Extracellular vesicle-based biosensors for Alzheimer's disease: A new frontier in precision diagnostics.

Alzheimer's disease (AD), the most prevalent neurodegenerative disorder, remains a diagnostic challenge due to its asymptomatic early stages and the lack of reliable, non-invasive biomarkers. Extracellular vesicles (EVs), including exosomes and microvesicles, are nano-sized membrane-bound particles released by cells into biological fluids and have emerged as promising carriers of disease-specific biomarkers. These vesicles reflect the physiological and pathological state of their parental cells, encapsulating proteins, lipids, and nucleic acids relevant to AD pathology, such as amyloid-β, tau,

Modulating metal-organic frameworks by surface engineering of stearic acid modification for follicular drug delivery and enhanced hair growth promotion.

Cyclodextrin metal-organic frameworks (CD-MOF) as delivery carriers have gained great attention in the biomedical field. However, limited by challenges of moisture-sensitive nature, the design and application of CD-MOF-based hair follicle delivery for androgenic alopecia (AGA) has rarely been explored. We developed the metal-organic frameworks as hair follicle-targeted delivery system (SA-MOF), stearic acid (SA) was used to modify metal-organic frameworks to form a protective hydrophobic layer on the surface and provide the additional hair growth-promoting effect. Bacterial Membrane Vesicles as Heteroantigen Reservoirs for Potentiated Colorectal Cancer Immunotherapy.

Cancer immunotherapy, including immune checkpoint blockade (ICB), often has limited efficacy due to inadequate neoantigen exposure. Although neoantigen vaccines can enhance immune sensitization, their clinical application is hindered by high identification costs and tumor heterogeneity. Alternatively, heteroantigen presentation by cancer cells offers a promising strategy to improve immunotherapy. Herein, hyaluronic acid (HA)-modified bacterial membrane vesicles were developed as heteroantigen reservoirs (HLGV) and then further loaded with PD-L1 siRNA (siPD-L1) for immune checkpoint blockade. Engineered Exosome-Loaded Silk Fibroin Composite Hydrogels Promote Tissue Repair in Spinal Cord Injury Via Immune Checkpoint Blockade.

Spinal cord injury (SCI) is a severe central nervous system disorder characterized by a high rate of disability and limited axonal regeneration. Excessive post-injury inflammation often leads to further neuronal damage. Immune checkpoint (IC) genes, which regulate immune cell activity, play a critical role in modulating post-injury inflammation and thus influence neural repair and functional recovery. In this study, analysis of the GEO database reveals that the IC gene T cell immunoglobulin and mucin domain-containing protein 3 (Tim3) is highly expressed in microglia following SCI, contributin

Suppression of Liver Fibrogenesis with Photothermal Sorafenib Nanovesicles via Selectively Inhibiting Glycolysis and Amplification of Active HSCs.

As the major driving factor of hepatic fibrosis, the activated hepatic stellate cells (aHSCs) rely on active glycolysis to support their aberrant proliferation and secretion of the extracellular matrix. Sorafenib (Sor) can combat liver fibrosis by suppressing HIF-1α and glycolysis, but its poor solubility, rapid metabolism, and low bioavailability restrict such a clinical application. Here, Sor was loaded onto polydopamine nanoparticles and then encapsulated by a retinoid-decorated red blood cell membrane, yielding HSC-targeted Sor nanovesicles (PDA/Sor@RMV-VA) with a high Sor-loading capacity

Tumor Microenvironment-responsive Nanocatalyst for Targeted Chemodynamic Cancer Therapy.

To address the challenges of insufficient hydrogen peroxide (H2O2) levels, rapid Fe3+ precipitation, and a slow Fenton reaction cycle, tumor-activated, self-accelerating CDT nanocatalysts are synthesized, comprising poly (lactic-co-glycolic acid) (PLGA)-encapsulated Ca-Fe peroxide clusters and polyarginine (R). Nanocatalysts are camouflaged with cancer cell membranes (CCM) to enhance tumor targeting. Additionally, polyarginine tailored the PLGA responsiveness to low H2O2 levels (50-100 µm). H2O2 triggered the degradation of PLGA, releasing CaFe clusters to produce Fe3+/Fe2+ and additional H2O2

Reliable high-PAP-1-loaded polymeric micelles for cancer therapy: preparation, characterization, and evaluation of anti-tumor efficacy.

