Ubiquitination of MHC class II molecules regulates B-cell development and response to antigens in mice.

Dendritic cells (DCs) and B lymphocytes produce major histocompatibility complex class II molecules (MHCIIs) in large amounts to maximize the display of peptides and fulfill their antigen-presentation functions. The surface expression of MHCIIs in these cells is regulated via the ubiquitination of a single conserved lysine residue in the cytoplasmic tail of all known β-chains. This modification is carried out mainly by the MARCH1 E3 ubiquitin ligase. In MARCH1-deficient DCs, the lack of MHCII ubiquitination results in its excessive accumulation at the plasma membrane, disorganizing lipid rafts

Type 2 diabetes impairs macrophage antimicrobial capability and increases susceptibility to sepsis.

Sepsis remains one of the primary causes of mortality in intensive care units, with an overall death rate of approximately 20%. Type 2 diabetes (T2D) exacerbates the incidence of infections, leading to increased long-term morbidity and mortality associated with sepsis. Macrophages are critical for the defense and clearance of invading pathogens during sepsis. Nevertheless, limited research has addressed the impact of T2D on macrophage dysfunction in sepsis conditions. In this study, we observed a significant impairment in the phagocytic and intracellular bacterial killing abilities of macropha

circACTR2 facilitates M1 macrophage activation and exacerbates renal inflammation in mice with chronic kidney disease via RUNX1/HuR/miR-9-5p.

This study elucidates the regulatory role and molecular mechanism of the circular RNA actin-related protein 2 homolog (circACTR2) in macrophage phenotypic transformation and renal injury in chronic kidney disease (CKD). A mouse model of CKD was established via adenine administration. An in vitro inflammatory model was generated using lipopolysaccharide-stimulated RAW264.7 macrophages. Results revealed that circACTR2 expression was significantly elevated in CKD models. Silencing of circACTR2 attenuated renal dysfunction, ameliorated histological injury, and reduced inflammatory cytokines in CKD

Single-cell analysis identifies CPT1a-associated metabolic remodeling in human NK cells during COVID-19.

Natural killer (NK) cells are critical for early antiviral immunity, yet their metabolic regulation during acute human viral infection remains incompletely understood. We analyzed NK cell activation and metabolic reprogramming in 47 vaccinated individuals with mild breakthrough SARS-CoV-2 infection and 20 matched healthy control subjects. COVID-19 patients exhibited elevated plasma interferon α and NK cell activation markers (CD69, CD38), alongside increased basal STAT5 phosphorylation, consistent with IL-15-mediated signaling. Functionally, NK cells from infected subjects displayed heightened

Mitochondrial potential reflects T cell fitness and function during cancer immunotherapy.

Checkpoint inhibitors have transformed cancer treatment, yet predicting responses remains challenging. Mitochondrial quality decreases in tumor infiltrating lymphocytes and correlates with impaired antitumor immunity in animal models. Mitochondrial membrane potential (MMP) increases with T cell activation and may also indicate cellular dysfunction. Here, we assessed the MMP of tumor-associated T cells as an indicator of cell phenotypes and immunotherapy responses in non-small cell lung carcinoma and clear cell renal cell carcinoma patients. Primary tumors were collected followed by analysis of

Sleep interruption aggravates sepsis by rewiring the macrophage immune response.

Sepsis is the leading cause of death in hospitals and is very common in intensive care units (ICUs). Sleep is frequently interrupted in the hospital setting, especially within the ICU. Patients who sleep poorly have worse outcomes, such as increased mortality and longer hospital stays; however, the molecular basis remains poorly understood. In this study, we utilized a mouse model to investigate the impact of sleep interruption on subsequent sepsis. We found that sleep interruption aggravated sepsis, as evidenced by higher mortality rates (88% in mice with interrupted sleep vs 57% in mice with

NDRG3 is essential for sustaining antigen-driven T cell responses by protecting against restimulation-induced cell death.

