Genetic association studies are popular for identifying genetic variants, such as single nucleotide polymorphisms (SNPs), that are associated with complex traits. Statistical tests are commonly performed one SNP at a time with an assumed mode of inheritance such as recessive, additive, or dominant genetic model. Such analysis can result in inadequate power when the employed model deviates from the underlying true genetic model. We propose an integrative association test procedure under a generalized linear model framework to flexibly model the data from the above three common genetic models an

Longitudinal measurement of biomarkers is important in determining risk factors for binary endpoints such as infection or disease. However, biomarkers are subject to measurement error, and some are also subject to left-censoring due to a lower limit of detection. Statistical methods to address these issues are few. We herein propose a generalized linear mixed model and estimate the model parameters using the Monte Carlo Newton-Raphson (MCNR) method. Inferences regarding the parameters are made by applying Louis's method and the delta method. Simulation studies were conducted to compare the pro

OBJECTIVE: To increase the knowledge of rumination and its associations with stressful events, we explored the relationships between 4 types of rumination (brooding, reflection, intrusive, and deliberate rumination) in a sample of 750 adult participants who experienced a highly stressful event. We also explored the predictive value of the different types of rumination on posttraumatic stress symptoms and posttraumatic growth 6 months after the highly stressful event occurred.
METHOD: Participants completed the Ruminative Response Scale and the Event-Related Rumination Inventory. Brooding and

Computational models represent a highly suitable framework, not only for testing biological hypotheses and generating new ones but also for optimising experimental strategies. As one surveys the literature devoted to cancer modelling, it is obvious that immense progress has been made in applying simulation techniques to the study of cancer biology, although the full impact has yet to be realised. For example, there are excellent models to describe cancer incidence rates or factors for early disease detection, but these predictions are unable to explain the functional and molecular changes that

OBJECTIVES/HYPOTHESIS: To assess the risk of aspiration using a novel valve circuit that dynamically modulates endotracheal tube cuff pressure during the ventilatory cycle using bench and live animal models.
STUDY DESIGN: Animal model.
METHODS: The bench model consisted of a cuffed endotracheal tube inserted into an artificial trachea. Leakage of liquid around the cuff was measured after 4 hours of constant or dynamic modulation of cuff pressure at variable peak end expiratory pressures. In the porcine model, eight animals were ventilated with the modulating valve circuit and compared to eig

We study effect of weak noise on the dynamics of a hair bundle model near the excitability threshold and near a subcritical Hopf bifurcation. We analyze numerically noise-induced structural changes in the probability density and the power spectral density of the model. In particular, we show that weak noise can induce oscillations with two distinct frequencies in both excitable and limit-cycle regimes. We then applied a recently developed technique of stochastic sensitivity functions which allows us to estimate threshold values of noise intensity corresponding to these transitions.

BACKGROUND: The prevalence of obesity among high school students has risen in recent decades. Many high school students report trying to lose weight and some engage in disordered eating to do so. The obesity proneness model suggests that parents may influence their offspring's development of disordered eating. This study examined the viability of a modified obesity proneness model in a high school population.
METHODS: Cross-sectional survey data from a random cluster sample of 1533 students in grades 9-12 from a Florida school district were analyzed using structural equation modeling. Variabl

BACKGROUND: With continuous glucose sensors (CGSs), it is possible to obtain a dynamical signal of the patient's subcutaneous glucose concentration in real time. How could that information be exploited? We suggest a model-based diagnosis system with a twofold objective: real-time state estimation and long-term model parameter identification.
METHODS: To obtain a dynamical model, Bergman's nonlinear minimal model (considering plasma glucose G, insulin I, and interstitial insulin X) is extended by two states describing first and second insulin response. Furthermore, compartments for oral glucos

An increasing number of students with dyslexia enter higher education. As a result, there is a growing need for standardized diagnosis. Previous research has suggested that a small number of tests may suffice to reliably assess students with dyslexia, but these studies were based on post hoc discriminant analysis, which tends to overestimate the percentage of systematic variance, and were limited to the English language (and the Anglo-Saxon education system). Therefore, we repeated the research in a non-English language (Dutch) and we selected variables on the basis of a prediction analysis. T

The effects of a drug on the genomic scale can be assessed in a three-color cDNA microarray with the three color intensities represented through the so-called hexaMplot. In our recent study, we have shown that the Hough Transform (HT) applied to the hexaMplot can be used to detect groups of coexpressed genes in the normal-disease-drug samples. However, the standard HT is not well suited for the purpose because 1) the assayed genes need first to be hard-partitioned into equally and differentially expressed genes, with HT ignoring possible information in the former group; 2) the hexaMplot coordi

Genome wide association studies have identified several single nucleotide polymorphisms (SNPs) that are independently associated with small increments in risk of prostate cancer, opening up the possibility for using such variants in risk prediction. Using segregation analysis of population-based samples of 4,390 families of prostate cancer patients from the UK and Australia, and assuming all familial aggregation has genetic causes, we previously found that the best model for the genetic susceptibility to prostate cancer was a mixed model of inheritance that included both a recessive major gene

Genome-wide association studies often face the undesirable result of either failing to detect any influential markers at all because of a stringent level for testing error corrections or encountering difficulty in quantifying the importance of markers by their P-values. Advocates of estimation procedures prefer to estimate the proportion of association rather than test significance to avoid overinterpretation. Here, we adopt a Bayesian hierarchical mixture model to estimate directly the proportion of influential markers, and then proceed to a selection procedure based on the Bayes factor (BF).

BACKGROUND: Juveniles becoming overly intoxicated by alcohol is a widespread problem with consequences ranging from hangovers to deaths. We ask: could overshoots of intended levels of intoxication be triggered by misperceptions of the types found in recent studies of decision making in dynamic systems? It is well known that the dynamics of the blood alcohol concentration (BAC) is complicated by a temporary accumulation of alcohol in the stomach delaying the uptake into the blood stream. We hypothesize that juveniles use an overly simplified drinking strategy, where drinking is guided by the di

In the assessment of diagnostic tests the task may arise to show that a candidate test is non-inferior compared to a comparative (standard) test with regard to the diagnostic sensitivity or specificity. This setting is known as "one-sided equivalence" and has been applied to a single comparison between two diagnostic tests (Chen et al., 2003). Recently, the approach has been extended into a meta-analytical framework (EFSA, 2006), allowing for the difference between the sensitivity (or specificity) of two diagnostic tests to be estimated using information gathered through systematic literature

Based on the analysis of electromyographic (EMG) data muscles are often characterized as normal or affected by a neuromuscular disorder. Motor unit potential (MUP) characterizations comprised of the conditional probabilities of a MUP being detected from a muscle of each of the following categories: myopathic, normal, and neuropathic, were estimated. The sets of MUP characterizations of a set of MUPs detected in a muscle were averaged to produce a set of muscle characterization measures related to the probability of the muscle belonging to each category conditioned on the set of MUPs detected.

BACKGROUND: Alcohol dependence (AD) is often comorbid with other disinhibitory disorders that are characterized by poor decision making and evidenced by disadvantageous strategies on laboratory tasks such as the Iowa Gambling Task (IGT). In this study, a variant of the IGT is used to examine specific mechanisms that may account for poor decision making on the task in AD both with and without comorbid psychopathology.
METHODS: The community sample (n = 428) included 134 young adult subjects with AD and a history of childhood conduct disorder (CCD), 129 with AD and no history of CCD, 60 with a
