Fully and spontaneously degradable carboxymethyl chitosan/oxidized hyaluronic acid-based membrane promoting three-dimensional tissue remodeling in vitro and in vivo.

Remodeling three-dimensional (3D) tissues presents a significant challenge in the treatment of damaged body tissues. Although stem cell-based therapies show promise, current delivery strategies-such as cell sheets, pre-formed constructs, and conventional scaffolds-often face limitations due to complex fabrication processes, prolonged in vitro culture, unsustainable mechanical properties, and non-degradable materials that hinder cell-to-cell communication and complete tissue integration. In this study, we present a fully degradable polysaccharide composite membrane developed through the lyophil

Highly Mechanically Robust Human-Derived Hydrogels for Tissue Engineering Applications.

Hydrogels are widely used in tissue engineering (TE) for their ability to replicate the extracellular matrix, providing a supportive environment for cell proliferation and tissue regeneration. However, their inherently low mechanical strength often restricts their use in load-bearing biomedical applications. To address these limitations, this study aimed to develop the first human-derived double-network (DN) hydrogel by combining human methacryloyl platelet lysates (hPLMA) and the natural polysaccharide chitosan (CHT) through sequential covalent and ionic crosslinking. The resulting DN hydroge

Prevention of brain scarring during cranial reconstruction through a bioactive polymer coating.

Although cranioplasty is a long-established surgical procedure used to repair or reshape skull defects, it is not devoid of side effects arising from the formation of a scar tissue between dura, implant, and subcutaneous tissue. To minimize these complications, we developed a bioactive coating comprising poly(vinyl alcohol) (PVA) electrospun fibers loaded with a polyanhydride based on betulin disuccinate and a tricarboxylic derivative of poly(ethylene glycol) (PEG-DBB), suitable for the modification of the surface of titanium alloys. Although cell viability assays proved biocompatibility of fi

Green synthesis of caffeine-catalyzed citric acid-PPG/PEG crosslinked alginate hydrogel scaffolds for prospective biomedical applications.

Alginate, a natural polysaccharide [(1 → 4)-linked β-D-mannuronate and α-L-guluronate], is commonly used in hydrogel form in the biomedical field, including wound healing, drug delivery, and tissue engineering applications. The type and density of cross-linking are key parameters determining the physicochemical and mechanical properties of the hydrogel. However, most of the crosslinking reagents used are generally toxic, requiring extensive modifications and limiting their applicability. Alginate readily forms ionic hydrogels through coordination with divalent cations such as Ca2+ and Ba2+; ho

Extracellular matrix-targeted biomaterials and nanocarriers for pelvic-floor repair/regeneration in stress urinary incontinence: local modulation of the MMP/TIMP axis and NF-κB/MAPK-linked remodelling programs.

Stress urinary incontinence (SUI) is associated with pelvic-floor extracellular-matrix (ECM) remodelling, including an imbalance between matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) that can weaken periurethral load-bearing connective tissue. This Review frames SUI as a clinically relevant ECM-failure niche and synthesises ECM-targeted biomaterials and nanocarriers designed to rebalance the MMP/TIMP axis while enabling pragmatic pharmacodynamic readouts for early translation. We organise recent preclinical and early translational work across three modali

Immune and Stem Cell-Mediated Bactericidal Amplification and Bone Remodeling for Infection Clearance and Osteointegration.

Implant-related infections reduce mesenchymal stem cell accumulation near implant sites while suppressing the viability and osteogenic capacity of these stem cells. Here, we show that pH-responsive biomaterial interfaces exploit infection-induced acidification to coordinate antimicrobial action with stem and immune cell functional reprogramming. The system can release bactericidal peptides for direct bacterial elimination, whereas indirect bactericidal enhancement occurs in the presence of magnesium ions and cell-targeting peptides, which enable a positive modulation of immune responses and re

ECM-Mimetic Biomineralized Ti Implant Coating Mediates Quiescence of Osteoclast Precursor Cells to Restore Osteogenesis-Angiogenesis Coupling for Accelerating Osteoporotic Fracture Repair.

