Construction and validation model of necroptosis-related gene signature associates with immunity for osteosarcoma patients.

Osteosarcoma is the most common malignant tumor in children and adolescents and its diagnosis and treatment still need to be improved. Necroptosis has been associated with many malignancies, but its significance in diagnosing and treating osteosarcoma remains unclear. The objective is to establish a predictive model of necroptosis-related genes (NRGs) in osteosarcoma for evaluating the tumor microenvironment and new targets for immunotherapy. In this study, we download the osteosarcoma data from the TARGET and GEO websites and the average muscle tissue data from GTEx. NRGs were screened by Cox

Predictors of urinary tract infection based on artificial neural networks and genetic algorithms.

BACKGROUND: Among women who present with urinary complaints, only 50% are found to have urinary tract infection. Individual urinary symptoms and urinalysis are not sufficiently accurate to discriminate those with and without the diagnosis. METHODS: We used artificial neural networks (ANN) coupled with genetic algorithms to evolve combinations of clinical variables optimized for predicting urinary tract infection. The ANN were applied to 212 women ages 19-84 who presented to an ambulatory clinic with urinary complaints. Urinary tract infection was defined in separate models as uropathogen coun

Identification of putative exported/secreted proteins in prokaryotic proteomes.

The increasing number of bacterial genomes being sequenced fuels an equal demand for methods to rapidly analyze the proteomes of these organisms. One group of proteins of pressing importance is the exported/secreted proteins, given their dominant immunogenicity and role in pathogenesis. With this in mind, a weight matrix algorithm and two artificial neural networks, one based on amino acid position within the N-terminus and the other on amino acid frequency, were developed for identification of such proteins. The neural networks and a hybrid method, combining the weight matrix algorithm and th

Towards better heartbeat segmentation with deep learning classification.

The confidence of medical equipment is intimately related to false alarms. The higher the number of false events occurs, the less truthful is the equipment. In this sense, reducing (or suppressing) false positive alarms is hugely desirable. In this work, we propose a feasible and real-time approach that works as a validation method for a heartbeat segmentation third-party algorithm. The approach is based on convolutional neural networks (CNNs), which may be embedded in dedicated hardware. Our proposal aims to detect the pattern of a single heartbeat and classifies them into two classes: a hear

A screening algorithm for the efficient exclusion of biliary atresia in infants with cholestatic jaundice.

BACKGROUND: Neonates with cholestasis may undergo many tests before biliary atresia (BA) or an alternative diagnosis is reached, and delayed intervention may worsen outcomes. An optimal diagnostic approach to reduce risk, cost, and delay has yet to be defined. The purpose of this study was to develop an algorithm that rapidly and accurately excludes BA for infants with cholestatic jaundice. METHODS: A single-center retrospective comparison of diagnostic workup was made between cholestatic infants with BA, and those without BA who underwent hepatobiliary iminodiacetic acid (HIDA) scan during a

The use of advanced machine learning to predict outcomes after atezolizumab plus bevacizumab for advanced hepatocellular carcinoma: a retrospective cohort study.

BACKGROUND: Combination immune checkpoint inhibitors are recommended as first-line therapy for advanced hepatocellular carcinoma. However, only a third of patients respond to treatment, and improved approaches to predict response are required. Using baseline clinical data, we aimed to use advanced machine learning models to predict overall survival and progression-free survival in patients with advanced hepatocellular carcinoma receiving atezolizumab plus bevacizumab. METHODS: This retrospective cohort study was conducted at 24 centres across eight countries. Patients aged 18 years and older

Prediction intervals for random-effects meta-analysis based on confidence distributions and Edgington's method.

Statistical inference about the average effect in random-effects meta-analysis has been considered insufficient in the presence of substantial between-study heterogeneity. Predictive distributions are well suited for quantifying heterogeneity since they are interpretable on the effect scale and provide clinically relevant information about future events. We construct predictive distributions accounting for uncertainty through confidence distributions from Edgington's p-value combination method and the generalized heterogeneity statistic. Simulation results suggest that 95% prediction intervals

Predicting human metabolic rate based on multiple physiological parameters: a pilot study.

