Robust differentiation of human enteroendocrine cells from intestinal stem cells.

Enteroendocrine (EE) cells are the most abundant hormone-producing cells in humans and are critical regulators of energy homeostasis and gastrointestinal function. Challenges in converting human intestinal stem cells (ISCs) into functional EE cells, ex vivo, have limited progress in elucidating their role in disease pathogenesis and in harnessing their therapeutic potential. To address this, we employed small molecule targeting of the endocannabinoid receptor signaling pathway, JNK, and FOXO1, known to mediate endodermal development and/or hormone production, together with directed differentia

Why brain radiation therapy should take account of the individual structural and functional connectivity: Toward an irradiation "à la carte".

Although radiation therapy (RT) is a main treatment of brain tumors, delayed cerebral toxicity may lead to cognitive deteriorations with adverse effects on quality of life. Despite technological advances in RT, the concept of brain connectome has not yet been incorporated in the strategy of irradiation. Because white matter tracts represent the main limitation of neuroplasticity, tumor surgery is increasingly performed with awake cortical-subcortical mapping. Here, the purpose is to reinforce the link between cognitive neurosciences and neurooncology, which is critical for neurosurgeons but al

Linking Neuroinflammation and Neurodegeneration in Parkinson's Disease.

Neurodegenerative diseases such as Parkinson's disease (PD) and Alzheimer's disease (AD) impose a pressing burden on our developed and consequently aging society. Misfolded protein aggregates are a critical aspect of several neurodegenerative diseases. Nevertheless, several questions remain unanswered regarding the role of misfolded protein aggregates and the cause of neuronal cell death. Recently, it has been postulated that neuroinflammatory processes might play a crucial role in the pathogenesis of PD. Numerous postmortem, brain imaging, epidemiological, and animal studies have documented t

CD8+ T cell immunity in an encephalitis model of Toxoplasma gondii infection.

Toxoplasma gondii infection induces a robust CD8 T cell immunity in the infected host, which is critical for keeping chronic infection under control. IFNγ production and cytolytic activity exhibited by CD8 T cells are critical functions needed to prevent the reactivation of latent infection. Paradoxically, the susceptible mice infected with the parasite develop encephalitis irrespective of the presence of vigorous CD8 T cell immunity. Recent studies from our laboratory have demonstrated that these animals have defect in the memory CD8 T cell population, which become dysfunctional due to exhibi

Mutations in the FPIV motif of Newcastle disease virus matrix protein attenuate virus replication and reduce virus budding.

The FPIV-like late domains identified in the matrix (M) proteins of parainfluenza virus 5 and mumps virus have been demonstrated to be critical for virus budding. In this study, we found that the same FPIV sequence motif is present in the N-terminus of the Newcastle disease virus (NDV) M protein. Mutagenesis experiments demonstrated that mutation of either phenylalanine (F) or proline (P) to alanine led to a more obvious decrease in viral virulence and replication and resulted in poor budding of the mutant viruses. Additionally, evidence for the involvement of cellular multivesicular body (MVB

Preclinical models of stem cell-mediated analgesia and tissue repair: mechanisms, challenges, and future directions.

As an emerging biological therapeutic approach, stem cell therapy demonstrates broad application prospects in analgesia and tissue regeneration, particularly achieving significant advances in treating conditions such as spinal cord injury and intervertebral disc degeneration. In recent years, preclinical model studies have deepened our understanding of the mechanisms underlying stem cell-mediated pain relief and tissue repair, revealing their potential to regulate inflammatory responses, promote nerve regeneration, and repair damaged tissues through multiple pathways. However, the heterogeneit

Structure-function correlates of fibrinogen binding by Acinetobacter adhesins critical in catheter-associated urinary tract infections.

