Chlorogenic acid mitigates postharvest broccoli senescence through modulation of oxidative stress and ascorbate-glutathione cycle.

Postharvest broccoli is highly susceptible to water loss and wilting, accompanied by a color deterioration of the heads from dark green to yellow, severely compromising its commercial value. The effects and underlying mechanisms of chlorogenic acid (CGA) treatment on reactive oxygen species (ROS) metabolism and ascorbate-glutathione (AsA-GSH) cycle in broccoli were investigated. The results showed that CGA treatment efficiently mitigated the quality deterioration in broccoli, which was evidenced by the lower yellowing index, the higher h° value and chlorophyll level of the treated samples. Thi

Structural and biochemical basis of ROC-dependent activation of LRRK2.

Mutations in leucine-rich repeat kinase 2 (LRRK2) are the most common cause of familial Parkinson's disease, yet the molecular mechanism governing LRRK2 activation remains incompletely understood. LRRK2 is a large multidomain enzyme whose kinase activity is regulated by intramolecular interactions and by its Ras of complex proteins (ROC) GTPase domain. Here, we combine cryo-electron microscopy, X-ray crystallography, and structure-guided biochemical perturbations to define how ROC conformational switching regulates LRRK2 activation. Cryo-EM reconstructions reveal that monomeric full-length LRR

Structure-based design and immunogenicity of an H5 hemagglutinin stem nanoparticle vaccine candidate.

Highly pathogenic avian influenza viruses (HPAIVs), particularly the H5N6 subtype, pose a serious global public health threat. Conventional influenza vaccines primarily target the highly variable globular head of hemagglutinin (HA), resulting in limited breadth of protection. Here, we employed a structure-guided vaccine design strategy to develop a self-assembling nanoparticle immunogen targeting conserved epitopes within the HA stem region. The HAstem antigen was rationally engineered to incorporate major neutralizing epitopes and further stabilized through the introduction of a GCN4 trimeriz

MoS2-Based Thermoelectric Materials: Current Advances and Future Prospects in Micro-Thermoelectric Devices.

Thermoelectric (TE) materials, capable of directly converting waste heat into electricity and enabling solid-state cooling, hold great potential for self-powered microelectronics and miniature sensors in the Internet of Things (IoT) and wearable technologies. Two-dimensional (2D) MoS2 (molybdenum disulfide) stands out as a promising candidate due to its compatibility with CMOS (Complementary Metal Oxide Semiconductor) back-end-of-line processes, which are well-established in microelectronics fabrication. It is experimentally suggested that bilayer MoS2 exhibits an ultra-high-power factor (PF)

A protein-initiated polymerization cascade enables a self-eliminating powder tissue adhesive for diabetic ulcer repair.

Inspired by natural protein-based adhesives and protein-guided biosynthesis and assembly processes, this work proposes a strategy for constructing a bioactive dynamic adhesive through a protein-initiated polymerization cascade. In this system, ovalbumin (OVA) acts as a macromolecular director, and sodium lipoate (LANa) serves as a polymerizable bioactive monomer. The surfactant property of LANa disrupts the tertiary structure of OVA, exposing hydrophobic domains and thiol groups. LANa subsequently binds to these exposed hydrophobic microdomains to form OVA-LANa complexes, while the exposed thi

Plasma-Derived Exosomal hsa-miR-3677-3p Induces Ferroptosis in Neurons by Targeting ABCB8 in Perioperative Neurocognitive Disorders After Prostate Surgery.

Perioperative neurocognitive disorders (PND) are prevalent complications in elderly patients following surgery, characterized by cognitive decline and memory impairment. This study investigates the contribution of plasma-derived exosomal microRNA hsa-miR-3677-3p to PND pathogenesis via ABCB8 regulation and subsequent induction of neuronal ferroptosis. Exosomes were isolated from plasma of patients with delayed neurocognitive recovery (dNCR) and non-dNCR patients. Characterization confirmed successful exosome isolation, revealing distinct microRNA profiles between the two groups. MicroRNA seque

Invoking ferroptosis and photon-controlled pyroptosis via an integrated therapeutic system for triple-pathway tumor therapy.

