Biofilm formation during pneumococcal carriage imprints naturally acquired humoral immunity.

Streptococcus pneumoniae (Spn) colonization of the nasopharynx is a prerequisite for transmission and invasive disease. To investigate how repeated asymptomatic colonization shapes immunity and influences bacterial traits, we developed the Repeated Asymptomatic Murine Pneumococcal Colonization (RAMPC3) model using strains belonging to serotypes: 2 (D39), 3 (WU2), and 4 (TIGR4). Sequential colonization revealed strain- and exposure-order-dependent effects on bacterial burden, with initial colonization yielding robust carriage and subsequent exposures resulting in diminished burden and rapid cle

Isolation, identification, and biological characteristics of Mannheimia haemolytica from goats.

With the rapid expansion of the meat goat farming industry in China and intensified interregional movement of goats, respiratory diseases have become increasingly prevalent across domestic goat farms-particularly under suboptimal husbandry conditions-posing a significant challenge to effective disease prevention and control. Mannheimia haemolytica, a primary bacterial pathogen implicated in caprine respiratory disease, exhibits marked geographic variation in key phenotypic and genotypic traits, including serotype distribution and antimicrobial resistance profiles. To address this critical anim

Focused ultrasound-based acoustic holography for 3D cell patterning and biomimetic microenvironment construction.

Focused Ultrasound (FUS) provides a non-contact, directional acoustic field extensively employed in diverse biomedical applications. However, conventional concave transducers are physically constrained to a single fixed focus, lacking the flexibility for dynamic field modulation necessary to meet increasingly complex functional requirements. In this work, we report a focused ultrasound-based acoustic holography (FUS-AH) approach that enables concave transducers to achieve sophisticated spatial control and generate intricate multi-focal patterns. By systematically investigating the propagation

Curcumin ameliorates Salmonella-induced enteritis by restraining NF-κB signaling and restoring microbiota-SCFA homeostasis.

Foodborne Salmonella infection triggers excessive inflammation, mucosal injury and gut microbiota dysbiosis. Here, curcumin was evaluated as a dietary bioactive against Salmonella-induced enteritis using in silico target-network analysis, macrophage assays and a mouse infection model. Target-network analysis identified 79 shared targets associated with curcumin and intestinal inflammation. In vitro, macrophages were treated with curcumin at 40 and 80 μM before stimulation with lipopolysaccharide or Salmonella Enteritidis infection. In vivo, mice were challenged with S. Enteritidis and orally g

Global emergence of the chikungunya epidemic: A narrative review of the virus-kidney relationship in Pakistan and beyond.

Chikungunya, a vector-borne viral disease, has become a critical global health issue due to its capacity for widespread outbreaks, especially in tropical and subtropical regions, and its recent global expansion. The resurgence of Chikungunya virus (CHIKV) in Karachi, Pakistan, has amplified public health challenges, driven by factors such as urbanization, climate change, and socioeconomic vulnerabilities, including limited healthcare infrastructure. Clinically, the disease primarily manifests with fever, rash, and debilitating joint pain, which often leads to prolonged discomfort and decreased

HDAC11 deficiency improves muscle phenotype in a Duchenne muscular dystrophy murine model by reducing inflammation and fibrosis.

Duchenne muscular dystrophy (DMD) is a devastating genetic disorder caused by mutations in the dystrophin gene, which encodes for an essential protein to maintain muscle integrity. Loss of dystrophin leads to progressive muscle degeneration, chronic inflammation and replacement of muscle by fibroadipose tissue. To date there is no cure for DMD and the identification of novel molecular targets involved in disease progression is needed to design new therapies to slow DMD progression and prolong survival. Here, we show for the first time that genetic deficiency of HDAC11 in a mouse model of DMD h

Case study of resecabtagene autoleucel in a subject with diffuse cutaneous systemic sclerosis treated in the RESET-SSc trial.

Under compassionate use, CAR T cells have elicited durable remissions in patients with refractory systemic sclerosis (SSc). Here, we report on the safety, efficacy, and correlative data of the first SSc subject treated with a fully human CD19-CART cell therapy (resecabtagene autoleucel) in the RESET-SSc trial (NCT06328777). Nine days post-infusion, the subject experienced a brief grade 2 episode of cytokine release syndrome that resolved promptly with intravenous hydration and indomethacin. Immune-effector-cell-associated neurotoxicity syndrome was not observed. Seven days post-infusion, the s

Purified diet reduces intestinal IgA and alters the microbiota accordingly.

Diet can affect health directly or by altering the gut microbiota; thus, there are strong interrelationships between the gut immune system, gut microbiota and diet. This study examined the effects of ingesting AIN-93M purified diet (PD) on gut immune function and gut microbiota in DO11·10 mice, in which T cell-dependent and T cell-independent (TI) IgA can be analysed separately. Ingestion of the PD for 2 weeks reduced both T cell-dependent and TI secretory IgA in the faeces compared with non-PD, whereas the diet did not affect T cell-dependent and TI serum IgA. Ingestion of the PD had no effec

Obesity: pathophysiology and therapeutic interventions.

