The innate immune response and activation of coagulation in alpha1,3-galactosyltransferase gene-knockout xenograft recipients.

BACKGROUND: The role of the innate immune system in the development of thrombotic microangiopathy (TM) after alpha1,3-galactosyltransferase gene-knockout (GTKO) pig organ transplantation in primates is uncertain. METHODS: Twelve organs (nine hearts, three kidneys) from GTKO pigs were transplanted into baboons that received no immunosuppressive therapy, partial regimens, or a full regimen based on costimulation blockade. After graft failure, histologic and immunohistologic examinations were carried out. RESULTS: Graft survival of less than 1 day was prolonged to 2 to 12 days with partial regi

Targeted delivery of IDR-1018 via biomimetic magnetic nanovesicles suppresses neurovascular cell death and promotes angiogenic repair after neonatal hypoxic-ischemic brain injury.

Neonatal hypoxic-ischemic brain damage (HIBD) is characterized by severe endothelial injury, inflammation, and apoptosis, which together contribute to irreversible neurodevelopmental deficits. In this study, we developed a biomimetic magnetic nanovesicle platform (IDR-MNC@RMVs) loaded with the host defense peptide IDR-1018 as a targeted therapeutic strategy for neonatal HIBD. Under external magnetic guidance, IDR-MNC@RMVs preferentially accumulated in ischemic brain regions, where they markedly reduced neuronal and vascular injury, attenuated brain atrophy, and improved neurological recovery i

Repeated human cranial bone-derived mesenchymal stem cell transplantation improved electrophysiological recovery in a spinal cord injury rat model.

Mesenchymal stem cell (MSC)-based therapy has been applied in several clinical trials of spinal cord injury (SCI). We have successfully established MSCs from human cranial bone and developed a longitudinal neuromonitoring technique for rodents. In addition to single transplantation, the potential of multiple transplantations has been suggested as a new therapeutic strategy. However, there are no reports on the electrophysiological effects of multiple MSC transplantations in SCI using transcranial electrical stimulation motor-evoked potentials (tcMEPs). Here, we aimed to elucidate the efficacy

Endogenous Melanin and Hydrogen-Based Specific Activated Theranostics Nanoagents: A Novel Multi-Treatment Paradigm for Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a systemic autoimmune disorder characterized by excessive proliferation of rheumatoid arthritis synovial fibroblasts (RASFs) and accumulation of inflammatory cytokines. Exploring the suppression of RASFs and modulation of the RA microenvironment is considered a comprehensive strategy for RA. In this work, specifically activated nanoagents (MAHI NGs) based on the hypoxic and weakly acidic RA microenvironment are developed to achieve a second near-infrared fluorescence (NIR-II FL)/photoacoustic (PA) dual-model imaging-guided multi-treatment. Due to optimal size, the

Nano-calcipotriol as a potent anti-hepatic fibrosis agent.

Calcipotriol (CAL) has been widely studied as a fibrosis inhibitor and used to treat plaque psoriasis via transdermal administration. The clinical application of CAL to treat liver fibrosis is bottlenecked by its unsatisfactory pharmacokinetics, biodistribution, and side effects, such as hypercalcemia in patients. The exploration of CAL as a safe and effective antifibrotic agent remains a major challenge. Therefore, we rationally designed and synthesized a self-assembled drug nanoparticle encapsulating CAL in its internal hydrophobic core for systematic injection (termed NPs/CAL) and further i

Iron homeostasis landscape informs patient survival and neoadjuvant chemotherapy response in cancer.

BACKGROUND: Cancer cells frequently alter iron metabolism by increasing uptake or decreasing export, promoting oncogenesis and proliferation. However, the quantitative relationship between iron homeostasis and cancer remains underexplored. METHODS AND RESULTS: We simplified these alterations into measurable changes in the expression levels of iron transport-related genes. We investigated the expression of iron transport-related genes by comparing genomic data of 478 human cancer cell lines to 105 normal tissue samples, as well as analyzing clinically paired 6903 tumor to 709 normal samples. MSC-delivered high-affinity variants of soluble PD1 lead to tumor regression.

