Results following liver transplantation for hepatocellular carcinoma have been dismal, attributed largely to recurrent disease locally or at distance sites. Undetected micrometastases or tumor that embolizes at the time of liver transplant from manipulation of the liver may account for these recurrences. Although FGFR2 is a well-validated oncogenic target, no selective FGFR2 inhibitors have been approved for clinical use. In this study, we report the discovery of 2H-pyrazolo[3,4-d]pyrimidin-4-amine derivative as novel, irreversible FGFR2 inhibitors. Ruxolitinib is a Janus kinase inhibitor effective against graft-versus-host disease and hemophagocytic lymphohistiocytosis. However, clinical management is challenged by potential drug-drug interactions and adverse events. Although therapeutic drug monitoring (TDM) can optimize drug safety and efficacy, real-world pharmacokinetic data are limited. To address this gap, we established a simple and sensitive LC-MS/MS method with a total run time of 8.1 min. The method demonstrated linearity over a range of 0.5-100 ng/mL.
