Alzheimer's disease risk gene Wwox controls amyloid pathology through metabolic reprogramming.

Genome-wide association studies have identified multiple loci that contribute to late-onset Alzheimer's disease risk, yet the functional mechanisms of many candidate genes remain unclear. WW-domain containing oxidoreductase (WWOX) is one such gene associated with disease and downregulated in patient brains, but its role in neurodegeneration is poorly understood. Here, we use Drosophila models to investigate the cell-type-specific and mechanistic roles of Wwox in ageing and amyloid-β42 (Aβ42)-induced pathology. Neuronal and glial knockdown of Wwox reduced lifespan and increased sleep drive, ind…

Metabolic stress exacerbates Parkinson's disease pathology through mitochondrial dysfunction and ferroptosis.

Epidemiological studies have linked type 2 diabetes to Parkinson's disease (PD), but causality and mechanisms remain unclear. Here, using Me … ed to high glucose and MPP⁺. Our findings establish that glucose intolerance directly aggravates PD pathology via mitochondrial dysfunction and ferroptosis, suggesting that targeting this cell death pathway or improving glycemic control could offer therapeutic opportunities for PD, especially in patients with metabolic comorbidities.

Leucettine L41 Ameliorates Tau pathology and neuroinflammation in Parkinson's disease models.

BACKGROUND: Aberrant protein phosphorylation and sustained neuroinflammatory activation are key biochemical features of experimental parkinsonism. Dysregulated tau processing and excessive microglial reactivity have been implicated in motor dysfunction associated with dopaminergic injury. In the present study, we evaluated the biochemical and functional effects of Leucettine L41 using biochemical, cellular, and in vivo systems. RESULTS: In an in vitro tau aggregation assay, L41 reduced the ThT fluorescence, indicating the suppression of fibril formation. In SH-SY5Y neuronal cells, L41 decreas

Fumarate and Its Esters: Physiology, Pathology, Molecular Mechanisms, Diseases, and Therapy.

Fumarate is a critical intermediate of the tricarboxylic acid cycle that, beyond its metabolic role, has emerged as an important regulator of cellular signaling. Fumaric acid esters (FAEs), including dimethyl fumarate and monomethyl fumarate, have been utilized as therapeutic agents for immune-mediated diseases and are being explored for broader applications. Accumulating evidence indicates that both fumarate and FAEs modulate diverse biological processes through mechanisms such as protein succination and metabolic reprogramming. In this review, we discuss the current understanding of fumarate

Somatic CRISPR editing of Msh3 mitigates Huntington's disease pathology in mice.

Huntington's disease (HD) is a fatal, dominantly inherited neurodegenerative disorder caused by a CAG repeat expansion in Huntingtin (HTT) e … expansion. Intervention at all three ages slowed striatal CAG expansion, reduced nuclear huntingtin pathology and suppressed transcriptional dysregulation, with earlier intervention having greater impact. Msh3 knockout also suppressed the production of the exon 1 Htt1a transcript. The results of our study provide important preclinical information relevant t …

Scavenger receptors associate with Alzheimer's pathology, cholesterol metabolism and synaptic dysfunction.

This study investigated relationships between cerebrospinal fluid (CSF) scavenger receptors and early Alzheimer's disease (AD) pathology in cognitively unimpaired older adults at elevated familial risk, with complementary analyses in autopsied frontal cortex. In the PREVENT-AD cohort, CSF levels of the scavenger receptors MSR1, SCARB2, and SCARF2 were examined in relation to AD biomarkers and synaptic proteins. Complementary analyses, with the same scavenger receptors, used frontal cortex tissue from the Douglas-Bell Canada Brain Bank and targeted sterol/oxysterol mass spectrometry. Linear reg…

Deep learning-based computational pathology: Technologies, clinical applications, and future directions.

The adoption of whole-slide imaging is establishing a new paradigm in digital pathology. However, the translation of artificial intelligence (AI) from research to clinical practice faces significant hurdles, largely due to a misalignment between algorithmic advances and the practical demands of pathological diagnosis and prognosis. In this review, we propose a dual-perspective framework to systematically bridge this gap by linking core clinical tasks with cutting-edge deep learning methodologies. We present a comprehensive overview of the field from 2020 to 2025, analyzing how architectures su…

Injury-induced tau pathology promotes aggressive behavior in Drosophila without neurodegeneration.

