Immunosuppressive State May Lead to Brain Metastases in Lung Squamous Cell Carcinoma: Gene Expression and Immunohistochemical Analysis.

BACKGROUND/AIM: Brain metastases (BMs) are rare in lung squamous cell carcinoma (LUSC). Therefore, research on biomarkers or mechanisms that can be used to predict them is limited. To verify whether mRNA profiles can accurately predict BMs, we used a machine learning approach on 20 TNM-matched LUSC tissue samples. MATERIALS AND METHODS: We conducted pathway and immunohistochemical analyses to investigate the underlying mechanisms of BM. RESULTS: A total of 15 mRNAs linked to BM were identified. The 15-mRNA signature was highly accurate in predicting BM, with an area under the curve and accur

Pan-cancer landscape of basement membrane: multi-omics research and single-cell sequencing validation.

Epithelial carcinoma cells require penetration of the basement membrane (BM) to metastasize. The BM is a thin layer of extracellular matrix beneath epithelial and endothelial tissues. It acts as a structural barrier, preventing cancer cells from invading and undergoing endocytosis and exocytosis. Thus, understanding the relationship between the BM and tumor immunity can lead to new strategies for halting cancer progression and metastasis. Gene expression data of 33 cancers were obtained from the Cancer Genome Atlas database. The study analyzed the correlation between BM regulatory genes, copy

DDR1 Drives Collagen Remodeling and Immune Exclusion: Pan-Cancer Insights and Therapeutic Targeting in Pancreatic Ductal Adenocarcinoma.

Discoidin domain receptor 1 (DDR1), a collagen-binding receptor tyrosine kinase, plays a key role in extracellular matrix remodeling, tumor progression, and immune evasion. However, DDR1's comprehensive role across diverse cancers and its therapeutic potential in immune-resistant tumors remain poorly defined. We performed a pan-cancer analysis integrating bulk transcriptomic datasets, single-cell RNA sequencing, and pathway enrichment to evaluate DDR1 expression, genetic alterations, and its associations with immune cell infiltration and clinical outcomes. DDR1 was consistently overexpressed i

Interplay of PRMTs and Identification of Biomarkers Through Machine Learning Algorithms in Pan-Cancer, Highlighting PRMT3 as a Biomarker in Pancreatic Cancer.

Protein arginine methylation was a common post-translational modification, playing a key role in many biological processes and disease. But the regulatory mechanisms of protein arginine methyltransferases (PRMTs) in cancer were not well understood. This study aimed to examine the influence of PRMTs across various cancers and identify potential PRMTs-related prognostic biomarkers. Three machine learning algorithms were employed to identify potential PRMTs biomarkers across cancers. Additionally, drug prediction and molecular docking studies were carried out to discover new therapeutic targets.

Multifaceted investigations of PSMB8 provides insights into prognostic prediction and immunological target in thyroid carcinoma.

The Proteasome 20S subunit beta 8 (PSMB8) is an integral element of the immunoproteasome complex, playing a pivotal role in antigen processing. Despite its significance, the contributory role of PSMB8 in oncogenesis, particularly in thyroid carcinoma (THCA), has not been well-characterized. To address this gap in knowledge, our study endeavored to delineate the potential associations between PSMB8 and THCA. Transcriptomic profiles and clinical data of patients with THCA were retrieved from The Cancer Genome Atlas (TCGA) database to facilitate comprehensive analysis. Complementary resources fro

Analysis of PTPRZ expression in glioma and schwannoma - PTPRZ-long is highly expressed and shed in glioma.

Protein tyrosine phosphatase receptor type Z (PTPRZ) is a central nervous system-specific receptor that shows enhanced expression in gliomas. Our earlier work identified a soluble, proteolytically released form of this molecule (sPTPRZ) in cerebrospinal fluid (CSF), which appeared markedly increased in glioma cases relative to non-tumor controls, implying possible diagnostic value. The expression landscape of PTPRZ across other primary brain tumors, such as schwannomas or meningiomas, has not been systematically clarified. Here, transcriptome profiling of 443 surgically resected brain tumor sa

Yin Yang 1 (YY1) as a Central Node in Drug Resistance Pathways: Potential for Combination Strategies in Cancer Therapy.