Multidrug-resistant Acinetobacter baumannii infections are an urgent clinical problem and can cause difficult-to-treat nosocomial infections. During such infections, like catheter-associated urinary tract infections (CAUTI), A. baumannii rely on adhesive, extracellular fibers, called chaperone-usher pathway (CUP) pili for critical binding interactions. The A. baumannii uropathogenic strain, UPAB1, and the pan-European subclone II isolate, ACICU, use the CUP pili Abp1 and Abp2 (previously termed Cup and Prp, respectively) in tandem to establish CAUTIs, specifically to facilitate bacterial adher

Gestational hypoxia in late pregnancy differentially programs subcortical brain maturation in male and female rat offspring.

BACKGROUND: Hypoxia is associated with pregnancy complications, such as preeclampsia, placental abruption, and gestational sleep apnea. Hypoxic insults during gestation can impact the brain maturation of cortical and subcortical pathways, such as the nigrostriatal pathway. However, the long-term effects of in utero hypoxic stress exposure on brain maturation in offspring are unclear, especially exposure during late gestation. The purpose of this study was to determine the impact of gestational hypoxia in late pregnancy on developmental programming of subcortical brain maturation by focusing on

Predicting Phloem Loading Types with Electron Microscopy.

Phloem loading is a critical step in the transport of photosynthetically produced sugars from leaves to sink tissues. The type of phloem loading-active apoplasmic, active symplasmic, or passive symplasmic-can be inferred from the density and structure of plasmodesmata (PD) along the phloem-loading pathway. This protocol describes a method to predict phloem loading types using transmission electron microscopy (TEM) to quantify PD density. The method follows established standards for sample preparation, imaging, and data analysis, enabling comparative studies across species.

Atypical hemolytic uremic syndrome triggered by acute pancreatitis.

The clinical manifestations of atypical hemolytic uremic syndrome (aHUS) are hemolytic anemia, thrombocytopenia, and acute kidney injury. It is one of the Thrombotic microangiopathy (TMA), with triggers for activation of the complement bypass pathway, such as infection and pregnancy, present in 70-80% of patients. Treatment for this disease requires plasma exchange or eculizumab, we encountered a patient with aHUS induced by gallstone-pancreatitis, after the initial plasma exchange, the patient's condition was improved, but the subsequent incomplete treatment eventually led to severe sequelae.

Diagnostic approach to lower limb edema.

Patients with lower limb edema are frequently referred to vascular specialists for evaluation. Multiple etiologies must be considered and often more than one cause may be present. Notably, the role of lymphatic system regardless of the underlying pathology has been underestimated. A thorough history and physical examination and a carefully considered laboratory and imaging evaluation are critical in differentiating causes. In this opinion article, we propose a diagnostic algorithm that incorporates a systematic approach to the patient with leg swelling and provides an efficient pathway for the

Craniofacial Growth: Current Theories and Influence on Management.

Craniofacial development is a highly coordinated process under a tight genetic control and environmental influence. Understanding the core concepts of growth and development of the craniofacial skeleton and the impact of treatment on growth potential is vital to successful patient management. To maximize outcomes and minimize iatrogenic consequences, proper sequencing and timing of interventions are critical. The development of the craniofacial skeleton occurs as a result of a sequence of normal developmental events: brain growth and development, optic pathway development, speech and swallowin

Statins reduce spirochetal burden and modulate immune responses in the C3H/HeN mouse model of Lyme disease.

Lyme disease (LD) is a systemic disorder caused by Borrelia burgdorferi. Lyme spirochetes encode for a functional 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase (HMGR EC 1.1.1.88) serving as a rate limiting enzyme of the mevalonate pathway that contribute to components critical for cell wall biogenesis. Statins have been shown to inhibit B. burgdorferi in vitro. Using a mouse model of Lyme disease, we found that statins contribute to reducing bacterial burden and altering the murine immune response to favor clearance of spirochetes.

New insights into the pathophysiology of renovascular hypertension.