Titanium (Ti) implants are widely used for treating bone fractures, but their clinical success is often limited by high failure rates due to the excessive osteoclastogenic activation of osteoclast precursor cells (OPCs) at the implantation site, particularly in patients with osteoporosis. Notably, the excessive osteoclastic differentiation of OPCs potently disrupts the osteogenesis-angiogenesis coupling to promote osteoclast-mediated osteolysis while dismantling the pro-osteogenic type H vascular niches, resulting in elevated risk of implant destabilization. To address this issue, here we repo

Multifunctional injectable hydrogels with self-healing and electroconductive properties as a potential platform for neural tissue engineering: Integration of polypyrrole and IKVAV peptide into modified alginate-hyaluronic acid networks.

Injectable biomaterials have emerged as a promising option for minimally invasive tissue repair, especially in cases with small and irregular lesions. In combination with electroconductivity and self-healing abilities, these materials provide unique benefits for neural tissue engineering. They promote adaptive defect filling, assist in post-injection structural repair, and improve bioelectrical signaling. The current study focuses on the synthesis and characterization of injectable, self-healing, electroconducting hydrogels, which are intended to serve as a potential platform for addressing sm

A sustained living coating for infectious keratitis therapy via probiotic consumption and production effects.

Excessive inflammation and infection are critical factors leading to biomaterial failure and delayed wound healing. Surface modification of biomaterials with drug-loaded coatings can effectively address these issues. Traditional drug-loaded coatings often suffer from diminishing drug concentrations and the development of bacterial resistance. To address these issues, we developed a living coating using Lactobacillus plantarum as a metabolic factory for sustained anti-inflammatory and antimicrobial effects for up to 10 weeks. This innovative approach leverages continuous consumption and product

Protective effects of cross-linked poly(N-acryloyl glycine) nanoparticles in type 2 diabetes-induced complications: in vivo study.

Chronic hyperglycemia-induced ROS and systemic inflammation can lead to damage to peripheral and cardiac nerves as well as the autonomic nervous system. This damage progresses slowly and occurs over a long period of time. This collective damage to vital organs of the body leads to fatal effects such as cardiac arrest or extreme discomfort to patients, and can cause peripheral infections and neuropathic pain. There are limited therapeutic options available for progressive diabetic peripheral neuropathy (DPN) and diabetic cardiac neuropathy (DCN) limited to blood glucose management. Along this l

Bioactive molecule-derived covalent organic frameworks as biocompatible scaffolds for targeted AKR1B10 inhibition in alcoholic liver disease.

Alcoholic liver disease (ALD) incidence is rising globally, yet effective targeted therapies remain elusive. In recent years, bioactive molecules have gained prominence in liver disease research. However, no natural inhibitors targeting key drivers of ALD have yet to be reported. Herein, we first identified AKR1B10 as a key pathogenic driver through transcriptomic analysis of 33 ALD patients and 23 healthy controls. Through virtual screening, we discovered four inhibitors targeting this enzyme, including ellagic acid, vitamin B8, hematoxylin, and porphyrin. Inspired by these findings, we devel

Drug-coated balloon angioplasty for coronary and peripheral artery disease: latest evidence and clinical indications.

Drug-coated balloons (DCBs) are devices used for the treatment of both coronary artery disease (CAD) and peripheral artery disease (PAD). One of the hypothesized advantages of DCB angioplasty over stent implantation is that DCB angioplasty does not result in the presence of a permanent metallic scaffold in the vessel wall. However, DCB angioplasty also has some important limitations, such as a potentially lower efficacy compared with other modalities; therefore, the role of DCBs in the treatment of CAD and PAD is not fully defined. Over the past 20 years, many clinical trials have been perform

Decellularized extracellular matrix bio-patch with strong and biodegradable wet adhesive interface for sutureless soft tissue defect repair.

Decellularized extracellular matrix-based bio-patches offer superior biocompatibility and bioactivity, making them an ideal implantation material for soft tissue defect repair. However, the fixation of these patches typically relies on sutures or staplers, causing secondary tissue damage, stress concentration, and excessive inflammatory cell infiltration. Here, we develop a novel self-adhesive small intestinal submucosa patch (A-SIS) fabricated by in situ polymerizing a biodegradable hydrogel adhesive layer on the surface of the SIS patch. The A-SIS demonstrated increased tensile strength, bio

Photoresponsive Blood-Derived Protein Hydrogels Packed with Bioactive Carbon Dots Modulate Mitochondrial Homeostasis and Reprogram Metabolism for Chronic Wound Healing in Diabetes.