The mitochondrial potassium channel Kv1.3 is a critical therapeutic target, as its blockade induces cancer cell apoptosis, highlighting its therapeutic potential. PAP-1, a potent and selective membrane-permeant Kv1.3 inhibitor, faces solubility challenges affecting its bioavailability and antitumor efficacy. To circumvent these challenges, we developed a tumor-targeting drug delivery system by encapsulating PAP-1 within pH-responsive mPEG-PAE polymeric micelles. These self-assembled micelles exhibited high entrapment efficiency (91.35%) and drug loading level (8.30%). As pH decreased, the mice

From in vitro to in vivo: The Dominant role of PEG-Lipids in LNP performance.

Lipid nanoparticles (LNPs) are widely employed for delivering nucleic acids, such as mRNA, in both vaccines and therapeutic applications. These LNPs generally include an ionisable lipid (typically ∼ 50 %) to facilitate nucleic acid encapsulation and a PEG-lipid (typically ∼ 1.5 %) to enhance nanoparticle stability. To examine how the choice of PEG-lipid impacts LNP performance, we investigated the physicochemical characteristics and potency of LNPs prepared using two PEG-lipids with different acyl chain lengths: DMG-PEG 2000 and DSG-PEG 2000, containing 14 and 18 carbon tail lengths, respectiv

Enhanced delivery of podophyllotoxin for hepatocellular carcinoma therapy using polymersome as an anticancer delivery platform.

Podophyllotoxin (PPT), a bioactive compound, shows promise as a potential cancer treatment drug. Nevertheless, low solubility and bioavailability of PPT necessitate a drug delivery system to improve its effectiveness. PPT was extracted from Linum album and delivered into HepG2 cancer cells using mPEG-PCL nanoparticles. Copolymers were synthesized and confirmed by UV-Vis, FTIR, 1HNMR, XRD, FESEM analyses, and the other physicochemical properties were also characterized. The critical micelle concentration of the copolymers was calculated, and the ratio of 1:10 with a CMC of 0.055 µg. mL-1 was se

Circulating small extracellular vesicles in chronic kidney disease and vascular calcification: "Tiny packages with big biological mission".

Chronic kidney disease (CKD), a condition affecting over 850 million people worldwide, is a major global health issue. CKD leads to multimorbidities and disease complexity, expediting the aging process and increasing mortality rates-a phenomenon sometimes referred to as "renal senescence." The leading cause of death in patients with CKD is cardiovascular disease (CVD), accounting for one-third to one-half of all deaths, in stark contrast to cancer, which is the primary cause of death in the general population. While previous studies on kidney disease have focused extensively on urinary extrace

Targeting granulosa cells with engineered DFO nanoparticles for the treatment of chemotherapy-induced premature ovarian failure.

Premature ovarian failure (POF) is a prevalent and lethal adverse event that severely affects female cancer patients receiving chemotherapy. It is highly correlated with the collateral damage to ovarian granulosa cells caused by ferroptosis. The excessive iron accumulation in the intracellular labile iron pool (LIP) and the aberrant generation of reactive oxygen species (ROS) is the major cause of ferroptosis. No compounds with both ferroptosis inhibition functions and ovarian-targeting specificity that do not reduce systemic chemotherapeutic efficacy have been successful in treating the POF i

Surface-Engineered HA-PEG-ICG/PLGA Nanoprobes with Vessels Targeting for Lymphatic System Visualization.

Targeted imaging of the lymphatic system is essential for the early diagnosis and management of lymphatic disorders, such as lymphedema. In this study, we developed a lymphatic-targeted fluorescent nanoprobe by encapsulating indocyanine green (ICG) within poly(lactic-co-glycolic acid) (PLGA) nanoparticles, further surface-modified with hyaluronic acid-polyethylene glycol (HA-PEG) to enhance specificity (HA-PEG-ICG/PLGA NPs). The nanoparticles were synthesized via a microemulsion technique followed by surface cross-linking, and thoroughly characterized by ultraviolet-visible (UV-vis) spectrosco

Reengineering glycyrrhetinic acid into a therapeutic oligomer for targeted tumor therapy with cardioprotection.

To overcome the inherent limitations of conventional nanocarriers such as therapeutic inefficacy and suboptimal drug loading capacity, a novel "polymeric nanodrug" strategy was developed. This approach redefines nanodrug design by employing glycyrrhetinic acid (GA) simultaneously as a bioactive agent and a polymerizable targeting monomer to synthesize poly-glycyrrhetinic acid (PGA), achieving near-theoretical drug loading approaching 100%. Subsequent PEGylation yielded hepatocellular carcinoma (HCC)-targeting PGA-PEG-GA nanodrugs, which exhibited a 1.89-fold enhancement in antitumor efficacy c

Polysaccharide-coated mPEG-PLA nanoparticles enhance thymoquinone delivery and therapeutic efficacy against colorectal cancer.