During the response to infections and pathogenic challenges, T cells must expand profoundly and be resilient to repetitive restimulation to clear the ongoing assault and protect the host. This process of prolific expansion is tightly regulated to quickly provide a robust pool of pathogen-fighting T cells, yet also limit excessive expansion to prevent inadvertent damage to host tissues. Restimulation-dependent pro-growth and pro-death signals help regulate this delicate balance of T cell expansion to maintain both host and T cell homeostasis. We have discovered that NDRG3 is a critical determin

Metabolic adaptations of inflammatory macrophages govern ferroptosis susceptibility via the GCH1-BH4-iNOS axis.

In inflammatory tissue niches, macrophages encounter intense oxidative stress due to their own production of reactive oxygen and nitrogen species as part of antimicrobial defense. Our findings reveal that inflammatory macrophages deploy distinct, context-dependent redox-protective mechanisms to survive this self-inflicted stress, thereby avoiding ferroptotic cell death. Specifically, LPS-activated macrophages, M(LPS), rely on the GTP cyclohydrolase 1 (GCH1)-tetrahydrobiopterin (BH4) pathway for ferroptosis resistance, whereas LPS + IFN-γ-activated macrophages, M(LPS-IFN-γ), depend primarily on

Hidden effects of polymyxin B: Its role in activating TLR2-mediated inflammatory responses.

Polymyxins are cyclic cationic lipopeptide antibiotics that disrupt both the outer and inner membranes of Gram-negative bacteria. They also bind to bacterial lipopolysaccharides (LPSs), thereby inhibiting LPS recognition by Toll-like receptor 4 (TLR4) and preventing macrophage activation. However, the immunomodulatory effects of polymyxin B during infection, in which multiple host- and bacteria-derived factors coexist, remain unclear. In this study, we examined the impact of polymyxin B, a representative polymyxin antibiotic, on mouse macrophage responses in the presence of interferon γ (IFN-γ

Tristetraprolin protects against ozone-induced acute lung injury and inflammation in mice.

Tristetraprolin (TTP) is an anti-inflammatory protein that mediates messenger RNA (mRNA) decay of certain transcripts, especially those encoding proinflammatory cytokines. TTP modulates various pathological outcomes in diverse inflammatory diseases; however, its role in ozone (O3)-induced acute lung injury (ALI) has never been tested. Here, we hypothesized that the loss of TTP would exacerbate O3-induced ALI and that the systemic overexpression of TTP would mitigate O3-induced ALI. Accordingly, TTP-knockout (TTPKO), airway epithelial cell-specific TTP-deficient (EpiKO), myeloid cell-specific T

Rytvela, an allosteric modulator of the interleukin-1 receptor, prevents preterm birth and neonatal complications in mice while nifedipine is ineffective.

Preterm birth is the leading cause of neonatal morbidity and mortality. Management of preterm labor relies on the use of tocolytics, like nifedipine, which fail to prevent preterm birth and related morbidities. Interleukin-1β plays significant roles in the pathophysiology of preterm birth including inflammation and uterine activation. The interleukin-1 allosteric receptor antagonist rytvela effectively suppresses inflammation and prevents preterm birth. We compared side-by-side rytvela and nifedipine in affecting preterm birth and offspring outcomes in a treatment modality approach. In a model

Unconventional functions of natural killer cells in homeostasis and disease.

NK cells are classically defined by their rapid cytotoxicity against tumor cells and infected cells and by early inflammatory cytokine production. However, unconventional roles for NK cells as regulators of immunity and tissue homeostasis have recently been uncovered. Beyond their classical roles, NK cells can orchestrate leukocyte trafficking, curtail responses of other immune cells, remove protein aggregates, support pregnancy, and contribute to healthy tissue regeneration. We discuss the importance of these myriad functional activities of NK cells in infection, cancer, autoimmunity, atopic

Contributions of T-helper 9 cells in endometriosis-associated inflammation and lesion growth.