Objective.Accurate prediction of human metabolic rate (MR) is essential for thermal comfort evaluation. However, traditional static lookup table methods lack adaptability in dynamic scenarios. As a preliminary study, this study aimed to reveal the correlation mechanism between four physiological parameters (heart rate, mean arterial pressure, skin conductance level, and ear temperature) and MR, comparatively analyze the performance of different input parameter combinations constructed by linear regression, support vector regression, and two-layer neural network (TLNN), and ultimately evaluate

Improving severity grading of chemotherapy-induced myelosuppression in AML via data-driven and model-based deep learning.

Chemotherapy-induced myelosuppression in acute myeloid leukemia (AML) frequently leads to life-threatening complications, yet current assessment standards lack the specificity required for personalized risk prediction. We present MM-AI-AML, a two-stage framework merging mechanistic mathematical modeling (MM) with artificial intelligence (AI) to predict myelosuppression severity using pre-treatment clinical data. Initially, a dynamic model simulating post-chemotherapy kinetics across four blood cell lineages was developed to derive a quantitative severity indicator, providing objective labels f

Model-free prognostication of non-linear time series.

OBJECTIVE: The COVID-19 pandemic has highlighted the importance of studying the course of infectious progression. Similar needs exist for time series of other origins. While models are commonly devised and fitted to the observed data, we recently demonstrated the feasibility to directly evaluate the noisy non-linear time series that characterize the occurrence. However, for practical utility, analytics alone has limited value. The requirement of forecasting - at least in the short term - needs to be met. METHODS: We initially utilized normalized new infections per day (7-day moving average fo

A Field-Based Predictive Model for Evidence-Based Classification in Male Footballers With Cerebral Palsy.

Over a 6-year research period, the classification of footballers with cerebral palsy (CP) has identified the best activity limitation tests that best capture the influence of eligible impairments on performance, leading to the implementation of a new classification system (i.e., FT1-FT3 sport classes). This study aimed to (i) assess the sensitivity of these tests to discriminate among sport classes; and (ii) estimate predictors of activity limitation outcomes to enhance accuracy in class allocation. A total of 177 international-level male CP footballers (25.90 ± 6.13 years.; 67.70 ± 11.49 kg;

Development and validation of a malignant risk prediction model for breast cystic-solid lesions based on clinical and ultrasound features.

OBJECTIVE: Accurate differentiation of benign and malignant breast cystic-solid lesions (BCSLs) is crucial due to the significant differences in their treatment strategies. This study aimed to develop and validate a malignant risk prediction model for BCSLs based on clinical and ultrasound (US) features. METHODS: A total of 312 patients with BCSLs were enrolled and divided into a training cohort (218 cases) and a validation cohort (94 cases). The least absolute shrinkage and selection operator (LASSO) was used to screen the most critical clinical and US features, and a prediction model was con

miRNA Target Prediction: An Overview of the Past and Current Tools.

MicroRNAs (miRNAs) are among the most studied molecules in recent years, and since their discovery, many miRNAs have been identified across various species. As members of the non-coding RNA family, miRNAs are key players in post-transcriptional gene regulation. These molecules can inhibit translation or promote degradation of messenger RNA (mRNA) by binding to the 3' untranslated region (UTR) of mRNA, thereby influencing almost all biological processes. To identify a miRNA's biological role, it is essential to predict the target sites to which it binds, a goal made possible through bioinformat

A machine learning approach coupling immediate and lagged drivers to predict sand and dust storms in Northern China.

Under global climate change, sand and dust storms (SDS) in Northern China have exhibited new spatiotemporal evolution characteristics. To investigate SDS driving mechanisms, this study integrated multi-source remote sensing and reanalysis data (2005-2024), employing SHapley Additive exPlanations (SHAP), receiver operating characteristic curve analysis method, and the extreme gradient boosting machine learning model. The results indicated that SDS frequency increased significantly after 2015, with a 47.6% rise in annual events (from 4 to 6 events/year), while high-intensity events declined from
