvest and short shelf life. In this study, ChCAX3, a vacuolar cation/H+ exchanger, was identified as a key regulator of fruit ripening. ChCAX3 showed high expression in fruit and stem tissues and was upregulated by CaCl2 and ethephon treatments. however, repressed during fruit ripening by ChMYB16. Yeast complementation and Arabidopsis thaliana assays validated its Ca2+ transport activity. Overexpression of ChCAX3 in tomato increased fruit Ca2+ content, delayed coloratio

Decoding mitochondrial signaling: neutrophil-tumor cell crosstalk in orchestrating cancer progression.

BACKGROUND: Mitochondria regulate energy metabolism, redox balance, cell survival, and innate immune signaling. Neutrophils, the most abundant circulating innate immune effector cells, play an essential role in reshaping the tumor microenvironment and can exhibit both anti-tumor and tumor-supporting behaviors. Although mitochondrial processes are increasingly recognized as key drivers of neutrophil functional diversity and neutrophil-tumor cell communication, a systematic synthesis centered on mitochondria remains limited. MAIN BODY: The review opens with an overview of defining features of mi

Medicinal-plant immunity and specialized metabolism in plant-pathogen interactions: mechanisms and applications.

Medicinal-plant immunity and specialized metabolism are mechanistically linked, but rigorous receptor validation, causal multi-omics, and field trials are needed for reliable translational applications. Medicinal plants combine classical immune signaling with lineage-specific specialized metabolism. This review focuses on the immune systems of medicinal plants and on how pathogen perception reshapes the biosynthesis of pharmacologically valuable secondary metabolites. We synthesize evidence for pattern-triggered immunity (PTI) and effector-triggered immunity (ETI), emphasizing pattern-recognit

Ceramide-PKCζ signaling axis mediates burn-enhanced ectopic calcification during Achilles tendon healing.

Burn injury frequently leads to heterotopic ossification (HO) during tendon healing, but the underlying molecular mechanisms remain poorly understood. This study investigated the role of the ceramide-protein kinase C zeta (PKCζ) signaling axis in burn-enhanced ectopic calcification during Achilles tendon healing and evaluated the therapeutic potential of PKCζ inhibition. A combined burn injury and Achilles tendon puncture model was established in C57BL/6 mice, with sham control, tendon puncture, and burn combined tendon puncture groups. Micro-computed tomography analysis revealed significantly

CAMK2 Expression and Its Regulation on Testosterone Synthesis in Mouse Testis.

Leydig cells are testosterone synthesis cells in testes, a process tightly regulated by luteinizing hormone (LH) through the activation of steroidogenic enzymes such as steroidogenic acute regulatory protein (StAR) and 3-beta-hydroxy-Delta5-steroid dehydrogenase (3β-HSD). While calcium/calmodulin-dependent protein kinase 2 (CAMK2) is known to modulate diverse cellular processes, including hormone signaling, its role in testosterone production remains unclear. In this study, we investigated the expression and functions of CAMK2 in mouse testes, focusing on its potential involvement in testoster

Phenylalanine primes renal tubular redox vulnerability and calcium oxalate crystal retention through a PPARδ-CPT1A fatty-acid oxidation axis.

Calcium oxalate (CaOx) nephrolithiasis is usually framed as a physicochemical disorder of urinary supersaturation, yet stable crystal retention also requires a permissive tubular epithelial state. Here, we tested whether circulating phenylalanine (Phe) functions as a systemic priming factor that sensitizes renal tubular epithelium to CaOx crystal retention under lithogenic stress. Targeted amino-acid metabolomics in patients with CaOx stones, two-sample Mendelian randomization, glyoxylate-induced mouse nephrolithiasis, HK-2 cell injury assays, renal transcriptomics, pathway-rescue experiments

Mitochondria-associated programmed cell death in colorectal cancer: molecular mechanisms, metabolic plasticity, and therapeutic opportunities.

Mitochondria integrate bioenergetics, redox homeostasis, calcium signaling, metabolite synthesis, organelle quality control, innate immune sensing, and regulated cell death. In colorectal cancer (CRC), mitochondrial function is not simply suppressed by aerobic glycolysis; rather, tumor cells dynamically redistribute flux between glycolysis, oxidative phosphorylation, glutaminolysis, fatty-acid metabolism, and the mevalonate pathway to meet stage- and treatment-specific demands. This metabolic plasticity determines whether mitochondrial stress is buffered or converted into a lethal signal. Here…

Oxaliplatin-Induced Liver Toxicity: Hepatic Transglutaminase 7 Upregulation Associates with Oxidative Stress, Inflammation, and Apoptosis.

Transglutaminases (TGs) are calcium-dependent enzymes that cross-link proteins, contributing to apoptosis, extracellular matrix (ECM) stabilization, and inflammation. While TG2 has been extensively studied in hepatic injury, the role of TG7 in oxaliplatin-induced liver responses remains unclear. Oxaliplatin, a third-generation platinum chemotherapeutic, effectively treats solid tumors but can induce hepatic stress through oxidative and pro-inflammatory signaling. Adult rats received intraperitoneal oxaliplatin (10 mg/kg weekly) for 6 weeks. qRT-PCR, immunohistochemistry (IHC), immunofluorescen

Pre-Parathyroidectomy PTH as an Integrated Biomarker of Glandular Remodeling and Skeletal Turnover in Secondary Hyperparathyroidism.

Secondary hyperparathyroidism (SHPT) is a major component of chronic kidney disease-mineral and bone disorder (CKD-MBD), reflecting progressive disturbances in mineral metabolism, endocrine signaling, skeletal remodeling, and parathyroid-gland biology. Traditionally, preoperative parathyroid hormone (PTH) has been used primarily as a biochemical threshold for surgical referral. However, persistent PTH elevation in advanced CKD-related SHPT may reflect more than isolated endocrine activity; available evidence suggests it integrates parathyroid-gland remodeling, receptor resistance, skeletal tur

NADPH oxidase 2 inhibitor GSK2795039 exerts antiplatelet and antithrombotic activity.

Reactive oxygen species (ROS) production by NADPH oxidase (NOX) is a key promoter of platelet activation, making NOX inhibition an attractive antiplatelet strategy. This study evaluated the effects of the novel NOX2 inhibitor, GSK2795039, on human platelet functional responses and ROS-mediated signaling pathways. GSK2795039 effectively suppressed both extracellular and intracellular ROS production in collagen-stimulated platelets. Consequently, GSK2795039 treatment significantly inhibited collagen-induced platelet aggregation, dense-granule ATP release, and α-granule P-selectin exposure. Furth