Colorectal cancer (CRC) remains a major therapeutic challenge due to systemic toxicity and poor tumour selectivity of conventional treatments. Thymoquinone (TQ), a natural anticancer compound, faces limited clinical utility because of poor solubility and bioavailability. To overcome these challenges, a fucoidan (FC)-coated methoxy poly(ethylene glycol)-poly(lactic acid) (mPEG-PLA) nanoparticle system was developed for targeted TQ delivery. The optimised FC-coated TQ nanoparticles (∼105 nm) exhibited high encapsulation efficiency (82.3 ± 0.77%) and low polydispersity (<0.2), enabling passive tu

Dual-targeted exosome nanoplatform co-delivering doxorubicin and Fuzi Lizhong Tang bioactives for synergistic DLBCL therapy and chemotherapy-induced diarrhea management.

The management of chemotherapy-induced diarrhea (CID) in diffuse large B-cell lymphoma (DLBCL) requires strategies that concurrently address intestinal protection and tumor-targeted therapy. In this study, we developed a multifunctional nano-delivery system based on natural exosomes to co-encapsulate doxorubicin (DOX) and bioactive components derived from the traditional Chinese medicine Fuzi Lizhong Tang (gingerol and atractylenolide), aiming to synchronize DLBCL-targeted chemotherapy with CID alleviation. Exosomes (TEX) isolated from the human DLBCL cell line OCI-Ly3 were loaded with DOX and

Inulin and hyaluronic acid-based oral liposome for enhanced photo-chemotherapy against orthotopic colon cancer and its reversal effects on tumor hypoxia and intestinal microbiota.

Colon cancer has a complex microenvironment and course, and conventional chemotherapy is hindered by low permeability and immunosuppression at the cancer site, leading to poor efficacy. Integrating intestinal environment regulation and molecularly targeted drugs are more attractive strategies. This study aimed to developed an oral colonic targeted delivery system (5-Flu/MET@MSNs/Ce6@HIL) using hyaluronic acid (HA) and inulin (IN) as key components. IN is a polymer with colon-specific targeting capabilities, while HA targets CD44 on the surface of colon cancer cells. We used IN and HA-modified

Multilayered Nanocarriers as a New Strategy for Delivering Drugs with Protective and Anti-inflammatory Potential: Studies in Hippocampal Organotypic Cultures Subjected to Experimental Ischemia.

Oxidative stress and neuroinflammation play a pivotal role in pathomechanisms of brain ischemia. Our research aimed to formulate a nanotheranostic system for delivering carnosic acid as a neuroprotective agent with anti-oxidative and anti-inflammatory properties to ischemic brain tissue, mimicked by organotypic hippocampal cultures (OHCs) exposed to oxygen-glucose deprivation (OGD). In the first part of this study, the nanocarriers were formulated by encapsulating two types of nanocores (nanoemulsion (AOT) and polymeric (PCL)) containing CA into multilayer shells using the sequential adsorptio

An anthocyanins-loaded extracellular vesicles coated with fucoidan nano-delivery system for enhancing dietary defense against inflammation-related colon injury.

Anthocyanins (ACNs) are water-soluble natural pigments exhibiting multiple physiologically active properties. However, their limited stability and bioavailability restrict their use in functional foods and pharmaceutical field applications. In this study, a FU-MEV-ACNs nano-delivery system was successfully constructed by encapsulating ACNs within milk-derived extracellular vesicles (MEV) via ultrasonication, followed by surface coating with fucoidan (FU). As anticipated, FU-MEV-ACNs exhibited a small particle size (approximately 190.8 nm) and uniform distribution (PDI: 0.167 ± 0.017). Under si

NIR-II light-driven in situ nanovaccine for cancer immunotherapy via lymph node migration-mediated accumulation.

Background: Tumor vaccines that combine tumor antigens with immune adjuvants offer a promising strategy for cancer immunotherapy. A particularly effective approach involves the in situ generation of tumor antigens within the tumor microenvironment and their subsequent delivery to the lymph nodes alongside immune adjuvants. However, Co-delivery of antigen and adjuvant on a single nano-platform remains facing some key problems, such as a single nanomedicine cannot simultaneously achieve antigen production, capture, and delivery to lymph nodes, as well as leakage of adjuvant due to destruction of