BACKGROUND AIMS: Immune checkpoint therapies aim to restore anti-tumor immunity by blocking inhibitory signals that suppress T-cell activation. The most effective current strategies use humanized antibodies targeting PD1 or its ligand PDL1. However, due to their large size, antibodies often exhibit limited tissue diffusion, resulting in poor penetration into solid tumors. METHODS: To address this challenge, we developed a novel checkpoint inhibitor approach that uses mesenchymal stromal cells (MSCs) to deliver a high-affinity, soluble PD1 receptor (sPD1-HAC). RESULTS: We found that sPD1-HAC

Scalable Engineering of Bio-Manufactured Extracellular Vesicles for Selective Delivery in Ovarian Cancer Patient-Derived Models.

Ovarian cancer remains challenging to treat because of late diagnosis, heterogeneity, and poor response to existing therapies. Here, we present a streamlined and scalable extracellular vesicle (EV) engineering approach that enables efficient cargo packaging and enhances targeted cargo delivery to ovarian tumors in vivo. Systematic comparison of vesicle-anchoring domains identifies an optimized scaffold for loading bioluminescent cargo into EVs. We characterized EV production from five cell lines commonly used for biopharmaceutical manufacturing and selected and stably engineered ExpiCHO cells

Hypoxic-inflammatory preconditioning endows BMSC-derived appoptotic extracellular vesicles with potent efficacy against IVDD via cell activation and mitochondrial homeostasis regulation.

Intervertebral disc degeneration (IVDD) is the primary cause of chronic low back pain, with the senescence of nucleus pulposus cells (NPCs) as its core driving mechanism. Mitochondrial homeostasis acts as a critical mediator linking cellular stress responses to the senescence program of nucleus pulposus cells. Recent studies have indicated that the transplantation of apoptotic extracellular vesicles (ApoEVs) derived from the apoptotic mesenchymal stem cells (MSCs) represents a novel direction for tissue regeneration therapy. Given that the pathological microenvironment of IVDD exhibits hypoxic

Involvement of Embryo-Derived and Monocyte-Derived Intestinal Macrophages in the Pathogenesis of Inflammatory Bowel Disease and Their Prospects as Therapeutic Targets.

Inflammatory bowel disease (IBD) is a group of intestinal inflammatory diseases characterized by chronic, recurrent, remitting, or progressive inflammation, which causes the disturbance of the homeostasis between immune cells, such as macrophages, epithelial cells, and microorganisms. Intestinal macrophages (IMs) are the largest population of macrophages in the body, and the abnormal function of IMs is an important cause of IBD. Most IMs come from the replenishment of blood monocytes, while a small part come from embryos and can self-renew. Stimulated by the intestinal inflammatory microenviro

Reprogramming of iPSCs to NPCEC-like cells by biomimetic scaffolds for zonular fiber reconstruction.

Ectopia lentis (EL), characterised by impaired zonular fibers originating from non-pigmented ciliary epithelial cells (NPCEC), presents formidable surgical complexities and potential risks of visual impairment. Cataract surgery is the only treatment method for EL, but it leads to the loss of accommodative power of the lens post-operatively. Furthermore, the challenge of repairing zonular ligaments in situ remains a significant global issue. Ocular tissue and aqueous humour samples from patients with EL were subjected to RNA sequencing and Olink high-throughput proteomic analysis, revealing the

Immune responses in mildly versus critically ill COVID-19 patients.

The current coronavirus pandemic (COVID-19), caused by SARS-CoV-2, has had devastating effects on the global health and economic system. The cellular and molecular mediators of both the innate and adaptive immune systems are critical in controlling SARS-CoV-2 infections. However, dysregulated inflammatory responses and imbalanced adaptive immunity may contribute to tissue destruction and pathogenesis of the disease. Important mechanisms in severe forms of COVID-19 include overproduction of inflammatory cytokines, impairment of type I IFN response, overactivation of neutrophils and macrophages,

Intraocular Penetration of a vNAR: In Vivo and In Vitro VEGF165 Neutralization.