Endometriosis is an inflammatory gynecologic disease characterized by ectopic growth of endometrial-like tissue, resulting in pelvic pain and infertility. T-helper 9 (Th9) cells play a known role in various chronic inflammatory diseases. Despite parallels between endometriosis and Th9-driven diseases, their role in endometriosis has not been extensively explored. We investigated Th9 cell involvement in endometriosis pathophysiology using human tissue samples, in vitro experiments with human-derived Th9 cells, and in vivo experiments to shed insight on the impact of adoptively transferred Th9 c

Invariant chain controls HLA-DQ folding and shapes the HLA class II proxiome.

The invariant chain (Ii) trimerizes in the endoplasmic reticulum and swiftly associates with nascent alpha and beta chains of major histocompatibility complex class II (MHCII) molecules. Ii was ascribed many roles pertaining to the antigen presentation capacity of MHCII molecules. The lack of Ii in knockout mice revealed its isotype- and cell type-dependent impact over the folding and trafficking of MHCII molecules. In humans, the role of Ii was mainly assessed in transfected cells. Here, using the CRISPR-Cas9 system, we addressed the impact of inactivating the Ii genes on the expression and f

Highly focused human CD8+ T-cell response in the lower airways during acute influenza infection.

Lung tissue-resident CD8+ T cells facilitate viral clearance and protective immunity to influenza viruses in animal models. Their role during acute human infection is not clear. Here we use bronchoalveolar lavage samples collected from human subjects naturally infected with influenza B virus to show that influenza-specific CD8+ T cells are expanded in the lower airways during acute infection and target only a few epitopes from phylogenetically conserved internal influenza virus proteins. The lower airway influenza-specific CD8+ T-cell immunodominance hierarchy is different from the hierarchy o

The emerging roles of NFIL3 in immunity.

NFIL3 (E4BP4) is a basic leucine zipper transcription factor best known for its roles in circadian regulation. It functions primarily as a transcriptional repressor through competitive DNA binding and the recruitment of histone-modifying complexes. More recently, NFIL3 has emerged as a critical regulator of immunity. It is essential for the development of multiple immune cell subsets, including NK cells and CD8+ dendritic cells, and modulates the activity of many others. For instance, NFIL3 influences B cell class switching and cytokine production in T helper subsets and macrophages. Its expre

The autoantibody β1-AA contributes to N/OFQ-exacerbated ventricular arrhythmias following acute myocardial ischemia.

Ventricular arrhythmias are a primary cause of sudden cardiac death after acute myocardial infarction (AMI). Both β1-adrenergic autoantibodies (β1-AA) and nociceptin/orphanin FQ (N/OFQ) are implicated in AMI, but their potential interaction in ischemic arrhythmogenesis remains unclear. This study investigated the interplay between N/OFQ and β1-AA and the underlying mechanism. A rat model of AMI was established by ligating the left anterior descending coronary artery. Rats were divided into Sham, coronary artery occlusion (CAO), CAO+NOP receptor antagonist (C-24), CAO+β1-AA+metoprolol, CAO+β1-A

Mechanism of SGLT2 inhibitors in improving immune imbalance in diabetic mice with sepsis via regulating macrophage M2 polarization through the APOC1/HMGB1/TLR4 axis.

This study aimed to investigate the mechanism by which the sodium-glucose cotransporter-2 (SGLT2) inhibitor dapagliflozin improves immune imbalance in sepsis with diabetes, focusing on its regulation of macrophage M2 polarization through the APOC1/HMGB1/TLR4 axis. A mouse model of type 2 diabetes was established through intraperitoneal injection of streptozotocin, after which sepsis was induced by cecal ligation and puncture. For in vitro studies, RAW264.7 macrophages and bone marrow-derived macrophages were exposed to high glucose and lipopolysaccharide. Molecular docking was employed to exam
