Biomaterial-based joint lubricants for osteoarthritis: emerging multifunctional designs and therapeutic strategies.

Osteoarthritis is a prevalent degenerative joint disease in which impaired lubrication can accelerate disease progression, making restoration of joint lubrication an important therapeutic avenue. This review summarizes emerging biomaterial-based joint lubricants and multifunctional designs that integrate lubrication with biological regulation and tissue regeneration. We highlight advances in hydrogels, microspheres, nanoparticles, and emulsions, focusing on material architectures, lubrication mechanisms, tribological performance, and biological functions including anti-inflammatory and antioxi

An engineered human placental organoid microphysiological system in a vascular niche to model viral infection.

The placenta forms the maternal-fetal interface to protect the developing fetus from xenobiotics or pathogens. However, the understanding of complex placental features and responses to pathogens are hindered due to the lack of near-physiological models. Here, we present an engineered human placental organoid microphysiological system (MPS) incorporated with vascular endothelium, which allows to recapitulate early placental features in a vascular niche. The MPS comprises a customized insert-based organ chip and a rocker, enabling in situ differentiation and formation of placental organoids from

PEG-Collagen Interpenetrating Networks Support Enhanced Vasculogenic Self-Assembly and Impact Cell-Mediated Remodeling.

The biophysical cues of natural and synthetic hydrogels, including stiffness and the rate of cell-mediated degradation, are often tuned to better understand how to form vessel networks in tissue constructs. Interpenetrating networks (IPN) combine the bioactivity and fibrillar architecture of naturally derived hydrogels and the tunability of synthetic hydrogels. We developed a poly(ethylene glycol) (PEG)-collagen (type I) IPN to investigate the interactive effects of stiffness, the rate of proteolytic degradation, and a fibrillar collagen network on the formation of microvascular networks and c

Functionalized peptide hydrogel to generate human insulin-producing cells in vitro.

Type I diabetes is a chronic disease that affects people worldwide. When insulin administration is no longer effective, transplantation of pancreatic islets represents an alternative for diabetics. However, islet grafting carries limitations, which include poor availability of donors, surgery risks, and lifelong immunosuppressive therapy. To address this, novel approaches, such as the use of soft hydrogels as vehicles of cells are being developed for tissue grafting applications. Self-assembling peptide hydrogels (SAPHs) are biocompatible and versatile materials widely used for both, three-dim

Kinetic degradation and biocompatibility evaluation of polycaprolactone-based biologics delivery matrices for regenerative engineering of the rotator cuff.

Whereas synthetic biodegradable polymers have been successfully applied for the delivery of biologics in other tissues, the anatomical complexity, poor blood supply, and reduced clearance of degradation byproducts in the rotator cuff create unique design challenges for implantable biomaterials. Here, we investigated lower molecular weight poly-lactic acid co-epsilon-caprolactone (PLA-CL) formulations with varying molecular weight and film casting concentrations as potential matrices for the therapeutic delivery of biologics in the rotator cuff. Matrices were fabricated with target footprint di

Three-Dimensional Printing Biologically Inspired DNA-Based Gradient Scaffolds for Cartilage Tissue Regeneration.

Cartilage damage caused by aging, repeated overloading, trauma, and diseases can result in chronic pain, inflammation, stiffness, and even disability. Unlike other types of tissues (bone, skin, muscle, etc.), cartilage tissue has an extremely weak regenerative capacity. Currently, the gold standard surgical treatment for repairing cartilage damage includes autografts and allografts. However, these procedures are limited by insufficient donor sources and the potential for immunological rejection. After years of development, engineered tissue now provides a valuable artificial replacement for ti

Long-Term Response to a Bioactive Biphasic Biomaterial in the Femoral Neck of Osteoporotic Rats.

Osteoporosis often leads to fragility fractures of the hip, resulting in impaired quality of life and increased mortality. Augmenting the proximal femur could be an attractive option for prevention of fracture or fixation device failure. We describe a tissue engineering based strategy to enhance long-term bone formation in the femoral neck of osteoporotic rats by locally delivering bioactive molecules; recombinant human bone morphogenic protein-2 (rhBMP-2), and zoledronic acid (ZA) by using a calcium sulfate/hydroxyapatite (CaS/HA) biomaterial. A defect was created by reaming the femoral neck …

Innovations in Airway Surgery.

The management of pediatric airway stenosis has evolved considerably over time. At the outset, dilation was the mainstay of management. In the 1900s, open surgery in the form of cricoid expansion procedures or resection procedures was the primary treatment with subsequent development of the slide tracheoplasty. Now in the twenty-first century, advances in endoscopic management, balloon dilation, and stenting, along with the advent of external scaffolds and tissue replacement continue to advance pediatric airway surgery.