Over the past few decades, obesity has transitioned from a localized health concern to a pressing global public health crisis affecting over 650 million adults globally, as documented by WHO epidemiological surveys. As a chronic metabolic disorder characterized by pathological adipose tissue expansion, chronic inflammation, and neuroendocrine dysregulation that disrupts systemic homeostasis and impairs physiological functions, obesity is rarely an isolated condition; rather, it is frequently complicated by severe comorbidities that collectively elevate mortality risks. Despite advances in nutr…

A redox-responsive NIR fluorescent nanoprobe for tumor microenvironment-activated surgical navigation with submillimeter precision.

Surgical precision in tumor resection critically relies on real-time intraoperative imaging, yet conventional probes face limitations in specificity and spatiotemporal control. Here, we present a tumor microenvironment (TME)-activated near-infrared (NIR) fluorescent nanoprobe (DNS-DYE/PEG-NI) that integrates dual responsiveness to hypoxia and glutathione (GSH) for submillimeter-level surgical navigation. The system comprises a GSH-activatable NIR fluorophore (λex/em = 679/730 nm) quenched by 2,4-dinitrobenzenesulfonyl (DNS) moieties and hypoxia-sensitive amphiphilic PEG-NI micelles. Upon tumor

Understanding the influence of transfusion and blood loss on tranexamic acid concentration in scoliosis surgery with blood loss.

AIMS: Tranexamic acid (TXA) stabilizes clot formation by inhibiting fibrin degradation and improves postoperative outcomes. However, rare adverse events (e.g., thrombosis, seizures) warrant dose-risk evaluation. This study examines how perioperative blood loss and transfusion practices affect TXA concentrations during paediatric scoliosis surgery. METHODS: Forty-three patients undergoing scoliosis surgery with TXA were retrospectively analysed. The study assessed the impact of packed red blood cell (PRBC) transfusion on plasma TXA levels and whether maintaining concentrations ≥10 μg/mL correl

Harnessing the dual immunomodulatory function of myeloid-derived suppressor cells to reshape the inflammatory microenvironment for osteoarthritis therapy.

Osteoarthritis (OA) pathogenesis is profoundly influenced by dysregulated immune dynamics, where persistent interleukin-17 (IL-17)/T helper 17 (Th17) cell mediated inflammation coordinates with failed regenerative processes to perpetuate joint destruction. Here, we unveil the role of myeloid-derived suppressor cells (MDSCs) as dual-phase regulators that paradoxically orchestrate both inflammatory escalation and tissue repair in OA progression. Intra-articular administration of MDSCs in OA mice amplified IL-17 dependent inflammatory cascades and chemokine-driven leukocyte recruitment, revealing

Surgical technique for volar plate, collateral ligament reconstruction, and neglected dislocation of the proximal interphalangeal joint in the fifth finger: A case report.

INTRODUCTION AND IMPORTANCE: Chronic unreduced dislocations of the proximal interphalangeal joint are uncommon and management principles for these injuries have not been defined. The dislocation can be either volar or dorsal and closed reduction is rarely successful due to soft tissue contractures. CASE PRESENTATION: A 61-year-old woman presented with a 17-year-old deformity of her left fifth finger, sustained from a bathroom slip. Initially, the patient complained of pain and deformity. After failed of conservative treatment, patient was referred for surgical intervention. Physical examinati

Hypermethylation of the p16INK4a Promoter as an Early Epigenetic Event in Nodular Hyperplasia Associated with Secondary Hyperparathyroidism.

Secondary hyperparathyroidism (SHPT) associated with chronic kidney disease (CKD) is characterized by parathyroid hyperplasia, which progresses from diffuse-type to nodular-type lesions. Nodular hyperplasia in SHPT is often considered to exhibit monoclonal proliferation, suggesting a shift toward neoplastic behavior, but the molecular mechanisms underlying this transformation remain poorly defined. In this study, we analyzed 340 surgically resected parathyroid glands from long-term dialysis patients who met clinical indications for parathyroidectomy. Based on histological architecture, lesions

Progress on the HIF-1α/VEGF/VEGFR2 signal pathway in hepatic alveolar echinococcosis.

Alveolar echinococcosis (AE), a lethal parasitic zoonosis mimicking malignant tumors, progresses via hepatic infiltration and metastatic spread, causing multiorgan failure. Despite its clinical resemblance to cancer, molecular drivers of its aggressiveness remain poorly defined. Recent studies highlight perilesional angiogenesis as pivotal for lesion invasiveness, mediated by VEGF-driven pathological vascularization. VEGF not only fuels parasitic proliferation by creating nutrient-rich microenvironments but also engages crosstalk with host-parasite interactions, including immune evasion by Ech

Pathologically Confirmed Dual Coronavirus Disease 2019-Associated Tracheobronchial Aspergillosis and Pulmonary Mucormycosis in a Non-Endemic Region: A Case Report.

Background: Coronavirus disease 2019 (COVID-19) has led to the expansion of the spectrum of invasive fungal infections beyond traditional immunocompromised populations. Although COVID-19-associated pulmonary aspergillosis is increasingly being recognised, COVID-19-associated mucormycosis remains rare, particularly in non-endemic regions. Concurrent COVID-19-associated invasive tracheobronchial aspergillosis and pulmonary mucormycosis with histopathological confirmation is exceedingly uncommon and poses significant diagnostic and therapeutic challenges. Case presentation: We report the case of