Tumor drug resistance, a major cause of treatment failure, involves complex multi-gene networks, remodeling of signaling pathways, and interactions with the tumor microenvironment. Yin Yang 1 (YY1) is a critical oncogene overexpressed in many tumors and mediates multiple tumor-related processes, such as cell proliferation, metabolic reprogramming, immune evasion, and drug resistance. Notably, YY1 drives resistance through multiple mechanisms, such as upregulation of drug efflux, maintenance of cancer stemness, enhancement of DNA repair capacity, modulation of the tumor microenvironment, and ep

Contrasting effects of DNA demethylation on cancer-germline gene expression in breast cancer and leukemia cells.

Human germline gene expression is normally constrained to the germ cells, responsible for the production of sperm and oocytes. Cancer-germline (CG) genes, a subset of germline genes involved in testis development, are frequently aberrantly activated in cancer cells. The present study investigates the broader hypothesis that epigenetic modifications, specifically DNA methylation, can modulate the expression profiles of several CG genes in cancer and germ cells. Breast cancer (BC), normal breast (NB), and chronic myelogenous leukemia (CML) cell lines were treated with the DNA methyltransferase i

Discovery of actionable drug targets to enhance T-cell infiltration and immune checkpoint blockade efficacy in pleural mesothelioma.

BACKGROUND: Malignant pleural mesothelioma (PM) is an aggressive cancer with limited treatment options. Although first-line nivolumab plus ipilimumab improves outcomes for some patients, a majority fail to respond. Mechanisms of immune resistance in PM remain poorly understood, underscoring the need for new clinically actionable drug targets to overcome immunotherapy resistance. METHODS: We established an in silico pipeline to investigate the molecular basis of T-cell exclusion in bulk RNA-sequencing data from 448 patients across three immune checkpoint blockade (ICB)-naïve PM cohorts. We ass

Insulin signalling-associated cell fate promotes neoplastic invasiveness in non-functioning pituitary gonadotroph adenoma via cis-regulatory elements activation.

BACKGROUND: Non-functioning pituitary gonadotroph adenoma (NFGA) is the most prevalent subtype of pituitary adenomas with a tendency to develop into a refractory invasive type. Currently, effective pharmacotherapy for NFGA remains to be developed due to limited understanding of exact mechanisms of its invasiveness. METHODS: Here, we integrated scRNA-seq and scATAC-seq data from three NFGA patients to investigate the cellular heterogeneity underlying tumour invasion. An independent cohort with 210 patients and three primary cell lines was used to evaluate the association between insulin signal

CTHRC1: a key player in colorectal cancer progression and immune evasion.

The multifunctional secreted protein, collagen triple helix repeat containing 1 (CTHRC1), has recently emerged as a significant focus within oncology research. CTHRC1 expression in tumors is governed by a complex interplay of regulatory signals, including methylation, glycosylation, and notably, non-coding RNAs, which constitute its predominant regulatory mechanism. Colorectal cancer (CRC), a highly prevalent epithelial malignancy, sees CTHRC1 influencing tumor progression and metastasis through its modulation of several downstream signaling cascades, such as Wnt/PCP, TGF-β/Smad, and MEK/ERK p

Strong PSMA Expression by a Pituitary Macroadenoma on PSMA PET/CT.

An 81-year-old man was found to have an elevated PSA, prompting a prostate biopsy that revealed Gleason grade 7 (3+4) prostate cancer. For staging purposes, an 18 F-DCFPyL PSMA PET/CT was performed, which demonstrated focal uptake in the pituitary region corresponding to a suprasellar mass. Subsequent pituitary MRI confirmed the presence of a pituitary macroadenoma. The expanding global use of PSMA PET has increasingly revealed PSMA ligand uptake in a variety of non-prostatic benign and malignant conditions, creating false positive results. Our case highlights that pituitary macroadenoma may h
