Up-regulation of the chemokine CCL18 by macrophages is a potential immunomodulatory pathway in cutaneous T-cell lymphoma.

Mycosis fungoides (MF) is the most frequent form of cutaneous T-cell lymphoma (CTCL), which can deteriorate from patch stage to dermal-based tumors and systemic involvement in years. The interaction of chemokines in the skin with CTCL cells might have implications for the pathogenesis of the disease. In this study, we show by PCR analysis and immunofluorescence staining that the chemokine CCL18 is present in skin biopsy specimens of patients with MF and its precursor form parapsoriasis en plaque but not in healthy tissue. In addition, the serum levels of CCL18 were increased threefold in MF pa

Sialic Acid-Targeting Lectin-Conjugated Oxygen Nanosomes Alleviate Hypoxia and Promote Macrophage Activation in Pancreatic Cancer Cells.

INTRODUCTION: Pancreatic ductal adenocarcinoma (PDAC) is characterized by severe hypoxia and an immunosuppressive tumor microenvironment, which contribute to poor therapeutic responses. This study developed sialic acid-targeting Lectin-conjugated oxygen nanosomes (Lectin-ONs) to combine targeted oxygen delivery with hypoxia alleviation and macrophage activation. METHODS: SNA- and MALII-conjugated oxygen nanosomes (SNA-ONs and MALII-ONs) were prepared and characterized for physicochemical properties, oxygen-loading and release behavior, stability, cytocompatibility, and cellular binding. Their

SERPING1 facilitates colorectal liver metastasis by modulating epithelial-mesenchymal transition and tumor microenvironment remodeling.

BACKGROUND: Colorectal liver metastasis (CRLM) remains the primary cause of mortality in patients with colorectal cancer (CRC). Despite its clinical significance, the complex molecular networks and microenvironmental dynamics driving CRLM remain incompletely understood. Identifying robust prognostic biomarkers and elucidating their underlying mechanisms are of critical importance for advancing targeted interventions. METHODS: We integrated Weighted gene co-expression network analysis (WGCNA) with differential expression profiling to identify the CRLM-associated hub genes. The clinical relevan

The prognostic marker NRIP1 is associated with tumor progression and immune infiltration in acute myeloid leukemia.

Acute myeloid leukemia (AML) is a clinically aggressive hematologic malignancy characterized by high relapse rates and treatment resistance, highlighting the need for novel biomarkers to improve clinical outcomes. In this study, we explore the roles of nuclear receptor-interacting protein 1 (NRIP1) in AML, focusing on its associations with tumor progression and immune infiltration. Analysis of public AML gene expression datasets reveals that NRIP1 expression is significantly increased in AML patients. Those with high NRIP1 expression have markedly shorter overall survival than those with low e

The Influence of Melanoma Extracellular Vesicles on Benign Melanocytes: A Role for PRAME in Modulation of the Tumor Microenvironment.

Melanoma is an aggressive skin cancer with a high tendency for metastasis and resistance to conventional therapies. This study explores the role of preferentially expressed antigen in melanoma (PRAME), a cancer-testis antigen, in melanoma progression, focusing on its function in melanoma-derived extracellular vesicles (EVs) and its impact on benign melanocytes. We show that PRAME is highly expressed in melanoma cell lines, tissues, and patient plasma and is present in EVs. These EVs transfer PRAME protein and mRNA to benign melanocytes, leading to significant alterations in gene expression, in

Data-mining-based biomarker evaluation and experimental validation of SHTN1 for bladder cancer.

BACKGROUND: Gene therapy in bladder cancer (BLCA) remains an area ripe for exploration. Recent studies have highlighted the crucial role of SHTN1 in the initiation and progression of various cancers and SHTN1 may have interacted with the FGFR gene. However, its specific function in BLCA remains unclear. MATERIALS AND METHODS: We investigated the association between SHTN1 expression and prognosis, immune infiltration, and the tumor microenvironment (TME) across multiple malignancies using 433 BLCA samples from The Cancer Genome Atlas (TCGA). Differential gene expression analysis, functional an

The bidirectional crosstalk between metastatic uveal melanoma cells and hepatic stellate cells engenders an inflammatory microenvironment.