One-year follow-up study of iBTA-induced allogenic biosheet for repair of abdominal wall defects in a beagle model: a pilot study.

PURPOSE: We evaluated the usefulness of biosheet, an in-body tissue-engineered collagenous membrane, as a novel repair material for abdominal wall defects in a beagle model. METHODS: Biosheets were prepared by embedding molds into subcutaneous pouches in two beagle dogs for 2 months, with subsequent storage in 70% ethanol. The obtained biosheets (thickness 0.5 mm, size 25 cm2) were implanted to replace same-size defects in the abdominal wall of two beagles in an allogenic manner. RESULTS: The biosheets were not stressed during suturing and did not split; moreover, patch implantation into the

A study protocol for a multicentre randomised clinical trial evaluating the safety and feasibility of a bioengineered human allogeneic nanostructured anterior cornea in patients with advanced corneal trophic ulcers refractory to conventional treatment.

INTRODUCTION: There is a need to find alternatives to the use of human donor corneas in transplants because of the limited availability of donor organs, the incidence of graft complications, as well as the inability to successfully perform corneal transplant in patients presenting limbal deficiency, neo-vascularized or thin corneas, etc. We have designed a clinical trial to test a nanostructured fibrin-agarose corneal substitute combining allogeneic cells that mimics the anterior human native cornea in terms of optical, mechanical and biological behaviour. METHODS AND ANALYSIS: This is a phas

Synthetic Poly(Vinylalcohol)-Based Membranes for Cartilage Surgery and Repair.

Cell-based therapies for cartilage repair are continually being developed to treat osteoarthritis. The cells are either introduced directly by intra-articular injection or via a cell-seeded matrix scaffold. Here, poly(vinylalcohol)-based membranes are developed to be used for mesenchymal stem cell implantation in cartilage repair procedures, having controllable physicochemical properties such as porosity, mechanical strength, and permeability, and a unique self-sealing property. The membranes possess a bilayer structure with a less porous layer providing mechanical strength and selective perme

Engineering cells with intracellular agent-loaded microparticles to control cell phenotype.

Cell therapies enable unprecedented treatment options to replace tissues, destroy tumors and facilitate regeneration. The greatest challenge facing cell therapy is the inability to control the fate and function of cells after transplantation. We have developed an approach to control cell phenotype in vitro and after transplantation by engineering cells with intracellular depots that continuously release phenotype-altering agents for days to weeks. The platform enables control of cells' secretome, viability, proliferation and differentiation, and the platform can be used to deliver drugs or oth

Effect of boron oxide addition on fibre drawing, mechanical properties and dissolution behaviour of phosphate-based glass fibres with fixed 40, 45 and 50 mol% P2O5.

Previous studies investigating manufacture of phosphate-based glass fibres from glasses fixed with P2O5 content less than 50 mol% showed that continuous manufacture without breakage was very difficult. In this study, nine phosphate-based glass formulations from the system P2O5-CaO-Na2O-MgO-B2O3 were prepared with P2O5 contents fixed at 40, 45 and 50 mol%, where Na2O was replaced by 5 and 10 mol% B2O3 and MgO and CaO were fixed to 24 and 16 mol%, respectively. The effect of B2O3 addition on the fibre drawing, fibre mechanical properties and dissolution behaviour was investigated. It was found t

Mesenchymal stem cells - a promising perspective in the orofacial cleft surgery.

Autologous bone grafts provide the golden standard for closure of oronasal fistulas in the cleft palate. Augmentation may be performed also by homografts and various xenogenic or alloplastic materials to prevent morbidity at the donor site but they may cause many problems (transmission of infections, immune response etc.). All the mentioned approaches also often reveal recurrences of the fistulas and prolong suffering of the cleft patients. Combination of mesenchymal stem cells (MSCs) and so called "platelet gel" seems to be a perspective method in this way. The platelet gel contains hydroxyap

Amniotic fluid stem cells in a bone microenvironment: driving host angiogenic response.

The repair of skeletal defects remains a substantial economic and biomedical burden. The extra-embryonic fetal stem cells derived from amniotic fluid (AFSCs) have been used for the treatment of large bone defects, but mechanisms of repair are not clear. Here we studied the potential contribution of human AFSCs to the modeling of an ectopic bone. We found that AFSCs are not osteogenic in vivo, and, compared to bone marrow-derived stromal cells, recruit more host CD31 and VEGF-R2 positive cells. Finally, when AFSCs were co-implanted with human-bone forming cells, a normo-osteosynthesis occurred,