Antitumor agents that rely solely on apoptosis often fail to disrupt the complementary cell survival cascades. In this study, we developed a redox-responsive integrated therapeutic system (QSH) that exploited ferroptosis and pyroptosis to enhance tumor therapy. QSH consisted of a dihydroorotate dehydrogenase (DHODH in mitochondria) inhibitor (Q, a ferroptosis inducer) and a photosensitiser (IHcy, a pyroptosis trigger) linked by a disulphide bond. Upon entering cancer cells, QSH could effectively target mitochondria by leveraging the mitochondrial membrane potential. Within the highly redox-str

Intratumoral Hypoxia Triggers Mitochondrial BHLHE40 ROS Sensing Pathway to Promote Radioresistance in Triple-Negative Breast Cancer.

Intratumoral hypoxia is a hallmark of triple-negative breast cancer (TNBC) and induces complex biological responses, including treatment resistance and mitochondrial production of reactive oxygen species (ROS). However, the direct mechanisms through which hypoxia-induced ROS are sensed and contribute to therapeutic resistance remain elusive. ROS can regulate the function of transcription factors through oxidation of cysteine thiol groups, but the compartmentalization limits their physical interaction with transcription factors. Here, BHLHE40, a transcription factor traditionally recognized for

Proteasome Inhibition Amplifies Endoplasmic Reticulum (ER) Stress Responses: Comparative Proteomics of Chinese Hamster Ovary Cell Lines.

Chinese hamster ovary (CHO) cells are widely utilised in the biopharmaceutical industry to produce therapeutic proteins. Understanding the mechanisms of endoplasmic reticulum (ER) stress and its interplay with protein degradation pathways remains pivotal for improving production efficiency and product quality. In this study, we investigated the proteomic responses of CHO-K1 (non-producer), CHO DP-12 (IgG-producer), and NISTCHO (IgG-producer) cell lines under ER stress induced by a combination of the proteasome inhibitor MG132 and the glycosylation inhibitor tunicamycin. Viability, cell growth,

Broad Roles of Endoplasmic Reticulum Stress Sensors Activated by Diverse Pathophysiological Stimuli.

The endoplasmic reticulum (ER) stress response is a critical cellular program that maintains proteostasis and membrane homeostasis through the activation of the ER stress sensor proteins inositol-requiring enzyme 1 (IRE1), protein kinase R-like ER kinase (PERK), activating transcription factor 6 (ATF6), and old astrocyte specifically induced substance (OASIS) family proteins. These sensors, canonically understood as transducers of the unfolded protein response (UPR), respond to the accumulation of misfolded proteins in the ER lumen as a result of ER luminal Ca2+ depletion, defective disulfide

Invited review: Unraveling whey protein-flavor interaction mechanisms-Advances in instrumental, computational, and sensory approaches.

Whey protein is part of a multibillion-dollar industry driven by rising consumer demand; however, its broader use is limited by its high reactivity with flavorings and potential for off-note development. Therefore, understanding the mechanisms underlying whey protein-flavor interactions is crucial. Although whey proteins can influence multiple aspects of flavor, their greatest impact is on volatile aroma. Whey protein-volatile interactions primarily occur through hydrophobic interactions, in addition to hydrogen bonding, electrostatic interactions, and Van der Waals forces. Volatiles with reac

Cascade-Activatable Nanoprobes for NIR-II Fluorescence/Photoacoustic Dual-Modal Imaging of Atherosclerotic Plaques.