Uveal melanoma is the most common primary ocular neoplasm in adults. It is peculiar for its hematogenous dissemination and its high propensity to spread to the liver. Current treatments rarely prolong patient survival. We hypothesized that metastatic uveal melanoma cells modulate the function of surrounding hepatic stellate cells to facilitate their own growth and survival. This study was conducted to investigate the role of the hepatic microenvironment on uveal melanoma aggressiveness. We demonstrated that the paracrine signaling of surrounding hepatic stellate cells have more transcriptional

Tumor's other immune targets: dendritic cells.

The induction of apoptosis in T cells is one of several mechanisms by which tumors escape immune recognition. We have investigated whether tumors induce apoptosis in dendritic cells (DC) by co-culture of murine or human DC with different tumor cell lines for 4-48 h. Analysis of DC morphological features, JAM assay, TUNEL, caspase-3-like and transglutaminase activity, Annexin V binding, and DNA fragmentation assays revealed a time- and dose-dependent induction of apoptosis in DC by tumor-derived factors. This finding is both effector and target specific. The mechanism of tumor-induced DC apopto

Radiotherapy Metastatic Prostate Cancer Cell Lines Treated with Gold Nanorods Modulate miRNA Signatures.

MicroRNA (miRNA) modulation has been identified as a promising strategy for improving the response of human prostate cancer (PCa) to radiotherapy (RT). Studies have shown that mimics or inhibitors of miRNAs could modulate the sensitivity of PCa cells to RT. In addition, pegylated gold nanoparticles have been studied as a therapeutic approach to treat PCa cells and/or vehicles for carrying miRNAs to the inside of cells. Therefore, we evaluated the capacity of hypofractionated RT and pegylated gold nanorods (AuNPr-PEG) to modulate the miRNA signature on PCa cells. Thus, RT-qPCR was used to analy

A composition-tunable cold atmospheric plasma chip for multiplex-treatment of cells.

Cold atmospheric plasma treatment promises a targeted cancer therapy due to its selectivity and specificity in killing tumor cells. However, the current plasma exposure devices produce diverse and coupled reactive species, impeding the investigation of the underlying plasma-anticancer mechanisms. Also, the limited mono-sample and mono-dosage treatment modality result in tedious and manual experimental tasks. Here, we propose a cold atmospheric plasma chip producing targeted species, delivering multiple dosages, and treating multiple cell lines in a single treatment. Three modules are integrate

Macrophage colony-stimulating factor (CSF-1) is expressed by spontaneously outgrown EBV-B cell lines and activated normal B lymphocytes.

Human B lymphocytes activated by mitogens or infected by Epstein Barr virus (EBV) have previously been shown to release colony-stimulating activity (CSA) supporting the growth of normal human bone marrow progenitors. We established five different human EBV-B cell lines spontaneously outgrown from nonmalignant peripheral blood cells and long-term bone marrow cultures. CSA derived from all of these lines induces the growth of murine macrophage colonies, whereas virtually no human bone marrow cell progenitors were stimulated. As observed in the tumor cell line MIA PaCa-2, a 4.3-kilobase (kb) tran

Establishment of a peripheral T-cell lymphoma cell line showing amplification of the c-myc oncogene.

A new T-cell lymphoma cell line, designated T34, was established from freshly isolated lymph node tumor cells of a patient with non-Hodgkin's diffuse large cell lymphoma. The T34 cells, as well as the parental lymphoma cells, showed mature helper/inducer immunophenotypes in that they formed spontaneous sheep erythrocyte rosettes and reacted with OKT-3 and OKT-4 monoclonal antibodies. They were negative for OKT-6, OKT-8, terminal deoxynucleotidyl transferase, WT-1, and HLA-DR antigens. Molecular analysis revealed that the T34 cells contained 8- to 16-fold amplified c-myc DNA. The same genetic c

Characterization of a monoclonal antibody that reacts with activated/proliferating cells and subsets of leukemia cells.