Autologous platelet concentrates (APC) represent a class of personalized regenerative materials for vascularized tissue regeneration. However, shortcomings including poor controllability of gel formation, lack of reactive oxygen species (ROS) scavenging ability, and deficient anti-inflammatory capacity restrict the tissue healing outcomes of APC. This study proposes an APC-based synergistic platform (CurCDs@iPRF-MA) for the treatment of chronic wounds in diabetes. Such a platform is composed of injectable platelet-rich fibrin (iPRF), gelatin methacryloyl (GelMA), and a carbogenic nanodrug from

Poly(L-lactic acid)/gelatin microfiber membrane loaded with mitomycin C promoting bladder defect repair by anti-fibrosis and antibacterial action.

When the bladder is affected by factors such as tumors, tuberculosis, or trauma, its normal structure and function can be severely damaged. At this point, bladder replacement or enlargement surgery may become necessary to restore its structure and improve the patient's quality of life. Traditional bladder reconstruction surgery, specifically ileum augmentation cystoplasty, involves a complex surgical process, along with relatively large operative wounds and possible intestinal obstruction complications. There is also an urgent need for suitable bladder replacement materials. Over the past deca

Transplantation of olfactory ensheathing cells embedded in the GelMA-F127DA hydrogel promotes the repair of spinal cord injury.

Spinal cord injury (SCI) is a highly disabling condition. Olfactory ensheathing cells (OECs) are important transplantable cells for treating spinal cord injury (SCI). However, directly transplanting cells into the injured area may result in the loss of transplanted cells. Additionally, secondary inflammatory responses reduce the survival of transplanted cells. In this study, a hydrogel with favorable properties, including plasticity, strong adhesion, mechanical stability, shear-thinning behavior, and pore size suitable for OEC growth and axon extension, was fabricated from methacryloylated gel

Osteogenic Effects of Ion Released from Biodegradable Metallic Magnesium and Calcium Coating.

This study investigates the effects of ion release and pH elevation from a biodegradable metallic magnesium (Mg) -30 calcium (Ca) coating on osteogenesis using osteoblast-like cells. The coating, formed on titanium (Ti) via magnetron sputtering, has previously been shown to enhance osteogenesis by promoting calcite formation on the Ti surface upon degradation in vitro study. However, the individual and combined roles of released Mg2+, Ca2+, and pH elevation remain unclear. To clarify these effects, culture media supplemented with Mg2+ and Ca2+ salts are prepared. Mg2+ at 4-5 mm promotes early

Augmenting bioactivity of in-situ captured Gd3+/Dy3+co-doped bio-glass by bisphosphonated chitosan-reinforced polyvinyl alcohol hydrogel scaffold with osteogenic and angiogenic features.

Conventional treatments display the limited ability to treat critical sized bone defects. In this context, the multifunctional hydrogels deliver a new treatment methodology to treat the critical bone disorders. Herein, a three-dimensional porous composite hydrogel with superior mechanical strength and the potential to promote osteogenesis has been fabricated using binary doping of Gd3+/Dy3+ in S53P4 bio-glass integrated PVA/chitosan. The alendronate loaded 10GDBGPC hydrogel provided sustained drug release and their 21 days of immersion in simulated body fluid (SBF) promoted apatite formation.

Biofilm formation and role of other pathogenic factors in the virulence of Staphylococcus epidermidis clinical isolates.

Medical device-associated infections represent a significant healthcare challenge, as sterilization of the biomaterial often necessitates device removal. The most frequently isolated microorganism in these infections is Staphylococcus epidermidis, a skin commensal capable of causing a wide range of nosocomial infections. The primary virulence factor of S. epidermidis is biofilm formation, which decreases antibiotic efficacy and host immune response. However, additional factors play crucial roles in infection establishment. Understanding the interplay between virulence factors is essential to d