The rupture of atherosclerotic plaques is a crucial event leading to acute cardiovascular incidents like myocardial infarction and stroke, and accurate imaging of atherosclerotic plaques is of great significance. Currently, early and precise diagnosis of atherosclerosis (AS) is still a challenge because of the lack of suitable diagnostic methods and probes. Herein, we develop a novel cascade-activatable nanoprobe IR820-PDA@BSA-T, which can sequentially respond to AS-overexpressed protein disulfide isomerase (PDI) and high viscosity microenvironment, for second near-infrared window (NIR-II) flu

Conopeptides as Modulators of Pain and Inflammation in Chemotherapy-Induced Peripheral Neuropathy by Targeting α7 and α9 Nicotinic Acetylcholine Receptors.

Chemotherapy-induced peripheral neuropathy (CIPN) is a prevalent and debilitating complication of cancer treatment, characterized by sensory dysfunction, including allodynia and hyperalgesia. Despite its clinical significance, there are no FDA-approved preventive options for CIPN, and current symptom management remains limited in effectiveness. Recent insights into CIPN's underlying mechanisms have highlighted the roles of neuroimmune interactions and ion channel dysfunction, particularly involving nicotinic acetylcholine receptors (nAChRs). Notably, the α7 and α9 nAChR subtypes play a critica

Artificially Designed 2D Composite Material With Tailored Thermal Expansion Toward Temperature-Stable and Accurate Nanomechanical Pressure Sensors.

Two-dimensional (2D) materials are suitable for constructing resonant nanoelectromechanical systems (NEMS) toward advanced sensing applications, due to their ultrasmall mass, high Young's modulus, and large strain limit. However, there exists a fundamental limitation: the thermal expansion of 2D materials leads to intrinsic frequency susceptibility of 2D NEMS to temperature variation, which has significantly impaired their deployment in realistic sensing applications. Here, leveraging the layer degree of freedom in 2D materials, we realize a type of artificially designed 2D composite materials

An Anthracenyl-Functionalized Boron Dipyrromethene Based Glutathione-Responsive Nanoplatform for Efficient Photodynamic Therapy Enhanced by Perfluorooctanoic Acid.

Photodynamic therapy (PDT) relies on light-activated photosensitizers to generate cytotoxic reactive oxygen species (ROS) for cancer treatment. However, its efficacy is severely limited by tumor microenvironment (TME) features such as hypoxia and elevated antioxidant levels (e.g., glutathione, GSH). To address these challenges, a GSH-responsive nanoplatform (enBDP-F NPs) was developed by integrating a heavy-atom-free photosensitizer (enBDP) with perfluorooctanoic acid (PFOA). The donor-acceptor-structured enBDP, featuring an anthracene-modified BODIPY core, enables efficient ROS generation via

Closed-Loop Recyclable Solid Polymer Electrolytes via Dynamic Lipoate Cross-Linking.

Growing demand for electrochemical energy storage has intensified concerns over battery end-of-life disposal, driving an urgent need for recyclable electrolyte materials. Here, we report a photo-curable, chemically recyclable solid polymer electrolyte platform built on a bio-derived difunctional cross-linker, poly(ethylene glycol) bis(lipoate) (PEG-LP2), prepared from naturally occurring α-lipoic acid. Visible-light-driven copolymerization of PEG-LP2 with poly(ethylene glycol) methyl ether acrylate in the presence of an ionic liquid (BMITFSI, 45 wt%) yields flexible, ionically conductive membr

Redox State of Glutathione and Cysteine in Plasma Following Acute Stroke.

Ischemic stroke is a major cause of long-term disability and death, with oxidative stress contributing substantially to post-ischemic injury. Reperfusion restores oxygen supply but simultaneously increases reactive oxygen species (ROS), amplifying secondary neuronal damage. This study examined time-dependent changes in systemic thiol redox status following transient middle cerebral artery occlusion (tMCAO) in rats. Plasma concentrations of cysteine (CySH), cystine (CySS), glutathione (GSH), and glutathione disulfide (GSSG), along with corresponding CySS/CySH and GSSG/GSH ratios and redox poten