This report describes a murine IgG2A monoclonal antibody, called L22, derived by immunizations with an Epstein-Barr virus-negative large cell lymphoma B cell line. The antigen detected by L22 is not present on normal peripheral blood cells, but is present on cells stimulated by various mitogens. The proportion of L22+ cells correlates closely with blastogenesis and 125I-uridine uptake. L22 precipitates a 89,500-dalton antigen under reducing conditions, and a 180,000-dalton antigen under nonreducing conditions. The immunoreactivity, molecular weight of the antigen, sequential immunodepletion, a

Emodin suppresses the nasopharyngeal carcinoma cells by targeting the chloride channels.

Emodin is a natural anthraquinone derivative isolated from the Rheum palmatum. Recent studies demonstrated that emodin has anti-cancer activity in different kinds of human cancer cell lines. However, the underlying mechanism has not been very well studied. Our previous studies showed chloride channels is an important target of anti-cancer drugs. Therefore, the purpose of this research was aimed to explore the role of chloride channels involving in the anti-cancer activity of emodin. The proliferation, cell cycle arrest and apoptosis of poorly differentiated human nasopharyngeal carcinoma cells

C/EBP-beta, C/EBP-delta, PU.1, AML1 genes: mutational analysis in 381 samples of hematopoietic and solid malignancies.

Mutations of transcription factors are associated with the pathogenesis of cancer. Genomic DNA from 381 cancers and cell lines representing leukemias, lymphomas and a variety of solid tumors were examined for mutations of genes coding for the C/EBP-beta, C/EBP-alpha, PU.1, and AML1 transcription factors using single strand conformation polymorphism (SSCP) and direct DNA sequencing. Mutation of C/EBP-beta (a chronic myelogenous leukemia cell line, Kcl22) and C/EBP-delta (a Burkitt's lymphoma cell line, Raji) were found. Interestingly, the sample with a C/EBP-beta alterations had two missense (P

Characterization of a new human transitional cell carcinoma cell line from the renal pelvis, RTCC-1/KMC.

Tumors of the renal pelvis and ureter are relatively common malignancies in Taiwan. Studying the genetic or biochemical aberrations is a feasible pursuit that has great potential to further our understanding of urothelial cancer and may provide clinically valuable information. We now report a new long-term culture (RTCC-1/KMC) of human TCC derived from the renal pelvis, which is aimed to be used as a target for those studying in this field. The cultured cells exhibited anchorage independence and loss of contact inhibition. Chromosomal analysis revealed an aneuploidy line with a modal number of

Suppression of AKT anti-apoptotic signaling by a novel drug candidate results in growth arrest and apoptosis of hepatocellular carcinoma cells.

Hepatocellular carcinoma (HCC) is the third most common cause of cancer fatalities worldwide, with limited treatment options and five year survival rates of between <5 and 15%. To address this medical need, we conducted a screen of a drug-like small molecule library for HCC-selective cytotoxins. We report here the identification of a disubstituted aminothiazole termed HBF-0079, with remarkable selective toxicity for HCC-derived cell lines versus non-HCC liver lines and most other cancer lines. HBF-0079 caused irreversible growth arrest and apoptosis of the HCC lines Huh7, Hep3B, HepaRG as well

Targeting RAB7 in human B lymphoma by a small molecule inhibitor arrests tumor cell growth.

RAB7, encoded by RAB7A in humans and Rab7 in mice, is a small GTPase that catalyzes endosome maturation. It mediates NF-κB activation through the assembly of intracellular membrane signalosomes in stimulated normal B cells and plays a B cell-intrinsic role in the antibody response in mice. Here we show RAB7A transcripts are expressed in primary diffuse large B-cell lymphomas (DLBCLs), and that RAB7 protein expression is heightened in activated human tonsil B cells as well as in DLBCL and Burkitt lymphoma cell lines. Treating these cell lines with CID1067700, a selective small-molecule RAB7 inh
