CCR5 chemokine receptor gene evolution in New World monkeys (Platyrrhini, Primates): implication on resistance to lentiviruses.

The CC chemokine receptor 5 (CCR5) gene of primates encodes a transmembrane protein involved in cellular signaling of some cell types of the immune system. Numerous studies have shown that this peptide is used by lentiviruses in conjunction with the CD4 receptor to mediate binding and entry in target cells of human and non-human primates. New World monkeys (NWM), differently from their African counterparts, have no description of in natura lentivirus infection. Some evidences suggest that a blockage occurs at the viral entry step of infection. To investigate this possibility, we have cloned an

Spatial population structure in a patchily distributed beetle.

The dynamics and evolution of populations will critically depend on their spatial structure. Hence, a recent emphasis on one particular type of structure--the metapopulation concept of Levins--can only be justified by empirical assessment of spatial population structures in a wide range of organisms. This paper focuses on Aphodius fossor, a dung beetle specialized on cattle pastures. An agricultural database was used to locate nearly 50 000 local populations of A. fossor in Finland. Several independent methods were then used to quantify key processes in this vast population system. Allozyme ma

Systematic review of economic evaluations in thalassaemia screening programmes globally: developing guidance for low- and middle-income (LMIC) settings.

OBJECTIVES: Thalassaemia, a genetic blood disorder, is a major public health burden. Most affected individuals reside in low-and-middle-income countries (LMICs). Screening programmes can reduce incidence, but in resource-constrained settings cost-effectiveness is important. This work aimed to investigate how economic evaluations of thalassaemia screening programmes have been conducted globally, to identify best practices for future evaluation suited to a LMIC context. DESIGN: Systematic literature review. DATA SOURCES: The original review was undertaken between May and July 2023; an update w

Genetic susceptibility to depression and risk of cardiometabolic diseases: a systematic review and meta-analysis of 21 Mendelian randomisation studies.

BACKGROUND: Depression is a risk factor for cardiometabolic diseases, but the extent to which this is attributable to genetic (rather than environmental) factors is unclear. Mendelian randomisation (MR) uses genetic variants to infer causality. We aimed to investigate the bidirectional association between respective genetic susceptibility to depression and cardiometabolic diseases. METHODS: We conducted a systematic review and meta-analysis of MR studies (unidirectional or bidirectional) in adults that assessed associations between genetic variants related to depression and any cardiometaboli

Clinical, biochemical, and genetic perspectives in hepatic glycogen storage diseases: HEPA-GSD study.

Glycogen storage diseases (GSDs) are a group of inherited metabolic disorders resulting from genetic deficiencies in enzymes involved in glycogen synthesis or breakdown, transport proteins, or proteins that regulate glycogen metabolism. Liver GSDs are further subdivided into subtypes based on the specific enzyme deficiency. This multicenter, cross-sectional, descriptive retrospective study, conducted across 24 centers, aimed to document the clinical and genotypic findings in patients with hepatic GSD. The study included 282 patients (152 males, 53.9%) from 278 unrelated families. At the time o

The 'other' colonic polyposis syndromes-evolving insights and guidance for endoscopists.

PURPOSE: Polyposis syndromes contribute to a significant proportion of colorectal cancer (CRC) diagnoses. While Lynch syndrome (LS) and familial adenomatous polyposis (FAP) are well characterised, a growing number of rarer polyposis syndromes are increasingly recognised in clinical practice. These conditions often present with overlapping phenotypes and variable penetrance, posing diagnostic and management challenges for the practising clinician. The evidence base remains limited, with current guidance largely derived from small cohort studies and expert opinion. This narrative review summaris

Nutrigenomic profiling identifies ZIP10 (SLC39A10) as a regulator of erythroid zinc homeostasis with genetic associations to anemia risk.

Erythroid progenitors undergo dynamic morphological changes and robust heme biosynthesis during differentiation. Zinc is essential for erythropoiesis, yet the mechanisms linking zinc availability to heme biosynthesis and anemia risk remain unclear. This study aimed to define zinc-responsive pathways in differentiating erythroid progenitors and to evaluate the translational relevance of SLC39A10 (ZIP10) genetic variants to hematological health. To elucidate the molecular basis of zinc's role in erythropoiesis, we performed transcriptomic profiling of zinc-restricted G1E-ER4 cells during differe

Integrating 730,947 exome sequences with clinical literature improves gene discovery.

Accurate estimates of allele frequencies aid in genetic discovery, including rare disease diagnosis, common disease investigations, and population genetics. Here, we present the Genome Aggregation Database version 4 (gnomAD v4), including 730,947 with exome sequences, a fivefold increase over previous releases. We demonstrate that statistical power to detect strong selective constraint continues to increase with sample size. We develop a new loss-of-function annotation pipeline, which learns genomic features predictive of nonsense-mediated decay and splicing effects from selection signals, ach

Molecular profiling reveals the malignant potential in solid pseudopapillary neoplasms of the pancreas.

Solid pseudopapillary neoplasm of the pancreas (SPN) is a rare tumor primarily affecting middle-aged women, typically characterized by indolent behavior but occasionally demonstrating malignant potential through invasive growth and metastasis. To elucidate the molecular mechanisms driving this heterogeneity, a multi-omics approach was applied to analyze paired metastatic lesions, primary tumors, and normal pancreatic tissues. Methylation profiling via the Illumina Infinium Methylation EPIC BeadChip identified 2425 differentially methylated positions (DMPs) in metastatic versus primary lesions,

The Complicated Genetics behind Uncomplicated Type B Aortic Dissection.

BACKGROUND: Up to 50% of uncomplicated type B aortic dissection (unTBAD) cases progress to become complicated with a mortality rate of up to 42% within 5 years of onset. Morphological and clinical parameters have previously been defined for the decision of surgical intervention in unTBAD to improve clinical outcomes. The analysis of genetic variants in this risk stratification has demonstrated a lack of evidence to influence clinical decision-making. METHODS: A comprehensive literature review was conducted using multiple electronic databases. A selection of genes recognized in thoracic aortic

Bidirectional two-sample Mendelian randomization analysis reveals a causal effect of chronic pain on chronic kidney diseases and renal function.

OBJECTIVE: A growing body of research has shown a connection between chronic pain and chronic kidney disease (CKD). However, it is unclear if these correlations point to a cause-and-effect link. Our goal is to investigate the causal link between renal function or CKD and chronic pain. MATERIALS AND METHODS: Using genome-wide association study (GWAS) datasets on these traits, we performed bidirectional two-sample Mendelian randomization (MR) analyses in this work to evaluate genetic linkages and possible causal links between chronic pain and CKD or renal function.
