Clarity and inclusivity as precursors to translating yoga health evidence: a diverse cross-sectional study on barriers, facilitators, and associations of yoga.

BACKGROUND: Translating evidence-based interventions (EBIs) to advance health behaviors and well-being takes 15-17 years. This translational gap is lengthened for specific EBI and populations, such as yoga for people of color, with higher weight, or who live in rural areas. Yoga principles have positively impacted myriad populations and outcomes and align with scientific rigor, but misinformation persists. Applying a translational lens to input from under-represented yoga end-users may speed the gap. PURPOSE: To examine how underrepresented yoga end-users perceive yoga with consideration to y

Evaluating the Environmental Impact of Clinical Research: A Full Life Cycle Analysis of a French Academic Randomised Clinical Trial.

Climate change poses the greatest threat to human health in the 21st century. The healthcare sector contributes approximately 5% of global greenhouse gas emissions and has a significant environmental impact. Although clinical trials are crucial for identifying effective and safe treatments and preventing disease, their environmental impact is poorly documented. Our study aimed to assess the environmental impact of a publicly funded, academic clinical trial by adapting life cycle assessment (LCA) methodology to clinical research. We performed a retrospective, simplified, full LCA using the EF 3

Metabolic crosstalk in breast cancer in patients with Type 2 diabetes mellitus: pathophysiology and nanotherapeutic opportunities for clinical translation.

The intersection of Type 2 Diabetes Mellitus (T2DM) and breast cancer defines a complex onco-metabolic axis where chronic hyperglycaemia, hyperinsulinemia, and oxidative stress synergistically drive malignant transformation, metabolic reprogramming, and chemoresistance. Despite epidemiological evidence linking these conditions, clinical management typically addresses each disease individually according to established protocols; however, the presence of T2DM significantly alters tumour biology, therapeutic response, and outcomes, necessitating greater consideration of metabolic context in breas

Superoxide Dismutase-Centered Modulation by Curcumin in Cardiovascular Diseases: Mechanistic Insights and Translational Implications.

Cardiovascular diseases (CVD) remain the leading global cause of morbidity and mortality, driven in part by dysregulated redox homeostasis and chronic inflammation. Superoxide dismutase (SOD), a key enzymatic defence against reactive oxygen species (ROS), plays a central role in maintaining cardiovascular integrity through regulation of oxidative stress across cytosolic (SOD1), mitochondrial (SOD2) and extracellular (SOD3) compartments. Impairment of SOD function contributes directly to endothelial dysfunction, myocardial injury and vascular remodelling. Curcumin (Cur), a pleiotropic polypheno

From FLOW to translational positioning in chronic kidney disease: mechanistic complementarity of SGLT2 inhibitors and GLP-1 receptor agonists.

Chronic kidney disease is a central node of the cardio-kidney-metabolic continuum and carries substantial residual cardiovascular and kidney risk. Sodium-glucose cotransporter 2 inhibitors (SGLT2i) have reproducibly reduced kidney disease progression and heart failure risk in CKD outcomes trials, whereas GLP-1RAs have established cardiovascular benefit across cardiometabolic populations. The FLOW trial (Evaluate Renal Function With Semaglutide Once Weekly) established hard kidney-outcome evidence for semaglutide in type 2 diabetes with CKD, although whether this renal benefit represents a broa

From molecular networks to translational intervention: current progress in the mechanisms of gemcitabine resistance in bladder cancer.

Bladder cancer remains one of the most common malignancies of the urinary tract. Although the treatment landscape has expanded rapidly in recent years, gemcitabine still occupies a central position in intravesical treatment for non-muscle-invasive bladder cancer, in perioperative systemic therapy for muscle-invasive disease, and in platinum-based first-line regimens for advanced urothelial carcinoma. Yet the long-term benefit of gemcitabine is frequently curtailed by primary non-response or acquired resistance. In practice, this problem is often recognized only after radiographic progression o

Mesenchymal stem cell-derived secretome in traumatic brain injury: Stage-specific paracrine mechanisms and translational challenges.

Traumatic brain injury (TBI) is a stage-dependent disorder that evolves from acute neuroinflammation, blood-brain barrier (BBB) disruption, oxidative stress, excitotoxicity, and apoptosis to subacute neurovascular remodeling and chronic impairment of neuroregeneration and circuit repair. Current clinical management improves survival by stabilizing physiological status and limiting secondary injury, but it rarely rebuilds damaged neural networks or restores long-term neurological function. Mesenchymal stem cells (MSCs) have therefore attracted attention as regenerative candidates, with increasi

Growth factor applications and clinical translation: advances and challenges.