Variable new antigen receptor domain (vNAR) antibodies are novel, naturally occurring antibodies that can be isolated from naïve, immune or synthetic shark libraries. These molecules are very interesting to the biotechnology and pharmaceutical industries because of their unique characteristics related to size and tissue penetrability. There have been some approved anti-angiogenic therapies for ophthalmic conditions, not related to vNAR. This includes biologics and chimeric proteins that neutralize vascular endothelial growth factor (VEGF)165, which are injected intravitreal, causing discomfort

Stable and efficient intraocular gene transfer using pseudotyped EIAV lentiviral vectors.

BACKGROUND: We have developed minimal non-primate lentiviral vectors based on the equine infectious anaemia virus (EIAV). We evaluated the in vivo expression profiles of these vectors delivered regionally to ocular tissues to define their potential utility in ocular gene therapy. METHODS: EIAV vectors pseudotyped with VSV-G or rabies-G envelope proteins were delivered subretinally, intravitreally or into the anterior chambers (intracameral administration) in mice. Reporter gene (eGFP) expression was analysed using in vivo retinal imaging or histological examination of eyes and brains at inter

Potential therapeutic implications of new insights into respiratory syncytial virus disease.

Viral bronchiolitis is the most common cause of hospitalization in infants under 6 months of age, and 70% of all cases of bronchiolitis are caused by respiratory syncytial virus (RSV). Early RSV infection is associated with respiratory problems such as asthma and wheezing later in life. RSV infection is usually spread by contaminated secretions and infects the upper then lower respiratory tracts. Infected cells release proinflammatory cytokines and chemokines, including IL-1, tumor necrosis factor-alpha, IL-6, and IL-8. These activate other cells and recruit inflammatory cells, including macro

Ultrasound-Driven Cu-Nanobots for Tumor Deep Penetration and Induction of Cuproptosis in Prostate Cancer Therapy.

Cuproptosis has attracted considerable attention in cancer therapy due to its unique ability to circumvent drug resistance, while the in vivo application is hindered by systemic toxicity, rapid clearance, and limited tumor tissue penetration. Widely recognized for its noninvasive application, ultrasound has emerged as a promising method for enhancing drug tumor penetration. In this study, we developed cuproptosis-oriented Nanobots to achieve deep penetration into prostate tumors under ultrasound irradiation. Asymmetric Cu-Nanobots (∼200 nm) were prepared by ultrasound polymerization using hexa

Targets for pharmacological intervention of endothelial hyperpermeability and barrier function.

Many diseases share the common feature of vascular leakage, and endothelial barrier dysfunction is often the underlying cause. The subsequent stages of endothelial barrier dysfunction contribute to endothelial hyperpermeability. Vasoactive agents induce loss of junctional integrity, a process that involves actin-myosin interaction. Subsequently, the interaction of leukocytes amplifies leakage by the leukocyte-derived mediators. The processes mainly occur at the postcapillary venules. The whole microvascular bed, including the capillaries, becomes involved in vascular leakage by the induction o

Bioactive Hydrogels and Scaffolds for Oral Mucosal Regeneration After Oral Squamous Cell Carcinoma Therapy: A Comprehensive Review.

Oral squamous cell carcinoma (OSCC) therapy frequently produces acute and chronic injury to the oral mucosa, including surgical lining defects and radiochemotherapy-associated oral mucositis (OM). Beyond pain and ulceration, these injuries compromise nutrition, speech, oral hygiene, and feasibility of dental/implant rehabilitation, and may disrupt oncologic treatment delivery. The oral cavity imposes stringent constraints on regenerative biomaterials-continuous salivary flow, high microbial load, and repeated mechanical shear-such that clinical success depends on reliable mucoadhesion/wet adhe