The microtubule-associated protein tau is implicated in neurodegenerative diseases, but its physiological roles remain poorly understood. Here, we find that panneuronal expression of human tau (HsTau) in Drosophila coupled with injury triggers elevated aggression in male flies, which was not observed in flies expressing nonphosphorylatable tau. These behavioral manifestations result from activation of dopaminergic circuits without neurodegeneration. Using in vitro reconstitution assays, we find that phosphorylated HsTau maintains microtubule binding but loses its ability to suppress catastroph

Synucleins in Neural Physiology: Understanding Endogenous Function to Better Contextualize Pathology.

Perhaps most well-known for its penetrant role in synucleinopathies, alpha-synuclein's nonpathological function remains incompletely characterized. Most widely regarded as a putative presynaptic protein, a growing body of work over the last few decades demonstrates that alpha-synuclein participates in a broad and more diverse set of neuronal functions. Alpha-synuclein is a small, intrinsically disordered protein comprising three functionally distinct regions that mediate membrane binding, vesicle clustering, and protein-protein interactions. At synapses, alpha-synuclein participates in multipl

Adaptive diagnostic reasoning framework for pathology with multimodal large language models.

BACKGROUND: Artificial intelligence enhances pathology screening efficiency, yet clinical adoption remains limited because most systems operate as opaque black boxes. We aim to resolve this opacity by establishing a framework that generates transparent, evidence-linked reasoning to support diagnostic auditing. METHODS: We present a framework that shifts off-the-shelf multimodal large language models from passive pattern recognition to active diagnostic reasoning. Using small labeled subsets from breast and prostate cancer datasets, we employ a two-phase self-learning process to derive diagnos…

FAM134B-mediated ER-phagy degrades APP and suppresses Alzheimer's disease pathology.

Endoplasmic reticulum autophagy (ER-phagy) is a selective autophagy pathway in which receptor proteins target ER membranes and proteins for … . This transcriptional suppression impairs ER-phagy, leading to APP accumulation and exacerbated AD pathology. AAV-mediated hippocampal expression of wild-type, but not LIR-mutant, FAM134B in 5XFAD mice restores ER-phagy, enhances APP clearance, reduces Aβ deposition, preserves synaptic and myelin integrity, and improves cognitive performance. These finding …

Semi-Supervised Domain Adaptation with Latent Diffusion for Pathology Image Classification.

Deep learning models in computational pathology often fail to generalize across cohorts and institutions due to domain shift. Existing approaches either fail to leverage unlabeled data from the target domain or rely on image-to-image translation, which can distort tissue structures and compromise model accuracy. In this work, we propose a semi-supervised domain adaptation (SSDA) framework that utilizes a latent diffusion model trained on unlabeled data from both the source and target domains to generate morphology-preserving and target-aware synthetic images. By conditioning the diffusion mode…

Prior Infection with Torque Teno Virus Mitigates Influenza Pathology in Mice.

Respiratory infections caused by influenza viruses are frequently associated with coinfection by other infectious agents. Torque teno viruse … amination of lung tissues revealed that prior TTSuV1 infection significantly attenuated IAV-induced pathology in coinfected mice. Coinfected animals also exhibited a tendency toward reduced IAV replication in the lungs as measured by qPCR, TCID50 and HAs compared to mice infected with IAV alone, accompanied by lower levels of virus-specific antibodies to IA …

Causal spectral segmenter: counterfactual graph reasoning for weakly supervised pathology segmentation.

MOTIVATION: Weakly supervised pathology segmentation aims to alleviate the reliance on costly pixel-level annotations, yet remains challenging due to severe tissue heterogeneity, complex morphological patterns, and strong contextual confounding in histopathology images. Existing transformer-based methods often learn spurious correlations between lesions and surrounding tissues, while graph-based approaches tend to suffer from feature over-smoothing, resulting in degraded boundary delineation and localization accuracy. RESULTS: To address these challenges, we propose a novel Causal Spectral Se…

Metachronous Cardio-Cerebral Infarction: A Rehabilitation Conundrum in Dual Pathology Recovery.

Cardio-cerebral infarction, the simultaneous (synchronous) or sequential (metachronous) occurrence of acute ischemic stroke and acute myocardial infarction, poses significant challenges in both acute care and rehabilitation. We describe a 52-yr-old man who presented with acute myocardial infarction followed by ischemic stroke diagnosed within hours, representing metachronous cardio-cerebral infarction. After initial thrombolysis and delayed percutaneous coronary intervention, he demonstrated significant cognitive and language impairments that limited participation in a standard outpatient card