BACKGROUND: Growth factors (GFs), as key molecules regulating cellular processes, demonstrate therapeutic potential in fields such as regenerative medicine, oncology, and metabolic regulation. However, inherent pharmacokinetic limitations, off-target effects, and safety concerns significantly hinder clinical translation. Although in-depth studies on mechanisms of action, smart delivery systems, and regulatory networks have improved GFs' stability, targeting, and safety - leading to clinical approvals of some related drugs - high costs, complex large-scale production processes, and the intricac

Single-cell and spatial transcriptomics analysis of osteoarthritis: pathway regulation, cell interaction networks, and therapeutic translation.

BACKGROUND: Osteoarthritis (OA) transcends the outdated paradigm of mere "wear and tear" cartilage loss. It is now recognized as a complex, whole-organ disease driven by multifaceted interactions among diverse cell populations across synovium, cartilage, subchondral bone, and infrapatellar fat pad. The profound heterogeneity within these tissues and the spatiotemporal dynamics of pathological processes have, until recently, remained a "black box," limiting our understanding of disease initiation and progression. The convergence of single-cell RNA sequencing (scRNA-seq) and spatial transcriptom

Overcoming Immunological Barriers in MSC-Derived Insulin-Producing Cells through CRISPR-Based Hypoimmunogenic Engineering and Translational Perspectives for Type 1 Diabetes.

Mesenchymal stromal cell (MSC)-derived insulin-producing cells (IPCs) represent an emerging strategy for β-cell replacement in type 1 diabetes mellitus (T1DM) owing to their differentiation potential, intrinsic immunomodulatory properties, and lower tumorigenic risk compared with pluripotent stem cell-derived platforms. However, accumulating evidence indicates that differentiation-associated immunogenicity, context-dependent immune recognition, and recurrent autoimmune responses may substantially limit long-term graft survival and therapeutic durability following transplantation. Advanced microfluidic and 3D cell culture platforms for modeling vascularization in diabetic foot ulcers: A systematic review of translational challenges and perspectives.

The healing process of diabetic foot ulcers (DFUs) presents a slow pattern with an increased risk of infections, ischemia, and thrombosis correlated with high levels of reactive oxygen species production. Vascular injury is one of the factors contributing to the difficulty of wound healing in diabetic patients. Although the understanding of the pathophysiology of DFUs has significantly increased in recent years, associated treatments still have a high level of failure, leading to high morbidity rates, mortality, and amputations. Three-dimensional (3D) cell culture platforms offer a new approac

Translational Evaluation of a Disodium Adenosine Monophosphate (AMP2Na)-Based Topical Formulation for Physiology-Aligned Skin Rejuvenation: Integrated In Vitro, Ex Vivo, and Clinical Evidence.

Skin aging stems from intrinsic decline and external stressors that induce oxidative stress and mitochondrial damage, ultimately lowering cellular energy production and slowing epidermal turnover to cause wrinkles, dryness, and pigment imbalances. While disodium adenosine monophosphate (AMP2Na) is hypothesized to enhance cellular adenosine triphosphate production and restore epidermal metabolism, its broader anti-aging effects have remained underexplored. To address this, a multi-tiered study integrating in vitro, ex vivo, and clinical investigations was conducted. Specifically, a 12-week expl

Bone marrow-derived mesenchymal stem cells for ischemic stroke: mechanisms, delivery strategies, clinical evidence, and translational challenges.

Ischemic stroke remains a major global health burden because of its high incidence, disability, and mortality. Current reperfusion therapies are constrained by narrow therapeutic windows and have limited capacity to reverse established neuronal injury or promote long-term functional recovery, highlighting the need for strategies that support neural repair and neurovascular remodeling. Bone marrow-derived mesenchymal stem cells (BMSCs) have been widely investigated as candidate therapeutic products for ischemic stroke because of their paracrine, immunomodulatory, trophic, angiogenic, and microe

Multifunctional Hybrid Nanomedicines for Neurodegenerative Diseases: A Paradigm Shift from Rational Design to Clinical Translation.

Effective drug delivery to the central nervous system remains a persistent challenge owing to the blood-brain barrier (BBB), heterogeneous pathological microenvironments, and the limited capacity of conventional carriers to achieve precise spatiotemporal control over drug release. Multifunctional hybrid nanomedicines (MHNs), defined here as engineered nanosystems that integrate two or more functional modules, such as targeted delivery, controlled release, and/or theranostic capabilities within a single platform, offer a transformative approach to neurotherapeutic design. Biomaterials and Nanoparticle-Based Therapeutics in Neurodegenerative Diseases: Bridging the Gap Between Innovation and Translation.

Neurodegenerative diseases, including Alzheimer's, Parkinson's, Huntington's, amyotrophic lateral sclerosis, and multiple sclerosis, represent a growing global health crisis characterized by irreversible neuronal loss, protein aggregation, chronic neuroinflammation, and mitochondrial dysfunction. Central to their therapeutic intractability is the blood-brain barrier (BBB), a highly selective neurovascular interface that excludes nearly 98% of conventional pharmacological agents from the central nervous system (CNS). Nanoparticle- and biomaterial-based delivery platforms have emerged as promisi