In recent years, the pathophysiology of renovascular hypertension has been reviewed, and the classic concept that activation of the renin-angiotensin system is solely responsible for the development and maintenance of renovascular hypertension has been challenged. In fact, experimental evidence indicates that other systems, such as the lipoxygenase pathway, may have a more critical role in the long-term maintenance of high blood pressure after renal artery stenosis. Herein we discuss the intrarenal mechanisms that control pressure-induced natriuresis under physiologic conditions and the role o

Emerging nanoparticle-based strategies to provide therapeutic benefits for stroke.

Functional neurological recovery remains the primary objective when treating ischemic stroke. However, current therapeutic approaches often fall short of achieving optimal outcomes. One of the most significant challenges in stroke treatment is the effective delivery of neuroprotective agents across the blood-brain barrier to ischemic regions within the brain. The blood-brain barrier, while essential for protecting the brain from harmful substances, also restricts the passage of many therapeutic compounds, thus limiting their efficacy. In this review, we summarizes the emerging role of nanopart

Lactylation modification: a new bridge connecting macrophage metabolism and pulmonary hypertension.

Pulmonary hypertension (PH) is a multifactorial, progressive disease characterized by pulmonary vascular remodeling and right heart failure, representing a substantial global health burden. Dysregulated interactions among multiple cell types within the pulmonary vascular microenvironment, particularly macrophage polarization imbalance with a predominant transition toward the M2 phenotype, play a critical role in driving pro-fibrotic and pro-remodeling processes in PH. Lactylation, a novel post-translational modification that covalently links lactate to lysine residues, translates glycolytic me

RUNX1 aberrations in blast-phase chronic myeloid leukaemia induce the RNA-binding protein SPATS2L, which promotes growth, survival and stress granule assembly.

RUNX1 is a critical regulator of hematopoiesis. In CML, somatic RUNX1 mutations and RUNX1::MECOM/EVI1, are associated with TKI resistance and progression to BP-CML. To examine post-transcriptional dysregulation downstream of RUNX1 aberrations, we used orthogonal organic phase separation to characterise the RNA-binding proteome of RUNX1 deficient BP-CML cells. RUNX1 depleted BP-CML cells exhibited significant alterations to RBPs involved in stress response pathways and translation/ribosome-biogenesis. Furthermore, RUNX1 depletion or expression of RUNX1::EVI1 in BP-CML cells induced expression a

The neonatal lung microbiome: a dynamic determinant of respiratory health, disease, and novel therapeutics.

The neonatal lung, once considered sterile, is now recognized to harbor a dynamic and complex microbiome that plays a critical role in respiratory health and disease. This review synthesizes current evidence on the composition, development, and functional impact of the lung microbiome in neonates, with a focus on its involvement in key respiratory disorders such as bronchopulmonary dysplasia, respiratory syncytial virus infection, neonatal acute respiratory distress syndrome, cystic fibrosis, and asthma predisposition. We place particular emphasis on the bidirectional communication along the g

Role of GLP-1 receptor agonists in the prevention and treatment of obesity-related cancer.

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have become pivotal in treating type 2 diabetes and obesity due to their ability to improve glycemic control and promote weight loss. Beyond their metabolic benefits, emerging evidence suggests that GLP-1 RAs may exert anticancer effects by modulating critical biological pathways involved in tumorigenesis, including insulin signaling, inflammation, and cellular proliferation. Current evidence derived from observational and secondary analyses suggests potential protective effects of GLP-1 RAs against several cancers-including prostate, breas

Dietary Fiber Levels Modulate Intestinal Mucosal Architecture and the Microbiome-Metabolome Axis to Support Immune Homeostasis in Brooding Wanxi White Geese.

Dietary fiber is a critical determinant of intestinal health, yet its optimal inclusion level for WWG during the critical brooding period remains undefined. This study aimed to evaluate the effects of varying dietary CF levels (approximately 3%, 5%, and 9%) on the intestinal morphology, immune function, and microbiome-metabolome axis of brooding WWG. A total of 120 one-day-old goslings were randomly assigned to the three dietary treatments for a 28-day trial. Histological analysis revealed that the 9% CF diet significantly improved gut morphology, yielding superior villus-to-crypt ratios in th
